1. Analysis of HBV precore 1896 site mutation and its HBV genotype and other laboratory features

Objective: To analyze the relationship between hepatitis B virus (HBV) gene mutation at 1896 in precore region with genotype and replication of HBV and the liver function of patients. Methods: HBV precore 1896 site mutation,the genotype of HBV and serum content of HBV DNA were determined by PCR in 60 patients positive of HBV DNA. Chemiluminescence miacropaticle immunoassay (CMIA) was used for detection of serum HBeAg and HBeAb. Liver function parameters were obtained by routine biochemistry method. Results: The alanine aminotransferase (ALT) level in HBV with 1896 site mutation was significantly higher than that in the wildtype virus. Site mutation at 1896 had no correlation with HBeAg, HBV genotype and HBV DNA content. HBV DNA content in patient with genotype C was significantly higher than that with genotype B(P<0.05). Conclusion: Mutation at 1896 in precore region of HBV gene is related to aggravation of hepatocyte injury. The serum content of HBV DNA is higher in genotype C than that in genotype B, indicating genotype C HBV has more active replication activity than genotype B.

3.Matrix metalloproteinase-9 and ischemic stroke

The pathophysiological mechanism of ischemic stroke is very complex.Many intracellular and extracellular factors play the important roles in it.Studies have shown that the expression of matrix metalloproteinase-9 (MMP-9) has the changes of quantity,time and space after stroke.These changes are closely associated with the occurrence and development of ischemic stroke.The experimental studies focusing on MMP-9 also have made corresponding effects.The in-depth study of the mechanism of action of MMP-9 is expected to provide a new approach for the diagnosis and treatment of ischemic stroke.
4.Effect of the PGE_2 Combined with TNF-? to the IP3-Ca~(2+) Pathway Mediated by Membrane TNF-?R of Fibroblasts

Objective To study the effect of different prostaglandin E2 (PGE2) combined with tumor necrosis factor-?(TNF-?) to the IP3-Ca2+ pathway mediated by membrane TNF-?R of fibroblasts. Methods The lung fibroblasts of breed mouse were primarily cultured. 10 ?g/L TNF-? and 10 ?g/L TNF-? combined with PGE2 of different doses were added to culture medium of fibroblasts. At some observation time,the expression of TNF-?R on the cellular membrane of fibroblasts was detected by the method of immunohistochemistry,the level of IP3 in cells was detected by the method of radioimmunoassay and the contents of Ca2+ in fibroblasts by the method of flow cytometry. Results With doses of PGE2 increasing at 30 s time point and 60 s time point,the cpm value of IP3 of fibroblast decreased gradually. The cpm value of IP3 significantly increased when the dose of PGE2 was 1.0 ?g/L,but obviously lower when the dose of PGE2 was 2.0 ?g/L at 120 s time point. The expression of TNF-?R and the contents of Ca2+ decreased in a dose-dependent way with the dose of PGE2 increasing. Conclusion Certain dose of PGE2 could suppress the effect that TNF-? enhance the cell proliferation of fibroblast by the IP3-Ca2+ pathway mediated by membrane TNF-?R of fibroblasts.
5.Analysis of curative effects of Pudilan Xiaoyan Koufuye on patients with eOLP and ROU

Objective To investigate the clinical effect of the treatment of erosive oral lichen planus and recurrent aphthous ulcer by using Pudilan Xiaoyan Koufuye.Methods The clinical data of 138 cases of oral mucosal diseases were retrospectively analyzed from September 2015 to June 2016 in our hospital, eOLP patients 50 cases, 11 cases in the control group one and the control group two, 28 cases in the observation group; ROU patients 88 cases, 21 cases in the control group one and the control group two, 46 cases in the observation group.The control group one was treated with topical western medicine, while the control group two was treated with Pudilan Xiaoyan Koufuye,and the observation group was treated with local topical spray and Pudilan Xiaoyan Koufuye, compared the total effective rate, VAS score and mucosal congestion in different groups of patients with oral diseases, and to observe the adverse reactions of the patients during the treatment.Results After one week of treatment, the total effective rate of treatment was significantly different between eOLP patients and ROU patients (P<0.05),the total effective rate of the observation group was higher than that of the control group one and the control group two ( P <0.05 ) .After two weeks of treatment, the total effective rate of eOLP patients was significantly different, and the total effective rate of observation group was higher than that of control group one (P<0.05),there was no significant difference among the ROU patients.The degree of mucosal hyperemia and the VAS score in the observation group were significantly different from those in the control group one and control group two in the patients with eOLP and ROU ( P <0.05 ) .No significant adverse mucosal staining occurred in all groups of patients, and there was no discontinuation due to obvious adverse reactions.Conclusion Pudilan Xiaoyan Koufuye adjuvant treatment of eOLP and ROU patients with short-term effect is significant, can improve the total effective rate of treatment, and no serious adverse reactions, high safety.
6.Effect of hyperthermia-induced MMP-9 on expression of claudin-1 in in vitro blood brain barrier model

