1.Controversies over the treatment for stage Ⅲ A-N2 non-small cell lung cancer and related advances

Stage Ⅲ A non-small cell lung cancer (NSCLC) has high heterogeneity and there are some controversies over the treatment of this disease,especially for patients with stage ⅢA-N2 NSCLC.This article investigates whether preoperative or postoperative radiotherapy can improve the survival of patients with stage ⅢA-N2 NSCLC and evaluates the effect of surgical treatment.
2. HIV-1 integrase inhibitory activity of endophytic fungi from Aquilaria sinensis (Lour.) Gilg

OBJECTIVE: To investigate the HIV-1 integrase inhibitory activity of endophytic fungi from medicinal plant Aquilaria sinensis (Lour.) Gilg. METHODS: HIV-1 integrase inhibitory activity of the isolated endophytic fungi was determined by HIV-1 integrase strand transfer inhibitory activity assay, and the active metabolites of the endophytic fungi with the greatest potential were studied by activity tracking. RESULTS: Among 78 strains of endophytic fungi isolated from A. sinensis, nine strains (11.54%) showed strong inhibitory activity against HIV-1 integrase. Three compounds were obtained from the fermentation of strain HN-AS-8 which was identified to be Chaetomium globosum by HIV-1 integrase strand reaction. Compound 1 inhibited HIV-1 integrase with IC50 value of 35.4 μmol · L-1, and compounds 1 and 3 were for the first time isolated from Chaetomium sp. CONCLUSION: Potential anti-HIV-1 metabolites exist in the endophytic fungi from A. sinensis, which can be new resources for new anti-AIDS drugs.
3.Protection effects of metformin on biological behaviour of human vascular endothelial cells under inflammatory conditions

Background Studies showed that inflammatory process participates in the pathogenesis anddevelopment of diabetic retinopathy targeting retinal vascular endothelial cells (RVECs).A growing body of evidence revealed that metformin reduces the risk of micro-and macro-vascular complications by protecting blood-brain barrier,however,whether it plays a protective effect on human retinal vascular by similar mechanism is still unelucidated.Objective This study was to investigate the effects of metformin on the proliferation,migration and secreting monocyte chemotactic protein-1 (MCP-1) and interleukin-8 (IL-8) of human retinal vascular endothelial cells (RVECs) under the stimulation of tumor necrosis factor-alpha (TNF-α).Methods RVECs were cultured and divided into normal control group,metformin (5 mmol/L) group,TNF-α 2.5 ng/ml group,and TNF-α+metformin (5,10,20 and 40 mmol/L,respectively) groups.Corresponding drugs were added into medium according to grouping for 24 hours.Cell numbers were calculated before and after treatment.The metabolic activity (absorbancy) of RVECs was measured with MTS assay.Cell migration of RVECs was assessed with transwell migration assay.The MCP-1 and IL-8 concentrations in the cell supernatant were detected by ELISA assay.Results The number of the cells was significantly different among the normal control group,metformin group,TNF-α group,and TNF-α+metformin (5,10,20 and 40 mmol/L,respectively) groups (F =189.31,P < 0.01).The metabolic activities of RVECs were 0.32 + 0.02,0.32±0.03,0.97 ± 0.02,0.90 ± 0.05,0.76 ± 0.15,0.74 ± 0.05 and 0.41 ± 0.03;migrated cell numbers were (1 214±49),(1 200±45),(1 648±43),(1 309±48),(1 279±73),(961±60) and (942±106)/field;the concentrations of MCP-1 were (0.385 ±0.050),(0.362±0.060),(2.285 ±0.200),(1.131 ±0.180),(0.622 ± 0.120),(0.537±0.090) and (0.492±0.130) μg/ml,and those of IL-8 were (0.385±0.080),(0.390±0.120),(1.123±0.130),(0.899±0.180),(0.680±0.060),(0.417±0.090) and (0.335±0.100) μg/ml in the normal control group,metformin group,TNF-α group,and TNF-α + metformin (5,10,20 and 40 mmol/L,respectively) groups,showing significant differences among the groups (F =73.31,103.89,150.92,268.32,all at P< 0.01).The cell number,cell metabolic activity,migrated cell number,and MCP-1 and IL-8 levels in the cell supernatant were evidently increased in the TNF-α group compared with the normal control group,and those in the TNF-α+10 mmol/L metformin group,TNF-e +20 mmol/L metformin group and TNF-α+40 mmol/L metformin group were significantly decreased in comparison with the TNF-α group (all at P<0.05).Conclusions Metformin can inhibit TNF-α-induced proliferation,migration and MCP-1 and IL-8 secretion of the cells,and therefore plays a protective role on RVECs in the inflammatory environment.
4.Determination of Methotrexate in Human Serum by HPLC

