1.Observation of curative effect of treatment of varicose veins of lower extremity with polyurethane foam sclerosing agent under fluoroscopic guidance

Objective To investigate and analyze the effect of polyurethane foam sclerotherapy on varicose veins of lower extremity under the guidance of fluoroscopy. Methods 90 patients with varicose veins treated in our hospital from February 2015 to May 2017 were selected and randomly divided into the control group and the experimental group, with 45 patients in each group. The patients in the control group were given conventional high ligation and stripping of the great saphenous vein. The patients in the experimental group were treated with fluoroscopic guided polyurethane foam sclerotherapy. The therapeutic effects of the 2 groups were compared and analyzed. Results After operation, 2 patients relapsed in the experimental group, and the recurrence rate was 4.4%. In the control group, 6 patients relapsed. The recurrence rate (13.3%) in the control group was significantly higher than that in the experimental group, with statistical difference. After the corresponding treatment, the intraoperative blood loss in the experimental group was (10.23±1.52) mL, and the bleeding volume in the control group was (32.19±2.34) mL. The bleeding volume in the experimental group was significantly less than that in the control group, with statistical difference. In addition, the average length of stay, length of operation and length of incision in the experimental group were significantly better than those in the control group, with statistical difference. Conclusion The clinical effect of fluoroscopy guided lauromacrogol foam sclerosing agent in the treatment of varicose vein of lower limb is better, the recurrence rate is low, can significantly reduce the amount of bleeding, shorten operation time, has clinical significance.
2.Relationship between AKT and PKCθ and morphine-induced inhibition of differentiation of T helper cells

Objective To evaluate the relationship between protein kinase B (AKT) and protein kinase (PKCθ) and morphine-induced inhibition of differentiation of T helper (Th) cells.Methods Peripheral venous blood samples were taken from 20 healthy volunteers..Peripheral blood mononuclear cells (PBMCs) were extracted by density gradient centrifugation method.CD4+ T lymphocytes extracted were purified by magnetic bead separation.CD4+ T lymphocytes were randomly assigned into 5 groups (n =3 each) using a random number table.CD4 + T lymphocytes were incubated routinely in group C.CD4 + T lymphocytes were incubated in the presence of PMA 25 ng/ml + ionomycin 1 μg/ml (group PI),PMA 25 ng/ml + ionomycin 1 μμg/ml + morphine 50 μμg/ml (group M),or PMA 25 ng/ml + ionomycin 1 μμg/ml + morphine 50 μg/ml + naloxone 50 μμg/ml (group N).The cells were incubated for 4 h in the incubator containing 5% CO2 at 37 ℃.The expression of interferon (IFN)-γ and interleukin-4 (IL-4) was detected by flow cytometry.The expression of IFN-γ and IL-4 was used to reflect the percentage of Th1 and Th2 cells,respectively.The ratio of Th1/Th2 was calculated.The expression of AKT,phosphorylated AKT (p-AKT),PKCθ and phosphorylated PKCθ (p-PKCθ) was detected by Western blot,and the ratio of p-AKT/AKT and p-PKCθ/PKCθ was calculated to reflect the activities of AKT and PKCθ,respectively.Results Compared with group C,the percentage of Th1 and Th2 cells and ratio of Th1/Th2 were significantly increased in PI,M and N groups,the activities of AKT and PKCθ were increased in PI and N groups,and the activity of PKCθ was increased in group M (P < 0.05).Compared with group PI,the percentage of Th1 cells,ratio of Th1/Th2 and activity of AKT were significantly decreased in group M,the ratio of Th1/Th2 was decreased in group N (P <0.05),and no significant change in the activity of PKCθ was found in M and N groups (P > 0.05).Compared with group M,the percentage of Th1 cells,ratio of Th1/Th2 and activity of AKT were significantly increased (P < 0.05),while no significant change in the activity of PKCθ was observed in group N (P > 0.05).Conclusion The mechanism by which morphine inhibits the differentiation of Th cells through activating opioid receptors is related to inhibition of AKT activation,but not related to PKCθ.
3.Shuxuening injection combined with deproteinized extract of calf blood on serum IGF-1, IL-1 and ICAM-1 in patients with cerebral infarction recovery stage

Objective To investigate Shuxuening injection combined with deproteinized extract of calf blood on serum IGF-1, IL-1 and ICAM-1 level in patients with cerebral infarction recovery stage.Methods 120 patients with cerebral infarction were collected.According to the different drug treatment, 60 cases in each group were given corresponding drug treatment, on the basis of control group, the calf blood extract injection was given in experimental group.After the end of treatment, all patients blood rheology, IGF-1, IL-1 and ICAM-1 levels were tested.Results Compared with control group, the indexes of the patients in experimental group improved more significantly, whole blood viscosity, red blood cell pressure volume, platelet aggregation rate decreased significantly(P<0.05); serum IGF-1 levels in experimental group decreased significantly(P<0.05).The levels of serum IL-1, ICAM-1 in experimental group decreased significantly(P<0.05).Conclusion Shuxuening injection combined with deproteinized extract of calf blood can significantly reduce serum IGF-1, IL-1 and ICAM-1 levels in patients with cerebral infarction recovery stage, improve blood flow, reduce blood viscosity.
4.Myocardial uptake of thiopental in the isolated rabbit heart:the study of pharmacokinetics and pharmacodynamics

