1.Purification and enzyme activity assay of filamentous temperature-sensitive protein Z in Streptococcus suis serotype 2

We conducted purification of filamentous temperature-sensitive protein Z of Streptococcus suis serotype 2 (S.suis 2) and measured its GTPase activity.The ftsz gene in the genome of the Chinese 05ZYH33 strain of S.suis 2 was successfully amplified using PCR,and then the ftsz gene was cloned into prokaryotic expression plasmid pET28a,and the recombinant plasmid pET28a-ftsz was transformed into E.coli BL21.After induction by IPTG,the isolated FtsZ protein was analyzed with SDS-PAGE.Then the recombinant protein was purified by Ni2+-NTA affinity chromatography.The rabbit serum was harvested after immunization with recombinant FtsZ protein,and was analyzed by indirect ELISA and Western blotting.The GTPase activity of FtsZ was measured with the malachite green method.Results showed that successfully constructed recombinant plasmid pET28a-ftsz and the recombinant protein with high purity was obtained;Western blot result indicated that FtsZ could react with the His-tag antibody and the rabbit serum;the polyclonal antibody titer of the rabbit serum reached 1 ∶ 13 107 200;FtsZ have GTPase activity.We successfully prepared S.suis 2 recombinant protein FtsZ having GTPase activity and high titer antiserum would be useful for the further study of S.suis 2 cell division mechanism.
2.Latent classes of self-disclosure and their psychological distress difference in patients with gynecologic cancer

Objective To explore the self-disclosure characteristics of gynecologic cancer patients,and compare psychological distress of each latent class.Methods A total of 177 gynecologic cancer patients from 8 tertiary hospitals were investigated by demographic questionnaire,the Distress Disclosure Index (DDI) and the Distress Thermometer (DT).Results The result showed that 3 latent classes model of self-disclosure was supported,including" High level-willing to disclosure to various people" (39.55%)," Medium level-willing to disclosure to spouse" (20.90%) and " Low level-not willing to disclosure to anyone" (39.55%).Significant differences were found in the effect of residence (x2 =9.341,P<0.05),education level (x2=16.862,P<0.05) and cancer type(P=0.009) on the latent class among these groups.Moreover,the psychological distress scores of the 3 latent classes were 6.61± 1.78,4.59± 1.57 and 3.67± 1.14,and the differences were statistically significant (x2 =83.56,P<0.05).Conclusion The self-disclosure of gynecological cancer patients can be divided into three classes and their psychological distress is different.So the specific intervention methods can be developed to improve the level of self-disclosure and psychological distress of gynecological cancer patients.
3.The relationship between spontaneous apoptosis and radiation-induced apoptosis of laryngeal carcinoma.

OBJECTIVE:
To study the change of laryngeal carcinoma cell apoptosis before and after radiotherapy.
METHOD:
Living tissue of diseased region obtained from 27 cases of laryngeal carcinoma before and during radiotherapy 10, 30 and 60 Gy were used in this study. With terminal deoxynucleotidyl transferase mediated dUTP biotin nick end labeling (TUNEL), the spontaneous and radiation-induced apoptotic rate (AR)were examined.
RESULT:
The AR of living tissue of laryngeal carcinoma before and during radiotherapy 10, 30 and 60 Gy were (21.0 +/- 0.7)%, (60.0 +/- 1.5)%, (42.0 +/- 1.3)%, (25.0 +/- 1.9)%, respectively. The radiation-induced apoptotic rate was significantly higher than that of spontaneous apoptotic rate (P< 0 . 5). In laryngeal carcinoma, spontaneous apoptosis was positively correlated with the radiation-induced apoptosis.
CONCLUSION
Detecting of AR of laryngeal carcinoma cells before and during radiotherapy may be helpful to predict the sensitivity of radiotherapy in laryngeal carcinoma patients.

