2.Effects of GATA6 silencing mediated by lentivirus on apoptosis of human liver cancer Huh-7 cells
Chinese Journal of Pathophysiology 2015;(5):938-942
AIM:To explore the effect of GATA6 gene silencing on apoptosis of hepatocellular carcinoma Huh-7 cells.METHODS:RNA interference vectors of the target gene GATA6 mediated by lentivirus were constructed in vitro to transfect the hepatocellular carcinoma cell line Huh-7.The apoptotic rate of transfected cells was measured by flow cytome-try.The protein expression of GATA6, NF-κB and Bcl-2 in transfected cells was determined by Western blotting.RE-SULTS:The transfection efficiency was 57.4%.The mRNA and protein expression of GATA6 reduced significantly after the carcinoma cell line Huh-7 being transfected by RNA interference vectors mediated by lentivirus. The apoptotic rate of the carcinoma cells with silent GATA6 gene was significantly increased (P<0.05).The protein expression levels of NF-κB and Bcl-2 were also significantly decreased.CONCLUSION: Lentiviral vector-mediated RNA interference of GATA6 has an inhibitory effect on the expression of the gene itself, and promotes the apoptosis of hepatocellular carcinoma cells.Regu-lation of the apoptosis-related protein expression by the NF-κB signaling to influence the proliferation and apoptosis of hepa-tocellular carcinoma cells might be one of the possible mechanisms.

CONCLUSION:The technology is reasonable in formulation and satisfactory in molding, and it can serve as theoretical basis for the production of Angelica tablets.
5.Portal azygous devascularization for the treatment of patients with portal hypertension
Chinese Journal of General Surgery 1997;0(04):-
Objective To evaluate therapeutic effects of portal azygous devascularization on patients with portal hypertension. Methods In this study, data of 212 patients with portal hypertension who underwent devascularization were analyzed retrospectively. Among them 52% patients had a history of variceal bleeding. Preoperative Child A accounted for 15% patients, B for 57% and C for 27%. Emergent operation and selective operation were performed in 11% and 89% cases, respectively. Hassab procedure was performed in 176 cases (83%), whereas modified Sugiura procedure in 36 cases (17%). Results Portal vein pressure decreased from (40?6) cm?H 2O to (36?7) cm?H 2O. The re-bleeding rate of 3, 5,10 years were 2%, 6%, 11%, and the survival rates of 3, 5, 10 years were 97%, 92% and 89%, respectively. Conclusions Devascularization is an effective procedure in controlling bleeding with low incidence of liver failure, operation mortality and encephalopathy. Emergent operation should be avoided as it causes high morbidity and mortality. The decreased portal pressure following devascularization may result in a reduced blood supply to the liver. Persistent exasperation of the liver function caused by chronic hepatitis and the development of hepatocellular carcinoma are the two major factors endangering long-term survival.

To investigate the prevalence and the antibiotic resistance of bacteria isolated from 25 miniature pigs. 45 bacterial strains were isolated, which were identified by biochemical assays, amplification of 16S rRNA genes by PCR and sequence analysis, and were evaluated for resistance to 30 antibiotics. The identification results showed that these bacteria belonged to Campylobacter (Campylobacter jejuni), Helicobacterium (Helicobacter pylori), Klebsiella (Klebsiella pneumoniae), Escherichia (Escherichia coli, Escherichia fergusonii), Pseudomonas (Pseudomonas aeruginosa), Stenotrophomonas (Stenotrophomonas maltophilia), Staphylococcus (Staphylococcus aureus, Staphylococcus haemolyticus, Staphylococcus simulans), Streptococcus (Streptococcus pneumoniae, Streptococcus suis, Streptococcus vestibularis, Streptococcus mitis, Gemella measles, Aerococcus viridans) and Bacillus (Bacillus subtilis, Bacillus licheniformis, Bacillus alvei, Bacterium megaterium). These bacteria were all susceptible to aztreonam and cephalothin. However, the resistence to furazolidone was found. Microbial population carried by miniature pigs in China had characters of diversity. Results of this study provided scientifical accordance for the microorganism monitoring of miniature pigs in China.

