1.Optimization of Extraction Process for Sanren Tongbian Mixture by Orthogonal Test

Objective:To optimize the extraction technology for Sanren Tongbian mixture. Methods:The water amount,decoction time and decoction frequency as the evaluation factors,and the content of amygdalin and the paste-forming rate as the evaluation indi-ces,the extraction process of Sanren Tongbian mixture was optimized by L9 (34 ) orthogonal design. Results: The optimal extraction conditions were as follows:adding 8-fold amount of water, decocting twice with 1. 5h for each time. Conclusion: The study provides scientific basis for the research of the extraction technology of Sanren Tongbian mixture, and the further experiments proved that the op-timized process is stable and feasible.
2.Separation and Preliminary Identification of Spoilage Organisms in Transmutative Soy Milk

In this paper, three spoilage organisms were separated from five transmutative soy milks, and all the three spoilage bacteria could survive condition of both 1?105Pa,30min and 300mg/kg Nisin. Morpha character, physiological and biochemical characteristics, and a phylogenetic analysis based on 16S rDNA gene sequences reveal that these three strains are Bacillus licheniformis, Bacillus pumilus and Brevibacillus borstelensis respectively. GenBank accessions for these three strains are EF439666-EF439668。
3.Reproductive Outcomes in Women with Prior Cesarean Section Undergoing In Vitro Fertilization: A Retrospective Case-control Study

The impact of prior cesarean section (CS) on the pregnancy and neonatal outcomes of in vitro fertilization and embryo transfer (IVF-ET) was investigated.A retrospective analysis was performed on 144 patients with prior CS between January 2013 and December 2015.The pregnancy,delivery,and neonatal outcomes of patients who had previous CS delivery and received IVF-ET were analyzed.The control group comprised 166 patients who had only previous vaginal delivery (VD) and received IVF-ET during the same period.The results showed that the basal follicle stimulating hormone level,estradiol level on human chorionic gonadotropin (hCG) day,gonadotrophin dosage,duration of stimulation,retrieved oocytes,fertilization rate,high-quality embryo rate,multiple birth rate,abortion rate and ectopic pregnancy rate had no significant difference between the two groups (P>0.05).The pregnancy rate (40.28% vs.54.22%) and implantation rate (24.01% vs.34.67%) were significantly lower (P<0.05),and the ratio of embryo difficulty transfer (9/144 vs.0/166) was significantly higher in CS group than in VD group.The risk of pernicious placenta previa and postpartum hemorrhage in twin deliveries was significantly increased in CS group as compared with that in VD group (P<0.05),and gestational age and neonatal birth weight were significantly reduced in twin deliveries as compared with singleton deliveries in both groups (P<0.05).It was suggested that the existence of CS scar may impact embryo implantation and clinical pregnancy outcome,and increase the difficulty of ET.We should limit the number of transfer embryos to avoid multiple pregnancies and strengthen gestational supervision in patients with cesarean scar.
4.Clinical application of anterolateral femoral skin flap pedicled with high site cutaneous branches of the transverse branch of lateral circumflex femoral artery

Objective To report the clinical effect of free transplanting for soft tissue defects pedieled with high site cutaneous branches of the transverse branch of lateral cirumflex femoral artery.Methods Cu- taneous branches of the descending branch of lateral circumflex femoral artery were found small or abscent in 7 patients.The anterolateral femoral skin flap was pedicled with high site cutaneous branches of the transverse branch to repair the soft tissue defects of the arm,hand,leg and foot,rather than with the descending ones. The size of the flap ranged from 15 cm?6 cm to 28 cm?13 cm,with part muscle valve,iliotibal tract and lat- eral femoral cutaneous nerve.The fractures were performed with internal or external fixation.Results All of the anterolateral femoral skin flap survived well postoperatively in the 7 cases and had good appearance and sensation at one stage.The function of the repaired extremities recovered well.Conclusion The anterolat- eral femoral skin flap pedicled with high site cutaneous branches of the transuvrse branch of lateral circumflex femoral artery has many advantages of good blood supply and large size.The flap was secluding,and can be taken with some muscle and lateral femoral cutaneous nerve.When cutaneous branches of the descending branch of lateral circumflex femoral artery is small or abscent,the anterolateral femoral skin flap with high site cutaneous branches of the transverse branch of lateral circumflex femoral artery is an optimal alternative.
5.Long non-coding RNA TTTY15 expression in osteosarcoma and its effect on viability and invasion ability of osteosarcoma cells

