1.Progress on Optical Probes for Hydrogen Sulfate Anion Sensing

The hydrogen sulfate plays an important role in biological and environmental areas and can contaminate the environment, which will cause harm to human body.Thus it is of primary importance to detect hydrogen sulfate with high selectivity and sensitivity.Among these methods for detection of hydrogen sulfate, the optical chemosensor based on molecular recognition is desirable with unique advantage.Anions optical sensing systems are predominantly attractive due to their simplicity, high degree of specificity, low detection limits, easy on-line analysis and especial colorimetric recognition and in situ detection.Herein, the progress during the last decades of optical chemosensors and probes for hydrogen sulfate based on the recognizing mechanism is summarized.The further research orientations are also prospected.
2.Effects of thyroxine on serum lipid and bone metabolism in elderly patients with subclinical hypothyroidism

Objective To investigate the effects of L-thyroxine(LT4) on serum lipid and bone metabolism in aged patients with subclinical hypothyroidism (SH). Methods Seventy-eight patients with diagnosed SH were randomly assigned to receive LT4 (therapy group) or placebo(control group) for one year. Mean dose for LT4 replacement was (80.1?37.2) ?g/d. The changes of clinical symptoms, body mass index (BMI) and serum levels of total cholesterol(TC), triglycerides(TG), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol(LDL-C), apolipoprotein A 1(ApoA 1), apolipoprotein B 100 (ApoB 100 ), calcium ion(Ca 2+ ), phosphorus ion (P 3+ ) and alkaline phosphatase (AKP) and bone mineral density (BMD) were measured before and after therapy. Results After LT4 therapy, clinical symptoms were ameliorated. 3.The application of comprehensive echocardiographic parameters in assessment of left ventricular diastolic function in patients with hypertension

Objective:To discuss the clinical assessment value of comprehensive echocardiographic parameters in detecting the left ventricular diastolic function in patients with hypertension. Methods: 60 hypertensive patients (30 cases without left ventricular hypertrophy (non-LVH), 30 cases with left ventricular hypertrophy(LVH) were divided into hypertension group, and 60 healthy subjects were in control group. All of the subjects were underwent M type, color ultrasound and tissue Doppler imaging (TDI). And to compare their comprehensive echocardiographic parameters.Results: Compared with the healthy control group, the IVSd, LVPWd, LVMI, LAD and LVD of hypertension group were significantly enlargement; the s', e', E peak and LVEDP of hypertension group also significantly increased, while a' and A peak were significantly reduced; compared with non-LVH, e', E peak and LVEDP of LVH group were significantly increased, and a' and A peak were significantly reduced, and these differences were statistically significant (t=-4.39,t=-4.39,t=5.47,t=-8.02, t=6.20,t=18.95, t=16.12;P<0.01).Conclusion: The comprehensive echocardiographic parameters can evaluate the extent of damage for left ventricular diastolic function of patients with hypertension, and they can be as the assessment standard of left ventricular hypertrophy. In clinical practice, TDI can be used as a new means in the evaluation of ventricular diastolic function.
4.Discussion on Mechanism and Effect of Mica Granules on Rats' Stomach Mucosa Injury Induced by Non-water Ethanol

[Objective] To study the protection and mechanism of mica granules to rats stomach mucosa injury caused by non-water ethanol.[Method] Randomly divide 48 healthy SD rats into high-,middle-and low-dosed groups of mica granules,Simida group,model control group and blank control group as well,i.e.6 groups.After fasting for 24h,separately make stomach perfusion of mica granules and Simida to all rats in advance;after 3h,make stomach perfusion of non-water ethanol 1ml/100g for stomach mucosa injury;45m later,take proper blood of lower vena cava,sacrifice them for stomach,then respectively test all mucosa injury index,SOD,MDA,and GSH-Px content in serum and mucosa,observe pathological changes to evaluate mucosa injury degree and medical function.[Result] In mica groups,the SOD and GSH-Px were markedly higher than that in model control group,but MDA was lower than later(P
5.Sixteen Cases of Primary Small Vessel Vasculitis with Involvement of Lungs

