2.The application of comprehensive echocardiographic parameters in assessment of left ventricular diastolic function in patients with hypertension

Objective:To discuss the clinical assessment value of comprehensive echocardiographic parameters in detecting the left ventricular diastolic function in patients with hypertension. Methods: 60 hypertensive patients (30 cases without left ventricular hypertrophy (non-LVH), 30 cases with left ventricular hypertrophy(LVH) were divided into hypertension group, and 60 healthy subjects were in control group. All of the subjects were underwent M type, color ultrasound and tissue Doppler imaging (TDI). And to compare their comprehensive echocardiographic parameters.Results: Compared with the healthy control group, the IVSd, LVPWd, LVMI, LAD and LVD of hypertension group were significantly enlargement; the s', e', E peak and LVEDP of hypertension group also significantly increased, while a' and A peak were significantly reduced; compared with non-LVH, e', E peak and LVEDP of LVH group were significantly increased, and a' and A peak were significantly reduced, and these differences were statistically significant (t=-4.39,t=-4.39,t=5.47,t=-8.02, t=6.20,t=18.95, t=16.12;P<0.01).Conclusion: The comprehensive echocardiographic parameters can evaluate the extent of damage for left ventricular diastolic function of patients with hypertension, and they can be as the assessment standard of left ventricular hypertrophy. In clinical practice, TDI can be used as a new means in the evaluation of ventricular diastolic function.
3.Effects and Mechanisms of Needle Pricking Therapy and Rotating-pulling Manipulation on Blood Velocity of Patients with Vertebral Artery Type of Cervical Spondylosis

Objective: To compare the effects of needle pricking therapy plus rotating-pulling manipulation and rotating-pulling manipulation on blood velocity,angiokinesis,and blood viscosity in patients with vertebral artery type of cervical spondylosis(CSA),and discuss its mechanisms.Methods: 216 patients were randomly divided into 2 groups according to numeric representation,108cases in the treatment group were treated by needle pricking therapy plus rotating pulling manipulation,and 108 cases in the control group were treated by rotating pulling manipulation.The peak systolic blood velocity(Vpeak),end diastolic blood velocity(Vmin),pulsatile index(PI)and resistant index(RI)of the bilateral vertebral arteries(VA)and basilar arteries(BA)were examined by transcranial doppler sonography(TCD).The values of whole blood apparent viscosity and plasma viscosity of both groups were recorded.The indexes before and after treatment of both groups were compared.Results: The therapeutic effects of the above-mentioned indexes of the treatment group were significantly superior to those of the control group(P<0.05).In comparison with pre-treatment,the whole blood apparent viscosity(high,medium and low shear rates)and plasma viscosity of the treatment group post-treatment were obviously reduced(P<0.01,P<0.05).Conclusion: Needle pricking therapy combined with pulling-rotating manipulation can significantly improve VATCS patients' clinical symptoms,which may be closely related to its effects in lowering vascular blood resistance and blood viscosity and increasing cerebral blood supply.
5.Establishment and clinical application of triplex taqman probes real-time PCR for pathogenesis of hand foot mouth disease

Objective To establish the triplex Taqman probes real-time RT-PCR method for simultaneously detecting of EV71,CA16 and EV.Methods Retrospective study.Specific primers and probes were designed based on conserved regions of EV71,CA16 and EV.The sensitivity,specificity and reproducibility were assessed by the optimized reaction system.A total of 176 throat swabs as the experimental group were collected from children with suspected hand foot mouth disease (HFMD),who admitted from April 2012 to July 2012 in Nanjing Children's Hospital affiliated to Nanjing Medical University.During this time,10 cases of healthy children,10 cases of outpatients with flu-like symptoms and 90 cases of inpatients in pneumology department of our hospital were recruited as control group,whose throat swabs were also collected.All of 286 samples were tested by the triplex Taqman probes real-time RT-PCR for simultaneously detecting EV71,CA16 and EV.SPSS13.0 was used to analyze the results.Results The sensitivities of the triplex Taqman probes real-time RT-PCR was 1.0 × 103 copies per milliliter for EV71,CA16 and EV.It showed 100% specificity for 9 enterovirus and 3 non-enterovirus.Analysis with 1.0 × 103-1.0 × 105 copies per milliliter constructed plasmids demonstrated high reproducibility with coefficient of variation of 0.44%-1.04% for EV71,0.38%-0.73% for CA16,and 0.46%-0.90% for EV.More over 176 samples collected from children with suspected HFMD were detected by triplex Taqman probes real-time RTPCR and real-time RT-PCR.The results showed 97.2% (171/176)agreement and 0.94 Kappa value with high concordance.Conclusions The triplex Taqman probes real-time RT-PCR detecting EV71,CA16 and EV simultaneously has been established successfully.The assay,with high sensitivity and specificity,provide good basis for the rapid clinical diagnosis of EV71,CA16 and EV and open up broad prospects for clinical and relevant researches.
6.lnfluence factors of dry eye among aerospace science and technology staff and the health guidance

