1.Construction of Subtractive cDNA Library of K562 Cells

Objective To construct the suppression subtractive library of hemin-inducing K562 cells for identifying the cDNA genes of globin synthesis regulatory factors expressed in K562 cells induced by hemin. Methods K562 cells were cultured under different concentration of hemin, both the percentage of positive benzidine staining cells and hemoglobin content were measured, the most reasonable concentration of hemin was chosen for inducing incubation of K562 cells. The poly (A) positive RNA (mRNA) was isolated from the hemin-induced K562 cells (tester) and non-induced K562 cells (driver) respectively, and double-strand cDNA molecules were synthesized by reverse transcription. After two times subtractive hybridization followed by two times polymerase chain reaction (PCR) amplification, the forward subtracted PCR products were ligated with pGEM T-Easy vector and the subtracted cDNA library was constructed. The library clones were selected by blue-white screening. The plasmid DNAs of the single positive colony were purified and digested by EcoRⅠ, and the inserts in plasmid were amplified by PCR. Results The maximum of positive benzidine staining cells percentage and hemoglobin content of K562 cells were obtained in 50?mol/L of hemin inducing condition. The suppression subtractive library of hemin-inducing K562 cells was constructed after K562 cells were induced by 50?mol/L of hemin. The library identification showed that the positive clones contain inserts in different length respectively. Conclusions The suppression subtracted cDNA library of hemin-inducing K562 cells were successfully constructed. This subtracted cDNA library combined with bioinformatics analysis can be used to explore structures and functions of hemin-inducing expression genes in K562 cells.
2.Dynamic study on fluoride release from glass ionomer cement in distilled water and artificial saliva

Objective In this study,the fluorine release behavior of six glass ionomer cements (GIC) samples was investigated in distilled water and artificial saliva.Methods The fluorine release behavior in GIC was measured by gas chromatography.Results Various GIC materials had the maxium fluorine release rate in the beginning,and the rate decreased rapidly.After 56 days,although the fluorine release quantity was in a trend of decreasing,the release process can be retained during a long period.For No3,No5,and No6 sampler,the fluorine release quantity in artificial saliva was lower than distilled water,whereas no significant difference was observed for Nol and No2 materials.The No4 sample produced a larger fluorine release quantity in artificial saliva.It can also be conducted that the immersible time of GIC and the fluorine release quantity met the equation of log ( Y ) =a + b x log ( t ).Conclusion The fluorine release behavior of six GIC matched the logarithmic regulation.
3.Analysis of surveillance results of human brucellosis in Anyang City, Henan Province in 2019

Objective:To understand the distribution characteristics of human brucellosis in Anyang City, Henan Province, and to provide scientific basis for control of brucellosis.Methods:The epidemic data and population data of human brucellosis in 2019 in Anyang City were collected from the Chinese Disease Control and Prevention Information System, and the epidemiological and clinical data were collected by Anyang Center for Disease Control and Prevention. Descriptive epidemiological method was used to sort out and analyze the epidemiological and clinical characteristics of brucellosis.Results:In 2019, 128 cases of brucellosis were reported in Anyang City, with an incidence rate of 3.12/100 000 (128/4 096 193). Among them, 85 cases were male and 43 cases were female, the sex ratio of male to female was 1.98∶1.00; the youngest was 1 year old, and the oldest was 83 years old, there were 100 cases between 40 and 69 years old, accounting for 78.12% (100/128); the main occupation was farmer (119 cases), accounting for 92.97% (119/128); a total of 96 cases had contact history with sheep, accounting for 75.00% (96/128). The peak of incidence was from April to August (81 cases), accounting for 63.28% (81/128). Eight counties (cities and districts) in Anyang City were all affected, and the top three counties (districts) were Yindu District (15.39/100 000, 42/272 983), Longan District (5.16/100 000, 12/232 421) and Neihuang County (4.02/100 000, 27/672 396). The median time from onset to diagnosis was 32.18 d, ranging from 3 to 350 d, 72 cases were ≥ 31 d, accounting for 56.25% (72/128).Conclusions:In 2019, the main population of brucellosis in Anyang City is male middle-aged and elderly farmers, and the main infection route is contact transmission. It is suggested to strengthen the implementation of prevention and control measures of brucellosis.

10.Esophagofundostomy in combination with esophagogastric devascularization for the treatment of portal hypertension

Objective To evaluate the effect of esophagofundostomy combined with devascularization for the treatment of portal hypertension.Methods From February 2009 to August 2013,30 cases (research group) underwent esophagofundostomy combined with devascularization,while 56 cases (control group) were treated by devascularization only.Splenectomy was performed in all patients.Results There was no perioperative mortality in either group.The difference of postoperative serum bilirubin,albumin and blood platelet was not statistically significant (t =1.13、0.23、1.53,all P > 0.05) ; The incidence of hepatic encephalopathy in research group and control group was 3% and 11%,respectively,three years mortality was 14% and 24%,respectively (x2 =0.61、1.22,all P >0.05).The rebleeding rate in research group was lower than control group,there was a significant difference (x2 =4.61,P < 0.05).Research group was superior to control group in improving esophageal-gastric varices and there was a significant difference (P < 0.05).Conclusions Compared with devascularization,esophagofundostomy combined with devascularization is more effective in reducing esophageal-gastric varices and prevention of rebleeding.
