1.Study on differential estrogen receptor beta expression of mandibular condylar chondrocyte in temporomandibular joint osteoarthritis.

OBJECTIVETo test the differential Estrogen Receptor (ER) beta levels of mitochondrial proteome of mandibular condylar chondrocyte in the rat model of temporomandibular joints osteoarthritis (TMJOA).
METHODS25 SD rats were divided into experimental group (15 rats) and control group (10 rats) randomly. TMJOA models were created in left sides of TMJ of 15 SD rats by the partial resection of the articular disc. The experimental rats were killed 3 months after operation. After the chondrocytes culture, Immunohistochemistry, semiquantitative RT-PCR and Western blot were used to test the differential ER beta expression levels in mitochondrial proteome of mandibular condylar chondrocytes. Mitochondrial proteins identification was carried out by two-dimensional electrophoresis and peptide mass fingerprint (PMF).
RESULTSPMF showed that one of the differential proteins was ERP. Immunohistochemistry and Western blot results suggested the significant difference of ERbeta protein levels between operation-treated and control group. The operation-treated group had lower ERbeta levels (P < 0.01).
CONCLUSIONIt has been demonstrated that ERbeta protein levels were decreased in mitochondria of TMJOA mandibular condylar chondrocytes, which suggests a role for mitochondrial ERbeta in the effects on TMJOA. The pathological role of ERbeta in the regulation of TMJOA progress remains to be elucidated.

2.Analysis of the electromyographic characteristics on shoulder fatigue caused by lifting load.

OBJECTIVETo analyze and evaluate the fatigue of shoulder skeletal muscle caused by different lifting loads with surface electromyography (sEMG).
METHODSAccording to the loading standard of1 Repetition Maximum (1RM), ten male volunteers performed 3 tasks of upper limb flexion, i.e. 10%, 50% and 90%-1RM. During action process, the signals of Upper Trapezius (UT), Lower Trapezius (LT), Serratus Anterior (SA) and Anterior Deltoid (AD) were recorded by sEMG. The Mean Amplitude (MA) served as an index to evaluate the changes in skeletal muscle fatigue.
RESULTSThe scores of Borg were 15.6, 15.9 and 15.2 for 3 loads of 10%-1RM, 50%-1RM and 90%-1RM, respectively (P > 0.05). The mean amplitudes (MAs) of Upper Trapezius, Lower Trapezius, Anterior Deltoid and Serratus Anterior in shoulders increased obviously. Under the load intensity of 10%-1RM, the MAs of Upper Trapezius and Anterior Deltoid increased significantly (P < 0.05), which were 0.898 and 0.736, respectively. After the exhaustion, the contribution of mean amplitude in shoulder muscle did not change significantly (P > 0.05).
CONCLUSIONThe low-load action for long time can induce easily the fatigue of upper trapezius and anterior deltoid.

3.Relationship between choroidal thickness and idiopathic macular epiretinal membrane

·AIM: To investigate the relationship between the thickness of choroid and the development of idiopathic macular epiretinal membrane (IMEM). ·METHODS: A retrospective analysis on 48 cases (48 eyes) of idiopathic macular epiretinal membrane patients (the IMEM group) was taken in our hospital from January 2014 to December 2016, and 50 right eyes in 50 healthy persons with physical examination were selected the control group, comparison on subfoveal choroidal thickness (SFCT) levels of sicked eyes, normal eyes in IMEM group and the control group were made, postoperative SFCT level change of sicked eyes in IMEM patients and normal eyes were investigated via follow-up, and analysis on correlation between postoperative choroidal thickness and the best corrected visual acuity was taken. ·RESULTS: The SFCT of sicked eyes in IMEM group 362.22±40.75μm was significantly lower than that of the contralateral eyes (410.56 ± 38.45μ m) and the right eyes of the control group (420.73±39.63μ m), and data of the contralateral eyes was lower than right eyes of the control group, distinct difference was shown between groups(P<0.05). The IMEM group's SFCT of sicked eyes and normal eyes at postoperative 1wk had no significant difference with that before operation (P>0.05), and at postoperative 1mo,SFCT of sicked eyes and normal eyes evidently increased, showing sharp difference compared with that before operation (P<0. 05). After that, the SFCT value stabilized, but there was no obvious difference between the sicked eyes and healthy eyes at postoperatively 1mo (P>0.05). The numbers of patients whose postoperative BCVA ≥ 0. 5 with different preoperative SFCT values had statistically significant differences (P<0.05), and those with BCVA ≥0.5 of SFCT values> 380μ m were significantly higher than those with <320μ m and 320μ m-380μ m groups. Fisher exact probability analysis showed that the differences were significant. Pearson analysis showed that there was a positive correlation between the postoperative choroid thickness and the best corrected visual acuity of IMEM group (r=0.629,P<0.05). · CONCLUSION: Choroidal thinning may be an important cause of IMEM, and preoperative choroidal thickness also has an influential effect on postoperative visual recovery.
4.Relationship between interleukin-6 gene-572C/G polymorphism and chronic periodontitis

