2.Analysis of postoperative complications after transrectal ultrasound-guided 24-core transperineal prostate biopsy

Objective To analyze the complication rates in 210 patients undergoing transrectal ultrasound-guided 24-core transperineal prostate biopsy.Methods Clinical data of 210 patients who underwent transrectal ultrasound-guided 24-core transperineal prostate biopsy from 2010 to 2012 were collected.A retrospective analysis of complications after prostate biopsy was conducted.Results The main postoperative complications were hematuria,bloody stool,dysuria,urinary retention,fever (>38℃),urinary infection,vasovagal syncope,and their complication rates were 43.3 % (91/210),10.9% (23/210),23.3% (49/210),25.2% (53/210),14.8% (31/210),21.9% (46/210),0.48% (1/210) respectively.In addition,5 patients (2.5%) were hospitalized due to biopsy related complications.Conclusions Transrectal ultrasound guided 24-core transperineal prostate biopsy is an relatively safe and effective puncture method with common minor complications and less severe complications.Adequate attentions should be paid to its complications.
3.Effect of eleutheroside B/E on proliferation and expression of PPARγand TGF-β1 in HBZY-1 cells cultured with high glucose

[ ABSTRACT] AIM:To explore the effects and mechanism of eleutheroside ( ETS) B or E on the proliferation of HBZY-1 cells treated with high glucose.METHODS:The HBZY-1 cells were cultured under high glucose condition.The 4th generation of HBZY-1 cells was used for determining the optimal cell density, which was consistent with the growth reg-ulation curve of the cells.The cells were divided into 6 groups:low glucose ( LG) group, high glucose ( HG) group, high glucose plus ETS-B/E ( low dose, medium dose and high dose) groups, and high glucose plus losartan ( LTG) group.Af-ter all cells were treated with the corresponding drugs at 24 h, 48 h and 72 h, the inhibitory rate of the proliferation was measured, and the expression of TGF-β1 and PPARγwas detected by immunocytochemistry and Western blotting.RE-SULTS:The best cell density was 2 000 cells/well, which was complied with the basic rules of the cell growth, and high glucose significantly promoted the HBZY-1 cell proliferation.At each time point, the inhibitory effects of ETS-B/E were significantly different between HG group and LTG group on the proliferation of the HBZY-1 cells ( P<0.05) .The expres-sion of TGF-β1 was significantly inhibited, and the expression of PPARγwas significantly promoted by ETS-B/E ( P<0.05).ETS-E showed stronger effect than ETS-B (P<0.05) in a concentration-and time-dependent manner.CONCLU-SION:ETS-B/E significantly inhibits the proliferation of HBZY-1 cells under high glucose condition by decreasing TGF-β1 expression and promoting PPARγexpression.
4.Investigation on reference range of anti-mutated citrullinated vimentin antibody among healthy individuals in Northeast area of Sichuan

Objective To establish the reference range of anti-mutated citrullinated vimentin antibody (anti-MCV ) among healthy individuals in Northeast area of Sichuan .Methods The Alegria enzyme-linked immunosorbent assay system and the matched anti-MCH reagent test kit were adopted to detect the serum anti-MCV concentration in 141 serum samples collected from healthy individuals .Results The intra-batch and inter-batch precisions and the functional sensitivity of the Alegria enzyme-linked immunosorbent assay system all met the laboratory requirements .The concentration of serum anti-MCV was 12 .10(6 .70-41 .30) U/mL in the healthy female group and 11 .10(6 .00-18 .30)U/mL in the healthy male group ,there was no statistical difference be-tween these two groups(Z= -0 .991 ,P=0 .322);the detection results of serum anti-MCV in the age groups of <50 years old and ≥50 years old had no statistical differences (Z= -0 .588 ,P= 0 .558) .The reference range of anti-MCV in the investigated healthy individuals was 0 .00-18 .14 U/mL .Conclusion The reference range of anti-MCV has no correlation with gender and age . Establishing the reference range of anti-MCV suitable for the healthy population in the local area has very important clinical applica-tion value .
5.Application of multi-tumor marker protein chip for tumor screening

