1.Proteasome inhibitor improves expressions of CHIP and HSP70 in ischemia reperfusion rats

Objective To explore the influence of proteasome inhibitor, MG-132 (N-benz0y1oxycarbonyl(Z)-Leu-Leuleucina1) on the expressions of carboxyl terminus of the Hsc70-interacting protein (CHIP) and heat shock protein 70 (HSP70) in ischemia reperfusion (I/R) rats and investigate the underlying mechanism of myocardial protection by the inhibitor. Methods Totally 54 adult SD rats were randomly divided into 3 groups, sham group (n=6), I/R group (n=24) and treatment group (I/R+T, n=24). The later 2 groups were further equally divided into 3 subgroups according different time of reperfusion. Left anterior descending (LAD) coronary artery was ligated for 30 min and then released for 2 h or 24 h or 7 d in different subgroups. Five minutes before reperfusion, MG-132 at dose of 0.75 mg/kg was given intravenously in I/R+T group,but I/R group and sham group were given the same volume of normal saline. The levels of CHIP and HSP70 at mRNA and protein level were detected by real-time PCR and Western blotting respectively. The correlation between CHIP and HSP70 expression was analyzed. Objective To study the expression of long non-coding RNA (LncRNA) stomach cancer-associated transcript 16 (STCAT16) in gastric cancer tissues and its effects on the proliferation, migration and invasion of gastric cancer cells.Methods The different expression of STCAT16 in 32 cases of gastric carcinoma and corresponding adjacent tissues was detected by real-time fluorescence quantitative polymerase chain reaction (PCR).The STCAT16 overexpression plasmid and empty vector was separately transfected gastric cancer cell line AGS with low expression of STCAT16.The cell proliferation of empty vector group, non-transfection group and STCAT16 analogue transfection group was evaluated by cell counting kit-8 (CCK-8) assay at 0, 24, 48, 72 and 96 hours after transfection.The colony forming ability was tested by colony formation assay.The cell invasion ability was measured by Transwell chamber assay and migration ability was tested by scratch-wound assay.The effects of STCAT16 on tumorigenicity in vivo were verified by tumorigenicity experiments in nude mice.T-test and one-way analysis of variance were performed for data analysis.Repeated measures analysis of variance was used to compare the repeated measured data among groups.Chi square test and Fisher exact probability method were used for comparison of counting data.Results The expression of STCAT16 in gastric cancer tissues was low (0.87±0.19), while it was high in corresponding adjacent tissues (2.32±0.37), and the difference was statistically significant (t=-20.859, P<0.05).The expression of STCAT16 of STCAT16 analogue transfection group was higher than that of empty vector group (3.43±0.25 vs 1.00±0.06), and the difference was statistically significant (t=-16.795,P<0.05).Compared to empty vector group and non-transfection group, the cell proliferation decreased in STCAT16 analogue transfection group at 72 and 96 hours after transfection (1.41±0.07, 1.42±0.08, 1.03±0.09, and 1.72±0.11, 1.78±0.14, 1.24±0.08, respectively), and the differences were statistically significant (t=15.043,5.358, 12.193 and 8.109, all P<0.05).The results of colony formation assay indicated that the colony forming ability of gastric cells in STCAT16 analogue transfection group was lower than that in empty vector group (97.3±9.1 vs 185.0±20.1) and non-transfection group (97.3±9.1 vs 138.0±11.1), and the differences were statistically significant (t=11.634 and 4.417,both P<0.05).The results of Transwell assay showed that the number of AGS cells passing through the membrane of STCAT16 analogue transfection group was significantly less than those of empty vector group and non-transfection group (151.0±28.1 vs 228.0±38.2 and 151.0±28.1 vs 199.3±17.9), and the differences were statistically significant (t=4.823 and 4.747,both P<0.05).After transfection for 48 hours and 72 hours, the scratch-wound repair rate of STCAT16 analogue transfection group decreased, compared with those of empty vector group and non-transfection group ((52.67±6.11)%, (53.33±5.51)%, (42.67±4.72)%, and (90.67±2.51)%, (90.60±5.41)%, (69.67±1.52)%, respectively), and the differences were statistically significant (t=5.773, 5.955, 21.000 and 5.881, all P<0.05).The results of tumorigenicity in nude mice showed that compared with those of empty vector group, the tumor size of STCAT16 analogue transfection group was smaller at one-, two-, three-and four-week ((0.42±0.10) cm3 vs (0.16±0.05) cm3, (0.66±0.13) cm3 vs (0.34±0.05) cm3, (1.25±0.22) cm3 vs (0.54±0.13) cm3, (2.54±0.46) cm3 vs (0.78±0.41) cm3)), and the differences were statistically significant (t=3.175, 4.190, 7.996 and 9.705, all P<0.05).Conclusions STCAT16 is lowly expressed in gastric cancer tissues.The proliferation, migration, invasion ability and tumorigenicity in nude mice of gastric cancer cell is inhibited after upregulating the expression of STCAT16.
3.Study on the clinical value of combined detection of HE4,CA199 and CA125 in diagnosis of ovarian cancer and benign ovarian cysts

