1.Research progress of bone marrow mesenchymal stem cells in the treatment of retinopathy

Stem cell therapy for retinal disease is under way,and several clinical trials are currently recruiting. These trials use human umbilical cord tissue-derived stem cells and bone marrow-derived stem cells to treat visual dis-orders such as age-related macular degeneration,and retinitis pigmentosa. Mesenchymal stem cells( bone marrow mes-enchymal stem cells,BMSCs) belong to adult pluripotent stem cells,capable of unlimited self-renewal,have the poten-tial multi-directional differentiation,and more recent researches,mainly through such cells can differentiation into reti-nal characteristics and morphology similar to or the same structural characteristics of functional impairment, which is widely used in the treatment of retinal diseases. In recent years′researches,whether in the vivo or in vitro experiments, BMSCs have the potential to differentiate into nerve cells offering the selectivity of treatment in retinal injury and disea-ses. So,this view discussed the biological characteristics of BMSCs and bone marrow in the mechanism of action of reti-nopathy,relevant treatment.
2.Reliability and Validity of Chinese Revision of the Anxiety Sensitivity Index-Revised

Objective: To develop the Chinese version of the Anxiety Sensitivity Index-Revised and examine its replicability, reliability and validity. Methods: This study translated the Anxiety Sensitivity Index-Revised ( ASI-R) and examined the factor analytic structure of anxiety sensitivity in a large sample of middle school students aged 12-18 years (n = 1556) . Factor analysis of the ASI-R items resulted in a hierarchical construct with three lower-order factors loading on a single higher-order factor. Results: after revision, the number of items of the inventory reduced from 36 to 15. Anxiety sensitivity seems to be a hierarchically organized construct with one higher-order factor (anxiety sensitivity ) and three lower-order factors: Physical concern, Cognitive concern (lose of control) , Social concern, which three factors contributed to 52. 29% of total variance. Factor loadings of items were 0. 63 ~ 0. 98 , the internal correlation between items were 0. 37 ~ 0. 53 , the correlation between factors to the total score were 0. 72 ~ 0. 84, internal correlation among factors were 0. 68 ~ 0. 78. internal consistency of each factor 0. 73 ~ 0. 78. the split-half reliability was 0. 71 ~0. 76, that of test-retest was 0. 70 ~0. 78. Conclusion: The questionnaire has satisfying reliability and validity.
3.Prevention and control of nosocomial infection in NICU:current status and future challenges

Nosocomial infection (NI), a major cause of morbidity and mortality in neonates, is the common challenges faced by health workers worldwide. This paper was to review the epidemiology, major challenges, prevention and control mea-sures of NI in neonatal intensive care unit. The present situation of NI in China and the corresponding countermeasures are also discussed.
4.Design and application of disease transfer table based on standardized communication between the ;maternity ward and the operating room

Objective To design the disease transfer table of standardized communication between the maternity ward and the operating room,and explore the application effect of disease transfer table based on situation (S)-background (B)-assessment (A)-recommendation (R) communication in cesarean delivery of maternal. Methods 570 pregnant women and 30 medical staff from two wards of maternity ward were selected as research object, the first ward was the control group, the second ward was observation group. The control group used traditional way of disease transfer,and the observation group used SBAR transition table for disease transfer, compared the incidence of adverse events after caesarean section of puerperal and the satisfaction of disease transfer of medical staff from two groups. Results The incidence of wound bleeding, postpartum hemorrage caused by uterine inertia, wound infection of observation group was 2.06%(6/291), 1.71%(3/291), 1.37%(4/291), which was lower than 3.94%(15/279), 3.58%(10/279), 4.66%(13/279) of control group (χ2=4.41, 4.17, 5.31,P < 0.05). The satisfaction of comprehensive, state of the iuness, work acceptance, transfer efficiency of medical staff was (4.75 ± 0.12), (4.82 ± 0.55), (4.86 ± 0.67), (4.76 ± 0.22) points, which were higher than (3.04 ± 0.29), (3.11 ± 0.35), (3.09 ± 0.32), (3.01 ± 0.42) points of control group (t=2.72-3.12,P < 0.05). Conclusions The SBAR condition transition table can improve the efficiency and quality of disease transfer between maternity ward and operating room, ensure the care safety of cesarean section maternal.
5.Intra-amniotic endotoxin and postborn hyperoxic exposure upregulate transforming growth factor-?1 and ?-smooth muscle actin in newborn mice lung

