1.Anatomical and Histological Study of Anterolateral Ligament of Knee

Objective To observe the anatomical and histological features of anterolateral ligament (ALL)in the knee of Chinese adults,so as to identify the existence of ALL and provide an anatomical foundation for clinical reconstruction.Methods Ten adult knee specimens were randomly selected to be dissected,and the femoral,tibial and meniscus attachment points of the ALL were observed.The length,width and thickness were measured using the vernier caliper after the dissection.Three specimens were subjected to histological staining in the end.Results (1)ALL originated from the lateral femoral condyle—the same point of the lateral collateral ligament femoral side or the distal-anterior side,with its body divided into two branches,located in the tibia and the lateral meniscus respectively.The starting point of tibial side ALL was located at the mid-point of Gerdy's tubercle to fibula head,below tibial cartilage edge,with the meniscus point located in the lateral meniscus anterior horn and body junction area.(2) The average length of ALL is 38.89 ± 4.67 mm.The width in the femur,tibial attachment point was fan-shaped spread connected with sclerotin,being the narrowest at the joint line.The width at the femur,tibial attachment point and the joint line was 8.49 ± 1.36 mm,8.15 ± 1.38 mm and 6.49 ± 1.09 mm respectively,with the thickness of 1.33 ± 0.38 mm.The distance from tibia attachment points to the Gerdy's tubercle,fibular head and tibia cartilage margin was 22.59 ± 3.04 mm,21.15 ± 2.78 mm and 5.76 ± 0.57 mm respectively.(3) HE staining showed that ALL was dense connective tissue consisting of parallel arranged collagen fibers,while S-100 staining indicated that ALL contained sensory motor nerve fibers.Conclusion ALL is independent of the joint capsule and originates from the femoral lateral condyle.Its body is divided into two branches,located in the tibia and the lateral meniscus respectively.
2.Morphological changes of enteric deep muscular plexuses interstitial cells of Cajal in rats with multiple organ disfunction syndrome

Objective: To observe the morphological changes of enteric deep muscular plexuses interstitial cells of Cajal (ICC-DMP) in rats with multiple organ dysfunction syndrome (MODS). Methods: Forty Wistar rats were randomly divided into control group and MODS model group. The enteric ICC-DMP network was observed using c-kit immunohistochemical staining with whole-mount preparation technique and confocal laser scanning microscopy , and the ultraslructural features of ICC-DMP was evaluated using transmission electron microscope. Results: Compared with those in control group, the distributions and densities of intestine ICC-DMP in MODS group were significantly decreased (P < 0. 05) , the ICC-DMP network was disrupted and the ultrastructural features of ICC-DMP were severely damaged. Conclusion: The ICC-DMP network was severely damaged in rats with MODS, and the mechanism of gastrointestinal dysmotility in MODS may be related to the morphological changes of ICC-DMP.
3.Construction of a rabbit model of knee prosthesis infection:environmental factorsin vivo affect Staphylococcus epidermidis and biofilms

BACKGROUND:Bacterial biofilm is the main cause of the infection of the prosthesis.In vitro experiments confirmed that hypertonic sodium chloride and ethanol can apparently promote the formation of staphylococcal biofilms. There are no reports on the effects of ethanol and hypertonic environment surrounding the prosthesis on the formation of biofilms.
OBJECTIVE: To evaluate the effects of different environment factors surrounding the prosthesis on the growth of Staphylococcus epidermidis and bacterial biofilm formation after replacement.
METHODS: White rabbit models infected with Staphylococcus epidermidis on the prosthesis were established, and were randomly divided into hypertonic sodium chloride, ethanol and control groups (n=15). The bacteria were injected with 0.1 mL 4% sodium chloride and 4% ethanol into the knee of rabbits in the hypertonic sodium chloride and ethanol groups. The rabbits were injected with 0.1 mL 0.9% sodium chloride in the control group. Three rabbits were sacrificed at 2, 4, 6, 8 and 16 days after inoculated with bacteria. Synovial fluid, prosthesis and tissue surrounding infection were obtained. Bacterium was cultured to extract total RNA. The ica operon transcription levels were detected in the gene levels. Adhesion of bacteria on the surface of the prosthesis was observed using a scanning electron microscope. Tissues surrounding the prosthesis were observed using hematoxylin-eosin staining.
RESULTS AND CONCLUSION:Histological examination revealed that inflammatory cel infiltration was observed in al the rabbits at 4 days after injection. Colony formation was found at 16 days after injection. At 6 days after injection, inflammatory cel infiltration was observed in the ethanol and control groups. Scanning electron microscope showed that compared with the control group, the bacteria adhered to the prosthetic surface became more in the hypertonic sodium chloride and ethanol groups at 6, 8 and 16 days (P < 0.05). At 6, 8 and 16 days, the expression of icaA mRNA was significantly higher in the hypertonic sodium chloride and ethanol groups than in the control group (P < 0.05). These data showed that the environment factors could affect the growth of Staphylococcus epidermidis and bacterial biofilm formation.
4.Effect of allitridum on remodeling of the transient outward potassium current of ventricular myocytes of spontaneously hypertensive rats.

