1. Research Progress of Liquid Crystalline Nanoparticles as Drug Delivery System

Liquid crystalline nanoparticles are composed of a certain concentration of amphiphilic lipid self-assembly in water. The attractiveness of this formulation is linked to the nanostructural versatility, compatiblity, digestiblity and bioadhesive properties of their lipid constituents, and the capability of solubilizing and sustaining the release of amphiphilic, hydrophobic and hydrophilic drugs. Liquid crystalline nanoparticles, as a novel drug delivery system, have great promise in drug delivery. The carrier materials, preparation methods, characterization and applications of liquid crystalline nanoparticles were reviewed based on relevant articles published in recent years to provide reference for further study of liquid crystal nanoparticles.
2. Preparation and in Vitro Evaluation of Triptolide-Loaded Cubic Liquid Crystals

OBJECTIVE: To prepare and characterize triptolide cubic liquidcrystal,study the rheological properties and in vitro transdermal properties. METHODS: Cubic liquid crystal (V2) was prepared by using phytantriol-carbitol-water three-component system. Using the ternary phase diagram method to optimize the target area. V2 was characterized by small-angle X-ray scattering, infrared spectroscopy and differential scanning calorimetry. Rheological properties of V2 was studied by a DHR-2 rheometer. In vitro drug release studies were conducted by using improved Franz diffusion cell method. RESULTS: SAXS is shown as double-diamond lattice cubic liquid crystal; rheological parameters are good; DSC results show V2 can be converted into a hexagonal phase at 65.5 ℃ and converted into a layered liquid crystal at 90 ℃, and the addition of the drug does not change the liquid crystal structure. In vitro transdermal cumulative permeation amount is about 1.54 times that of the gel, and the sustained release of triptolide can be as long as 48 h. CONCLUSION: The prepared triptolide liquid crystal has good appearance and suitable rheology, and its cumulative permeation amount and infiltration rate of V2 are superior to the hydrophilic gel, which provides theoretical reference for triptolide percutaneous administration.
3.Protective effect of isoflurane preconditioning on myocardium against ischemia-reperfusion injury in isolated rat hearts

Objective To investigate the protective effect of isoflurane preconditioning on myocardium against isehemia-reperfusion(I/R)injury and the possible mechanism.Methods Fifty male SD rats weighing 250- 300 g were randomly divided into 5 groups(n=10 each):Ⅰ control group(C);Ⅱ I/R group and 3 isoflurane preconditioning groups with 0.5%(Ⅲ),or 1.0%(Ⅳ)or 2.0% isoflurane(Ⅴ).The animals were anesthetized with intraperitoneal pentobarbital 40 mg?kg~(-1).The hearts were immediately excised and placed in cold K-H solution.The aorta was canuulated and heart retrogradely perfused with K-H solution aerated with 95% O_2 and 5% CO_2 at 37℃ and 10 kPa in a langendorff apparatus.Left ventricular end-diastollc pressure(LVEDP)and left ventrieular systolic pressure(LVSP)were measured from a fluid-filled latex balloon in the left ventricle.The isolated hearts were made globally ischemic for 30 min followed by 60 rain reperfusion in group Ⅱ-Ⅴ.In the 3 isoflurane preconditioning groups the hearts were perfused with K-H solution saturated with 0.5% or 1.0% or 2.0% isoflurane for 15 rain followed by 15 rain washout before ischemia.The cardiac function variables including LVEDP,LVSP dp/dt_(min),dp/dt_(max) and HR were measured after epuilibrium(baseline values),immediately before ischemia,at the end of 30 min ischemia and 60 min reperfusion.The infarct size and cytochrome C level in cytoplasm and mitochondria of myocytes were measured.Results I/R significantly increased LVEDP and decreased LVSP,dp/dt_(min),dp/dt_(max) as compare with control group.Sevoflurane preconditioning significantly attenuated the depression of cardiac function caused by I/R.Only LVEDP was significantly higher during reperfusion period in the 3 sevoflurane preconditioning group than in the control group but there was no significant difference in LVSP,dp/ dt_(min),dp/dt_(max) between control group and the 3 preconditioning groups.The infarct size was significantly smaller in the 3 preconditioning groups than in I/R group.Cytochrome C level was significantly increased in cytoplasm but decreased in mitochondria in I/R group as compared with control group.Sevoflurane preconditioning significantly ameliorated the release of cytochrome C from mitochondria to cytoplasm in the 3 sevoflurane preconditioning group.Conclusion Isoflurane preconditioning can protect the heart against I/R injury by attenuation of the release of cytochrone C from mitochondria to cytoplasm.
4.A Study on the awareness of HIV/AIDS and risk behaviors among migrant workers at construction sties in Beijing

Objective To investigate the awareness of HIV/AIDS and risk behaviors among migrant workers at construction sites in Beijing. Methods The methods of cluster sampling was employed,a questionnaire survey was administered among 1000 migrant workers from two construction sites and a face-to-face interview with the informed consent was conducted in a limited scope.Results There was considerable disparity in the awareness of the routes of AIDS transmission;66.8% of the migrant workers were aware that the condom could reduce the risk of infection,but 51% of them had never used it.Only 2.3% of the migrant workers had sexual intercourse with female sex workers.Conclusion Although the migrant workers under survey have some basic knowledge on HIV/AIDS,their essential understanding was mostly vague,and financial condition have restricted to a great extent their risk behaviors.
5.Determination of the Physical Chemistry Constants of Total Cumarin

