1.Research on Suicide Attempts and Suicide Attitude Status of Medical University Undergraduates-to Take Kunming Medical University as an Example

Objectives To study the suicide attempts and suicide attitude status and the characteristics of Medical University undergraduates, provide specific recommendations for mental health education and counseling. Methods The investigation was conducted among 567 undergraduates and 130 graduate students in Kunming Medical University with Self-designed Suicide Attempts Investigation and Suicide Attitude Questionnaire (QSA), and the results of different groups were compared. Results 14.46%undergraduates had suicidal ideation, 4.23%undergraduates had suicide attempts, but there were no significant different between female and male. The suicide attitude in general was contradictory and neutral. Sex,academic qualification,grade, family location and economic status variables had influence on suicide attitude. Conclusions Although the medical university undergraduates disapproved of the suicidal behavior, but it still reflected the pursuit and respect for the humanistic concern and value of life to a certain extent.Suicidal attitude had significant different in demographic variables.
2.The effect of exogenous IGFBPrP1 on collagen content of liver tissue in mice

Objective To investigate the effect and mechanism of exogenous IGFBPrP1 on collagen content in liver tissue. Methods Twenty-four male C57BL/6 wild-type mice were randomly divided into three groups: Control group (n=8), rmIGFBPrP1 2 weeks group (n=8) and rmIGFBPrP1 4weeks group (n=8). Both hematoxylin-eosin (HE) staining and picric acid-Sirius red staining were performed. The protein expression of IGFBPrP1, Collagen Ⅰ , Collagen Ⅲ, FN, TGF-β1, Smad3 andp-Smad2/3 was evaluated by immunohistochemistry or Western blot. Results Exogenous IGFBPrP1 can cause pathological changes in liver tissue. Collagen content was significantly increased by hematoxylin-eosin (HE) staining and picric acid-Sirius red staining (P<0.05). The protein expression of IGFBPrP1, FN and Collagen Ⅰ was gradually increased after rmIGFBPrP1 injection for 2 weeks and 4 weeks by Western blot (P<0.01). The protein expression of Collagen Ⅲ was obviously increased in the rmIGFBPrP1 2 weeks group and rmIGFBPrP1 4 weeks group by immunohistochemistry, and the level in the 4 weeks group was higher than that in the 2 weeks group (P<0. 01). The protein expression of TGF-β1, Smad3 and p-Smad2/3 in liver tissue was significantly increased after rmIGFBPrP1 injection in a time-dependent manner by both immunohistochemistry and Western blot (P<0. 01).Conclusion Exogenous IGFBPrP1 can cause a marked increase in collagen content and the excessive deposition of ECM through the TGF-β1/Smad3 pathway in liver tissue.
3.High risk factors of hematogenous metastasis in non-surgical thoracic esophageal squamous cell carcinoma after chemoradiotherapy and its influence on survival and prognosis

Objective:To analyze the risk factors of first-episode hematogenous metastasis in patients with thoracic esophageal squamous cell carcinoma (ESCC) who received non-surgical treatment after radiotherapy and chemotherapy, and its impact on survival and prognosis.Methods:The clinical data of 230 ESCC patients who met the inclusion criteria and received radical radiotherapy in Tengzhou Central People′s Hospital and Affiliated Hospital of Xuzhou Medical University from January 2011 to October 2018 were retrospectively analyzed.Logistic regression analysis and survival were used to analyze the risk factors and prognosis of blood group metastasis after treatment.Results:In 230 patients with thoracic esophageal cancer, 70 cases (30.4%) developed hematogenous metastasis for the first time.Compared with patients without hematogenous metastasis, the median overall survival was 15 months and 20 months (χ 2=7.249, P=0.007), and the median progression free survival was 9 months and 13 months (95% CI was 7.2-10.8 months and 10.8-15.2 months, respectively χ 2=21.664, P<0.001). Logistic multivariate analysis showed that there was significant difference in the occurrence of hematogenous metastasis among different N stages (χ 2=30.764, P<0.001). N stage was an independent factor for judging hematogenous metastasis, and the increased N stage increased the risk of hematogenous metastasis (OR value were 6.000, 12.629 and 48.167, respectively; 95% CI were 1.712-21.025, 3.546-44.976 and 10.848-213.858, respectively; all P<0.05). The overall survival time of patients with concurrent chemoradiotherapy before hematogenous metastasis was longer than that of patients with sequential chemoradiotherapy and radiotherapy alone (χ 2=10.002, P=0.007). Stratified analysis showed that adjuvant chemotherapy after concurrent chemoradiotherapy could prolong the overall survival of patients with N2 and N3 (χ 2=11.025, P=0.001). Conclusion:N staging is an independent factor to judge the hematogenous metastasis.ESCC patients with hematogenous metastasis after chemoradiotherapy have poor prognosis.N2, N3 patients with concurrent chemoradiotherapy after adjuvant chemotherapy have clinical benefits.
4.The application of MRI in sporadic inclusion-body myositis

