1.Role of JNK signaling pathway in hyperoxia-induced expression of connective tissue growth factor in A549 cells

Objective To investigate the role of c-Jun N-terminal kinase (JNK) signaling pathway in hyperoxia-induced expression of connective tissue growth factor (CTGF) in A549 cells.Methods The cultured A549 cells were seeded in 6-well culture plates at a density of 1 × 105 cells/ml (0.5 ml/well,24 wells in total) and were randomly divided into 4 groups (n =6 each) using a random number table:control group (group C);95% oxygen group (group 95% O2);room air plus SP600125 (JNK inhibitor) group (group C+SP);95% oxygen plus SP600125 group (group 95% O2+SP).SP600125 5 μmol/L was added to each well in C+SP and 95% O2+SP groups.The A549 cells were incubated for 48 h in an incubator filled with room air (C and C+SP groups) or 95% oxygen (95% O2 and 95% O2+SP groups).The expression of phosphorylated JNK (p-JNK) and CTGF mRNA was determined using Western blot and realtime reverse transcriptase polymerase chain reaction,respectively.Results Compared with group C,no significant change was found in the expression of p-JNK and CTGF mRNA in group C+SP (P>0.05),and the expression of p-JNK and CTGF mRNA was significantly up-regulated in group 95% O2 (P< 0.05).Compared with group 95% O2,the expression of p-JNK and CTGF mRNA was significantly down-regulated in group 95% O2+SP (P<0.05).Conclusion JNK signaling pathway activation is involved in up-regulation of hyperoxia-induced CTGF mRNA expression in A549 cells.
2.Evaluation on in vitro release of gastric floating sustained-release preparation of Tripterygium wilfordii by HPLC fingerprints

Objective To investigate the drug in vitro release behavior of gastric floating sustained-release preparation of Tripterygium wilfordii and establish their quality evaluating methods.Methods HPLC was Employed to gain the fingerprints of releasing medium of preparations.Triptolide was selected as marker to calculate the linear equation concerning peak area then the relative drug release and f2 simila-rity factor value were acquired.Results Various components in gastric floating sustained-release tablets and pellets of T.wilfordii could not release synchronously while sustained-releasing but all of them in the gastric floating sustained-release capsules of T.wilfordii prepared by using multiparticulate site-controlled release technology could release synchronously while sustained-releasing.Conclusion The quality and in vitro release behavior of gastric floating sustained-release preparation of T.wilfordii could be evaluated more scientifically and comprehensively by using HPLC fingerprints method.
3.Clinical and Laboratory Findings and Ultrasound Assessment in the Differential Diagnosis of Central Precocious Puberty and Premature Thelarche

Objective The differential diagnosis of female precocious puberty (PP) and premature breast development (PT) was made by ultrasound and related hormone levels,so as to provide the basis for clinical diagnosis.Methods In this study,from 2015 to 2016 due to early breast enlargement (age < 8 years) to our hospital pediatric patients with children,according to the diagnostic criteria were divided into PP group and PT group.Ultrasonography was performed on all patients and graded according to the criteria.At the same time,the levels of clinical and hormones were compared between the two groups.ROC analysis was used to identify the significant differences.Results In this study,a total of 60 cases of female children,including PP and PT children were 30 cases.Ultrasonic breast grading was positively correlated with age,BA,BD,LH level,FSH level and E2 level,but there was no correlation between BA/CA.There was no statistical difference between the breast ultrasound and the breast diameter (P < 0.05).The levels of LH and FSH,basal LH/ FSH,LF and FSH peak and LH/FSH peak in the PP group were significantly higher than those in the PT group (P < 0.05).The ROC analysis found 100% sensitivity and 85% specificity for LH/FSH peak >0.25 for diagnosis of PP,with a 72% sensitivity and a specificity of 65% for basal LH > 0.1 mIU/ml.The SHBG level was the only one that did not overlap [PP:80.6 (62.3-95.4) vs PT:114.5 (107.6-121.5)].There was no significant difference in kisspeptin,leptin and neurokinin B between the two groups (P > 0.05).Conclusion Breast ultrasound by grading and measurement of breast buds can be effective in evaluating the development of female precocious puberty breast development,but can not identify PP and PT.The study found that SHBG concentration and peak ratio of LH/FSH can effectively identify PP and PT,but kisspeptin,neurokinin B and leptin is not enough to identify PP and PT.
4.Effect of Tanggankang on the Expression of CREB in Diabetic-fatty Liver Rats

Objective To observe the effect of Tanggankang on the expression of CREB in diabetic-fatty liver rats;To investigate the mechanism of Tanggankang. Methods STZ and feeding high fat forage induced diabetic rats were randomly divided into model group, Huganning group, Tanggankang high dose group, Tanggankang medium dose group, Tanggankang low dose group, 10 rats in each group, with 10 normal rats as control. After 12 weeks of drug intervention, rats were killed, the livers were removed, and the expression of CREB was detected by Western blot. Results Compared with normal group, the level of CREB of model group was significantly decreased (P<0.01). Compared with model group, Tanggankang low, medium and high dose group significantly decreased the level of CREB (P<0.01). Conclusion Tanggankang may play an active role in glucose and lipid metabolism by affecting the expression of CREB.