Chinese Journal of General Surgery 2009;24(7):536-538
Objective To evaluate the treatment and prevention measure for intrahepatie diffuse biliary stricture after orthotopic liver transplantation. Methods The clinical data of 21 patients with intrahepatic diffuse biliary stricture after orthotopic liver transplantation from January 2002 to December 2007 in Beijing Center for Organ Transplantation were retrospectively analyzed. A prospective clinic study was launched in order to prevent microthrombosis in the microcireulation of bile duct during warm and cold iscbemia in non-heart-beating donor since January 2006. Urokinase perfusion of the artery system was conducted during graft harvesting and reperfusion at the end of graft trimming to reduce the incidence of intrahepatic diffuse biliary stricture. Results Among 21 patients with intrahepatie diffuse and mixed type biliary stricture, 16 patients underwent liver retransplantation, and the other 5 patients died of primary graft failure while waiting for retransplantation. The incidence of intrahepatic diffuse biiiary stricture was 5.9% in non-urokinase perfusion group. On the contrary, the incidence rate of intrahepatie diffuse biliary stricture was 1.4% in urokinase perfusian group (x2 = 5.98, P < 0. 05). Conclusions Liver retransplantation is effective for refractory biliary stricture in liver transplant recipients. The incidence of intrahepatic diffuse biliary stricture is reduced in non-heart-beating donor by using urokinase perfusian.

Objective To explore the related factors of school refusal behavior in child and adolescent. Methods Investigated 141 students who had school refusal behavior in the department of psychiatry as the case group. 109 students were chosen randomly from 3 different grades in different schools as the control group. Two experienced psychiatrists made diagnosis according to DSM-Ⅳ criteria, the case group completed the direct factor investigation of School Refusal and the general condition questionnaires. The control group completed the general condition questionnaire only. Results 61 (43.7%) of the case group were diagnosed mental disorder according to DSM-Ⅳ ,17(28.91% ) of them were diagnosed mood disorder, which was the most in all kinds of mental disorder in case group. The results showed that age (P = 0.0003 ), location (P = 0. 035 ) and family income (P < 0. 01 )were the most important factors for school refusal behavior in child and adolescent. The opinions of child and parent on the school refusal factors were quite different (P < 0. 05 ). The former three direct reasons of school refusal were study pressure (6.47%) ,much more examinations and homework( 6.03% ) ,and willing to stay at home only (5.63%). Conclusion Mental disorder is the main causation of school refusal behavior. Age, location and family income are related closely to school refusal behavior. Study pressure, peer difficulty and poor parent-child relationship are direct factors of school refusal behavior.

Chinese Journal of Trauma 2011;27(4):370-374
Objective To study the mRNA and protein expressions of the neurofilament in the region of acute spinal cord injury (SCI) of rats after olfactory ensheating cells (OECs) transplantation combined with methyprednisolone administration and investigate the molecular mechanisms of OECs transplantation combined with methyprednisolone administration in promoting the recovery of the spinal cord. Methods Acute spinal cord injury was established in SD rats ( T10 ) by using NYU instrument. The rats were randomly divided into control group, SCI group, DF12 group, OECs transplantation group, methyprednisolone administration group and OECs + methyprcdnisolone group. The mRNA and protein expressions of the neurofilament in the SCI regions of rats in different groups at different time points were detected by using immunohistochemistry, RT-PCR and Western blotting. Results A significant increase of mRNA and protein expressions of the neurofilament could be found in the other five groups compared with the control group at days 7, 14 and 28 after SCI. The mRNA and protein expressions of the neurofilament in the injury region of the OECs group, the methyprednisolone group and the OECs + MP group were more significantly increased than that of the SCI group and the DF12 group. The expression of the neurofilament in the injury region of the OECs + MP group was more significantly increased than that of the OECs group and the MP group ( P < 0. 05). Conclusions OECs transplantation or methyprednisolone administration can induce the mRNA and protein expressions of the neurofilament. Meanwhile, OECs transplantation combined with methyprednisolone administration can significantly increase the mRNA and protein expressions of the neurofilament, as may be one of mechanisms promoting spinal cord repair.