Objective To study the effect of hyperthermia-induced MMP-9 on expression of claudin-1 in in vitro BBB model.Methods BMEC and astrocytes were cultured.An in vitro BBB model was established with its permeability assayed by LY test.The BBB model was randomly divided into 37℃ group,39℃ group,37℃+MMP-9 group and 39℃+MMP-9 group.The permeability of BBB model was recorded at 30,60 and 90 min respectively.Expression of claudin-1 in BMEC was detected by Western blot with immunocytochemical staining.Results The permeability of BBB model was higher in 39℃ group than in 37℃ group (P<0.05) and was significantly higher in 39℃ +MMP-9 group than in 37℃ group (P<0.05).The number of claudin-1 positive BMEC was smaller in 39℃ +MMP-9 group than in 37℃ group at the same time points (P<0.05) and in 39℃ +MMP-9 group than in 39℃ group at 90 min (75.0±12.8/HP vs 97.0±6.1/HP,P<0.05).The expression level of claudin-1 was lower in 39℃ group than in 37℃ group (P<0.05) and was significantly higher in 39℃ +MMP-9 group than in 37℃ group (P<0.05),which further increased with the increasing time.Conclusion Hyperthermia can downregulate the expression of claudin-1 in in vitro BBB model and upregulate the permeability of in vitro BBB model.Adding MMP-9 can further aggrevate the damage of in vitro BBB model,indicating that hyperthermia can aggrevate the damage of in vitro BBB model through MMP-9.
7.Comparison of effect of standard Gamma nail and proximal femoral nail anti-rotation for geriatric intertrochanteric fractures

[Objective]To investigate and compare the effects of standard gamma nail (SGN) and proximal femoral nail anti-rotation (PFNA) in the treatment of osteoporotic intertrochanteric fractures in the elderly.[Method]From May 2002 to September 2005,55 patients with intertrochanteric fractures were treated with SGN.Sixty-eight patients were treated with PFNA from November 2005 to June 2008.The effects of the two implants about perioperative complications,bone healing and the function of the operated hip joint were compared.[Result]No statistically significant difference was found between the two groups in blood loss,blood transfusion,infection,deep venous thrombosis and hospital stay.One patient developed femoral head avascular necrosis postoperatively in Gamma nail group.The incidence of postoperative hip pain in Gamma nail group was higher than that in the PFNA group,but there was no significant difference in the Harris score.[Conclusion]Gamma nail and PFNA nail are reliable methods to treat geriatric intertrochanteric fractures,with advantages of minimal iatrogenic injury,stable fixation and few complications.
8.The influence of peritoneal dialysate on peritoneal macrophage releasing TNF-?

To investigate the damage on macrophage of the commercial peritoneal dialysis solution(CDS). Methods Macrophages were seperated from peritoneal fluid remained overnight of seven CAPD patients and TNF-a level of supernatant was determined and compared with those macrophages from uremic patients not yet recieving peritoneal dialysis. Results TNF-a levels of different glucose concentration decreased obviously in experimental group compared with control group, especially lower in 2.5% and 4.25% group. Conclusion In vivo experiment confirms that CDS possesses a long time inhibition on macrophage and this inhibition varies with different glucose concentrations.
9.Longitudinal stress and strain of normal human vascular system of the extremities, and the blood vessels following venous repair

Objective To investigate the longitudinal residue strain and stress-strain relation of normal human vascular system of the extremities and the relation between the different defect length and the option of repair, as well as a comparison of the different selection of the methods of repair judged from the longitudinal biomechanical effect. Methods A mechanical analysis including stretch experiment and longitudinal stretch ratio measurement were undertaken to evaluated the longitudinal residue strain and stress-strain relation of arteries and veins of the extremities in human. The results of different methods for repair of the defect of the same kind of vessels were compared with Student-t test. The length precinct to repair arterial injuries between end-to-end anastomoses and vein graft through 95% distribution region was evaluated. Results The longitudinal stretch ratio of arterial system of the extremities decreased from the proximal to the distal ends of the vascular branches and the stress-strain relation curve of artery turned gradually to the right side, while that of veinous system exhibited a contrary picture. 10.Effects of norepinephrine on the activity of NF-?B in cardiomyocytes

AIM: To study the effects and mechanisms of norepinephrine (NE) on the activity of nuclear factor-kappa B (NF-?B) in cardiomyocytes. METHODS: Using the cultured neonatal rat cardiac myocytes, the levels of reactive oxygen species and the nuclear DNA binding activity of NF-?B were measured in cardiomyocytes before and after stimulated with NE alone, NE+prazosin+propranolol, and NE+vitamin E, respectively. RESULTS: NE increased significantly the levels of reactive oxygen species and NF-?B activity in cardiac myocytes. These effects of NE were attenuated by vitamin E pretreatment, as well as prazosin and propranoloe. CONCLUSION: The effects of NE on cardiomyocytes might be exerted partly through NF-?B activation, and associated with NE-induced overproducion of reactive oxygen species via the adrenergic receptor pathway.