Objective To investigate the characteristic MRI features of sporadic inclusion?body myositis(sIBM). Methods Clinical and MR imaging data of 6 patients with sIBM diagnosed by muscle biopsy from May 2013 to November 2014 were retrospectively analyzed. All patients showed insidious onset of lower limb muscle weakness and diagnosed as sIBM by muscle biopsies. All patients were evaluated by the score of the severity of fatty infiltration, inflammation and atrophy in MRI. Results All patients were observed fatty infiltration with different degrees. The fatty infiltration in thighs was characterized in a decreasing order of frequency:gluteus maximus (6 cases), vastuslateralis (6 cases), vastusintermedius (6 cases), vastusmedialis (6 cases), sartorius (5 cases), adductor magnus (5 cases), rectus femoris (4 cases), semi?membranosus (4 cases), semi?tendinosus (4 cases), biceps femoris (4 cases), gracilis (3 cases), adductor longus(2 cases).The fatty infiltration in thighs was characterized in a decreasing order of severity:vastuslateralis (3.2 points), vastusintermedius (3.2 points), vastusmedialis (3.0 points), adductor magnus (3.0 points), gluteus maximus (2.7 points), bicepsfemoris (2.2 points), semi?membranosus (2.1 points), semi?tendinosus (2.1 points), rectus femoris (1.5 points), sartorius (1.3 points), gracilis (0.8 points), adductor longus (0.7 points). All patients showed the features of distal distribution andsymmetry. Inflammation was observed in 3 patients. 1 patient only involved the vastuslateralis, the other 2 patients were observed muscle inflammation with different degrees in 12 muscles. Atrophy was observed in 5 patients. The atrophy in thighs was characterized in a decreasing order of frequency:vastuslateralis (5 cases), vastusintermedius (5 cases), vastusmedialis (4 cases), adductor magnus (4 cases), semi?membranosus (2 cases), rectus femoris (1 cases), sartorius (1 cases) and gluteus maximus;there was no atrophy in adductor longus, gracilis,semi?tendinosus, biceps femoris. Conclusion The MRI characteristic manifestations of sIBM is fatty infiltration and atrophy in the distal portion, particularly involving the vastuslateralis, vastusintermedius, vastusmedialis and adductor magnus.
5.Effect of Er:YAG laser on the dynamic changes in subgingival flora in type 2 diabetic patients with periodontitis.

OBJECTIVETo observe the changes in subgingival microflora before and after Er:YAG laser treatment on diabetic patients with periodontitis, and to compare with the subgingival microflora of chronic periodontitis.
METHODSSubgingival plaque of 13 pairs of teeth (26 sites) was selected from type 2 diabetic patients at pretreatment, one month post-treatment, and three months post-treatment. Subgingival plaque was also obtained from 11 cases of moderate to severe chronic periodontitis with similar severity of periodontitis. The DNA of the subgingival plaque samples was extracted. Whole bacterial 16S rDNA gene fragments separated by denaturing gradient gel electrophoresis. Specific DNA bands were then chosen for retrieval and sequencing.
RESULTSThe gene sequencing results of the special DNA bands of subgingival plaque samples show that the pathogenic bacteria of both diabetic periodontitis and simple chronic periodontitis were Prevotella intermedia and Tannerella forsythia, respectively. The composition of the subgingival microflora before and after laser treatment changed. Some DNA bands, including that of Tannerella forsythia, disappeared or weakened one month after treatment. A new strip appeared, which belonged to Actinomyces sp.
CONCLUSIONThe profiles of the subgingival microflora changed after treatment, and one month was indicated as an important stage. Er:YAG laser may have an important function in delaying microflora recolonization.

6.Effects of different concentrations of irbesartan on the differentiation and mineralization of preosteoblasts