OBJECTIVE:To determinate the serum concentration of methotrexate by HPLC. METHODS:The samples were separated on a Waters C18 column at a column temperature of 30 ℃. The mobile phase consisted of acetonitrile-phosphate buffer(pH 7.2,12∶88) at a flow rate of 1.0 mL?min-1. The detection wavelength was set at 303 nm. RESULTS:The linear range of methotrexate was 0.5~80 ?g?mL-1(r=0.999 4). The methodological recovery rates were all above 90%; the intraday RSD was 3.03 %~3.52 % and the inter-day RSD was 2.57%~4.05%. CONCLUSION:The method is proved to be simple,rapid,accurate and sensitive,and it is suitable for the determination of serum concentration and pharmacokinetic study of methotrexate.
5.Surveillance on multidrug-resistant organism infection in patients with malignant tumor

Objective To understand multidrug-resistant organism (MDRO) healthcare-associated infection(HAI) in patients with malignant tumor, so as to provide basis for making HAI prevention and control measures.Methods Targeted surveillance method was used to survey MDRO HAI in patients with malignant tumor in a hospital from January 1 to December 31, 2014, WHONET 5.3 software was used to analyze the data.Results A total of 54 056 patients with malignant tumor were surveyed, HAI occurred in 3 542 (6.55%) patients, 847(23.91%)of which were MDRO HAI.Most patients(54.55%) with MDRO HAI were >60 years old.A total of 2 606 bacterial strains were isolated from patients with HAI, 847 (32.50%) of which were MDROs, and most were extended-spectrum beta-lactamases (ESBLs)-producing Escherichia coli (n=459, 54.19%).The major site of MDRO HAI was lower respiratory tract (47.34%).The top 3 departments of detection of MDROs were departments of thoracic surgery(17.12%), colorectal and anal surgery(9.92%), and general surgery (8.26%).Conclusion Incidence of MDRO HAI is higher in patients with malignant tumor, surveillance of high risk population and monitoring of antimicrobial resistance of pathogens should be strengthened, so as to reduce the occurrence of HAI.
6.RP-HPLC Determination of the Content of Ephedrine?HCl in Xiaoyan Zhike Plaster

Objective To establish a RP-HPLC method to determine the content of ephedrine?HCl in Xiaoyan Zhike Plaster. Method The sample was prepared according to 2005 edition of Chinese Pharmacopocia, the column of Inertsil-3 ODS (4.6 mm?250 mm, 5 ?m) was used. The mobile phase was composed of methanol-0.1% of phosphoric acid (5∶95). The flow rate was 1 mL/min, with UV detection at 207 nm. Results The linear range was 0.041~0.41 ?g (r=0.999 8). The lowest limit of detection was 0.025 6 ?g. The average recovery and the precision was 99.55% and 2.58%. Conclusion The method is simple, sensitive and accurate, and can be used for the quality control of Xiaoyan Zhike Plaster.
7.RP-HPLC Determination of Geniposide in Xiaoyan Tuire Patch

Objective To establish a RP-HPLC method to determine the content of geniposide in Xiaoyan Tuire Patch. Method The column of Agilent ZORBAX SB-C18 (4.6 mm?250 mm 5 ?m) was used. The mobile phase was composed of water-acetonitrile-phosphoric acid (86∶14∶0.01). The flow rate was 1 mL/min, with UV detection at 238 nm, and the temperature was 30 ℃. Results The linear range was 0.07~0.70 ?g (r =0.99997). The average recovery was 98.60% and the RSD was 1.96%. Conclusion The method is simple, sensitive and accurate, and can be used for the determination of Xiaoyan Tuire Patch.
8.Analysis of surveillance results of human brucellosis in Anyang City, Henan Province in 2019

