1.Surgical skills of total orthotopic liver transplantation in rats

Objective:To explore the surgical skills of establishing stable total orthotopic liver transplantation models in the rats by two cuff technique.Method:Fifty orthotopic rat liver transplantations were performed by two cuff technique.Results:The median anhepatic phase of recipients was 17 minutes. The postoperative survival rate after 24 hours was 90%, after one week 50%.Conclusion:Self-made blood vessels cuff and amendment in donor-taking and donor-tailoring skills may attribute to establishment of stable rats total orthotopic liver transplantation models.

3.Study on the Direct Powder Compression Technique of Angelica Micropowder

OBJECTIVE:To study the direct powder compression technology of Angelica micropowder. METHODS: The formula of Angelica micropowder was optimized by mixture uniform design with tablet weight variation and friability as restrictive conditions, and a regression equation was established based on the comprehensive grading on the hardness and disintegration to figure out the optimal solution. DPS software was applied in the calculation. RESULTS: Prepared by the direct powder compression technology, the optimal formula of Angelica micropowder was obtained as follows: the hardness of the optimized tablet could reach as high as 75 N and the disintegrating time was about 10.24 min, with smooth and intact surface. CONCLUSION:The technology is reasonable in formulation and satisfactory in molding, and it can serve as theoretical basis for the production of Angelica tablets.
4.Effects of GATA6 silencing mediated by lentivirus on apoptosis of human liver cancer Huh-7 cells

AIM:To explore the effect of GATA6 gene silencing on apoptosis of hepatocellular carcinoma Huh-7 cells.METHODS:RNA interference vectors of the target gene GATA6 mediated by lentivirus were constructed in vitro to transfect the hepatocellular carcinoma cell line Huh-7.The apoptotic rate of transfected cells was measured by flow cytome-try.The protein expression of GATA6, NF-κB and Bcl-2 in transfected cells was determined by Western blotting.RE-SULTS:The transfection efficiency was 57.4%.The mRNA and protein expression of GATA6 reduced significantly after the carcinoma cell line Huh-7 being transfected by RNA interference vectors mediated by lentivirus.The apoptotic rate of the carcinoma cells with silent GATA6 gene was significantly increased (P<0.05).The protein expression levels of NF-κB and Bcl-2 were also significantly decreased.CONCLUSION: Lentiviral vector-mediated RNA interference of GATA6 has an inhibitory effect on the expression of the gene itself, and promotes the apoptosis of hepatocellular carcinoma cells.Regu-lation of the apoptosis-related protein expression by the NF-κB signaling to influence the proliferation and apoptosis of hepa-tocellular carcinoma cells might be one of the possible mechanisms.
5.Portal azygous devascularization for the treatment of patients with portal hypertension

Objective To evaluate therapeutic effects of portal azygous devascularization on patients with portal hypertension. Methods In this study, data of 212 patients with portal hypertension who underwent devascularization were analyzed retrospectively. Among them 52% patients had a history of variceal bleeding. Preoperative Child A accounted for 15% patients, B for 57% and C for 27%. Emergent operation and selective operation were performed in 11% and 89% cases, respectively. Hassab procedure was performed in 176 cases (83%), whereas modified Sugiura procedure in 36 cases (17%). Results Portal vein pressure decreased from (40?6) cm?H 2O to (36?7) cm?H 2O. The re-bleeding rate of 3, 5,10 years were 2%, 6%, 11%, and the survival rates of 3, 5, 10 years were 97%, 92% and 89%, respectively. Conclusions Devascularization is an effective procedure in controlling bleeding with low incidence of liver failure, operation mortality and encephalopathy. Emergent operation should be avoided as it causes high morbidity and mortality. The decreased portal pressure following devascularization may result in a reduced blood supply to the liver. Persistent exasperation of the liver function caused by chronic hepatitis and the development of hepatocellular carcinoma are the two major factors endangering long-term survival.
6.Clinical efficacy of sentinel lymph node biopsy for oral cancer: A systematic review

Objective:To evaluate the clinical sensitivity and specificity of sentinel lymph node biopsy (SLNB) for oral cancer.Methods:By searching Medline,CENTRAL,EMBSE,CBM,CNKI,WANFANG,etc,an studies about SLNB for the patients with oral cancer array for lymph node metastasis were retrieved.The quality of included studies was evaluated by QUADAS items.The data were analyzed by statistic software Meta-DiSc 1.4.Results:25 studies including 934 subjects were analyzed in the current systematic review.The pooled values of sensitivity,specificity were 91% (95% CI 88%-94%) and 100% (95 % CI 99%-100%) respectively.The area under summary receiver operating characteristic (SROC) curve (AUC) of SLNB for cervical lymph node metastasis was 0.99.Conclusion:SLNB procedure has high sensitivity and specificity in the diagnosis of cervical lymph node metastasis,it can be used as an useful method for cervical lymph node metastasis screening in patients with oral cancer.
7.Identification and susceptibility test of bacteria isolated from miniature pigs in China

