1.Primary suggestions on the anti-viral drugs treatment in chronic HBV carriers

Objective To establish a standard anti-viral treatment for chronic HBV carriers. Methods With reference to the domestic and abroad literatures, the diagnostic and therapeutic standards were set and prepared to be verified. ResultsThe diagnostic standards included:(①ALT 30 -40 IU/L: add 2 scores. (②Age:30 -39 ,add 1 score; ≥40,add 2 scores. (③Ultrasound examination: obviously abnormal,add 1 score;deteriorated during follow up:add 2 scores. ④Fibrosis markers positive: add 1 score. (⑤History of HCC in vertical relatives:add 1 score. ⑥ Existence of conditions which can induced elevation of ALT such as fatty liver or alcoholic liver or BMI > 23 kg/m2,minus 2 scores. The therapeutic standards included:(①≥5 scores: should be treated;(②3 -4 scores: may be treated or just follow up;(③≤2 scores;follow up and reexamine every 6 months. ConclusionsThe above standards are preliminary suggestions which should be checked and rectified by liver biopsy to achieve a more accurate and more practical standard.
2.Investigation of nurses′learning motivation and learning satisfaction with continued education and the influencing factors

Objective To investigate nurses′ learning motivation and learning satisfaction with continued education and analyze its influencing factors. Method One hundred and seventy-nine nurses were enrolled in the survey and the general information questionnaire, education participation scale and continuing education learning satisfaction scale were used in the investigation. Results The average score by nursing education participation scale was (3.26 ± 0.59), and that of continuing education learning satisfaction scale was (2.74 ± 0.48). There were statistically significant differences in age, length of employment, professional title, marital status, employment mode between the scores by the two scales (P < 0.01). Conclusions Nurses′ learning motivation and learning satisfaction with continuing education are in the medium level. Pertinent guidance such as bringing continuing education in depth and width is suggested to improve satisfaction and motivations of nurses.
4.Construction of eukaryotic expression system of a cell surface molecule——human CD40 ligand

Objective To construct human eukaryotic expression system expressing T cell surface molecule CD40L and identify the expression product in SCCL cell line h446. Methods The expression vector pcDNA3.1-CD40L was constructed by recombinant DNA technology, then transfected into h446 cell line of SCCL by lipidosome methods, and the expressed membrane protein was identified by flow cytometry. Results The pcDNA3.1(+)-CD40L was constructed successfully and the coding region was inserted into the vector correctly. The membrane protein was observed by flow cytometry on transfected h446 cell line of SCCL. Conclusion Human CD40L was expressed successfully in eukaryotic system, which is very important in the anti-tumor vaccination study with a recombinant human CD40L in clinic.
5.Study on the relationship between adiponectin/leptin ratio and metabolic index in type 2 diabetes mellitus

Objective To investigate the relationship between adiponectin /leptin ratio and metabolic index in type 2 diabetes mellitus .Methods A total of 167 cases of type 2 diabetes in our hospital endocrinology clinic 59 people from January 2013 to 2014 were enrolled in the study ,all patients were divided into three groups accordius to fasting plasma glucose(FPG)(42 people< 7 .2 mmol/L as Group 1 of ,7 .2-9 .4 mmol/L as group 2 66 people> 9 .4mmol/L as Group 3) .Patients who have been excluded from treatment with insulin or insulin .Requiring patients to 10 - 12 hours of fasting ,taken fasting blood samples and samples will be sent to the hospital clinical laboratory measured by biochemical analysis way ,the patient of glycosylated hemoglobin ,fasting blood glucose ,blood lipid indexes for comprehensive testing and calculation of patient related indexes ,and carries on the analysis .Results the patient′s body mass index(BMI) ,fasting insulin (FINS) ,low‐density lipoprotein cholesterol(LDL) ,triglyceride(TG) ,leptin and adiponectin ,between the ages of no statistical significance exists ,male patients with adiponectin levels and TG ,BMI ,HDL and LDL has a relatively close relationship (positive ,P=0 .000 3) leptin levels and BMI was negatively correlated (P<0 .05) .The adi‐ponectin levels of female patients were closely related to TG ,LDL and HD (positive correlation ,P<0 .05) .Leptin levels and TG were associated with BMI (negative correlation) ,and A/L ratio was correlated with all the indexes (negative correlation) in male and female patients (P<0 .05) .Conclusion HOMA‐IR can not be used as an important indicator of insulin resistance ..A/L and it are more suitable for .
7.Expression of Twist gene in human hepatocellular carcinoma cell strains of different metastatic potential.

In order to investigate the role of Twist gene in the metastasis of hepatocellular carcinoma (HCC), total RNA was respectively extracted from three HCC cell strains with different metastatic potentials, HepG2, MHCC-97L and MHCC-97H. The first strand cDNA was synthesized by reverse transcription, which was then used as template to perform fluorescent quantitative polymerase chain reaction (FQ-PCR). The quantity of Twist gene expression was normalized by that of the housekeeping gene, GAPDH for each sample. ANOVA was used to estimate the relationship between Twist gene and metastasis potential of HCC. The results showed that the normalized initial cDNA concentrations of Twist gene in HepG2, MHCC-97L and MHCC-97H were (9.45+/-0.25)x10(-4), (1.82+/-0.41)x10(-3), (3.06+/-0.62)x10(-3), respectively. FQ-PCR revealed significant differences in the expression level of Twist among HCC cell strains with different metastatic potentials. It was concluded that high expression level of Twist was closely associated with more aggressive behaviors of HCC. Twist provides a novel indicator for HCC metastasis.