Objective To investigate the expressions of transforming growth factor-?1(TGF-?1)and ?-smooth muscle actin(?-SMA)in the lungs of mice with intra-amniotic endotoxin priming and exposed to 60% hyperoxia after born in order to elucidate the possible relationship with bronchopulmonary dysplasia(BPD).MethodsFifty C57 pregnant mice were divided into 2 groups:lipopolysaccharide(LPS,40 ?g/L)group and saline solution group,and then received an intra-amniotic injection of corresponding solution on E15.The neonatal mice of each group were randomized to be set in 60% oxygen exposure or in room air.So there were 4 subgroups,LPS+air,LPS+hyperoxia,saline+air and saline+hyperoxia groups.On days 1,3,7,10 and 14 after birth(8 rats each time point),the lung histological changes was assessed with hematoxylin and eosin(HE)staining for radial alveolar counting(RAC).The expressions of TGF-?1 and ?-SMA proteins were detected by immunohistochemical and immunofluorescence staining,and the expressions of TGF-?1 and ?-SMA mRNA by real-time polymerase chain reaction(RT-PCR).ResultsIn the LPS+hyperoxia group and saline+hyperoxia group,RAC began to decrease on day 3,and then further declined in a time-dependent manner.Compared with saline+hyperoxia group,LPS+hyperoxia group had significantly lower RAC(P
6.Expression of TRAP1 in esophageal carcinoma and its influence on the prognosis

Purpose To investigate TRAP1 expression in esophageal cancer and its relationship with clinicopathological features and prognosis.Methods Expression levels of TRAP1 in 60 pairs of cancer and adjacent normal tissues were detected by immunohistochemistry,and the relevance between TRAP1 and clinicopathological features was evaluated.Kaplan-Meier and Cox proportional regression analyses were performed to determine the association of TRAP 1 expression and survival of the patients.Results The positive expression rate of TRAP1 was 55.0%,and the amount of relative protein transcript level was 2.7 ± 1.1 in the cancer tissue.The positive expression rate of TRAP1 was 11.7%,and the relative expression protein was 0.5 ± 0.4 in the adjacent normal tissue.The expression level of TRAP1 in cancerous tissue was significantly higher than that of tissue adjacent to carcinoma.There was no statistically significant difference between the expression levels of TRAP1 with sex,age,location,and degree of differentiation (P > 0.05).There was statistically significant relationship between recurrence,metastasis and TNM stages with the expression of TRAP1 levels (P < 0.05).The average survival time of TRAP1-negative patients was 69.0 months (95% CI:60.2-77.9) while the TRAP1-positive patients was 34.2 months (95% CI:24.4-44.1).High levels of TRAP1 were correlated with decreased survival of postoperative esophageal cancer patients (P < 0.05).Conclusion TRAP1 is overexpressed in esophageal cancer and associated with the progression and prognosis of esophageal cancer.
7.Effects of triptolide on the expression of androgen receptor in human prostate LNCaP cells and its mechanism of action.

To study the regulation of androgen receptor (AR) expression in human prostate cancer LNCaP cells by triptolide (TP) and the possible mechanism, by using qRT-PCR and Western blot, the AR mRNA and protein levels in TP treated LNCaP cells were detected, and the AR protein level in TP and NF-κB inhibitor treated LNCaP cells was also detected; a series of pGL3-AR promoter reporter gene vectors were built using restriction-free cloning method, and the vectors were employed to investigate the effects of TP on the transcriptional activity of AR promoter in LNCaP cells; the upstream proteins which may play regulatory roles were detected using western blot assay. After treated LNCaP cells with TP for 48 h, AR mRNA and protein expressions decreased with increasing TP concentration. The expression of AR target gene PART1 and prostate specific antigen (PSA) was also downregulated by TP treatment; a series of pGL3-AR promoter reporter vectors were constructed and validated by sequencing and luciferase activity; the results of dual luciferase reporter assay showed that TP downregulated AR at the transcriptional level; PI3K/AKT/NF-κB pathway which is associated with AR promoter activity was drowregulated by TP. In conclusion, our results demonstrated that the transcriptional activity of AR in LNCAP cells was downregulated by TP, and PI3K/AKT/NF-κB pathway may be involved in the regulation mechanism.
8.Correlation between vertebral artery dominance and posterior circulation ischemic stroke

Objective To investigate the correlation between vertebral artery dominance (VAD) and posterior circulation ischemic stroke (PCIS).Methods The consecutive patients with acute ischemic stroke hospitalized from November 2013 to October 2015 were collected.All patients underwent MRI and magnetic resonance angiography (MRA).The dominant vertebral artery was defined as having the larger diameter if left and right diameter difference ≥0.3 mm or the vertebral artery connected to the basilar artery in a more straight fashion if the diameter difference < 0.3 mm.They were divided into either an anterior circulation ischemia stroke (ACIS) group or a PCIS group according to the classification criteria of Oxfordshire Community Stroke Project (OCSP).A multivariate logistic regression analysis was performed to investigate the correlation between VAD and PCIS.Results A total of 226 patients withacute ischemic stroke were enrolled,including 172 patients (76.1%) in the ACIS group and 54 (23.9%) in the PCIS group.Fifty-four patients (23.9%) had VAD,including 38 (70.3%) on the left and 16 (29.7%) on the right.The proportion of the patients with VAD in the PCIS group was significantly higher than that in the ACIS group (55.6% vs.14.0%;x2 =39.115,P < 0.001).Multivariate logistic regression analysis showed that VAD was an independent risk factor for patients occurring PCIS (odds ratio,13.60,95% confidence interval 6.90-27.01;P<0.001).Conclusions VAD is closely associated with the occurrence of PCIS,and it is an independent risk factor for PCIS.
9.Use of optimized superovulation conditions for C57 BL/6 J mouse purification