Objective To investigate the relationship between interleukin-6(IL-6) gene -572C/G polymorphism and the risk of chronic periodontitis disease. Methods IL-6-572C/G genotype polymorphism was examined in 93 patients with chronic periodontitis and 96 control subjects by PCR restriction fragment length polymorphism(PCR-RFLP) method. Results IL-6-572GG genotype and G allele were more frequent in chronic periodontitis patients than that in control subjects (P < 0. 05). -572CC, CG and GG genotype frequency were 52.7%, 40. 9 %, 6.4 % in chronic periodontitis patients and 67.7 %, 31.3%, 1.0 % in control subjects, respectively. -572CG + GG geuotype and G allele were more frequent in patients than that in control subjects (P < 0. 05). Compared with CC genotype, the odds ratio for chronic periodontitis was 1.88(95% CI: 1.04-3.40, P<0.05) for CG+GG genotype. Conclusions The frequency of-572CG+GG genotype was higher in patients with chronic periodontitis compared with the control subjects. IL-6-572C/G polymorphism may be a genetic susceptibility factor for chronic periodontitis in Chinese Hans population.
5.Correlation between a blood pressure variability-based scoring system and target organ damage in patients with hypertension.

OBJECTIVETo investigate the relationship between a blood pressure variability (BPV)-based scoring system (BPVSS) and the target organ damage in patients with hypertension.
METHODSWe selected 95 consecutive inpatients with essential hypertension admitted between January and June, 2015 in the Department of Cardiology of Guangzhou General Hospital of Guangzhou Military Command. The BPV indices were analyzed for their correlation with the parameters of target organ damage (IVSd, LVPWd, baPWV_L/R, and IMT_L/R). The patients with a BPVSS of 3.9 or higher (control, 43 cases) and those with a lower BPVSS (observation group, 52 cases) were compared for differences in IVSd, LVPWd, baPWV_L/R, IMT_L/R and the proportion of carotid plaques.
RESULTSSimilar with the traditional BPV indices, BPVSS was negatively correlated with IMT_L/R (r=-0.278/-0.324, P<0.05). BPVSS was also negatively correlated with IVSd (r=-0.241), LVPWd (r=-0.223), and baPWV_L/R (r=-0.468/-0.373) (P<0.05). IVSd, LVPWd, baPWV_L/R and IMT_L/R were all significantly higher in the observation group than in the control group (t=2.307, 2.516, 3.250/2.790, and 2.372/3.425, respectively; P<0.05). The proportion of carotid plaques in the observation group was significantly higher than that in the control group (Χ(2)=27.833, P<0.001).
CONCLUSIONBPVSS indicates the severity of target organ damage in patients with hypertension. A greater BPV is correlated with a lower BPVSS score and more severe damages of the heart and blood vessels.

6.AM1241 preconditioning reduces microglial activation and injury induced by lipopolysaccharide

Objective To investigate the effect of preconditioning with cannabinoid CB2 receptor agonist AM1241 on microglial activation and injury induced by lipopolysaccharide ( LPS) and interferon-γ (IFN-γ). Methods The microglial cells were chosen and assigned to control group,AM1241 treatment group, LPS/IFN-γ inducement group and AM1241+LPS/IFN-γ treatment group. Cells of control group were cultured in normal medium;cells of AM1241 treatment group were preconditioned with AM 1241 for 2 h, and then the medium was changed with normal medium;cells of LPS/IFN-γ inducement group were exposed to the medium containing 1 μg/mL LPS plus 50 U/mL IFN-γ for 24 h;cells of AM1241+LPS/IFN-γ treatment group were preconditioned with AM1241, then the medium were changed with normal medium for 2 h, and at last, cells of this group were exposed to 1 μg/mL LPS plus 50 U/mL IFN-γ for 24 h. Microglial metabolism was assessed by MTT assay;NO release was measured by Reagent Kit;microglial shapes were observed through microscope. Results CB2 receptor agonist preconditioning can up-regulate the microglial CB2 receptor expression markedly;cell metabolism of AM1241+LPS/IFN-γ treatment group (92.55 ±8.37%) was obviously higher than that of LPS/IFN-γ inducement group (75.04±3.01%, P<0.05);AM1241+LPS/IFN-γ treatment group (43.44±5.52 μmol/L) released significantly less NO than LPS/IFN-γ inducement group (90.87±4.28 (μmol/L, P<0.05). Cells of the LPS/IFN-γ inducement group were destroyed seriously with enlarged soma and thickened and shortened pseudopodium;cells of the AM1241+LPS/IFN-γ treatment group were destroyed slightly with slightly enlarged soma and thickened and shortened pseudopodium. Conclusion Preconditioning with cannabinoid CB2 receptor agonist AM1241 reduces microglial activation and injury induced by LPS plus IFN-γ.
7.Expression of a disintegrin and metalloprotease 8 during the drug -induced liver in mice

