1.Correlation between interleukin-1 and the obesity of polycystic ovary syndrome

Objective To investigate the correlation between interleukin-1β(IL-1β),interleukin-1 receptor antagonist(IL-1 ra)and the obesity of polycystic ovary syndrome(PCOS).Methods(1)From Oct.2006 to Jan.2007,118 PCOS patients were enrolled in this study in Peking University Third Hospital,which were divided into 56 patients in obese PCOS group and 62 patients in non-obese PCOS group according to the WHO International Obesity Task Force Asia-Pacific criteria[body mass index(BMI)25 kg/m2].The polymorphism of IL-1β gene promoter region,exon-5 and intron 2 of IL-1 ra gene were detected by PCR.(2)Twenty-nine obese PCOS patients and 31 non-obese PCOS patients were selected randomizedly serum levels of IL-1β,IL-1 ra were measured by ELISA,in the mean time,serum levels of fasting glucose,fasting insulin and the total white blood cell,hypersensitive C-reactive protein levels were measured.Results(1)Genetic test:the frequency of TT genotype and T allele of IL-1β promoter region(-511)in obese PCOS patients were significantly higher than those in non-obese patients(44.6% vs.11.3%,63.4% vs.39.5%,all P <0.05).The frequency of IL-1 ra Ⅰ / Ⅴ genotype and Ⅴ allele of IL-1 ra gene were 19.6% and 9.8% in obese PCOS patients,which were significantly higher than those in non-obese group(3.2% and 1.6%,P <0.05).(2)Serological test:serum level of IL-1β and IL-1ra of(149 ±36)and(284 ±97)ng/L in obese PCOS group which were significantly higher than those in non-obese PCOS group[(96 ± 42)and(208 ± 84)ng/L,P < 0.05].Fasting blood glucose,fasting insulin and hypersensitive C-reactive protein and white blood cell count were(5.1 ± 0.7)mmol/L,(17 ± 9)mU/L,(1.5 ± 0.6)mg/L and(7.0 ± 2.3)× 109/L in obese PCOS group,which were significantly higher than in non-obese PCOS group[(4.9 ±0.5)mmol/L,(11 ±8)mU/L,(0.9 ±0.4)mg/L and(5.9 ±1.3)× 109/L,P<0.05].(3)The correlation between interleukin and BMI: serum levels of IL-1β(r =0.673)and IL-1 ra(r =0.557)were positively correlated with BMI in PCOS patients(P < 0.05).Conclusions Inflammatory factors IL-1β and IL-1ra had correlation with obesity of PCOS patients,PCOS patients who carried T allele of IL-1β gene promoter region(-511)and Ⅴ allele of IL-1ra gene were high risk of obesity.
2.Development of Sine Wave Magnetic Field Generator and Effects on Potassium Channel Current

Purpose: A kind of sine wave magnetic field generator with the extremely low frequency (ELF) is developed, the frequency and intensity of the magnetic field can be adjusted, and has been easily used in Bio-experimental study. Methods: At first ,we design a DC power supply which provide DC voltage to other chips,then the signal generated by single chip ma-chine system and the power amplifier combined by discrete components, it provides current 0 mA~136 mA.The current coil is designed by experiment's required,the size or turns is changed, the magnetic field intensity is changed too. Results: The fre-quency of the device output is 0 kHz-25 kHz with the intensity in the range of 0 mT~10 mT within which the continuous lin-earity is adjustable. Power sources are less than 30 W ,which can work for long.This device performs reliably and the measured values in accord with the theoretical values and has been used in the experiment of magnetic biological effect. Conclusions: The experiment results indicate that there exist the effects of ELF sine wave magnetic field upon potassium current of neurons, and that such bioeffects vary with the intensity and frequency of the field.Also,there may exist window effect areas.
3.Chronic invasive fungal rhinosinusitis complicated with infratemporal fossa fungal infection: 2 cases report.

Two cases of chronic invasive fungal rhinosinusitis were reported. One patient healed while another died with suspicious residual fungal infection. The clinical symptoms of infratemporal fungal infection include maxillofacial pain, with or without fever. Acute or chronic inflammation of soft tissue could be revealed pathologically. Proof of pathogen may not be found in histopathological slice while serological detection may reveal positive evidence. Surgical resection, good drainage and postoperative antifungal therapy could cure or promptly relieve the symptoms of the disease.

4.Gender difference in emotion regulation:a resting EEG study

Objective Frontal resting EEG asymmetry was used to evaluate the gender effect of emotion regulation.Methods The resting EEG was recorded in 30 healthy volunteers (19-26 years old) by the guide 40 amplifier,and off-line analysis on EEG data were conducted by scan4.3 software.The statistical analysis were performed using SPSS 17.0.Results In normal human,frontal resting EEG alpha power was statistically significant(F=4.918,P =0.035).Asymmetry and gender interaction group (F =0.668,P =0.421),electrode point and gender interaction group (F =0.283,P =0.756),electrode point and asymmetry interaction group (F =1.418,P =0.260),electrode point,asymmetry and gender interaction group (F =1.164,P =0.327) had no statistical significance (P > 0.05).The comparison of male and female asymmetry degree of each electrode point (F4/F3 group t =0.465,F8/F7 groupt=0.809,FP2/FP1 group t=1.542) had no statistical significance (P>0.05).Conclusion Normal human brain hemispheres exist asymmetry.The asymmetry does not exists gender effect.In non-task state,frontal resting EEG alpha power asymmetry can not be recognized as the indicator of emotion regulation ability.
5.Effect of Passive Position Acupuncture on Cerebral Palsy