Objective To determine pH value, the solubility in different solvents and the oil-water partition coefficients of cumarin. Method UV spectrophotometry was used to determine the content of cumarin. Shake-flask method was developed to determine the oil-water partition coefficients of cumarin. Result The higher solubility values of cumarin were 2 044,2015 and 1969 ?g/mL in acetone , methanol and ethanol, it was almost indiscerptible in water, the limit solubility was 1096.9 ?g/mL, which become stable while the whisk time reached to 8 hours. The oil/water detached coefficient was 11.210, which was higher in acid solvent. Conclusion The method is simple, rapid and reliable.
6.Corneal sensitivity and dry eye after LASIK

AIM: Firstly to evaluate the changes of corneal sensation and its effect to dry eye syndrome after laser in situ keratomileusis (LASIK); secondly to evaluate the relation between the changes of corneal sensation and the laser ablation depth. ·METHODS: 30 patients (60 eyes) had bilateral LASIK with a superior-hinged flap. Every patient underwent preoperative and postoperative (after one week, one month and three months) evaluations, including corneal sensation, Schirmer's basic tear secretion test, tear breakup time (TBUT), corneal fluorescein stainting, and a questionnaire evaluating dry eye obtained from each patient.·RESULTS: Compared with preoperative level, after one week and one month, the difference of corneal sensation was very significant (P< 0.01); after three months, there was no statistical difference (P > 0.05). After one week, there was no statistical difference (P > 0.05) in the subjective dry eye symptoms, after one month, the difference was significant(P=0.025 <0.05) and after three months, the difference was very significant (P=0.001 < 0.01). Compared with preoperative level, the basic tear secretion decreased significantly after one week and one month(P< 0.01), it did not return to the baseline level after three months (P < 0.01). Tear breakup time decreased significantly after the surgery, and after three months, there was still statistical difference compared with preoperative level (P < 0.01); with cornea fluorescence staining, we found corneal epithelial defect was most serious after one month after LASIK, and did not return to the baseline level after three months (P < 0.01). Linear regression analysis and relevant were used to evaluate the relation between the changes of corneal sensation and the laser ablation depth (r=0.798, P < 0.01). ·CONCLUSION: The central corneal sesation decreased noticeable after LASIK, and it recovered three months postoperatively, but the indicators of dry eye syndrome did not return to their preoperative levels following it. We have found that the decreasation of corneal sensation had positive correlation with the laser ablation depth.
7.Relationship between Vascular Endothelial Growth Factor and Pool Prognosis in Nephroblastoma

Objective To study the connection between vascular endothelial growth factor(VEGF) and pool prognosis in nephroblastoma.Methods Serum VEGF was measured with immuneohistochemical(ELISA) methods in 35 children of nephroblastoma at preope-ration,1 week,2 weeks after surgery respectively. Simple visiting happened after surgery 4,8,16,32 weeks.The countrol group was 35 healthy children. Result Before surgery,the median VEGF in the children was (89.47?22.45) ?g/L.After surgery 1 week,2 weeks,levels in the children fell significantly to (2.75?0.31) ?g/L,(1.52?0.18) ?g/L.There was a significant difference compared with preoperation ( t =4.125 P
8.Opportunities and challenges in gene therapy of liver cancer

Gene therapy has emerged as an efficient modality to treat human diseases.This method is based on the transfer of genetic material to tissues to induce a curative effect.Gene therapy vectors are molecular constructs used to facilitate the penetration of genomic sequences inside the cells.Viral vectots have however several limitations when administered directly to the patient.They may cause significant toxicity by activating innate immunity or by eliciting an adaptive immune response against viral proteins.In addition,targeting the vector to the desired site is an issue when given systemically.The use of cells as vehicles for gene therapy vectors has many advantages.The combination of cell-viro-gene therapy has been thought as a new and promising strategy for therapy of cancer.The targeting vector to cancer stem cells will become a new direction in the field of gene therapy.In this article,we will introduce progressions,limitations and future directions of gene therapy of liver cancer.

10.Expression and Clinical Significance of PPARγ in Bladder Urothelial Cancer

Objective To investigate the expression and clinical significance of peroxisome proliferator activated receptor γ(PPARγ)in bladder urothelial cancer tissues.Methods Parafflin‐embeded specimen of bladder urothelial cancer tissues from 50 cases and normal tissues near the bladder urothelial cancer from 5 cases were harvested from the Pathology Department of the Renmin Hospital of Wuhan University between 2006 and 2009.Those cases had complete pathological and clinical data.The ex‐pression of PPARγ was detected by immunohistochemical SP method.Quantitative analysis of the PPARγ was measured by high definition pathological graphics context report system (HPIAS‐1000).One‐way analysis of variance and SNK (q)tests were used to analyze the mean density and the positive area rate of the immunohistochemical results.All data were processed by SPSS 13.0.Results The expression of PPARγwas significantly higher in bladder urothelial cancer tissues than in para‐carcinoma tis‐sues(P<0.05).Correlation between expression of PPARγ with TNM stag of bladder urothelial cancer was as follows :Positive rate of PPARγin the tissues with primary tumor size ≥3 cm was 72.4% ,significantly higher than 33.3% in the tissues with tumor size <3 cm(P<0.05);positive rate of PPARγin the cases with lymph node metastasis was 72.7% ,significantly higher than 46.4% in the cases without lymph node metastasis(P<0.05);positive rate of PPARγin patients at stage T3‐4 group was 75.0% ,significantly higher than 41.9% and 45.5% in patients at clinical stage T1 and T2(P<0.05);positive rate of PPARγin patients with poor differentiation was 68.2% ,significantly higher than 42.9% in patients with high or middle differentiation group(P<0.05).Conclusion PPARγ plays an important regulating role in the onset and progress of bladder urothelial cancer ;PPARγexpression level was correlated with primary tumor size ,pathological types and differentiation degree ,lymph node me‐tastasis and clinical stage.This result suggested that PPARγ was closely correlated to metastasis of bladder urothelial cancer.