Objective To study the expressed dynamic change and roles of a disintegrin and metalloprotease 8 (ADAM8) during drug -induced liver injury induced by acetaminophen (AAP).Methods Mice were randomly divided into two groups:normal group and experimental group. The mice in experimental group were respectively drawn blood by remo -ving the eyeballs and serum was separated to detect the activity of aspar -tate aminotransferase (AST) and alanine aminotranferease (ALT) at 6, 24, 42 and 72 h after intraperitoneal injection with 550 mg? kg-1 AAP. The activity of serum AST and ALT in the mice of normal group was also detected.The expression of hepatic ADAM8 at protein and mRNA levels was detected by Western blot and RT -PCR in the mice of normal group and experimental group at different time points after AAP injection . Results Serum AST and ALT activity was first increased and then decreased after AAP injection in the mice.The expression of ADAM8 were significantly up -regulated (P <0.05) at 6 h, reached peak level at 24 h (P <0.05).Then gradually down -regulated and recovered near the normal level at 72 h after AAP injection.Conclusion ADAM8 were remarkably differently expressed during drug -induced liver injury induced by AAP, which may play an important role in promoting liver damage during drug -induced liver injury induced by AAP.
8.Effects of microplastics exposure on learning and memory in mice and its mechanism

Objective To investigate the effect of microplastic(MPs)exposure on learning and memory in mice,and its mechanism by observing the protein expression of brain-derived neurotrophic factor(BDNF)/tyrosine receptor kinase B(TrkB)/N-methyl-D-aspartate receptor subtype 2B(NR2B)signaling pathway and neurogenesis.Methods Forty male C57BL/6 mice were randomly divided into control group(Ctrl)and microplastics exposure group(MPs).Mice in MPs group were treated with 0.3 mg/(kg·d)microplastics,administered by gavage at a volume of 200 μl for 30 consecutive days.Morris water maze test was used to evaluate the spatial learning and memory ability of mice.Western blotting was used to detect the protein levels of BDNF,TrkB and NR2B in hippocampus of mice.Immunofluorescent staining was used to observe the number of doublecortin(DCX)and neuronal nuclei antigen(NeuN)positive cells in the hippocampus of mice to evaluate hippocampal neurogenesis.Results Compared with the control group,the ability of learning and memory decreased significantly in MPs group mice(P<0.01).The expression levels of BDNF,TrkB and NR2B in the hippocampus of MPs group mice were significantly lower than those of control group(P<0.05).The number of DCX and NeuN positive cells in the hippocampus of MPs group was significantly lower than that of control group(P<0.01).Conclusion MPs exposure induces learning and memory impairment which may be related to inhibiting BDNF/TrkB/NR2B signaling pathway and reducing hippocampal neurogenesis.
9.Multicenter cross-sectional study of anemia in patients with gastric and colorectal cancer before and after the operation.

OBJECTIVESTo determine the prevalence of cancer-related anemia and iron deficiency anemia (IDA) in patients with gastric and colorectal cancer in North of China.
METHODSA cross-sectional study of 262 inpatients diagnosed with gastric or colorectal cancer admitted to eight general hospitals in Beijing from August 2009 to December 2009 was performed. The blood samples were took on the day after admission and the seventh day after operation for the tests of hemoglobin, serum iron and ferritin. The morbidity of cancer-related anemia and IDA before and after the surgery was also compared respectively.
RESULTSThe preoperative morbidity of cancer-related anemia was 36.6% in 131 patients with gastric cancer, and the morbidity of IDA was 52.1%. The mean age of the anemic patients was higher than that in cases without anemia [(62 ± 11) yrs vs. (57 ± 12) yrs, P < 0.05]; the postoperative morbidity of IDA increased to 72.6% (P < 0.05). In the 131 cases with colorectal cancer, the preoperative incidence of cancer-related anemia and IDA was 37.4% and 61.2%, respectively. About 45% of the cases with anemia had a tumor in the right colon. Postoperative incidence of IDA was significantly higher than that before the surgery (76.7%, P < 0.05). Only 10.3% of the anemic patients were treated with chalybeate therapy before surgical procedures, and the proportion was 22.7% after the operation. More than 50% of anemic patient received blood transfusion.
CONCLUSIONSCancer-related anemia is a common clinical manifestation in patients with gastrointestinal cancer, and anemia occurs more frequently in elder and patients with right colon tumor. The treatment to cancer-related anemia is insufficient and a systematic therapy is needed to be established.

Objective To study the optimal waist circumference (WC) cut-off values for identifying metabolic risk factors in middle-aged and elderly subjects in Shandong Province of China.
Methods A total of 2 873 men and 5 559 women were included in this cross-sectional study. Metabolic syndrome (MetS) was diagnosed according to the definition of Chinese Diabetes Society in 2004. The relation between WC and MetS was analyzed by multivariate logistic regression analysis. The optimal WC cut-off values were identified using the area under the ROC curve and the different diagnostic criteria for central obesity were compared.
Results The WC was the risk factor for MetS independent of BMI, blood glucose, blood lipid, and blood pressure. The optimal WC cut-off value was 83.8 cm and 91.1 cm for identifying MetS in women and men, respectively. Compared with 80 cm and 85 cm for women and men, 85 cm and 90 cm had a higher Youden index for identifying all metabolic risk factors and MetS in women and men.
Conclusion The appropriate WC cut-off value is 85 cm and 90 cm for identifying central obesity and MetS in women and men in Shandong Province of China.