ObjectiveTo observe the clinical effect of compositive treatment on cerebral palsy children with rapid acupoint acupuncture at passive position.Methods37 children were treated with compositive treatment including acupoint acupuncture and physical training.Other 21 who treated with medicine were included as control.Results3 cases were cured and 33 cases were markedly effective in treatment group,which was better than that of control(P<0.05).ConclusionThe therapy composed of rapid acupoint acupuncture and physical training can significantly improve the function of children with cerebral palsy.
6.Clinical observation on radiofrequency scalpel in the treatment of rhinophyma

Objective To assess the efficacy of radiofrequency scalpel in the treatment of rhinophyma.Methods Twenty-three patients diagnosed with rhinophyma were randomly divided into radiofrequency group (13 patients) and routine surgery group (10 patients).Hypertrophic nasal tissue was removed by a radiofrequency scalpel in the radiofrequency group,and by a scalpel followed by dermabrasion in the routine surgery group.Several parameters,including operation time,intraoperative blood loss,surgical wound healing time,postoperative erythema duration,scarring incidence rate and appearance improvement value,were compared by t test or chi-square test between the two groups.Results All the patients completed the treatment.Compared with the routine surgery group,the radiofrequency group showed shorter operation time ((24.4 ± 2.8) vs.(44.0 ± 4.7) minutes,P < 0.01)and less blood loss ((10.4 ± 1.4) vs.(39.0 ± 5.7) ml,P < 0.01).No significant difference was observed in the surgical wound healing time,erythema duration,postoperative appearance improvement value and scarring incidence between the two groups.Conclusion Radiofrequency scalpel is effective and safe in the treatment of rhinophyma with shorter operation time and less intraoperative blood loss.
7.Research on quality parameters of scutellariae radix (formula particles) using on-line NIR in pilot with different extraction process.

The on-line monitor for the changes in the content of baicalin in Scutellariae Radix formula particles during the extraction process was conducted by using near infrared spectroscopy (NIR). High performance liquid chromatography (HPLC) was used as a reference method. Kennard-Stone (KS) was used to divide sample sets, so as to compare different pretreatment methods. The synergy interval partial least squares (SiPLS) was used to screen out modeling wave band to establish partial least-squares models. The relative error method was applied to predict forecast set samples of Scutellariae Radix in three extraction phases. The results showed that the model established by Savitzky-Golay smoothing with 11 points (SG11 points) was the best, with the root mean square with cross validation (RMSECV), root mean square error of correction (RMSEC) and root mean square error of prediction (RMSEP) of baicalin of 0.092 7, 0.134 4 and 0.114 8, respectively, the determination coefficient R2 of greater than 0.99, and the relative error of baicalin content of less than 5%. This indicates that the on-line near infrared reflectance spectroscopy could be applied in on-line monitor and quality control of the extraction process of Scutellariae Radix formula particles.

8.The effects of casticin on phosphorylation of FoxO3a in stem-like cells derived from ovarian cancer SKOV3 cell line

Background and purpose:Ovarian cancer is associated with a high recurrence and mortality due to the existence of cancer stem cells. The purpose of this study was to investigate the effect of casticin (CAS) on the capability of self-renewal in ovarian cancer stem cell like cells (OCSLCs) derived from SKOV3 cell line. Methods:SKOV3 cell line cells were cultured in vitro, and OCSLCs were obtained and amplified through suspended culture with conditioned medium of the stem cells. The phosphorylation level of FoxO3a was analyzed using Western blot. The protein expression of FoxO3a was inhibited by FoxO3a speciifc siRNA transfection, and then the ratio of sphere-formation was detected. Results: Compared with parental cells, OCSLCs over-expressed phosphorylated FoxO3a (pFoxO3a) and had elevated ratio of sphere-formation [(3.1±0.3)% vs (34.8±6.8)%, P<0.05]. CAS significantly inhibited the capability of sphere-formation in OCSLCs and down-regulated the expression level of pFoxO3a. And the transfection of FoxO3a speciifc siRNA suppressed the protein expression of FoxO3a and attenuated the inhibitory effect of CAS on the sphere-formation of OCSLCs. Conclusion: Reduced expression level of pFoxO3a is involved in the effect that CAS inhibits sphere-formation of OCSLCs derived from SKOV3 cell line.
9.Expression and effect of HSV-TK gene in osteosarcoma cells

Purpose To observe the killing effect of HSV-TK/GCV system on osteosarcoma cells. Methods The eukaryotic exprssoin vector (tgCMV-HyTK) containing HSV-TK gene was transduced into human osteosarcoma cell lines SAOS-2 by DOTAP. Total RNA extracted from HSV-TK expressing cells (SAOS-2TK) were tested by RNA dot blot analysis. A chemosensitivity of SAOS-2TK cells to GCV and “bystander effect” were measured by means of MTT colrimetric assay. Results HSV-TK gene was expressed in SAOS-2TK cells. SAOS-2TK cells were susceptible to the cytotoxicity of GCV. A meaningful “bystander effect” was demonstrated. Conclusion HSV-TK/GCV system may kill selectively the human osteosarcoma cell line SAOS-2.