8.Quality of abstracts for randomised controlled trials published in Journal of Clinical Anesthesiology based on CONSORT for Abstracts

Objective To assess the quality of randomized controlled trials(RCT)abstracts published in Journal of Clinical Anesthesiology by CONSORT for Abstracts.Methods Articles invol-ving human RCTs published from 2012 to 2013 were reviewed and searched through WanFang Med Online.Trials involving animal experiments,in vitro,RCTs without abstract,and meta-analyses were excluded.According to the CONSORT checklists for abstracts,the quality of abstracts for RCT were assessed.Results In total,392 RCT abstracts were analysed.The median word counts of ab-stracts was 364 (IQR 306-444),sample size was 60 (IQR 40-80).Almost all abstracts provided an appropriate description of conclusions (100%), numbers randomized (99.0%) and objective (99.0%).The majority of abstracts described interventions (94.6%)and participants (82.4%).Re-quirements present in less than 50% of the abstracts were details regarding trial design (46.2%)and harms (48.7%).The descriptions of randomization (13.3%),blinding (1.8%),methods-outcome (3.6%)and results-outcome (9.7%)were very low.Moreover,title,recruitment and numbers ana-lysed were not reported.Conclusion The quality of RCT abstracts and adherence to the CONSORT checklist for abstracts remains poor,and the CONSORT for Abstracts should be endorsed to improve the quality of RCT abstracts as early as possible.
9.The mediating role of mindfulness level on relationship between perceived stress and sleep problems

Objective To investigate the relationships among perceived stress,mindfulness level,and sleep problems,and to indicate the mediating role of mindfulness level in the relationship between perceived stress and sleep problems.Methods For 179 undergraduate students,the Five Facet Mindfulness Questionnaire (FFMQ),Chinese Perceived Stress Scales (CPSS) and Athens Insomnia Scale (AIS) were used to measure.Results The scores of the scales were (120.43 ± 1 1.23) for FFMQ,(40.17 ± 6.86) for CPSS and (5.28 ± 4.04)for AIS.Pearson correlation analysis showed that perceived stress was negatively correlated to mindfulness level (r =-0.602,P =0.000) and positively correlated to sleep problems (r =0.459,P =0.000) and that there was significant negative correlation between mindfulness level and sleep problems (r =-0.427,P =0.000).Hierarchical linear modeling showed significant partial mediation of mindfulness level on the association between perceived stress and sleep problems.Conclusion Mindfulness level partly mediated the positive link between perceived stress and sleep problems.
10.Toll like receptor expression in spleen of rat skin transplantation model

Objective:The expression patterns of TLR4,9,10 in the rat spleen were compared between the rat model of acute skin-transplant rejection and physiologically normal rats ,and their physiological significance were accessed.Methods:6 Wistar and 6 SD rats were used as donors ,and another 24 SD rats were used as recipients which were divided into 4 groups of 6.The same back skin slices were transplanted from donor to recipient , Group A:allograft ( Wistar to SD );Group B:homograft ( SD to SD );Group C:autograft;and Group D:normal SD rats as control.Rat spleens were collected at 1,4,7,10,14,and 21 days after surgery ,and mRNA levels of TLR4,9,10 in splenic mononuclear cells were determined by real-time PCR and corresponding protein expression was accessed by immunohistochemistry.Results:TLR4,9,10 mRNA levels of Group A were significantly higher than that of Group B and C ( P<0.05);TLR4,9,10 mRNA levels showed no significant difference between Group B and C (P>0.05).TLR4,9,10 expressions were moderate (++) to strong (+++) in Group A,and generally weak (+) in Group B,C and D.Conclusion:The patterns of TLR4,9,10 mRNA and protein expression in the rat model of acute skin-transplant rejection indicate that the three genes are involved in acute transplant rejection;the three genes may be expressed coordinately;inhibition of the Toll-like receptors may provide therapeutic targets for acute transplant rejection.