AlM: To investigate and evaluate the prevalence of xerophthalmia among aerospace science and technology staff and its influencing factors for the eye health guidance.
METHODS: The staff underwent physical examination in our examination center from September 2013 to June 2014 were preliminary screened, and those patients with symptoms of dry eye were investigated with questionnaires and diagnosed by basic lacrimal secretion test ( Schirmer l ) , breakup time of tear film ( BUT ) test and fluorescein ( FL) staining test .
RESULTS: There were 606 complaints of dry eye symptoms of the 1 000 premiers, distributed in random (60. 6%), and 432 employees were diagnosed (43. 2%). The positive rate of dry eye was 71. 3%. The causes of xerophthalmia, besides of gender, age, also include a variety of factors such as air-conditioner, heating, video terminals, sleep time, etc.
CONCLUSlON: Dry eye, an epidemic disease, has a variety of symptoms and complex etiology, the main causes of aerospace science and technology staff with dry eye is working environment and some local factors. We can give appropriate health guidance according to the different factors. Meanwhile the relevant series of dry eye screening should also be taken as a routine examination in ophthalmic examination.
7.Chinese Mental Health Organizations in Medical and Educational Areas:A Comparative Investigation of Current Service,Management and Problems

Objective:To investigate current service,management and regulation conditions of Chinese mental health organizations in medical and educational areas.Methods:Professional staff from mental health organizations in 28 Chinese provinces,municipalities or autonomous regions took part in the investigation.Results:Most staff members in the two kinds of hospitals had medical background and bachelor degree.Most staff members in colleges and high schools had psychological or pedagogic background.Both medical and educational mental health organizations were faced with such problems as lacking of supervisions,unscientific management of human resources and lacking of ethical regulations.Conclusion:Chinese medical and educational mental health organizations should make fully use of their comparative advantages,and improve their professional service level and mutual cooperation.

9.Studies on the expression of Survivin in human leukemia K562 treated by cytarabine

Objective To investigate the influences of cytarabine on Survivin gene expression in human leukemia K562 cell line and discuss the mechanism of drug resistance in chemotherapy. Methods The IC50 of cytarabine was chosen by MTr assay. The K562 cells were exposed to certain concentration of cytarabine for about 24 hours and 48 hours. The expression levels of Survivin gene were detected by RT-PCR and Western-blot. Results After exposed to cytarabine for 24 hours and 48 hours, the Survivin mRNA level of K562 cells was significantly elevated about 1.92 and 3.38-fold, and the protein expression level was elevated about 1.92 and 2.64-fold. Conclusion An elevated expression of Survivin was tested in K562 cells treated by cytarabine. Consequently, the elevated expression of Survivin could resist apoptosis induced by chemotherapeutic agent which probably associated with chemotherapeutic drug resistance.
10.The mechanism through which low-intensity pulsed ultrasound promotes the differentiation of C2C12 myoblasts

Objective:To observe the effect of low-intensity pulsed ultrasound (LIPUS) at different intensities on the expression of adiponectin and its receptors in C2C12 myoblasts, and to explore the potential mechanism by which LIPUS promotes the differentiation of C2C12 myoblasts.Methods:C2C12 myoblasts cultured in vitro were randomly divided into a control group and U 0.1, U 0.3 and U 0.5 groups. The control group received sham LIPUS exposure, while the U 0.1, U 0.3 and U 0.5 groups were exposed to LIPUS at intensities of 0.1W/cm 2, 0.3W/cm 2 or 0.5W/cm 2 respectively, and 1MHz for 5 min daily for 5 days. Cell viability was measured using CCK-8 assays. Fluorescence quantitative reverse transcription-polymerase chain reactions were used to detect the mRNA expression of adiponectin, adiponectin receptor 1 (adipoR1) and T-cadherin in the cells. Western blotting was employed to assess the protein expression of adiponectin, adipoR1, T-cadherin, adenosine monophosphate activated protein kinase (AMPK), activated phosphorylated adenylate-activated protein kinase (P-AMPK), embryonic myosin heavy chain (eMHC) and myogenin (MYOG). The differentiation ability of the 4 groups was measured using cell immunofluorescence chemistry. Results:After the intervention the cell viability in the U 0.1, U 0.3 and U 0.5 groups was significantly higher than in the control group. Compared with the control group, the average mRNA expression of adiponectin and the receptors of adipoR1 and T-cadherin were up-regulated significantly in the U 0.3 and U 0.5 groups. The average adiponectin, adipoR1 and T-cadherin protein expressions, and the AMPK phosphorylation level in the U 0.3 and U 0.5 groups had increased significantly compared with the control group, but all were significantly lower than in the U 0.3 group. The average protein expression of eMHC and MYOG, and the C2C12 myoblast fusion indices of the U 0.3 and U 0.5 groups were significantly higher the control group′s averages. Conclusions:LIPUS can promote the differentiation of C2C12 myoblasts. It is most effective at 0.3W/cm 2, administered for 5min/d at 1MHz with a 20% duty cycle. Its regulatory mechanism may be related to up-regulation of the expression of adiponectin, the adipoR1 and T-cadherin receptors, and the activation of AMPK phosphorylation in C2C12 myoblasts.
