1.Expression of tissue factor in pulmonary artery after experimental acute pulmonary embolism

Objective To investigate the expression of tissue factor and explore its clinical significances in pulmonary artery after acute pulmonary thromboembolism.Methods Thirty-four Japanese white rabbits (Level Ⅱ animals) were randomly (random number) assigned into four groups:group A (specimen of pulmonary artery was taken 3 hours after pulmonary embolism,n =8),group B (specimen of pulmonary artery was taken 8 hours after pulmonary embolism,n =8),group C (specimen of pulmonary artery was taken 24 hours after pulmonary embolism,n =8) and control group (pseudo-operations were carried out without injecting autologous blood clots,n =10).The animal model of pulmonary thromboembolism was established by injecting autologous blood clots into jugular vein through a 5F catheter and confirmed by digital subtraction angiography.The mRNA expression of TF in different parts of pulmonary artery was assayed by RT-PCR.The q test was utilized if there was a significant difference in a given continuous variable among three groups analyzed by ANOVA.Results The TF expression in the specimen adjacentto emboli was stable at 3 h,8 h or 24 hours after embolism.The mRNA expression of TF at 3 h and 8 h after embolism was lower in specimen taken from distal-end of morbid pulmonary artery than those adjacent to emboli.While at 24 hours after embolism,there were similar mRNA expressions in specimen either adjacent or distal to emboli.Conclusions The high expression of tissue factor in pulmonary artery tissue adjacent to emboli could lead to locally increased coagulation activity,indicating the necessity of initiating anti-coagulation treatment as soon as possible after acute pulmonary embolism.
2.Changes in transforming growth factor-beta mRNA expressionin the lung in a dog model of cardiopulmonary bypass-induced acute lung injury

Objective To observe the changes in the transforming growth factor-beta 1 (TGF-β1 ) mRNA expression in the lung in a dog model of cardiopulmonary bypass(CPB)-induced acute lung injury. Methods Thirty-six healthy adult mongrel dogs of both sexes weighing 15-16 kg were randomly assigned into control group and CPB group ( n = 18 each) . Lung injury was produced by CPB according to the method described by Williams. Six animals were killed at each of the following time points: before CPB (T0 ) and 30 and 60 min after termination of CPB (T1 , T2) in each group. Lung specimens were obtained for microscopic examination and determination of TGF-β1 mRNA expression (by RT-PCR) and MDA content. The lungs were lavaged and the protein concentration in the brancho-alveolar lavage fluid (BALF) was determined and pulmonary permeability index (PPI) was calculated. Results Microscopic examination showed massive inflammatory cell infiltration, alveolar capillary dilatation, congestion, widened alveolar septum, massive RBC in the alveolar space and focal atelectasis in the lung in CPB group. The TGF-β1 mRNA expression and MDA content and PPI were significantly higher in CPB group than in control group. The TGF-β1 mRNA expression and MDA was positively correlated to PPI (MDA: r = 0.867, P < 0.01; PPI: r = 0.821, P < 0.01) . Conclusion TGF-β1 mRNA expression in the lung is significantly up-regulated after CPB and is an important factor contributing to CPB-induced acute lung injury.

Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) has become a widely used method in the field of microbial identification. Its obvious advantages including rapidity, great accuracy and high throughput attract many researchers to investigate its potential for usage in other microbiological fields. Currently, several studies have reported MALDI-TOF MS-based analysis of bacterial resistance to antibiotics, which can identify the specific heterogeneous spectrum peaks related to drug resistance through simple visual analysis of the spectrum or more complex analysis of the entire spectrum using informatics methods and statistical approaches. Therefore, MALDI-TOF MS has become a potential tool for detecting antibiotic resistance in bacteria. This review mainly summarized the progress in MALDI-TOF MS-based analysis of bacterial resistance to antibiotics.

5.Therapeutic Observation of Acupuncture plus Chinese Medicinal Fumigation for Chronic Pelvic Inflammatory Disease

Objective To observe the clinical efficacy of acupuncture plus Chinese medicinal fumigation in treating chronic pelvic inflammatory disease (CPID).Method Totally 120 CPID patients were randomized into group A, group B and group C, 40 cases in each group. Group A was intervened by acupuncture plus Chinese medicinal fumigation, group B was by dry acupuncture treatment, while group C was by Chinese medicinal fumigation alone. After 3 treatment courses, the clinical efficacies were observed, and the relapse rates among the cured cases in 8 months after the whole treatment were compared among the 3 groups.Result The total effective rate and recovery rate were respectively 95.0% and 70.0% in group A, versus 82.5% and 45.0% in group B, and 57.5% and 32.5% in group C, and the total effective rate and recovery rate in group A were significantly different from that in group B and group C (P<0.05). The treatment duration for the recovered cases in group A was significantly different from that in group B and C (P<0.05). The relapse rate in the recovered cases in the 8-month follow-up was 10.7% in group A, versus 44.4% in group B and 53.8% in group C, and the relapse rate in group A was markedly lower than that in group B and C (P<0.05).Conclusion Acupuncture plus Chinese medicinal fumigation is an effective method in treating CPID, and its advantages include content efficacy, short treatment duration, and low relapse rate, etc.
6.The inhibitory effect of Matrine on the proliferation of peripheral lymphocytes of patients with rheumatoid arthritis

