1.Meta analysis of the level of plasma D-dimer in patients with ovarian cancer

Objective To evaluate the correlation between plasma D dimer levels and ovarian cancer, as well as its pathologic staging.Methods According to the inclusion and exclusion criteria , articles published about the correlation between plasma D dimer levels and human ovarian cancer were searched by means of PubMed , Embase, CNKI and WanFang Data from their inception to December 31th , 2013.In addition , references′list of the included articles and their related citations in Pubmed were also reviewed for additional articles.After quality assessment of all studies , Meta-analysis and ANOVA were performed with RevMan 5.2 and SAS 9.1 for statistic analysis.Results A total of 13 articles were identified, including 679 cases of ovarian cancers , 685 cases of benign controls and 477 cases of healthy controls.Meta-analysis revealed that:(1) Plasma D dimer levels in ovarian cancer groups were higher than that in benign control groups ( Z =8.61, P <0.000 01 ) and healthy control groups ( Z =6.76, P <0.000 01);(2) Plasma D dimer levels in benign control groups were higher than that in healthy controls (Z=3.15, P=0.002); (3) Plasma D dimer levels in advanced ovarian cancer FIGO (Ⅲ-Ⅳ) were higher than that in early cancer FIGO (Ⅰ-Ⅱ) (Z=3.38, P=0.000 9);(4)But there were no difference between relapse groups and remission groups (Z=1.19, P=0.23).ANOVA revealed that:(1) There were significant difference of D dimer levels among ovarian cancer groups or benign groups , healthy control groups; ( 2 ) Significant increasing of D dimer levels was revealed by Pairwise ANOVA from healthy controls, benign controls to ovarian cancer.Conclusions Plasma D dimer levels were high in patients with ovarian cancer , especially in FIGO (Ⅲ-Ⅳ) ovarian cancer.No difference of D dimer levels were investigated between relapse phase and remission phase.
2.Value of Quantitative Detection of Urinary Endotoxin for the Diagnosis of Gram-negative Bacteria Infection in Urinary System

Objective To survey the value of quantitative detection of urinary endotoxin for the diagnosis of gram-negative bacteria infection in urinary system,in order to provide evidences for early diagnosis and treatment.Methods The levels of urinary endotoxin were detected by kinetic turbidimetric technique,at the same time urine was cultured and WBC,CRP were detected.Results In 379 patients with gram-negative bacteria infection in urinary system,132 patients were upper urinary tract infection and 247 were lower urinary tract infection.The level of WBC,CRP and endotoxin were 11.25(7.98,13.99)× 109/L,81.36(76.64,83.50)mg/L and 49.57(17.28,78.37)pg/ml in upper urinary tract infection patients and in lower uri-nary tract infection patients were 10.24(7.94,12.24)×109/L,70.46(70.46,77.32)mg/L and 21.18(21.18,37.31)pg/ml. The group of upper urinary tract infection was higher than lower urinary tract infection on CRP and endotoxin,and have sig-nificant difference between the two groups of patients (P<0.05).Analysis of ROC,the diagnostic Cut-off value of 39.7pg/ml,the sensibility and specificity of endotoxin for the diagnosis of gram-negative bacteria infection on upper or lower urinary system were 84.1% and 90.3%.Conclusion The levels of urinary endotoxin detected by kinetic turbidimetric technique is simple and efficient,and have significant diagnosis value of gram-negative bacteria infection on upper or lower urinary sys-tem.
3.Correlative study on interleukin-6,interleukin -8 and interleukin-10 with radiation-induced lung damage

Objective To study radiation -induced lung damage after lung ionizing radiation and the temporal and spatial release of pro -inflammatory cytokines of interleukin -6(IL-6),interleukin-8(IL-8) and interleukin-10(IL-10)in the irradiated lung tissue.Methods BALB/C male mice weighted around 25 g were randomly divided into two groups:radiation group ( R) and control group ( C) ,with 30 mice in R group and10 mice in C group.The thorax of mice was irradiated by 6 MV X-ray with 25 Gy in 5 fractions.The mice were sac-rificed at 12 weeks post irradiation.Lung tissues were collected and embedded in paraffin .After HE staining,lung histopathological changes were detected by immunohistochemistry to detect IL -6,IL-8 and IL-10 expression in lung tissue.Results Immunohistochemistry results showed that the expressions of IL -6,IL-8 and IL-10 were mainly expressed in macrophages and inflammatory cells .The results showed that the expressions of IL -6 and IL-8 in R group were significantly higher than that in C group .IL-10 expression level was lower than C group.Conclusion After 12 weeks exposing to radiation ,cytokines of IL-6,IL-8 and IL-10 in lung tissue are associated with radiation -induced lung injury .
4.Study on CT-guided collagenase injection in the treatment of herniation of L4-5 lumbar disc by posterolateral approach