Objective To understand the clinical characteristics of primary small vessel vasculitis with involvement of lungs in children and analyze the causes of misdiagnosis.Methods Clinical data from 16 patients with primary small vessel vasculitis were collected and their pathological,antineutrophil cytoplasmic antibody(ANCA),pulmonary lesion manifestations,image and misdiagnosis were further analyzed.Results Among 16 cases,9 cases were microscopic polyarateritis(MPA) and 4 cases were Wegeners granulomatosis(WG) and 3 cases were Churg-Strause syndrome(CSS).The ages of onset were from 3 to 14 years old with average age of 7.5 years old.Seventy-five percent(12/16) were ANCA positive.The patients with lung involvement had cough,chest pain,dyspnea,hemoptasis and asthma.Findings on lung X-ray films were variable.Some showed multiple patch shadows,others showed single or multiple masses or nodular shadows with or without cavity formation.After the treatments with corticosteroid and cyclophosphamide,10 cases turned better;including 8 cases with normal on lung X-ray films.Three cases given up;3 cases died.Conclusion It is very difficult to make diagnosis of primary small vessel vasculitis with involvement of lungs and should be paid more attentions to this issue.
6.High glucose induces apoptosis of human peritoneal mesothelial cells by caspase-3 activation

Objective To investigate the mechanism of high-glucose-induced injury to human peritoneal mesothelial cells(HPMC). Methods (1)The cultured HPMCs were exposed to culture medium containing different concentrations of glucose(1. 5% , 2. 5% , 4. 25% )for 48 hours and 4. 25% mannitol and normal culture medium were as control. Then apoptosis was observed by flow cytometry and caspase-3 activity was measured by ApoAlert?CPP33/Caspase-3 Assay kits. (2) The cultured HPMCs were exposed to 4.25% glucose culture medium containing different concentrations of caspases inhibitor, Z-VAD. fmk (25, 50, 100 ?mol/L) for 48 hours and 4. 25% glucose culture medium containing DMSO was as control. Then apoptosis was observed by flow cytometry and caspase-3 activity was measured by ApoAlert?CPP33/ Caspase-3 Assay kits as well. Results (1) Glucose increased caspase-3 activity in a concentration-dependent manner. Compared to control, caspase-3 activity was significantly higher in 4. 25% glucose group and 2. 5% glucose group, but not significantly different in 1. 5% glucose group and 4. 25% mannitol group. (2) Apoptotic rate of HPMC was significantly lower in Z-VAD. fmk group than that in control. Z-VAD. fmk decreased the number of apoptotic cells in a concentration-dependent manner. Also, caspase-3 activity of HPMC was significantly lower in Z-VAD. fmk group than that in control. Conclutions (1) High-glucose can induce apoptosis and caspase-3 activation of HPMC in a dose-dependent manner. (2) Z-VAD. fmk inhibits high glucose-induced apoptosis of HPMC in a dose-dependent manner. (3)High glucose induces apoptosis of human peritoneal mesothelial cells by caspase-3 activation.
7.Difference analysis of proteome between diabetic cataract and age related cataract