Objective To study the application of multi‐tumor marker protein chip for early tumor screening and diagnosis . Methods From Nov .2011 to Dec .2015 ,10 736 samples ,including people receiving physical examination and with high risk of canc‐er in Fengcheng Hospital were collected .Twelve tumor markers in serum(AFP ,CEA ,NSE ,CA125 ,CA153 ,CA242 ,CA199 ,PSA ,f‐PSA ,FER ,β‐HCG and HGH)were measured by multi‐tumor markers protein chip detective system ,and the results were analyzed . Results We found out 967 samples with positive markers in the 10 736 samples .Of which ,496 were male and 471 were female ,the positive rate were 4 .62% and 4 .39% respectively .Totally 473 were diagnosed with tumor confirming by clinical pathology ,postive diagnosis rate was 48 .91% .Conclusion The multi‐tumor marker protein chip (C12 system) can detect multiple tumor markers simultaneously to improve screening process and achieve rapid detection ,w hich has higher positive detectiong rate and clinical value on diagnosing malignant tumor in early stage .
6.Screening on the tumor related markers in the serum of the renal cell carcinoma patients by the SELDI-TOF-MS technique

Background and purpose:Surface enhanced laser desorption ionization time of flight mass spectrometry (SELDI-TOF-MS) is useful in helping to identify the molecular changes closely related to renal cell carcinoma. We explored the different expression of sera protein between human renal cell carcinoma patients and normal to screen renal cancer-specific biomarkers. Methods:The protein mass spectrometry of 28 cases with renal cell carcinoma and 28 normal persons were detected by WCX2 protein chip combining with SELDI-TOF-MS technique for screening the different proteins. Serum samples from 28 patients with clear renal cell carcinoma and 28 normal persons were used to detect biomarkers for clear renal cell carcinoma by SELDI-TOF-MS technique with WCX2 Proteinchip. Results:170 effective protein wave crests between 1.5?103-30?103(1.5-30 kD) were detected. Seven proteins were specifically detected in sera of patients with clear renal cell carcinoma, but not in normal donor. The proteins with MW 4.098,5.917,6.643 ?103 were down-regulated ,and four proteins with MW 5.572,6.344,6.529,8.518 ?103 were up-regulated. Conclusion:Detection of specific protein in human renal cell carcinoma sera is significant both for determination of clinical specific biomarkers and study of cancer development mechanism.
7.Transrectal 12-core biopsy of the prostate guided by transrectal ultrasonography

Objective To evaluate the clinical value of transrectal ultrasound guided systematic 12-sample needle biopsy in patients with elevated serum prostate-specific antigen and/or abnormal digital rectal examination findings. Methods The data of 220 patients who underwent 12-sample transrectal ultrasound guided needle biopsy (comprising conventional 6 biopsies and 3 biopsies in each lateral peripheral zone) of the prostate were retrospectively analyzed. Results Of the 220 patients,73 (33.2%) had prostate cancers on biopsy;the clinical stages were as follows:4 cases of stage T 1,21 of T 2,15 of T 3 and 33 of T 4.If the conventional 6-core biopsy alone was performed, the detection rate would be 31.4%,4 cases (3 of stage T 1 and 1 of stage T 2,all with tumor volume less than 0.5 ml) would be missed. The improvement by 12-core biopsy was marked in patients with stage T 1-T 2 prostate disease (16%,4/25).No serious complications were observed in patients with transrectal ultrasound guided systematic 12-core biopsy of the prostate. Conclusions The 12-sample procedure can improve the cancer detection rate of the early stage prostate cancer and tumor with volume less than 0.5 ml.Therefore,more attention should be paid to the lateral peripheral zone biopsy.
8.Positive impact of letting patients pick their doctors on medical quality

Firstly the paper expounds the fact that letting patients pick their doctors goes with the tide of social development: ①the connotation of medical quality determines the necessity of patients involvement; ②evaluation of medical quality has extended from self appraisal to social assessment; and ③the mechanism of letting patients pick their doctors conforms to the medical quality requirements of the times. Then the paper discusses the positive impact of letting patients pick their doctors on medical quality, including the choice of doctors by outpatients and the impossibility of doing so in medico technical and emergency departments or by inpatients. The paper also gives an account of the measures taken for safeguarding medical quality when adopting the practice of letting patients pick their doctors. Lastly the paper explains the boosting role of the practice and the difference between letting patients pick their doctors and letting them name particular surgeons for their operations.
9.Progress of the anti-tumor mechanisms of Smac mimics

The tolerance of tumor cells to treatment factors is the main reason to limit the therapeutic effect.The high expression of inhibitor of apoptosis proteins (IAPs) is an important factor in the development of therapeutic resistance in tumor cells.As an effective pro-apoptotic protein,the second mitochondria-derived activator of caspase (Smac) can significantly enhance the sensitivity of tumor cells to chemoradiotherapy and promote the apoptosis of tumor cells.However,exogenous Smac protein and its active groups can not enter the tumor cells and play their functions.Therefore,the researches on Smac mimics and their functional mechanism have become the focus of attention of researchers.In this paper,the research progress of Smac mimics and their anti-tumor mechanisms were reviewed.