Objective CT-guided collagenase injection through posterolateral path into the interior and superficial prominence of the herniated lumbar disc in the treatment of L4-5 lumbar disk herniation.Methods The approach of posterolateral path to the prominence through intervertebral foramen and epidural space of L4-5 was established according to the analysis of anatomical features and vertebral medical images. The accuracy and safety of the approach were evaluated by CT scanning after epidural injection with air. CT-guided injection was operated in 66 cases of prolapse of L4-5 intervertebral disc confirmed by CT. Results Technical success was 100%. Followed up for 3 months to 3 years, 62 cases (93%) were markedly suecessfud with the effective rate of 93%. Twenty-three had CT re-examination three months later showed the prominences becoming smaller or disappeared in 22 patients having a resolved rate of 95%.Conclusions CT-guided injection of collagenase is one of the effective methods for the treatment of prolapse of L4-5 intervertebral disc.
5.Clinical observation on treatment of late stage cancer patients with stanozolol and shenqi injection

Objective To study the effect of stanozolol and shenqi injection used in comprehensive treatment of late stage cancer.Methods 158 late stage cancer patients,all treated only by rountine method and supportive treatment were randomly divided into 2 groups.And at the same time,79 patients in the treatment group were treated with stanozolol,2mg,po,tid,and shenqi injection 250ml,ivdrip,qd for nine weeks.All patients were recorded weight and hematology biochemical indicator.Results The treatment group surpasses the control group obviously (P<0.01 ).Conclusion Stanozolol and shenqi injection can improve quality of life of late stage cancer patients and reduce dyscrasia syndrome.
6.The synergetic effect of combined astaxanthin and lithium chloride treatment on cognitive dysfunc-tion of chronic omethoate poisoned mice

Objective To investigate the synergetic effect of combined astaxanthin ( AST) and lith-ium chloride ( LiCl) treatment on cognitive dysfunction of chronic omethoate poisoned mice. Methods 8 mice were selected randomly as control group from 55 healthy adult male Kunming mice,and the rest were used to establish chronic organophosphate poisoning cognitive impairment models by injecting omethoate 5 mg/kg subcutaneously every day for 4 weeks. Totally 40 successfully established models were randomly divid-ed into model group,AST group,edaravone group,LiCl group and AST+LiCl group with 8 in each. Morris wa-ter maze test was used to examine the learning and memory ability of mice. Contents of reactive oxygen spe-cies (ROS) in hippocampus were measured by enzyme-linked immunosorbent assay (ELISA). Activity of superoxide dismutase ( SOD) in hippocampus was measured by colorimetric assay. Morphology of hippocam-pus area was observed by HE staining. The distribution and expression of p-PI3K,p-Akt,p-GSK3β and p-CREB were determined by immunohistochemical staining ( IHC staining) and Western blot. Results The average escape latency of 5 days in each group was statistically significant (F=1662.147, P<0.05) . The av-erage escape latency of 5 days in AST+LiCl group was significantly lower than that in model group ( all P<0.05) and was lower than other treatment groups. Compared with the control group (0.087±0.007,0.084± 0.009,0.097±0.002,0.076±0.012),the hippocampal neuronal injury in model group was serious,the expres-sions of p-PI3K (0.032±0.008),p-Akt (0.03±0.006),p-GSK3β (0.028±0.007) and p-CREB (0.020± 0.008) was significantly lower ( all P<0.05) . The injuries of hippocampal neurons in AST+LiCl group were slightly lighter than that in model group,and the expression of p-PI3K (0.067±0.008),p-Akt (0.065± 0.005),p-GSK3β (0.068±0.009) and p-CREB (0.062±0.008) in hippocampus was significantly higher than that in model group ( all P<0.05) . Conclusion Combined AST and LiCL treatment exerts neuroprotec-tive effect on cognitive dysfunction induced by chronic organophosphate poisoning via up-regulating the ex-pression of Akt/GSK3β/CREB.
7.Effects of Exhausted Treadmill Running on the Expression of Parvalbumin Positive Neurons and GluN2B-containing NMDA Receptors in the Prefrontal Cortex of Rats