Background With the changes of diet and living style,the diabetes has become the major diseases affecting human health.Diabetic cataract is a common complication of diabetes. Objective The present study was to investigate the difference of lens proteomics between diabetic cataract and age related cataract using two dimensional electrophoresis (2-DE) and mass spectrometry in order to postpone happening of diabetic cataract and offer the effective approach to the prevention and therapy of diabetic cataract. Methods The lenses were obtained from 8 diabetic patients and 12 age-related cataract patients during the surgery to extract the protein by lysis and centrifugation.The lens proteins were separated using immobilized pH gradients 2-DE.Image analysis was carried out using PDQuest Advanced-8.0.1 software package.Significant difference of the crystallines was identified by matrixassisted laser adsorption/ionization time of-flight-mass spectrometry (MALDI-TOF-MS) and peptide mass fingerprint combined with protein database. Results The maps of 2-DE showed that lens proteins of diabetic cataract and age related cataract were in the section of pH 5-9 with the relative molecular weight 14000-97000;while relative molecular weight of more abundant crystalline was localized at 20000-31000.About 3 differential protein spots were detected by image analysis software.Two crystallines,αB and βB1 crystallin,were identified using MALDI-TOF-MS.Conclusions Proteomic analysis of lens can be accomplished and the proteins can be well separated,moreover,differential proteins can be analyzed using 2-DE and mass spectrometry between diabetic cataract and age related cataract.These results indicate that αB and βB1 crystallin proteins accelerate the development of diabetic cataract.This technique offers a new avenue for clarity of lens proteins of diabetic cataract other than age related cataract.
8.Expression, purification and characterization of a thermostable lactate dehydrogenase from Thermotoga maritima.

The gene encoding thermostable lactate dehydrogenase (Tm-LDH) was cloned into the plasmid pHsh from Thermotoga maritima, and expressed in Escherichia coli JM 109. The recombinant protein was purified to homogeneity by a simple step, heat treatment. The recombinant enzyme had a molecular mass of 33 kDa. The optimal temperature and pH of Tm-LDH were observed 95 degrees C and 7.0. The purified enzyme had a half-life of 2 h at 90 degrees C, and exhibited better stability over a pH range from 5.5 to 8.0. The K(m) and V(max) values were 1.7 mmol/L, 3.8 x 10(4) U/mg of protein for pyruvate, and 7.2 mmol/L and 1.1 x 10(5) U/mg for NADH, respectively. The expression of Tm-LDH in T7 system could not obtain high efficiency, but it has been soluble over-expression in pHsh system and reached 340 mg/L. The superior stability and productivity of Tm-LDH will lay the foundation of its industrial-scale fermentation and application in the NAD regeneration.

9.Multi-slice spiral CT perfusion imaging of chronic obstructive pulmonary disease

Objective To evaluate the diagnostic value of multi-slice spiral CT(MSCT)perfusion imaging in chronic obstructive pulmonary disease(COPD).Methods Twenty COPD patients and20 volunteers underwent 8-row detector spiral CT(MSCT)perfusion imaging using cine scan mode with5 mm slice thickness.0.5 s rotation time and a total scan time of 45 s with 5 s intervals.60 ml contrast agent(300 mg I/ml)were administered at a rate of 4 ml/s from the forearm superficial vein.The imaging data were transferred to a workstation.A time-density curve and pseudo-color map were generated automatically with GE CT perfusion 3 software,the blood flow(BF),blood volume(BV),mean transit time(MTr)and permeability surface(PS)were measured.Results Time-density curve was flatter and the peak of the curve was obviously lower in COPD patients than the volunteers.The BF.BV.PS in COPD volunteers was(10.58 ±4.85)s and(4.50 ±1.71)s respectively.The BF,BV and PS in COPD patients Was lower than the volunteers,the MTY was higher(P<0.01).Conclusion MSCT perfusion imaging is helpful for the diagnosis of COPD.
10.Qualitative research on the factors affecting compliance of patients with maintenance hemodialysis

Objective To explore the factors affecting compliance of patients with maintenance hemodialysis and to provide reference for the improvement of patients' compliance.Methods Ten patients with maintenance hemodialysis were individually interviewed in this study.The data were analyzed based on phenomenological method.Results Three factors were found,including feasibility of doctor's advice,characteristics of patients such as awareness of disease related knowledge,recognition todisease progression and usual behavioral pattern,as well as social factors such as economic status,life pressure,family support,professional supervision and guidance.Conclusion In order to enhance patients' compliance,it is suggested to improve thefeasibility of doctors' advice and health education,as well as pay close attention to the influence of patients' recognition,psychological status and social factors on the compliance.