Objective To explore the clinical value of combined detection of human epididymis protein 4(HE4) ,carbohydrate an‐tigen 199(CA199) and carbohydrate antigen 125(CA125) in diagnosis of ovarian cancer and benign ovarian cysts .Methods The se‐rum levels of HE4 ,CA125 and CA199 were determined by using chemiluminescent immunoassay in 45 cases of patients with ovarian cancer(ovarian cancer group) ,60 cases of patients with benign ovarian cyst(benign disease group) ,and 30 cases of healthy individu‐al(control group) .Results The serum levels of HE4 ,CA199 and CA125 in the ovarian cancer group were higher than those in the ovarian cancer group and control group ,there were statistically significant differences(P<0 .05) .Compared with the control group , the serum level of CA125 in benign disease group was significantly higher(P<0 .05) ,while no statistically significant differences of serum levels of HE4 and CA199 were observed between the two groups(P>0 .05) .The sensitivity of combined detection of the three indicators increased to 82 .2% ,and the specificity remained at 90 .0% .Conclusion Combined detection of HE4 ,CA125 and CA199 might have clinical significance in early diagnosis and identification of ovarian cancer .
4.Distribution and Drug Resistance of Pathogens in a Hospital During Last Three Years

OBJECTIVE To investigate the distribution and resistance of commonly encountered pathogens,and provide reference of antimicrobial agents.METHODS For the clinical specimens during the last three years,flora identification and bacteriostatic test were operated with Microscan WalkAway-40.RESULTS Gram-negative bacteria were 252,507 and 742 strains,Gram-positive bacteria were 142,166,and 243 strains,fung were 0,26,and 229 strains from 2002 to 2004,respectively,the most commonly encountered Gram-negative bacteria were Escherichia coli,Klebsiella pneumoniae,Pseudomonas aeruginosa,Enterobacter cloacae,and Acinetobacter baumannii;the Gram positive bacteria were Staphylococcus aureus,S.epidermidis,S.haemolyticus,and Enterococcus.CONCLUSIONS It is important for reasonable drug application to know the distribution and resistance of commonly encountered bacteria.
5.Studies on HPLC Fingerprint of Triterpene Acids in Poria Peel

Objective To establish a method for HPLC fingerprint determination of the triterpene acids in poria peel.Methods Ethanol solvent and ultrasound method were used to extract the sample. The chromatographic conditions of RP-HPLC were:acefonitrile-0.5%phosphoric acid(linear gradient elution),the detection wavelength being 242 nm, temperature at 30℃and flow rate being 1.0mL?min-1. Ten batches of poria peel produced in different places were measured.The common model was set up to calculate the correlation coefficient, cos?between every batch vector and the total standard model vector, thus to define the similarity.Results There was much difference in the constituent content of poria peel produced in different places and batches.Conlusion The HPLC fingerprint of triterpene acids is stable and reprodacible,which is useful for standard cultivation and quality evaluation of the crud drug of poria peel.
6.The expressions of HSFl and XAF1 in endometrial carcinoma and their correlation analysis

ABSTRACT:Objective To test the expressions of heat shock transcription factor 1 (HSF1 )and XIAP-associated factor 1 (XAF1 )in different endometrial tissues,and analyze the association between their expressions and the clinicopathological features of this malignancy.Methods The expressions of HSF1 and XAF1 in 64 cases of endometria1 carcinoma (EC group)and 33 cases of normal endometrial tissues (NE group)were detected with immunohistochemistry S-P method.The correlation was observed.Results The positive expression rate of HSF1 was much higher in EC group than in NE group (76.6% vs .36.4%,P <0.05).The positive rate of XAF1 was 31.2% in EC group and 72.7% in NE group (P <0.05).The positive expressions of HSF1 and different subgroups of histological grade,myometrial invasion and lymph node metastasis were significantly different (P <0.05)in EC group.The positive expressions of XAF1 and different subgroups of histological grade,myometrial invasion,clinical stage and lymph node metastasis were significantly different (P < 0.05 )in EC group.There was a negative correlation between HSF1 and XAF1 in EC group (P <0.05).Conclusion In EC group,the high expression of HSF1 may inhibit the growth of XAF1 expression,cause excessive growth of cancer cells,reduce the apoptosis of cancer cells,and finally lead to the further development of tumors.
7.Effects of the fixed metallic dental materials on MRI images