Objective To define the best dose of PMSG/hCG for superovulation and optimize the biological purification system for genetic-modified mice of C57BL/6J background, we injected different dose of PMSG/hCG to promote superovulation, and carried out in vitro fertilization and embryo transplantation.Methods 5-week old female C57BL/6J mice were injected with 4 different doses(5 IU,7.5 IU,10 IU and 15 IU, respectively) of PMSG/hCG.Then the mice were mated with wild type male mice and embryos were obtained by cesarean section.In another approach, oocytes of superovulated mice were fertilized in vitro and then transplanted into surrogate female mice . Results In the cesarean section group, the highest rate of pregnancy was 89.00 ±19.05% in the 5 IU injection group.But the difference compared with other groups was not statistically significant.In the embryo transplantation group, the average number of oocytes from 10 IU injection group was 30.33 ±0.89, and the average number of 2-cell embryo was 23.78 ±0.19, significantly higher than those in the other three groups.Conclusions Administration of 5 IU PMSG/hCG will improve mating efficiency in cesarean section-based purification of C57BL/6J mice.10 IU PMSG/hCG is the best dose for superovulation of C57BL/6J mice.The number of 2-cell embryos is maximized when 10 IU PMSG/hCG is used in the embryo transplantation.
10.Effects of prolonged hyperoxia on pulmonary microvascular development and expression of Ehprin-B2 in neonatal mouse lungs

ObjectiveTo investigate the effects of long-term oxygen exposure on the pulmonary microvascular development and the expression of Ephrin-B2 of lungt issue in neonatal mice.MethodsForty-eight 2-day-old Kunming mice were randomly divided into hyperoxia group and air group, with 24 mice in each group. Mice in hyperoxia group were exposed to 70% oxygen to establish a model of bronchopulmonary dysplasia (BPD). Six mice from each group were sacrificed at 3, 7, 14 and 21 days of age, and lung tissue was collected for further test. The lung sections were stained with hematoxylin and eosin for histological evaluation, radial alveolar counts (RAC) and microvessel density (MVD) measurement by CD34 immunohistochemistry. Location and expression of Ephrin-B2 in lung tissue were measured by immunohistochemistry. Ephrin-B2 mRNA and protein levels were detected by fluorescent quantitative reverse transcriptase-polymerase chain reaction and Western blot, respectively. Two independent samplest-test was used for statistical analysis. Results(1) Pathological changes: The pathology of lung tissue in hyperoxia group showed typical BPD-like changes with advancing postnatal age, presenting mainly with simplified alveolar development and decreased microvessel number. Compared with the air group, RAC and MVD were significantly decreased in 14-day-old mice (6.067±0.432 vs 6.950±0.243,t=4.365,P<0.05; 4.133±0.476 vs 4.867±0.472,t=2.680,P<0.01) and 21-day-old mice in the hyperoxia group (8.050±0.362 vs 9.817±0.487,t=7.127,P<0.05; 4.333±0.532 vs 6.017±0.937,t=3.828,P<0.01). (2) Location and expression of Ephrin-B2: Ephrin-B2 was mainly expressed in alveolar epithelial cells, and weakly expressed in alveolar septum. Compared with the air group, the average optical density of Ephrin-B2 was significantly decreased in 7-day-old (0.146±0.013 vs 0.153±0.009), 14-day-old (0.140±0.007 vs 0.161±0.006) and 21-day-old mice in the hyperoxia group (0.138±0.008 vs 0.166±0.009)(t=-2.049,-9.442 and-10.087, allP<0.05). (3) Ephrin-B2 mRNA and protein levels: The Ephrin-B2 mRNA levels in 14-day-old (0.65±0.14 vs 1.05±0.16,t=4.609,P<0.01) and 21-day-old mice (0.57±0.09 vs 1.13±0.18,t=6.816,P<0.01) were significantly lower in hyperoxia group than in the air group. The Ephrin-B2 protein levels were also significantly lower in hyperoxia group than in the air group in 21-day-old mice (0.13±0.03 vs 0.29±0.08,t=4.587,P=0.000).ConclusionsOxygen-induced BPD model mice have simplified alveolar development, reduced MVD and decreased expression of Ephrin-B2 in lung tissue, which may play an important role in the pathogenesis of BPD.
