4.Cures of keloids by operation and closely following X-ray therapy

Objective To observe the effect of cures of keloids by operation and closely following X-ray therapy. Methods X-ray therapy was taken in no more than 24 hours after the keloids removing operations on 63 subjects, partly irradiating incision with 100~140kv X-ray,which was continued in the next three days,once for each day,till the total amount reached DT 1600~2000cGY. Results All postoperative incision healed in one-stage,all cases were followed up for 6~12 months, the new hyperplastic scar didn' t appeare on the incision of 57 subjects, while this happened on those of the other six with some improvement after operation than before operation. Rate of healing reached 90. 5% .Conclusion Cure of keloid by operation and closely following X-ray therapy can reach low recurrence and aesthetic effection,and it is worth popularizing in the clinic.
5.Observation on cellular immune response in mice induced by co-immunization of Mtb8.4 gene vaccine and plasmid encoding human interleukin 12

Objective To observe the specific cellular immune response induced by co-immunization of DNA vaccine of Mtb8.4 and plasmid encoding human interleukin 12(hIL-12)in mice.Methods Fifteen C57BL/6N mice were divided into following groups:Mtb8.4 gene vaccine plus plasmid of hIL-12,Mtb8.4 gene vaccine,BCG,empty vector alone and PBS.Mice were immunized intramuscularly in both hind limbs three times at the intervals of three weeks or once subcutaneously with 1?106 of viable M.bovis BCG Pasteur at the time of the first DNA immunization.The level of IFN-? in supernatant of spleno-lymphocyte cultures was measured by ELISA.CTL activities of spleno-lymphocyte were detected with LDH release assay.Results The levels of IFN-? and IL-2 in supernatant of spleno-lymphocyte cultures in the group of Mtb8.4 gene vaccine plus plasmid of hIL-12 were significantly higher than that of group of Mtb8.4 alone(P
6.CORRELATION BETWEEN THE EXPRESSION OF VASCULAR ENDOTHELIAL GROWTH FACTOR AND ITS RECEPTOR AND ANGIOGENESIS IN HUMAN COLORECTAL CANCER

To investigate the correlation between the expression of vascular growth factor (VEGF) and its receptor KDR and angiogenesis in human colorectal cancer. VEGF and KDR were detected in 68 cases colorectal cancer by immunohistochemical SABC technique. Microvessel density was determined by immunostaining for factor Ⅷ related antigen. Results showed that the positive rate of VEGF was 55 9% (38/68) in 68 cases of colorectal carcer, and VEGF was mainly located at the cytoplasm or the membrane of the colorectal cancerous cell; KDR positive rate was 45 6% (31/68), and KDR was located in the vascular endothelial cell of colorectal cancer tissues and their related Peri Cancerous tissues but also in the cytoplasm or the membrane of the colorectal cancerous cell. 7.HISTOLOGICAL CHANGES IN NORMAL RAT BRAIN AFTER GAMMA IRRADIATION

To study time and dose related changes after gamma knife radiosurgery (GKR) in rat brain, right caudate nucleuses of rats were irradiated by OUR XGD gamma units. The maximum dosages of 20,50,75 and 160Gy were given using a 4 mm collimator. HE, Nissl, Luxol fast blue staining and immunohistochemistry were utilized to observe histological changes.Histological changes in the right caudate nucleuses such as microvessel injury, which appeared as telangiectasia, vasodilatation, vessel wall thickening and edema, and astrocyte changes, which appeared as hypertrophy and proliferation, were observed at various time points after GKR. These findings demonstrate time dependent and dosage dependent changes are found in normal cerebral tissue after GKR. These results provide an explanation of the process taking place during the latent period before delayed radiation necrosis develops.
8.Study on inhibiting expression of IGF1R in hepatocellular carcinoma cells by constructing shRNA eukaryotic expression vectors

Objective To construct short-hairpin RNA (shRNA) eukaryotic express vectors targeting of insulin like growth factor-1 receptor (IGF1R) gene, and to explore the changes of adhesion, invasion and FAK protein expression of MHCC-97 H hepatocellular carcinoma cells with RNA interference. Methods The shRNA oligonucleotide fragments were designed and synthesized based on the sequence of IGF1R mRNA. Double strands were formed after annealing and inserted into pGCsi-U6-Neo-GFP vector. The recombinant was transformed into Stable 3, then plasmids were extracted and identified by restriction enzyme and sequencing analysis. The most effective vectors were selected by RT-PCR and Western blotting after transfecting 293T cells. The best one was used to transform MHCC-97H cells which were selected with G418 to obtain positive colons. The changes of adhesion, invasion and FAK protein expression in MHCC-97H cells were studied. Results The restriction enzyme analysis and sequencing analysis demonstrated that shRNA had been inserted into vectors, and their sequences were the same as the design. It was indicated by RT-PCR and Western blotting that the silencing efficacy of IGF1R was 88%. The ability of adhesion and invasion significantly decreased after IGF1R silencing in MHCC-97H cells, and so was the expression of FAK protein. Conclusion IGF1R pGCsi-U6-Neo-GFP shRNA can significantly decrease the abilities of adhesion and invasion in MHCC-97H cells, and inhibit the expression of FAK protein.
9.Expression difference of cerebral cortex and liver glucocorticoid receptors at protein level following severe closed traumatic brain injury in awake mice

Objective To study the relationship of expression of central cortex glucocorticoid receptor (GR) at protein level with GR expression in the liver at protein level and with changes of serum cortisol and adrenocorticotropic hormone (ACTH) following severe closed traumatic brain injury (TBI) in mice. Methods Severe TBI was established in awake mice by using a BIM-Ⅲ biomechanical machine. At 0.5, 2, 8, 24, 48 and 72 hours after TBI, the total cytosolic GR in the cortex and liver were detected with Western blotting. Levels of serum ACTH and cortisol were measured by ELISA technique and radio-immunological assay (RIA) respectively. Results The expression of GR both in the cortex and liver were obviously down-regulated at protein levels at 2-72 hours after TBI and increased slowly eight hours after injury. The GR in the liver showed no recovery at 72 hours after injury and that in the cortex was decreased continually at 24 hours after injury. Serum ACTH and cortisol levels were increased markedly compared with control group, when there were two different peaks in the observation curve.Conclusion There is glucocorticoid resistance both in the central and peripheral tissues after severe closed TBI in the awake mice, which changes in a time-dependent manner.
10.Diagnosis and treatment of prostate sarcoma (report of 4 cases)

Objective To study the management of prostate sarcoma. Methods Four cases of prostatic sarcoma were presented with review of the literature. Results Of the 4 cases,two were spindle cell sarcoma,one leiomyosarcoma and the other rhabdomyosarcoma.Combined modality therapies were used,including surgical,radiation,chemotherapy,Chinese herb medicine and immunotherapy.One case has been out of follow up and one has survived 22 months without tumor.The other two patients died within 7 and 19 months after operation. Conclusions Prostatic sarcoma is an aggressive malignancy with a bad prognosis.Individualized treatment should be formulated.