To investigate the prevalence and the antibiotic resistance of bacteria isolated from 25 miniature pigs. 45 bacterial strains were isolated, which were identified by biochemical assays, amplification of 16S rRNA genes by PCR and sequence analysis, and were evaluated for resistance to 30 antibiotics. The identification results showed that these bacteria belonged to Campylobacter (Campylobacter jejuni), Helicobacterium (Helicobacter pylori), Klebsiella (Klebsiella pneumoniae), Escherichia (Escherichia coli, Escherichia fergusonii), Pseudomonas (Pseudomonas aeruginosa), Stenotrophomonas (Stenotrophomonas maltophilia), Staphylococcus (Staphylococcus aureus, Staphylococcus haemolyticus, Staphylococcus simulans), Streptococcus (Streptococcus pneumoniae, Streptococcus suis, Streptococcus vestibularis, Streptococcus mitis, Gemella measles, Aerococcus viridans) and Bacillus (Bacillus subtilis, Bacillus licheniformis, Bacillus alvei, Bacterium megaterium). These bacteria were all susceptible to aztreonam and cephalothin. However, the resistence to furazolidone was found. Microbial population carried by miniature pigs in China had characters of diversity. Results of this study provided scientifical accordance for the microorganism monitoring of miniature pigs in China.
8.Modulatory effects of prostaglandin E1 on expression of NF-?B in acute lung injury

Objective To investigate modulatory effects of prostaglancdin E1 on the expressions of NF-?B and cytokines in serum, and to explore the protection mechanisms of PGE1 in ALI.Methods Forty-five male SD rats were randomly divided into three groups: group A(normal group,n=15), group B(LPS group,n=15) and group C(LPS +lipo-PGE1 group,n=15). The pathological changes and activity of NF-?B in the lung tissue, TNF-?, IL-10 and IL-12 concentration in serum at 1 hours,2 hours and 4 hours after intraperitoneal LPS were observed.Results Hyperemia and edema of lung tissue in the LPS +lipo-PGE1 group was obviously less than that in the LPS group. Activity of NF-?B and serum concentrations of TNF-? and IL-12 in the LPS +lipo-PGE1 treated group were more decreased than that in the LPS group, and serum IL-10 concentration was obviously increased in the LPS +lipo-PGE1 group. Conclusion PGE1 reduced LPS-induced ALI through doun-regulating NF-?B activity and inhibiting expression of harmful inflammatory cytokines.
9.Elimination Effect of Radix Pulsatillae Liquid Extracts on R Plasmid in Pseudomonas Aeruginosa

Objective To explore the elimination effect of radix Pulsatillae extracts on R plasmid of pseudomonas aeruginosa.Methods R plasmids of pseudomonas aeruginosa were isolated from the sputum of elderly patients suffering from respiratory infection,and the elimination test of radix Pulsatillae extracts on these plasmids was performed in vitro and in vivo.Results Being treated with the extracts for 24,48 and 72 h in vitro,the elimination rate of R plasmids was 0,1.8 %and 3.4 %respectively,while in SDS(sodium dode canesulphonate)control group the elimination rate was 0,0.8 %and 1.6 %respectively.Being treated with the extracts for 24,48 and 72 h in vivo,the elimination rate was 0,2 %and 12.8 %respectively,and 0 in the control group.Conclusions Both in vitro and in vivo,radix Pulsatillae extracts have an elimination effect on R plasmid of pseudomonas aeruginosa,and the effect in vivo is stronger than that in vitro.
10.Hotspot subjects in domestic studies on information behaviors

Objective To reveal the subjects and hotspots in domestic studies on information behaviors by analyzing the domestic papers on information behaviors. Methods The key words in domestic papers on information behaviors were analyzed by factor analysis and co-word clustering analysis with the structure of subjects in domestic studies on information behaviors elaborated. Results The domestic studies on information behaviors were focused on the fol-lowing 12 aspects, such as information literacy, network information behaviors, information behavior model, infor-mation dissemination, and information service in libraries. Conclusion The hotspots in domestic studies on informa-tion behaviors are characterized by the various objects, scattered subjects, and balanced degrees.
