1.Mechanism by which interferon reduces the resistance of MGMT positive glioma stem cells to temozolomide

OBJECTIVE:To investigate the role of interferon to increase the sensibilization of MGMT positive glioma stem cel s to temozolomide in vitro. METHODS:Glioma cel lines, U251 and SKMG-4, were induced by suspended cloning bal formation method to harvest MGMT positive glioma stem cel s, U251G and SKMG-4G. Cel counting kit-8 assay was used to detect the kil ing effect of interferonα/βcombined with temozolomide on MGMT positive glioma stem cel s. RT-PCR and western blot assay were employed to determine the expression of MGMT and nuclear factorκB in MGMT positive glioma stem cel s. RESULTS AND CONCLUSION:Western blot results showed positive expression of MGMT in U251G and SKMG-4G cel s at protein levels. After intervention with interferonα/β, the mRNA expression of MGMT and nuclear factorκB in SKMG-4G and U251G cel s was reduced significantly, and then further decreased after temozolomide treatment. These findings indicate that interferonα/βcan remarkably strengthen the kil ing effect of temozolomide on MGMT positive glioma stem cel s.

Objective To investigate the clinicopathologic relationship between serum platelet derived growth factor-BB(PDGF-BB)and IgA nephropathy in children.Methods The level of serum PDGF-BB was detected by double antibody enzyme linked immunosorbent assay(ELISA) in 15 cases healthy children and 30 cases IgA nephropathy.According to patholgical degree constituted by WHO in 1982,the IgA nephropathy group divided into 5 degrees:Ⅰ,Ⅱ,Ⅲ,Ⅳ,Ⅴ(Ⅰ,Ⅱ were light patholgic change group;Ⅲ,Ⅳ were moderate patholgic change group;Ⅴ was severe patholgic change group).The serum PDGF-BB in IgA nephropathy group and none-IgA nephropathy group,and in different renal pathology type IgA nephropathy group were analyzed.Data were analyzed by using SAS 6.12 software.Results The level of serum PDGF-BB were(247.35?55.79) ng/L in control group and(869.16?200.73) ng/L in IgA nephropathy group.It was higher in IgA nephropathy group than in control group,the difference was significant(P

Objective To investigate the predictive value of androge receptors in stage pT1 non-muscle-invasive bladder cancer.Methods A total of 196 patients with stage pT1 non-muscle-invasive bladder cancer who underwent a transurethral resection of the bladder in Baoji People's Hospital from February 2003 to June 2012 were recruited to carry a retrospective analysis.The mRNA expression of the androge receptors transcript variants 1 (AR1) and 2 (AR2) was measured by quantitative real-time PCR.Spearman's correlation coefficient was used to analysis the correlation of androge receptors mRNA level to KRT5 and KRT20 mRNA.Results Kaplan-Meier analysis indicated that the recurrence-free survival,progression-free survival and cancer specific survival of patients with high AR1 mRNA expression (≥35.47) were significant better than patients with low AR1 mRNA expression (P <0.05).Multivariate COX regression analysis revealed that high AR1 mRNA expression was an independent prognostic marker for recurrence-free survival and cancer specific survival (P < 0.05).Spearman rank correlation revealed a significant positive association between mRNA expression of AR1 and KRT5 (rs =0.3171,P < 0.001) as well as a negative association with multifocal tumors (rs =0.1478,P < 0.05).Conclusion Androge receptors mRNA expression can predict recurrence-free survival and cancer specific survival in patients with stage T1 nonmuscle-invasive bladder cancer.Further studies on the levels of androge receptors mRNA tend to be particularly important.
4.Effects of maternal serum on permeability of glomerular endothelial cell membrane.

The mechanism of injury on the human glomerular endothelial cells (ciGENC) induced by preeclampsia serum was investigated. Concentration of maternal serum sFlt-1 protein was detected by ELISA. Fluorescently-labeled bovine serum albumin infiltrating through lower chamber of Transwell was measured by multifunction microplate reader. Morphologic change of ciGENC was observed under inverted phase contrast microscope. The concentration of sflt-1 in preeclampsia groups was significantly increased as compared with control group (P<0.01). Permeability in preeclampsia groups was significantly increased as compared with control group (P<0.01). By contrast with severe preeclampsia group, the permeability of ciGENC monolayer in mild preeclampsia group was decreased significantly (P<0.05). Intervention of exogenous VEGF significantly decreased permeability of ciGENC in preeclampsia groups. It was concluded that sFlt-1 increased ciGENC permeability by damaging integrity of endothelial barrier function.

This study examined the effect of over-expression of sFlt-1 by trophoblasts on the barrier function of glomerular endothelial cells and the role of VEGF in this process in order to explore the pathogenesis of glomerular disease in preeclampsia. SFlt-1 expression in the human trophoblasts (TEV-1 cells) was enhanced by transfecting sFlt-1 plasmid DNA into TEV-1 cells. The monolayer barrier function of glomerular endothelial cells (ciGEnCs) was determined by measuring the fluorescence intensity of bovine serum albumin (BSA) that crossed the monolayer of glomerular endothelial cells. The results showed that the over-expression of sFlt-1 by TEV-1 cells led to the barrier dysfunction of ciGEnCs, and the exogenous VEGF could alleviate the ciGEnCs dysfunction resulting from the over-expression of sFlt-1 to a certain extent. It was concluded that the dysregulation of sFlt-1 and VEGF in preeclamptic pregnancy may contribute to the barrier dysfunction of glomerular endothelial cells, and VEGF may play an important role in maintaining the barrier function of glomerular endothelial cells, but it may not be the sole factor.
6.Effect of epigenetic modification of maspin on extravillous trophoblastic function.

