1.Neurobiological Mechanisms of Two Animal Models of Relapse

Relapse is the reinstatement of drug-seeking and drug-taking behaviors following a period of abstinence.It is one of the main characteristics of drug addiction and is also the major problem requiring immediate action for the treatment of drug addiction.In this review,in order to offer new ideas to eventually treat drug addiction,efforts are made to describe the establishment of two animal models of relapse-the drug self-administration (SA) and extinctionreinstatement procedure,and the drug-conditioned place preference(CPP) and extinction-reactivation procedure.Then attention is given to assess the criterion validity of the animal models of relapse and to explore the neurobiological mechanisms involved in relapse.
2.Restoration of skeletal muscle contusions by the transplantation of skeletal muscle satellite cells

Objective To probe the transplantation of skeletal muscle satellite cells of new-born rats for repairing skeletal muscle contusions. Methods Thirty-six Sprague-Dawley adult rats were separated at random into experimental and control groups (including a normal saline control group and a normal control group). Contusions were created on the left hind legs of rats in the experimental group. The cells were cultured and antibodies were identified through immunocytochemical staining and fluorescence labeling. In the experimental group, 0.2 ml of a muscle satellite cell suspension was slowly injected into the medial and lateral heads of the right calf gastrocnemius. The normal saline control group was injected with an equal volume of normal saline solution at the same sites. On the 5th, 10th and 15th day after the operation, EMG wave-amplitude and wave-width were recorded. The recovery rate of the muscle's wet weight and the gray value of a muscle fiber transverse section were determined.Two-factors analysis of variance was performed on the data. Results Before transplantation the muscle satellite cells were bright red after staining and their nuclei were blue after fluorescence labeling. After the operation the number of cells fluorescing and the strength of the fluorescence gradually decreased. The transplanted cells formed myotubes, melted into the muscle tissue and gradually took part in the restoration. The stained muscle satellite cell nuclei were free in intercellular space and melted into the basement membrane gradually. The outlines of the muscle tissue became progressively clearer. EMGs showed the fibrillation potentials as well as positive sharp waves reduced, but they too tended to become similar to those from the normal side. Restoration of EMG wave-amplitude and wave-width in the experimental group was better than in the saline control group and close to those in the normalcontrol group. The gray value of the muscle fiber transverse section area under HE staining was significantly lower at the 5th day post transplantation in both the experimental group and the saline control group than in the normal control group. At the 10th and 15th days post transplantation, the average gray value in the experimental group had increased significantly more than in the saline control group, and it was close to the normal control group average. In the experimental group the recovery rate of muscle wet weight was close to 1, but in the saline control group it was less than 1. Conclusions Transplanting skeletal muscle satellite cells of new-born rats can produce evident effects in renovating injured muscles.
3.Study of inflammatory indexes related to impaired vascular function in patients with impaired glucose tolerance

Objective To investigate the changes of inflammation levels in patients with impaired glucose tolerance(IGT) and the clinical significance.Methods A total of 30 IGT patients(IGT group) and 30 type 2 diabetes (T2DM) patients (T2DM group) were enrolled in current study.Meanwhile,30 health people were selected as control group (NGT group).The concentration of fasting plasma glucose (FPG),insulin resistance index (HOMA-IR),glycosylated hemoglobin (HbA1c),high-sensitivity C-reactive protein (hsCRP),cystatin C(Cys C) and fibrinogen(FIB) were measured.Results The levels of FPG,2 h glucose(2 h PG),HOMA-IR and HbA1c in three groups were significant (F =8.805,14.403,7.389,10.246 ; P < 0.05).The levels of CRP,Cys C and FIB in three group were also significant (F =6.766,4.733,11.683 ;P < 0.05).The levels of CRP,Cys C,FIB in IGT were (1.12 ±0.22) mg/L,(0.23 ±0.10) mg/L,(0.93 ±0.18) mg/L,higher than that in NGT group((0.70 ±0.16) mg/L,(6.02 ±0.49) g/L and (4.36 ±0.30) g/L).Meanwhile,the levels of CRP,Cys C and FIB in T2DM were significantly higher than of NGT(P < 0.05).Single factor correlation analysis showed that there was positive associationship between hs-CRP,Cys C and FIB with 2 h PG,HbA1 c and HOMA-IR(P < 0.05).Conclusion Endothelial dysfunction is seen in prediabetes stage,which plays an important role in the occurrence and development of vascular disease in impaired glucose regulation and diabetic patients.