AIM:To observe the expression of long noncoding RNA TTTY 15 in osteosarcoma tissues and cell lines and to explore its effect on the viability and invasion ability of osteosarcoma cell lines.METHODS:qPCR was used to detect the expression of TTTY15 in 11 cases of osteosarcoma and its adjacent tissues.The mRNA levels of TTTY15 in os-teosarcoma cell lines(143B,Saos2,MG-63,U2OS and HOS)and human osteoblast cell line hFOB1.19 were also tested. TTTY15 was down-regulated after transfected with small interfering RNA in MG-63 cells,the cell line with the highest level of TTTY15.The effect of TTTY15 knockdown on the viability of MG-63 cells was measured by CCK-8 assay.The cell cycle distribution was analyzed by flow cytometry.The effect of TTTY15 knockdown on the cell invasion ability was detected by Transwell assay.The levels of miR-216b-5p and FOXM1 mRNA were detected by qPCR, and the changes of the related proteins were determined by Western blot.RESULTS:Compared with the adjacent tissues,the expression of TTTY15 in-creased in the osteosarcoma tissues(P<0.01).Compared with the human osteoblast cell line,the expression of TTTY15 increased in the osteosarcoma cell lines(P<0.05), and the level of TTTY15 in the MG-63 cells was the highest(P<0.01).After knockdown of TTTY15 expression in the MG-63 cells,the cell viability was decreased(P<0.05),cell cycle progression was inhibited(P<0.01), and the cell invasion ability was decreased(P<0.01).The expression of miR-216b-5p was increased(P<0.01)and the expression of FOXM1 mRNA was decreased(P<0.01).The protein expres-sion of FOXM1,CDK4,cyclin D1,MMP-2 and N-cadherin was decreased,while the protein expression of E-cadherin was increased(P <0.05).CONCLUSION: The expression of TTTY15 is increased in the osteosarcoma tissues and cell lines.The low expression of TTTY15 inhibits the cell viability and invasion ability of osteosarcoma cells.The possible mechanism is that the knockdown of TTTY 15 expression results in the increase in miR-216b-5p expression and the down-regulation of FOXM1 expression.

Objective To observe the effects of 75 gram glucose oral tolerance test (75 g OGTT) and standard mixed meal test (SMMT) on insulin secretion function of the islets of Langerhans and plasma free fatty acid (FFA) in patients with type 2 diabetes mellitus.Methods Seventy-six patients with type 2 diabetes without using insulin and with no obvious complications were recruited for 75 g OGTT following overnight fasting on the first day and SMMT (bread 50 g,egg 50 g and milk 250 ml) on the 7th day.Blood specimens were collected from each patients before the tests and 30 min,60 min,120 min and 180 min after glucose or meal load to measure their levels of plasma glucose,serum insulin,C peptide,FFA and lipids (total cholesterol,triglyceride,high-density and low-density lipoprotein cholesterol).Results No difference in fasting plasma glucose,serum insulin,C peptide,FFA and lipids between 75 g OGTT and SMMT was found.Postprandial plasma glucose 30 min,60 min,120 min and 180 min after 75 g OGTT was significantly higher than that after SMMT,with (15.3?3.5) vs (9.9?3.4) mmol/L,(18.2?4.8) vs (12.8?4.0) mmol/L,(16.3?5.8) vs (12.2?4.9) mmol/L and (10.6?5.4) vs (9.5?4.5) mmol/L (F=28.1,P
7.Effects of microwave on the contents of MDA and SOD in renal cortex and testis of mice