We aimed to study the effect of allitridum (All) on the transient outward potassium current (Ito) of ventricular myocytes of spontaneously hypertensive rats (SHR). Totally 30 male SHRs were randomly divided into three groups: low-dose All group (7.5 mg·kg(-1)), high-dose All group (15.0 mg·kg(-1)) and normal saline group. The other 10 sex and age matched Wistar-kyoto rats (WKY) were also taken as control group (WKY group). All animals received i.p. administration for 8 weeks. The dual enzymatic method was used to separate single ventricular myocyte from animals. Patch-clamp technique was used to record Ito and analyze the effect of All on the current. It was shown that the left ventricular hypertrophy of SHR was reversed significantly by All. Furthermore, the density of Ito was recovered in both high and low dose All groups. The peak current densities of Ito were enhanced from 18.23±3.64 to 25.17±2.86 pA/pF (P<0.01) and 36.47±5.42 pA/pF (P<0.01) at +50 mV by All 7.5 mg·kg(-1) and 15.0 mg·kg(-1), respectively, which was not significantly different with WKY group. The effect was associated with positive shift of the steady-state, close-state inactivation, and shortened recovery from inactivation of Ito. It is concluded that All decreases the remodeling of Ito of ventricular hypertrophic myocytes of SHR.

5.Effects of high plasma triglyceride caused by ApoC Ⅲ transgene on ab-dominal aortic aneurysm induced by elastase in LDLR-/-mice

AIM:To investigate the effects of high plasma triglyceride (TG) caused by apolipoprotein C Ⅲ( ApoC Ⅲ) transgene on the occurrence and development of abdominal aortic aneurysm ( AAA) .METHODS:The animal models of hypercholesterolemia and hypercholesterolemia combined with hypertriglyceridemia were established by feeding high-fat diet to LDLR-/-and ApoC Ⅲ+LDLR-/-mice, respectively.AAA was induced in these mice by pancreatic elastase. By evaluating the incidence of AAA, relative maximal abdominal aortic diameter, disruption of the elastic lamellar structure and expression of matrix metalloproteinases ( MMPs) in the aorta walls of the AAA, the occurrence and development of AAA were compared in LDLR-/-and ApoC Ⅲ+LDLR-/-mice fed with either chow diet or high-fat diet.In addition, an in vitro TNF-α-induced aneurysmal microenvironment model on vascular smooth muscle cells ( VSMC) was used to study the impact of triglyceride-rich lipoproteins ( TRLs) from mice with normal or high contents of ApoCⅢon elastin protein expres-sion.RESULTS:Feeding the high-fat diet aggravated the severity of AAA in the LDLR-/-mice.ApoC Ⅲ+LDLR-/-mice fed with high-fat diet had less severe AAA than LDLR-/-mice fed with high-fat diet.TRLs inhibited degradation of VSMC elas-tin protein induced by TNF-α, and in vitro TRLs from the mice with high content of ApoC Ⅲ, compared to those with nor-mal content of ApoC Ⅲ, had better inhibitory effect on the degradation of elastin.CONCLUSION:High plasma TG caused by ApoC Ⅲtransgene alleviates AAA of the LDLR-/-mice induced by elastase and high-fat diet.The effect is probably attrib-uted to the hypertriglyceridemia caused by ApoC Ⅲtransgene.
6.Multiwalled carbon nanotubes improve the morphology of the femoral head of a rabbit model of steroid-induced necrosis of femoral head