BACKGROUND:Angiotensin II receptor antagonists have been found to exerct a stronger protective effect on bone than angiotensin converting enzyme inhibitors. OBJECTIVE:To investigate the effect of different concentrations of irbesartan (angiotensin II receptor antagonist) on the differentiation and mineralization of mouse preosteoblasts. METHODS:Mouse preosteoblast cel lines MC3T3-E1 in logarithmic phase were selected and cultured in the osteogenic induction medium containing 0 (control group), 0.001, 0.01, 0.1 mmol/L irbesartan, respectively. Ten days later, the cel differentiation was observed by alkaline phosphatase staining. The mineralization was observed by alizarin red staining after 21 days of culture. mRNA expressions of osteocalcin, alkaline phosphatase and Runt-associated transcription factor 2 in osteoblasts were detected by real-time PCR at 1, 4, 7, 14 and 21 days of culture. RESULTS AND CONCLUSION:The activity of alkaline phosphatase in al the irbesartan groups (0, 0.001, 0.01, 0.1) was higher than that in the control group (P<0.05), which was the most obvious in 0.01 mmol/L. The number and area of calcium nodules in each irbesartan group were significantly higher than those in the control group (P<0.05), especial y in 0.01 mmol/L. Compared with the control group, 0.01 mmol/L irbesartan significantly upregulated the mRNA expressions of osteocalcin, alkaline phosphatase and Runt-associated transcription factor 2 (P<0.05). These results suggest that 0.01 mmol/L irbesartan significantly promotes the differentiation and mineralization of osteoblasts.

AIM:To determine the contents of euphorbiasteroid and lathyrol diadetate benzozte in Semen Euphorbiae and Semen Euphorbiae Pulveratun from different habitats.METHODS:A kromasil C_(18) column(150 mm×4.6 mm,5 μm)was used with methanol-water(68:32)as the mobile phase and UV detection was at 280 nm.RESULTS:The linear ranges of euphorbiasteroid and lathyrol diadetate benzozte were from 0.200 μg to 1.200 μg and 0.206 μg to 1.651 μg(r = 0.999 7,r = 0.999 9).The contents of euphorbiasteroid and lathyrol diadetate benzozte in Semen Euphorbiae from different habitats ranged from 0.450 8% to 0.625 1 % and 0.692 3% to 0.752 9%.The contents of two kinds of compositions in Semen Euphorbiae Pulveratun ranged from 0.156 8% to 0.221 1% and 0.232 2% to 0.283 0%.CONCLUSION:The method for quantitating euphorbiasteroid and lathyrol diadetate benzozte in Semen Euphorbiae is accurate and reliable,and can be used to control the quality of toxicity Semen Euphorbiae and Semen Euphorbiae Pulveratun.

9.Effects of Total Saponins from Panax Japonicus on Acute Hepatic Injury Induced by Carbon Tetrachloride

Objective To discuss the protection mechanism of total saponins from Panax japonicus on acute hepatic injury induced by carbon tetrachloride. Methods HepG2 cells were used to establish CCl4-induced liver cell injury model in vitro experiments. Mouse model of acute liver injury was caused by 1%CCl4 oil on Balb/c. Mice were randomly divided into normal group, model group and medicine group. Mice in the medicine group were given a gavage with 20 mL/kg total saponins from Panax japonicus, while mice in the other two groups were given a gavage with the same amount of stroke-physiological saline solution. MTT method was used to detect the activity of hepatic cells. The pathological changes of mouse liver were examined by HE staining. RT-PCR was used to detect changes in the expression of transforming growth TGF-β, TNF-α, and TLR4 mRNA. Results Compared with normal group, less hepatic cells survived in model group (P<0.01);compared with model group, more hepatic cells survived in medicine group (P<0.01). HE staining showed that damages in liver tissues of medicine group significantly improved than those in model group. RT-PCR results showed that the levels of TGF-β, TNF-α, and expression of TLR4 mRNA increased more significantly than those in model group;the expression of TGF-β, TNF-α, and TLR4 mRNA in medicine group decreased more significantly than those in model group, with statistical significance (P<0.01). Conclusion The total saponins from Panax japonicus has a protective effect on liver injury induced by carbon tetrachloride.
10.Study on Adjuvant Immunoactivity of Polysaccharides from Panax Japonicus

Objective To study the adjuvant immunoactivity of polysaccharides from Panax japonicus by alcohol of different concentrations;To discuss its part with the strongest adjuvant immunoactivity. Methods Polysaccharides from Panax japonicus was sunk with alcohol of different concentrations, and 30%alcohol compound, 60%and 90%alcohol polysaccharide were obtained. Different segments of polysaccharide and OVA protein were injected to mice once a week for three times for immunity. Five days after the last immunity, the mice were executed to collect blood, and the antibody titer was determined. The three parts of alcohol compound were scanned by infrared spectrum to determine the type of polysaccharide preliminarily. Results Compared with the control group, the antibody titer of different segments of polysaccharide obviously increased, especially the polysaccharide sunk by 60%alcohol. Infrared spectrum analysis showed that polysaccharides from Panax japonicus contained pyranose ring structure. Conclusion Polysaccharides from Panax japonicus has significant adjuvant immunoactivity, and polysaccharide sunk by 60%alcohol has the strongest adjuvant effects.