Side population cells (SPs) are selected by the efflux features of fluorescent dye Hoechst33342.SPs have the characteristics of cancer stem cells,and play an extremely important role in the occurrence and development of tumors.At present,the SPs in common types of malignant tumor have been studied extensively at home and abroad.The study of SPs may shed some light on the diagnosis and treatment of cancer.
5.Functional change of neurotransmitters in central nervous system for outpatients with methadone maintenance treatment

Objective To detect neurotransmitters function in the brain and the brain functional index of outpatients with methadone maintenance treatment,and understand the change features of brain function.Methods Collect brain electrical power signals were collected from 58 outpatients with methadone maintenance treatment (MMT) and 44 healthy adults,and the correlations between the power of the neurotransmitters and the brain functional status were analyzed.Results (1) In MMT group,all neurotransmitters were lower generally than those in the control group,and the differences of GABA ((17.73±3.54) μV2 vs.(121.48±44.64) iμV2,P<0.01),Glu ((42.18±12.84) μV2 vs.(105.31 ± 34.95) μV2,P<0.05) were significant.(2) The relative powers of GABA ((17.10±51.72) μV2 vs.(78.67± 10.93) μV2,P<0.001) and Glu((30.48±21.61) μV2 vs.(69.23±42.26) μV2,P<0.001) reduced significantly,while 5-HT((297.18±31.54) μ V2 vs.(280.18±31.54) μV2,P<0.01),ACh ((235.08±37.72) μV2 vs.(217.23±40.60)μV2,P<0.05),NE((164.11±33.05) μV2 vs (146.39±30.80) μV2,P <0.01),DA((98.87±22.48) μV2 vs.(91.49±21.04)μV2,P<0.05) rised significantly in MMT group.(3)The global power was lower((1012.01± 195.09)μV2 Vs.(1775.94±458.99) μV2,P<0.01),and the excitement-inhibition index(2.19± 1.46 vs.0.99±0.47,P<0.001) and the relative entropy((89.45±9.71) % vs.(75.48±9.97) %,P <0.01)were higher than those in the controls.(4) In MMT group,there were positive correlation between all neurotransmitters and global power,as well as that between Glu and excitement-inhibition index (r=0.264,P<0.05),NE and Vasomotor index(r=0.269,P<0.05),but negative correlation between relative entropy and 5-HT(r=-0.276,P<0.05) as well as DA(r=-0.375,P<0.01)significantly.Conclusion The brain function of outpatients with MMT are lower than the normal significantly,and there are clearly imbalance between the excitement and inhibition among the brain neurotransmitters.
6.An analysis of HIV test results among pregnant women in Henan province in 2005-2007

Objective To understand the HIV infection status among pregnant women in Henan province and provide basis for preventing mother to child transmission of AIDS.Methods The HIV-test results obtained from January 2005 to May 2007 among pregnant woman in 31 counties(districts)with relatively high HIV prevalence were analyzed.Results A total of 720 605 pregnant woman were tested for HIV antibody,of whom 541 were identified HIV positive and the HIV-positive rate was 0.08%.Of the 364 HIV positive pregnant women 223(61.26%)decided voluntarily to terminate pregnancy,while 136(37.88%)chose to continue with their pregnancy,and the proportion of HIV positive pregnant women who continued with the pregnancy in the 3 different years was 31.51%(46/146),36.31%(57/157)and 54.10%(33/61),respectively,showing an yearly rising trend.Of the 284 HIV positive mothers 250(88.03%)received PMTCT intervention.Conclusion The rate of PMTCT in HIV-positive pregnant women can be further improved by increasing HIV testing among pregnant women and HIV positive women of child bearing age and by early identifying and managing them.
7.Determination of Plasma Concentration of Duloxetine by LC-MS

OBJECTIVE:To develop an LC-MS method for the determination of the concentration of duloxetine in human plasma.METHODS:Plasma sample(0.2 mL)was taken,with the protein precipitated twice using acetonitrile.The mobile phase consisted of 20 mmol?L-1 ammonium acetate(pH3.5)-methanol-acetonitrile(38∶20∶42).The sample was separated on a Thermo C18 column with plasma concentrations(time-concentration profile)of duloxetine in 10 healthy volunteers determined by LC-MS after taking a single oral dose of duloxetine enteric-capsule(22.4,44.8 or 67.2 mg).RESULTS:The calibration curve of duloxetine was linear over the concentration range of 0.78~100 ng?mL-1(r=0.999 5)with the recovery at 100.8%,the RSD of intra-day and inter-day at no more than 7.8% and 9.3%,respectively.CONCLUSION:The method developed in our study was proved to be accurate and sensitive,and suitable for the pharmacokinetic study of duloxetine.