BACKGROUND: Multiwaled carbon nanotubes can accelerate the proliferation and differentiation of osteoblasts, and exert a therapeutic effect on steroid-induced necrosis of femoral head (SNFH). OBJECTIVE:To investigate the function of multiwaled carbon nanotubes in the establishment of a rabbit model of SNFH. METHODS:Thirty-six New Zealand white rabbits were divided randomly into three groups. In treatment group, 16 rabbits were given intraglutealy injection of dexamethasone (2.5 mg/kg) every day and injection of 1 mL liquor of multiwaled carbon nanotubes (0.1 g/L) into the bilateral femur medulary space every week. In model group, 16 rabbits were given intraglutealy injection of dexamethasone (2.5 mg/kg) every day and injection of 1 mL normal saline into the bilateral femur medulary space every week. In control group, four rabbits were given intraglutealy injection of 2 mL normal saline every day and injection of 1 mL normal saline into the bilateral femur medulary space every week. RESULTS AND CONCLUSION: Four weeks after hormone injection, the trabeculae began to exhibit a smal amount of thinner fractures, an accumulation of fatty tissue in the bone marrow were obvious, bone marrow fat cels became bigger and microvascular thrombosis appeared in the model group, while there was no positive histopathological manifestation in the treatment group. This indicates that the multiwaled carbon nanotubes can extenuate pathological damage to the femoral head to a certain extent.
7.Effects of different ratios of medicine dosage for sevoflurane and propofol on postoperative cognitive function in aged rats

Objective To evaluate the effects of different ratios of medicine dosage for sevoflurane and propofol on postoperative cognitive function in aged rats.Methods Sixty healthy male Wistar rats,aged 18-20 months,weighing 600-650 g,were divided into 6 groups (n =10 each) using a random number table:control group (group C),sevoflurane anesthesia group (group S),propofol anesthesia group (group P) and different ratios of mmedicine dosage for sevoflurane and propofol groups (group SP1,group SP2 and group SP3).Normal saline 10 ml was infused intravenously in group C.Group S inhaled 3.1% sevoflurane.Propofol was infused intravenously at a rate of 40 mg · kg-1 · h-1 in group P.In group SP1,2.4% sevoflurane was inhaled,and propofol was intravenously infused at a rate of 10 mg · kg-1 · h-1.In group SP2,1.8% sevoflurane was inhaled,and propofol was intravenously infused at a rate of 20 mng · kg-1 · h-1.In group SP3,1.2% sevoflurane was inhaled,and propofol was intravenously infused at a rate of 30 mg · kg-1 · h-1.Duration of anesthesia was 2 h in all the groups.After loss of righting reflex,open reduction and internal fixation was performed after tibial fracture was induced in the other five groups except group C.At 1 day after anesthesia,Y-maze test was performed,and the time spent on the novel arm and total number of entries into each arm were recorded,and fear conditioning test was carried out for determination of the rate of freezing time to reflect the cognitive function.The rats were sacrificed after behavioral testing,and the hippocampus was removed for determination of occludin,matrix metalloproteinase-2 (MMP-2) and MMP-9 expression by Western blot.Results Compared with group C,the time spent on the novel arm was significantly shortened,the total number of entries into each arm and rate of freezing time were decreased,the expression of occludin was down-regulated,the expression of MMP-2 and MMP-9 was up-regulated in the other five groups,and the most unmarked change in the indices mentioned above was found in group SP2 (P< 0.05 or 0.01).Conclusion Different ratios of medicine dosage for sevoflurane and propofol can induce postoperative cognitive decline,and inhaling 1.8% sevoflurane and infusing propofol 20 mg · kg-1 · h-1 produce little influence on postoperative cognitive function in aged rats.
8.Study on the thickness-change of different thickness thermoplastic materials after thermoforming and saliva immersion.