Objective To investigate the effects of Matrine (Mat) on the proliferation and cell cycle of the peripheral blood lymphocytes of patients with rheumatoid arthritis.Furthermore,its mechanism of treatment for rheumatoid arthritis (RA) is explored by comparing with methotrexate (MTX).Methods MTr method was used to measure the light-absorption value of the cells treated with different doses of Mat and MTX at different time,and the inhibition rate(IR% ) was calculated.Flow cytometry was used to measure cell cycle of the cells treated with different doses of Mat and MTX for 48 h.Results ① Both MTX and Mat could inhibit the proliferation of PBL in patients with RA and the effect was dose-dependent.The IR%was increased (P<0.05).IR% at 48 h was higher than IR% at 24 h (P<0.01).The two medications had no significant difference (P>0.05).② Both MTX and Mat could change the cell cycle and acted on G1/S point.They both increased the number of cells in G0/G1 phase and decreased cells in S and G2/M phase (P<0.05).The two medications had no significant difference (P>0.05).Conclusion Mat inhibits the proliferation and proliferation-dependent processes of PBL in patients with RA.Its effect is comparable to MTX.Mat may have the potential in the trea-tment of RA.
7.Impact of portal hypertension and hypersplenism on prognosis of hepatocellular carci-noma after curative treatment

Objective: To investigate the impact of portal hypertention with hypersplenism of different severity and splenectomy on prognosis of hepatocellular carcinoma (HCC). Methods: We retrospectively analyzed the clinical data of 403 patients with HCC who met the Milan criteria and received radical treatment in Tianjin Third Central Hospital from January 2008 to January 2018. Cox propor-tional risk regression analysis was performed for parameters such as platelet levels (PLT), albumin-bilirubin (ALBI) grade, aspartate ami-notransferase-to-platelet ratio index (APRI), and post-sinusoidal resistance (PSR). HCC patients with severe hypersplenism were as-signed into two groups according to treatment method: radical treatment for HCC alone and radical treatment for HCC plus splenecto-my. Clinical data were compared, and the two groups were evaluated using the Kaplan-Meier survival analysis method. Results: Univar-iate and multivariate analyses showed that PLT was an independent risk factor for overall survival (OS) and disease-free survival (DFS) in patients with HCC. OS curves differed significantly with different PLT among patients with HCC (P=0.013). Furthermore, parameters of portal hypertension in cirrhosis, such as PSR, APRI, and ALBI grade, were risk factors for HCC prognosis. The degree of portal hyper-tension and hypersplenism, liver function, and tumor-node-metastasis stage did not differ between the two groups (P>0.05). Survival analysis showed significantly longer OS in the radical treatment plus splenectomy group (P=0.025). Following were the 1-, 3-, and 5-year survival rates: radical treatment alone group 100% , 98.2% , and 68.5% and radical treatment plus splenectomy group. 97.1% , 79.4%, and 56.8%, respectively. DFS did not differ between the two groups (P=0.326). Conclusions: Clinical parameters, such as PLT, PSR, APRI, and ALBI grade, are important prognostic factors in HCC patients with portal hypertension and hypersplenism. Radical treat-ment for HCC plus splenectomy can improve OS in HCC patients within the Milan criteria with severe hypersplenism.
8.Expression of Aurora-A in cervical cancer and precancerous lesions,and its correlation with HPV infec-tion