8.Preparation of Rifampicin Suppositories and Solubility Determination

OBJECTIVE: To prepare rifampicin suppositories and to determine its dissolution in vitro. METHODS: Rifampicin suppositories were prepared with gelatin and glycerine as base material, the dissolution of rifampicin suppositories was determined by UV spectrophotometry and which were compared with rifampicin capsules sold in the market. RESULTS: The linear range of rifampicin was 10.0~60.0?g?mL-1(r=0.9 999), and the average recovery was 99.87%(RSD=0.42%, n=6). The dissolutions of rifampicin suppositories and rifampicin capsules in vitro at 45min were(89.9?0.97)% and(79.8?1.14)%, respectively. CONCLUSION: Rifampicin suppositories were simple in preparative method, well-formed, low in cost, and with high dissolution in vitro.
9.Effect of lithium chloride on neuronal apoptosis and expression of P53 and nuclear factor kappa B after forebrain ischemia in gerbils

BACKGROUND: Recently, lithium was reported shown neuroprotective effect against apoptosis induced by a variety of insults in vitro and in vitro,but the precise mechanisms underlying its neuroprotective effect remain unknown.OBJECTIVE: To observe the effect of lithium chloride on neuronal apoptosis and the expression of P53 or nuclear factor kappa B (NF-κB) protein in the CA1 region of the hippocampus after global ischemia in gerbils.DESIGN: A randomized controlled experimental research.SETTING: Department of Anatomy of Nanjing Medical University.MATERIALS: Fifty-four healthy male gerbils weighing 50-70 g, clearing grade, were purchased from Experimental Animal Center of Zhejiang Province.METHODS: Totally 54 gerbils were randomly divided into three groups namely: sham-operation group (SH group), ischemia-reperfusion group (IR group) and lithium chloride group (LI group), with 18 in each group. SH group, IR group and LI group were further divided into 3 subgroups respectively (SH1d, SH3d, SH7d; IR1d, IR3d, IR7d; LI1d, LI3d, LI7d), according to the time of reperfusion, with 6 gerbils in each. Gerbils in LI group were injected intraperitoneally with lithium chloride 3 mEq /kg, once a day for 7consecutive days before operation. Normal saline was used instead of lithium in SH group and IR group as vehicle control. Forebrain ischemia was induced at 24 hours after the last injection of lithium chloride. After gerbils being anesthetized, the bilateral common carotid arteries were blocked with micro aneurysm clips for 5 minutes, and the micro aneurysm clips were removed and the cerebral blood flow restored. Sham-operation animals were underwent the same operation except occlusion of bilateral common carotid arteries. Gerbils in each group were killed at every time points.4 μm coronal sections at 1.7-4.0 mm visual cross were cut at the level of the dorsal hippocampus. The apoptosis cells were assayed with in situ Cell Death Detection Kit, and assay of positive cell in cell apoptosis, P53 and positive NF-κB was performed with immunohistochemistry staining. The total number of TUNEL positive cells, P53 or NF-κB positive cells per image (area of 1 mm2) was counted.MAIN OUTCOME MEASURES: Neuronal apoptosis and expression of P53 or NF-κB protein in the CA1 region after cerebral ischemic reperfusion.apoptosis cell in cerebral hippocampus CA1 region: No TUNEL positive cells were detected in SH group, a large majority of TUNEL positive cells were detected in the CA1 region in IR group on the 3rd day after reperfusion [(552.0±145.5, 142.4±103.5) pcs/mm2, t= 5.623, P < 0.01], and TUNEL positive cells declined on the 7th day after reperfusion. The numbers of TUNEL positive cells in the CA1 region of LI3d, LI7d group were significantly lower than those of IR3d, IR7d group [(408.0±119.8, 156.0±108.2) pcs/mm2,CA1 region: In IR group, the expression of P53 protein was increased on the 1st, 3rd and 7th day after reperfusion compared with that in SH group and cerebral hippocampus CA1 region: No NF-κB protein was expressed in SH group. In IR group, the expression of NF-κB protein was increased on the 1st day after reperfusion (78.5±25.2)/mm2, significantly increased on the 3rd day after reperfusion (176.5±35.5)/mm2 and on the 7th day after reperfusion, the expression of NF-κB protein disappeared. There were no significant statistical difference between LI group and IR group on the 1st day after reperfusion. The expression of NF-κB protein in LI group was significantly lower than that in IR group on the 3rd day after reperfusion [(64.5±30.8)/mm2,t=5.824, P < 0.01].CONCLUSION: Lithium chloride can significantly suppress neuronal apoptosis after global ischemia in gerbils. The down-regulation of expression of P53 or NF-κB protein is one of the mechanisms of the neuroprotective effect by lithium chloride.