OBJECTIVETo survey and compare the thickness-change of different thickness thermoplastic materials under different test condition and make sure the relationship between the thickness-change and the material initial thickness in order to provide a guide in selecting the suitable thickness thermoplastic in practice.
METHODSTo choose Biolon, the thickness include 1.0 mm, 0.75 mm, 0.5 mm. Used Electron Vernier caliper to measure the thickness-change of different thickness thermoplastic materials under different processing mode. The data was analyzed by SPSS 10.0.
RESULTSAfter thermoforming the thickness of thermoplastic became thinner, the thickness of Biolon 0.75 mm decreased by 0.14 mm, Biolon 1.0 mm decreased by 0.22 mm and Biolon 0.5 mm decreased by 0.14 mm. After saliva immersion the thickness became thicker. The thickness of Biolon 0.75 mm increased by 0.02 mm, Biolon 1.0 mm increased by 0.03 mm and Biolon 0.5 mm increased by 0.02 mm.
CONCLUSION1)The influence of different processing mode to the thickness-change had relation to the material initial thickness. 2)The Biolon 0.75 mm had certain superiority in thickness stability compared to the homogeneous brand through the above research.

9.Short-term follow-up study of Cercon all-ceramic crowns and bridges.

OBJECTIVECercon all-ceramic system was applied clinically for the first time to make all-ceramic crowns and bridges for the patients. The objective of this study was to evaluate the short-term follow-up results of this new system from both technical and clinical aspects.
METHODS15 units of Cercon all-ceramic crowns and bridges were fabricated and applied to 5 patients, all the cases were followed at 1, 3, 6 and 12 months' periods after cementation of the restorations.
RESULTSCercon all-ceramic system demonstrate natural and esthetical appearance. No fracture, broken and color change were found.
CONCLUSIONThe reliability of all-ceramic crowns and bridges made from Cercon system was greatly improved due to the strengthening effects of the zirconia copings and substructures, and the indication of this system can be extended to long span posterior bridges. In addition, the better esthetics compared to PFM technique makes it a comparatively ideal all-ceramic system for crowns and bridges.

10.Construction of acid-sensitive potassium channel-3 eukaryotic expression plasmid and its express in SH-SY5Y cells.

OBJECTIVETo construct the acid-sensitive potassium hannel-3(TASK3) eukaryotic expression plasmid and to establish a stable SH-SY5Y cell line expressing enhanced green fluorescent protein (EGFP)-tagged TASK3.
METHODSTASK3 coding region was subcloned into pEGFP-N1 plasmid to construct a recombinant vector alled pEGFP-TASK3. The correct recombinant expressing plasmid was transfected with X-feet transfection reagent to SH-SY5Y cells. The cell line stably expressiing EGFP tagged-TASK3 gene was established by screening with antibiotic G418 and fluorescence microscope. The expression and localization of the EGFP tagged-TASK3 fusion protein was detected by Western blot and confocal microscope. Exposure of the SH-SY5Y cell line expressing stably TASK3-eGFP fusion proteins was exposed to different pH media (7.0, 6.7, 6.4, 6.1) for 24 h, the cell viability was assessed with cell counting Kit-8 (CCK-8).
RESULTSAll the results of identification by PCR, digestion with restriction endonuclease and sequencing indicated that the recombinant eukaryotic expression plasmid pEGFP-TASK3 was constructed correctly. The stable SH-SY5Y cell line expressing EGFP tagged-TASK3 fusion protein was successfully established. Exposure of the wild type SH-SY5Y cells and the stable SH-SY5Y-GFP tag-TASK3 cell line to different pH media (7.0, 6.7, 6.4, 6.1) for 24 h, the cell viability of two group cells significantly reduced with pH declining, and the difference was statistically significant (P < 0.05). Compared with wild type SH-SY5Y cells, the cell viability of stable SH-SYSY-GFP tag-TASK3 cell line increased significantly with the same pH media, and the difference was statistically significant (P < 0.05).
CONCLUSIONThe eukaryotic expression vector pEGFP-TASK3 is successfully constructed and the cell line stably expressing TASK3-eGFP fusion is established which is important for their fundamental research and potential applications.