Objective To explore the effects of excise-induced fatigue on the microloop plasticity of prefrontal cortex through observing the expression of parvalbumin positive neurons in prefrontal cortexes of rats induced by exhaustive exercise,so as to find out the possible mechanism of the central regulation of exercise-induced fatigue by measuring the expression of NMDAR2B receptors.Methods Thirty-six Wistar rats were randomly divided into an exhausted group (E),a repeated exhaustion group (RE) and a control group (CG),each of 12.For group E,the adjusted Bedford incremental load of treadmill exercise program was employed:the initial treadmill speed was 8.2 m/min,lasting for 15 minutes,then increased to 15 m/min for another 15 minutes,and finally increased to 20 m/min till exhaustion.For RE group,they were given continuous treadmill exercises to exhaustion for consecutive 7 days.The immunofluorescence technique was used to observe the expression of PV+ interneurons after exhausted treadmill running.The Western blotting technique was used to determine the expression of NMDAR2B in the tissue of the prefrontal cortex.Results After the exhausted treadmill running,the expression of PV+ interneurons in the prefrontal cortexes of both E and RE groups increased significantly compared with the control group(P<0.01).The immunofluorescence results indicated that NMDAR2B positive neurons were seen in group E,but not obviously in group CG and RE.The Western blotting showed that compared with CG group the protein expression of NMDAR2B in prefrontal cortexes of group E was relatively high,and that of group RE was relatively low,but without significant difference (P>0.05).The running distance and prefrontal cortex NMDAR2B expression were found negatively correlated (P< 0.01).Conclusions Exhaustive exercises have an impact on the plasticity in rats' prefrontal cortex neural network through regulating the local loop of PV positive neurons.This plasticity of the prefrontal cortex is involved in the regulation of central fatigue.The present study might provide morphological basis for the research of central mechanism of the exercise-induced fatigue.

Objective:Helicobacter pylori is one of the human pathogens causing gastric ulcers and cancers.A key virulence factor of H.pylori is the Cag pathogenicity island,which encodes a type IV secretion system.HP0525 is an essential component of the Cag system and acts as an inner membrane associated ATPase.To construct recombinant plasmid containing hp0525 gene of Helicobacter pylori(H.pylori)NCTC 11637,and analysis of sequence nucleic acid,express it in E.coli BL21 and study the influence of the HP0525 protein on proliferation of SGC-7901 cells.Methods:H.pylor hp0525 gene was amplified from the genome DNA by PCR,then operated T-A cloning and sequenced.The hp0525gene fragment was inserted directionally into vector pMD18-T to construct recombinant clones of hp0525 and was sequenced.The recombinant plasmid was transformed into E.coli BL21for expressing under induction of IPTG.Purify the expressed protein by Ni2+-NTA column chromatography.Expressed product was analyzed by Western blot and MALDI-TOF.Added the purified protein into SGC-7901 cells,the effect of cell proliferation in SGC-7901 cells induced by the recombinant protein was observed by MTT.Results:A 993 base pairs long hp0525 gene,which encodes a product of 330 amino acid,was obtained using PCR method and was cloned into pMD18-T vector successfully.The sequence analysis for hp0525 showed that it shares 97%~99% homology with other strains in Gene bank.SDS-PAGE showed a protein band with a relative molecular weight of 36 000,which was consistent with the expectation.The expressed product reached a purity of 97% after Ni2+-NTA column chromatography.The protein after dialyzed and annealed,was co-cultured with SGC-7901 cells,The protien of different concentration co-cultured with SGC-7901 cells for different times,found that the protein in low concentration stimulates proliferation of cells,to achieve some concentration,it inhibits proliferation of cells along with multiplication of the concentration of the protein;The protein inhibits proliferation of cells relay on the extension of time and concentration.Conclusion:It is indicated that the correct hp0525 gene was obtained and expressed in E.coli BL21.High purified protein was obtained by Ni2+-NTA column chromatography,the protein can inhibits proliferation of SGC-7901cells,and T-ATPase activity which posed a basis for further researching on its biological function.
10.STUDY ON ANTIOXIDATIVE AND ANTITUMOR EFFECT OF SELENIUM CONTAINING POLYSACCHARIDE IN GANODERMA LUCIDUM IN MICE

Objective: To investigate the effects of selenium containing polysaccharide from Ganoderma lucidum on SOD, GSH Px activity, MDA content in blood and liver and anti tumor effect in tumor bearing mice, and relationship between antioxidative and anti tumor effect. Methods: The tumor bearing BALB c mice were injected with 2.5 g/L? 5.0 g/L and 10.0 g/L SeGLP 1 respectively for 14 days. Weight of tumor, both SOD and GSH Px activities and MDA content of blood and liver were determined. Results: SeGLP 1 could significantly inhibit the growth of Hca f, maximal inhibition rate was 44%, SeGLP 1 significantly increased SOD, GSH Px activity, decreased MDA content of blood and liver in tumor bearing mice. Conclusion: The anti tumor effect of SeGLP 1 may be due to enhanced antioxidative capacity in tumor bearing mice.