Objective The objectives of this study were to investigate the expression of Aurora-A pro-tein in cervical cancer and precancerous lesions,and its relationship with human papilloma virus( HPV) infection, and to analyze the role of Aurora-A in the pathogenesis of cervical cancer. Methods One hundred cases of cer-vical biopsy or surgical resection specimens were collected from high-risk HPV( HR-HPV) test. There were 20 cases of normal cervical tissues,20 cases of CIN grade 1 ( CIN1 ) ,20 cases of CIN grade 2 ( CIN2 ) ,20 cases of CIN grade 3(CIN3),and 20 cases of cervical squamous cell carcinoma. The expression of Aurora-A protein was detected by immunohistochemistry and the correlation between Aurora-A expression and HR -HPV infection was analyzed. Results Aurora-A was highly expressed in cervical intraepithelial neoplasia and cervical cancer (P<0. 05),and its positive expression rate increased with the degree of cervical lesions. There was a positive correlation between Aurora-A expression and cervical cancer(r=0. 475,P<0. 001). There was a positive cor-relation between Aurora-A expression and HR-HPV infection in CIN2 and CIN3(V=0. 591,P<0. 05). Con-clusion Aurora-A may be associated with the development of cervical cancer. Aurora-A can be used as an important biomarker for the early diagnosis of cervical intraepithelial neoplasia or cervical cancer. It is also a po-tential therapeutic target for cervical cancer.
9.Effect of Total Glucosides of White Paeony Capsules on Psychological Ethology and Hypothalamic Neuroendocrine System of Stress Rats

Objective To observe the effects of total glucosides of white paeony(GWP) on psychological ethology and neuroendocrine system of stress rats. Methods Ten male Sprague-Dawley rats were treated as normal control group,the others (n=40) were constructed by isolated custody and chronic unpredictable stimuli to build the model of stress rats.Stress rats were randomly divided into 4 groups:model control group,GWP high dose group and low dose(66,33 mg·kg-1) group,fluoxetine(5. 0 mg·kg-1) group,each group had 10 rats.The changes of rat behavior were monitored by locomotor activity system.The change of emotion was observed by tail suspension test.The pathology change of adrenal was observed by HE staining.The levels of CorT and ACTH in serum were measured by radioimmunoassay.The expression of CRH mRNA of hypothalamic were detected by RT-PCR. Results The activity of the level scoring and vertical scoring of GWP high dose group was obviously improved as compared to that in model control group,while static time was significantly shortened,Adrenal index of GWP groups were increased significantly(P<0.05),thymus index of the treatment group was declined(P<0.05),CorT and ACTH content were decreased (P<0.05) and hypothalamus CRH mRNA of GWP group was decreased(P<0.05). Conclusion The improvement of activity and related hormone anomalous changes were observed in rats treated with GWP.The mechanism of its actions might be involved adjusting the endocrine function,decreasing CorT and ACTH content and the expression of hypothalamic CRH mRNA of stress rats.
10.Analysis on Mechanism of Huangwu Ganfu Ointment in Relieving Knee Osteoarthritis Pain Based on Network Pharmacology and Experimental Verification

Objective To analyze the mechanism of Huangwu Ganfu Ointment in relieving pain of knee osteoarthritis(KOA)based on network pharmacology;To verify it in animal experiments.Methods The active components of Huangwu Ganfu Ointment were obtained by TCMSP database,PubChem database and SwissADME platform,the effective components were screened,and the targets were obtained from SEA database.KOA disease-related targets were obtained from GeneCards,OMIM and other databases,and the intersection targets were obtained.A effective component-target-disease network was constructed using Cytoscape 3.9.0 Software.Protein-protein interaction(PPI)network was constructed by STRING database and core targets were screened.GO and KEGG enrichment analysis of intersection targets were analyzed using DAVID platform.The KOA rat model with cold and damp syndrome was established,and the intervention of Huangwu Ganfu Ointment was carried out.The efficacy was observed and the core target expressions were detected.Results Totally 104 effective components were screened from Huangwu Ganfu Ointment,and 59 potential targets were obtained for treating KOA.PPI network interaction analysis obtained the important targets of IL6,IL1B and PTGS2.KEGG enrichment results showed that Huangwu Ganfu Ointment may involve 84 signaling pathways such as IL-17,TNF,TRP and NF-κB in the treatment of KOA,most of which were related to inflammation.The results of animal experiments showed that Lecuesne MG scores increased in the model rats(P<0.05),and paw withdrawal threshold(PWT)significantly decreased(P<0.05).Compared with model group,PWT in Huangwu Ganfu Ointment medium-and high-dosage groups were significantly recovered,and synovitis Krenn score decreased(P<0.05).The Mankin score of cartilage tissue of Huangwu Ganfu Ointment high-dosage group decreased(P<0.05).The contents of IL-6 and IL-1β in all Huangwu Ganfu Ointment groups decreased(P<0.01).Huangwu Ganfu Ointment medium-and high-dosage groups could down-regulate the expression of TRPV1 protein(P<0.05,P<0.01).Conclusion The mechanism of Huangwu Ganfu Ointment in alleviating the pain of KOA may be related to reducing inflammatory response,reducing the release of inflammatory factors of IL-1β and IL-6,alleviating inflammatory pain sensitivity of KOA,and down-regulating the expression level of TRPV1.