Objective: To observe the effects of microwave on the activity of SOD and the contents of MDA in renal cortex and testis of mice. Methods: Microwave generator(2 450 MHz, 10 mW/cm2) was used to expose mice; NBT,DTNB and TBA were used to mearure the activity of SOD and the contents of MDA in renal cortex and testis of the mice after microwave exposure.Results: The content of MDA in renal cortex and testis of the mice increased progressively on days 1,6,12 and reached the highest level on day 24 after the microwave exposure (P<0.01). The activity of SOD in renal cortex and testis of the mice decreased progressively on days 1, 6, 12 and reached the lowest level on day 24 after the microwave exposure (P<0.01). Conclusions: Microwave exposure can produce reactive oxygen free radicals and lead to depress SOD activity.

The genus Nodulisporium, is known to produce secondary metabolites with structural diversity. A new alkaloid, 2-hy- droxy-1,1-dimethyl-1,2,3,9-tetrahydro-4H-carbazol-4-one(1), was isolated from the extract of a fungal strain Nodulisporium sp. fermented with rice, together with three known phenols, tyrosol(2), hydroxytyrosol(3), and hydroxytyrosol acetate(4). Their structures were identified by detailed spectroscopic analyses.

9.A prospective observation for the patients with type 2 diabetes mellitus complicated with stroke

Objective To learn the stroke occurrence status and the influencing factors of the patients with type 2 diabetes,and to provide the basis for the prevention of stroke.Methods Patients with type 2 diabetes which were reported in 2011 year from the system of chronic disease monitoring information management of Zhejiang Province were selected and followed -up until October in 2014.Stroke occurrence were recorded and the influencing factors were analyzed.Results The incidence rate was 5.22% (59/1 129),and the annual incidence was 1.56%.Fifty six cases (94.92%) were ischemic stroke.Logistic regression analysis showed that older than 65 and hypertension were risk factors for type 2 diabetes complicated with stroke.The group of 65-and 75-were associated with higher risk of stroke than 45-(OR =3.38,95% CI:1.39-8.24;OR =7.77,2.14-28.24).Hypertensionin were the influencing factors of stroke among patients with type 2 diabetes (OR =10.92,95% CI:5.94-20.09).Conclusion Older age and hypertension were risk factors for type 2 diabetes complicated with stroke.Therefore effective control blood pressure among older patients with type 2 diabetes should be strengthed to prevent stroke.
10.Contamination status and molecular biological characteristics of Legionella in cooling water samples from different places in Wuxi city.

OBJECTIVETo investigate the contamination state of Legionella in cooling water samples from different places in Wuxi city and reveal the molecular biological characteristics of Legionella strains.
METHODS112 parallel water samples (500 ml each) were collected from 56 sites in Wuxi city during year 2009 - 2010. The samples were used for Legionella test and quantitative culture. The isolated Legionella strains were used for serotyping, pulsed-field gel electrophoresis (PFGE), sequence-based typing (SBT), and intracellular growth were tested.
RESULTSThe positive proportion of Legionella was 39. 3% (22/56) among all sampling sites. A total of 29 Legionella strains were isolated, and the serotypes include LP1, LP3, LP5 and LP6. LP1 serotype was the major one with a proportion of 65.5% (19/29). 29 Legionella strains got 17 PFGE types. There were 10 SBT types among 10 Legionella strains with different PFGE types. Comparing to LP1 strain (ATCC 33152), WX2011062 (LP6) and WX2011067 (LP5) had strong intracellular growth ability in mouse peritoneal macrophages J774 cell line (the amount of intracellular bacteria on day 0 after infection were (5.5 +/- 1.32) x 10(5), (3.9 +/- 0.60) x 10(5), (7.8 +/- 0.76) x 10(5) CFU/ml, respectively; the amount of intracellular bacteria on day 3 after infection were (58.3 +/- 1.61) x 10(5), (2700.0 +/- 655.74) x 10(5), (3066.7 +/- 208.17) x 10(5) CFU/ml, respectively).
CONCLUSIONThe Legionella contamination existed in cooling water samples from different places in Wuxi city. Legionella strains isolated showed high genetic variation. Some Legionella strains had vigorous intracellular growth ability.