4.Influencing factors of willingness to participate in time banking mutual care model among the elderly in urban communities in China: a literature review

With the accelerating process of aging, the burden of elderly care in China is increasing dramatically. However, the current care models were difficult to meet the multi-level and diversified needs of the elderly. Time banking mutual care model is proposed as an important auxiliary model. Since the elderly in urban communities are the major participants of time banking mutual care model, understanding their willingness to participate is very important for the effective implementation and sustainable development of time banking mutual care model. This paper summarizes the research progress and the major influencing factors of the willingness of the elderly to participate in time banking mutual care model, in order to provide theoretical support for alleviating the increasingly severe burden of elderly care in China and realizing active aging.
5.The antioxidant transcription factor Nrf2 and Rad51 interact in BLM reduced DNA damage

Objective :
To explore the mechanism of nuclear factor E2-related factor 2 (Nrf2) in bleomycin (BLM) -induced DNA damage of lung cancer cells.
Methods :
The fluorescence intensity of tissue reactive oxygen species (ROS) in human lung cancer cells A549 and H1299 was detected by a reactive oxygen species fluorescent probe ( DCFH-DA) .DNA damage of A549 and H1299 cells treated with 5 μg / ml BLM was detected by Western blot and micronucleus.Then,the Nrf2-low expression transient siRNA and the stably shRNA-transformed lung cancer cells were respectively constructed to detect the DNA damage after BLM.The interaction between Nrf2 and Rad51 in cells was detected by co-immunoprecipitation method.
Results :
Under the action of BLM,ROS levels in A549 and H1299 cells increased significantly,and Nrf2 entered the cell nucleus for activation(P<0. 01) .Under the effect of BLM,resveratrol ( Res) stimulated A549 and H1299 cells to up-regulate the protein expression of Nrf2 ( P < 0. 01) ,while the expression of DNA damage marker protein γ-H2AX decreased(P<0. 01) .On the contrary,after interfering the protein expression of Nrf2 in A549 and H1299 cells with siRNA,the protein expression of γ-H2AX in A549 and H1299 cells was significantly up-regulated under the action of BLM(P<0. 01) .Western blot results showed that the expression of homologous recombination repair protein Rad51 and the expression of Nrf2 protein in A549 cells with low Nrf2 expression were significantly down-regulated (P <0. 01) .
Conclusion
The antioxidant transcription factor Nrf2 in tumor cells can interact with Rad51 to regulate the expression of Rad51 and reduce BLM-induced cellular DNA damage.

Objective To evaluate the survival and prognostic factors of esophageal cancer treated with definitive ( chemo ) radiotherapy by applying novel radiation techniques including three-dimensional conformal radiotherapy (3DCRT) or intensity-modulated radiotherapy (IMRT). Methods Clinical data of 2762 patients with non-operated esophageal squamous cell carcinoma who underwent definitive ( chemo ) radiotherapy from 2002 to 2016 in 10 hospitals were retrospectively analyzed.The prognostic factors were also identified and analyzed. Results The median follow-up time was 60. 8 months. The 1-, 2-, 3-and 5-year overall survival (OS) of all patients was 71. 4%,48. 9%,39. 3%,and 30. 9%,respectively.The 1-,2-,3-and 5-year progression-free survival (PFS) was 59.5%,41.5%,35.2%,and 30%,respectively.The median survival was 23 months.The median time to progression was 17. 2 months.Multivariate analysis demonstrated that age, primary tumor location, clinical stage, tumor target volume, EQD2 and treatment mode were the independent prognostic factors for OS.Primary tumor location,clinical stage,tumor target volume and EQD2 were the independent prognostic factors for PFS. Conclusions In this first large-scale multi-center retrospective analysis of definitive ( chemo) radiotherapy for esophageal squamous cell carcinoma in China, the 5-year OS of patients with esophageal squamous cell carcinoma is significantly improved by 3DCRT, IMRT combined with chemotherapy drugs. However, the findings remain to be validated by prospective clinical trials with high-level medical evidence.