To study the dynamic process of myocardial uptake of thiopentai in the isolated rabbit hearts. Method: Thiopental at doses of 500?mol, 1500?mol and 500?mol was given sequentially to the perfused rabbit heart in a total time of 15 min. The outflow concentration of thiopental was measured with high performance liquid chromatography and the left ventricular +dp/dtmax served as a effective parameter. Resuh: The disposition and elimination of thiopental can be best described hy a two-compartment open model. It can disposed into myocardium rapidly (T_(1/2)?=0.5?0.1 min), but elimination was relatively slow (T_(1/2)?=25.3?10.1 min). The transfer rate was slower from peripheral to central compartment than from central to peripheral compartment. The tbeoritical maximum depressant effect of thiopental on + dp/dt (Emax) was 19.0 4-11.2 kPa.s~(-1) corresponding to 1/10 E_0. Conclusion: The myocardial uptake of thiopental can be fitted to a two-compartment open model with rapid disposition and relative slow elimination process.
5.The study of the relationship between erythrocytic membrane band 3 and acid-base changes in patients with cor pulmonale

The changes of erythrocytic membrane band 3 protein and intraery-throcytic and extrar rythrccytic gases and electrolytes were studied in 69 cases of cor pulmonale and 50 normal subjects.It was found that in the patients of cor pulmonale accompanied with type Ⅱ respiratory failure,the relative low level of erythrocytic membrane band 3 protein and the restriction of HCO-3/Cl-exchange were the factors to aggravate CO2 retention and respiratory acidosis,relative intraerythrocytic alkalosis resulted from the relative increase of intra-erythrocytic HCO-3([HCO-3]),and prompt adminstration of oxygen to cor pulmonale patients with hypoxemia could not only improve extraerythrocytic acid-base imbalance but also increase intraerythcocytic P5O2 and the tissue capacity to store oxygen.

To explore the effects of lipopolysaccharide (LPS), TNF ? and IL 1? on cyclooxygenase 2 (COX 2) expression and prostaglandins (PGs) in rat polymorphonuclear (PMN), measured COX 2 mRNA was determined by reverse transcription polymerase chain reaction and changes in PGs after stimulation of PMN with LPS, TNF ? and IL 1? for 24 hours were observed. The resting PMN were found to express COX 2 mRNA weakly, while it was significantly up regulated after the stimulation of LPS,TNF ?and IL 1?. The level of PGs was increased markedly at the same time. These results suggested that COX 2 might play an important role during inflammatory process involving PMN.

9.Effects of LPS,TNF-? and IL-1? on cyclooxygenase-2 expression in human umbilical vein endothelial cells

AIM: To explore effects of LPS,TNF-?,IL-1? on cyclooxygenase-2(COX-2) expression and prostaglandins(PGs) production in human umbilical vein endothelial cells (HUVEC). METHODS: HUVEC were stimulated with lipopolysaccharide(LPS),TNF-?,IL-1? for 24 hours. Using in situ hybridization,reverse-transcription polymerase chain reaction and immunohistochemistry, COX-2 expression in both mRNA and protein level were observed and prostaglandins in culture medium were measured. RESULTS: Resting HUVEC expressed a little COX-2 mRNA. Expression of mRNA and protein of cyclooxygenase-2 increased significantly post-stimulation with LPS,TNF-?,IL-1? and PGs raised markedly at the same time. CONCLUSIONS: Resting HUVEC expressed a little COX-2 mRNA, inflammatory stimuli induced COX-2 superexpression and PGs production. These results suggested that endothelial cells could involve in inflammation by controlling of COX-2.
10.Design and application of fall risk scale in hospitalized patients in neurology department

Objective To score the fall risk of hospitalized patients with neurologic disease using fall risk scale and observe the changes of fall risk score after this fall risk scale was used. Methods The fall risk scale in hospitalized patients in neurology department was designed. The fall risk of hospitalized patients in neurology department from January 2005 to December 2007 were assessed. Moreover, safety nursing mea-sures were brought into practice to prevent from falling down. The fall risk of hospitalized patients in neurolo-gy department from January 2002 to December 2004 were assessed too. Results Target patients enhanced the sense of preventing from falling down after the fall risk scale was used, and the incidence of falls in pa-tients was significantly decreased. Conclusions To assess the fall risk of hospitalized patients in neurology department and implement safety nursing measures among the target patients may be effective in preventing the patients from falling down.