In this paper,to evaluate the existence and extent of MRI artifacts caused by frequently used metallic dental materials and to compare the influence of different MRI sequences on artifacts. The artifact of Au-Pt-Pd alloy crown were the smallest and Au-Ag-Pd alloy crown, pure Titanium crown were smaller. But the artifact of Ni-Gr alloy crown were the largest.With the same scan sequences, the different metal materials can make different effects on MRI images with the different scan sequences, the MRI artifact is not obvious caused by the same metal materials. Artifacts produced by oral metallic materials related closely to the type of the materials.The high quality MRI can be gained by choosing proper metallic materials.
8.Effect of Different Package and Storage Conditions on Effective Components of Astragalus Pieces

This experimental study was aimed to find the effect of different package and storage conditions on the content of effective components of A stragalus pieces. A stragalus pieces were stored under different storage conditions by using different packaging materials and packaging methods. Every three months, the contents of Calycosin-7-glu-coside and astragaloside were determined according to the 2010 version of Chinese Pharmacopoeia. With the extend-edstorage time, the contents of two effective components were significantly decreased. After six-monthstorage, the contents were not consistent with the standard of the pharmacopoeia standards. Room temperature had relatively big influence on the loss of content. The plastic and aluminum paperpackagingwere better than kraft paper packaging. The content ofastragaloside using non vacuum packaging method was relatively higher than the vacuum packaging. Contentunder the conditions of cool storehouse and nonvacuum plastic bags was higher than other packagingmethod. And the changes of both contents were relatively stable. It was concluded that the A stragaluspieces should be packed with non vacuum plastic bags, and stored in a cool and dry place.
9.Research Progress of Traditional Chinese Medicine on Blood-brain Barrier

Blood-brain barrier (BBB) is a kind of important barrier system in the body. It can have a choice to prevent certain substances from entering the brain, in order to maintain the relative stability of internal environment of the central nervous system (CNS). However, it also became an important restriction factor for the treatment of CNS diseases. Traditional Chinese medicine (TCM) had good curative effects on several kinds of CNS diseases, which illustrated that effective composition of TCM can pass through or influence the function of BBB. This paper summarized the research progress of literatures on current pharmacological and clinical trials for the effect of TCM on BBB.
10.The research on interdisciplinary cooperative transformation learning modelin Disaster Nursing Course

Objective To study the application effect of interdisciplinary collaborative transformational learning mode in undergraduate nursing students′ disaster nursing teaching. Methods A total of 115 undergraduate nursing students in 2014 and 2015 were enrolled in this study. The subjects were divided into two groups: the control group (62 cases) and the experimental group (53 cases). The control group adopted the case teaching mode of full-time teachers, and the experimental group adopted the interdisciplinary collaborative transformation learning mode. The theoretical knowledge, skills, practical skills, critical thinking, humanistic accomplishment and nurse-patient communication skills of two groups of nursing students before and after intervention were compared. Results The scores of theoretical knowledge, operational skills, practical ability, critical thinking, humanistic accomplishment and nurse-patient communication before the intervention were (42.72±10.18), (35.72±7.11), (41.19±5.32), (298.09±3.98), (298.38±6.03), (52.87±6.02) points, (87.21±5.05), (87.02±4.81), (86.13±4.48), (378.47± 9.16), (327.15 ± 12.01) and (69.11 ± 3.38) points after intervention respectively. There was significant difference before and after intervention t=- 57.687-- 14.328, P<0.01). The scores of theoretical knowledge, operational skills, practical ability, critical thinking, humanistic accomplishment and nurse-patient communication before the intervention were (43.90±9.56), (38.21±8.87), (41.39±6.77), (296.76± 10.02), (296.89±6.37), (52.37±5.45) points, (82.27±6.46), (84.06±5.63), (78.21±6.99), (344.18±10.88), (315.16±12.82) and (64.45±3.37) points after intervention, respectively. There was significant difference before and after intervention (t=-38.221--9.823, P<0.01). There was no significant difference between the two groups before the intervention (P>0.05), and the score of the experimental group was significantly higher than that of the control group after intervention (t=- 18.107-- 2.997, P<0.01). Conclusions Introducing interdisciplinary cooperative transformation learning in disaster nursing teaching can promote the enhancement of nursing students'knowledge of disaster succeeding.