Objective:To understand the distribution characteristics of human brucellosis in Anyang City, Henan Province, and to provide scientific basis for control of brucellosis.Methods:The epidemic data and population data of human brucellosis in 2019 in Anyang City were collected from the Chinese Disease Control and Prevention Information System, and the epidemiological and clinical data were collected by Anyang Center for Disease Control and Prevention. Descriptive epidemiological method was used to sort out and analyze the epidemiological and clinical characteristics of brucellosis.Results:In 2019, 128 cases of brucellosis were reported in Anyang City, with an incidence rate of 3.12/100 000 (128/4 096 193). Among them, 85 cases were male and 43 cases were female, the sex ratio of male to female was 1.98∶1.00; the youngest was 1 year old, and the oldest was 83 years old, there were 100 cases between 40 and 69 years old, accounting for 78.12% (100/128); the main occupation was farmer (119 cases), accounting for 92.97% (119/128); a total of 96 cases had contact history with sheep, accounting for 75.00% (96/128). The peak of incidence was from April to August (81 cases), accounting for 63.28% (81/128). Eight counties (cities and districts) in Anyang City were all affected, and the top three counties (districts) were Yindu District (15.39/100 000, 42/272 983), Longan District (5.16/100 000, 12/232 421) and Neihuang County (4.02/100 000, 27/672 396). The median time from onset to diagnosis was 32.18 d, ranging from 3 to 350 d, 72 cases were ≥ 31 d, accounting for 56.25% (72/128).Conclusions:In 2019, the main population of brucellosis in Anyang City is male middle-aged and elderly farmers, and the main infection route is contact transmission. It is suggested to strengthen the implementation of prevention and control measures of brucellosis.
9.Endoplasmic reticulum stress inhibitor salubrinal enhanced radiosensitivity of head and neck carcinoma cells

Objective To explore the effect of salubrinal (sal,an endoplasmic reticulum stress inhibitor) on radiosensitivity of human head and neck squamous carcinoma cells (HNSCC).Methods Cells were divided into two groups of sal treatment and its control.For drug treatment group,cells were treated with 10 mmol/L sal for different time (12,24,36 h) and then irradiated.The levels of a core protein GRP78 of endoplasmic reticulum stress (ERS) in HNSCC (KB,Fadu,and Detroit 562 cells)were analyzed by Western blot assay at different time (0,20 min,1 h,3 h,6 h,12 h,24 h and 48 h) after irradiation.Cell survival was measured with colony formation assay.Results Western blot assay revealed that the protein levels of GRP78 in three kinds of HNSCC significantly increased from 20 min to 1 h and peaked at 3 h after radiation (t =12.72,13.37,18.31,P < 0.05).Compared with the control group,treatment of cells with sal decreased GRP78 protein levels (t =14.25,5.34,3.12,P < 0.05) in three cell lines and also significantly enhanced radiation damage and reduced cell viability.The sensitization enhancement ratios (SER) of sal in three cell lines were 1.16,1.05 and 1.06,respectively.Conclusions Rradiosensitivity of HNSCC could be effectively enhanced by sal treatment.
10.Detection and clinical significance of immune function in lung cancer patients

Objective To study the variety and significance of immune function in lung cancer patients.Methods 68 pretherapy lung cancer patients and 20 healthy volunteers with peripheral blood samples by flow cytometry.Results The proportion of CD+3T cells,CD+4T cells and the ratio of CD4/CD8 decreased in lung cancer patients,while the proportion of CD+8T cells and natural killer cells increased.Serum levels of TNF-α,IL-4,IL-6,IL-10 were significantly increased in patients compared with healthy controls,while the ratio of Th1/Th2 was decreased.(2)The natural killer cells level was negatively correlated with the CD3,CD4,CD8 levels respectively.The CD4 level was negatively correlated with the levels of IL-6,IL-10 respectively.(3)No statistically significant relationship was found between the peripheral blood lymphocyte,Th1-Th2 cytokines and pathological type,lymph node metastasis,distant metastasis.Conclusion Immune suppression was very common in lung cancer patients,particularly cell mediated immunity suppressed markedly.The proportion and function of CD+4T cells decreased significantly.which may resulted from the Th1,Th2 imbalance.Natural killer cells and CD+8T cells increased significantly,but could not prevent tumor development,which may resulted from these factors that suppress immune response in tumor microenvironment.