This study investigated the effect of epigenetic modification of maspin on extravillous trophoblastic function. The mRNA expression of maspin in placentae from normotensive and preeclamptic pregnant women was detected by RT-PCR. TEV-1 cells, a human first-trimester extravillous trophoblast cell line, were cultured and treated with CoCl(2) (300 μmol/L) to induce chemical hypoxia and with 5-aza (500 nmol/L) to induce demethylation. The mRNA expression of maspin in TEV-1 cells subjected to different treatments was determined by RT-PCR, and the proliferative and migratory abilities of TEV-1 cells were assessed by cell counting kit-8 (CCK-8) and Transwell assays. Our results showed that the maspin mRNA expression level in placentae from preeclamptic women was much higher than that from normotensive women. CoCl(2) or 5-aza could up-regulate the mRNA expression of maspin and significantly suppress the proliferation and migration of TEV-1 cells. It was concluded that the epigenetic modification in promoter region of maspin contributes to incomplete trophoblast invasion, which offers a novel approach for predicting and treating placental dysfunction.
7.Study on ameliorating measures for lactose intolerance to milk.

OBJECTIVETo find out the diet measurements for alleviating symptoms of lactose intolerance when people had had a drink of milk.
METHODSEight lactose intolerance (LI) subjects suffering from watery diarrhea were selected from 38 subjects by using hydrogen breath test (HBT) joined the test of drinking cow's milk along with various other foods designed by orthogonal trial. The 100 healthy college students were selected to observe symptoms after they ingested 25 g milk powder in different manners.
RESULTSThe breath-hydrogen concentration in groups with ingesting milk and various other foods was lower than those with ingesting milk only. Egg was better than mantou in alleviating symptoms. (F vaule of stool lactose and symptom score were 7.5, 9.0, respectively, P < 0.05). Whole wheat bread might aggravate flatus. The prevalence of the symptom in 100 college students by drinking milk in fasting state (27/98) was significant in comparing with those drinking milk in regular meal (13/98), chi2 = 6. 156, P < 0. 05.
CONCLUSIONThe study demonstrated that different kinds of food, and different milk drinking manners should affect the intolerance symptoms. We suggested: should not drink milk in fasting state, and drinking milk together with egg and staple food such as steamed bread, bread or gomuti should be the best choice for breakfast. Drinking milk with regular manner might alleviate the symptoms of intolerance. For avoiding lactose intolerance, whole wheat bread should not be taken along with milk.

8.Effects of remote ischaemic preconditioning on cardiac function in heart operation under thoracoscope

Objective To observe the effects of remote ischaemic preconditioning on myocardium in heart operation under thoracoscope.Methods One hundred and twenty patients (72 males,48 females,aged 41-69 years,weighing 49-68 kg,falling into ASA physical status Ⅱ or Ⅲ) who received heart operation under thoracoscope were randomly divided into 2 groups (n =60 each):remote ischaemic preconditioning group (group RIPC) and control group (group C).Comparing the pH value of arterial blood around the RIPC;observing the variation of LVEF and CI at before the operation,before leaving room and 24 h after operation,and the changes of myocardial enzymes (including cTnI,CK-MB and LDH) at basic level and 6,24 and 48 h after RIPC;comparing the basic state of the two group during the perioperation.Results Compared with group C,the pH value of artery blood was lower in group RIPC after RIPC(P<0.01).CI in RIPC group was higher than that in group C at 24 h after operation(P<0.05);there was no statistic difference in LDH between the two groups.The value of cTnI in RIPC group was lower than that in group C at 6 and 24 h after operation (P<0.05 or P<0.01).The value of CK-MB in group RIPC was lower than that in group C at 6,24 and 48 h after operation (P<0.05);there was no statistic difference in.LDH between the two groups.Basic conditions during and after the surgery in two groups had no statistic difference.Conclusion Remote ischaemic preconditioning can protect to myocardium during the heart operation under thoracoscope.
9.Effects of intravenous thrombolysis with urokinase for treatment of acute cerebral infarction due to vertebrobasilar ischemia

Objective To evaluate the effect of intravenous thrombolysis with urokinasc for treatment of acute cerebral infarction due to vertebrobasilar artery ischemia.Methods Thirteen pafients with a clinical diagnosis of acute cerebral infarction arising from vertebrobasilar artery ischemia received thmmbolytic therapy with urokinase(5×105-1.5×106U) infused intravenously within 1 h.The NIH Stroke Scale(NIHSS)was used to evaluate the patients'neurological function before and at 0,6,24 hand at 7 and 14 days after the treatment.Modified Rankin Scale(MRS)was used for neurologiical assessment 3 months after the treatments. Results The mean NIHSS score of the patients was 17.46±6.19 before treatment and rapidly decreased to 7.92±6.43 24 h after the thrombolytic therapy.Three months after the therapy,8 patients had favorable clinical outcomes defined as capability of independent living(MRS score of 0-2);2 patients had a poor outcomes with assisted living,and 3 patients died.Bleeding occurred in 4 cases including 2 cerebral hemorrhage cases. Conclusion Favorable clinical outcome can be achieved with intravenous urokinase therapy in the majority of patients with acute cerebral infarction due to vertebrobasilar ischcrnia.
