2.Studies on the expression of TNF alpha, IL-1 beta, IL-6 and IL-4, IL-10, IL-13 mRNA in lung tissue of rat with acute lung injury induced by oleic acid and lipopolysaccharide

Objective To study the role of pro inflammatory cytokines tumor necrosis factor?(TNF ?),interleukin 1beta(IL 1?),interleukin 6(IL 6) and anti inflammatory cytokines interleukin 4(IL 4),interleukin 10(IL 10),interleukin 13(IL 13) in acute lung injury(ALI). Methods Wistar Rats (200?50g) were randomly divided into the oleic acid(OA) and Lipoplysaccharide(LPS) induced lung injury experimental group and the only OA induced lung injury control group. The LPS(2mg/kg) was injected in rat's tail vein at the 1, 4, 12 and 24 hours after administration of OA(0.2ml/kg, iv). Reverse transcription polymerase chain reaction(RT PCR) were used to detecting the expression of TNF ?, IL 1?, IL 6 and IL 4, IL 10, IL 13 mRNA in the lung tissue of rats in two groups. Results The results showed that the peak expression of TNF ? and IL 1? mRNA at one hour after first hit by oleic acid, and IL 6 mRNA at four hours were observed. Others, such as IL 4, IL 10 and IL 13 mRNA, the highest levels in lung tissue at four hours after lung injury induced by oleic acid. Conclusions The results suggested that TNF ? and IL 1? are presenting early phase of ALI, and that IL 6 played its important role in delayed development. The over expression of anti inflammatory cytokines IL 4, IL 10 and IL 13 may be as a promoters rather than protectors in inflammatory amplification.
3.Effect of Ankle States on Peak Torque of Knee Flexion and Extension for Old Adults

Objective To explore the effect of state of the ankle on peak torque of knee flexors and extensors. Methods 18 adults in community over 65 years old were included. Their peak torque with knee flexion and extension was measured under 4 kinds of action of ankle. Results There was significant difference in peak torque neither flexion nor extension among 4 kinds of action of ankle (P>0.05). Conclusion It is unimportant of the state of ankle for knee training.
4.A primary study of evolution of hepatitis B virus based on motif discovery.

Hepatitis B is a serious infectious disease worldwide, and hepatitis B virus (HBV) is the direct cause of this disease. In recent years, as an essential part of its evolutionary process, HBV mutation has been extensively studied domestically and globally. However, the study on the conserved sequences in HBV sequences is still in its infancy. In this study, we applied multiple EM for motif elicitation (MEME) algorithm to discover HBV motif and proposed a new metric, conservative index (CI), to carry out phylogenetic analysis based on HBV sequences. Then, the constructed phylogenetic tree was subjected to reliability assessment. The results demonstrated that the new metric CI combined with the MEME algorithm can effectively help to discover motifs in HBV sequences and construct a phylogenetic tree based on them and to analyze the evolutionary relationship between HBV sequences; in addition, the possible ancestral sequences of samples may be obtained by conservative analysis. The proposed method is valuable for the exploratory study on large HBV sequence data sets.

5.Effect of hydromorphone postconditioning on myocardial ischemia-reperfusion injury in isolated rat hearts and the role of mitochondrial permeability transition pore

Objective To investigate the effect of hydromorphone postconditioning on ischemiareperfusion (I/R) injury in isolated rat hearts and the role of mitochondial permeability transition pore (mPTP).Methods Male Sprague-Dawley rats, aged 2-3 months, weighing 250-320 g, were used in the study.The rats were heparinized and anesthetized with intraperitoneal 10% chloral hydrate 350 mg/kg.The hearts were excised, and perfused in a Langendorff apparatus with K-H solution saturated with 95% O2-5% CO2 at 36.5-37.5 ℃.Forty isolated rat hearts were randomly divided into 4 groups (n=10 each)using a random number table: control group (group C), group I/R, hydromorphone postconditioning group (group H), and hydromorphone postconditioning + mPTP opener lonidamine group (group HL).Group C was continuously perfused with K-H solution for 120 min.Group I/R was perfused with K-H solution for 30 min, the perfusion was then suspended for 30 min, and group I/R was perfused with K-H solution for another 30 min.Group H was perfused with K-H solution for 30 min, the perfusion was then suspended for 30 min, and group H was perfused with K-H solution containing 0.3 μmol/L hydromorphone for 10 min, and then with K-H solution for 50 min.Group HL was perfused with K-H solution for 30 min, the perfusion was then suspended for 30 min, and group HL was perfused with K-H solution containing 0.3 μmol/L hydromorphone and 30 μmol/L lonidamine for 10 min, and then with K-H solution for 50 min.At 30 min of equilibration (T0), and 30 and 60 min of reperfusion (T2,3), left ventricular systolic pressure (LVSP), left ventricular end-diastolic pressure (LVEDP), left ventricular developed pressure (LVDP) , ±dp/dtmax, heart rate (HR), and coronary flow (CF) were measured.The concentrations of lactic dehydrogenase (LDH), creatine kinase-MB (CK-MB) , and troponin-T (Tn-T) in the coronary effluent were determined at T0 and T3.The coronary effluent was collected at T0 and 15 min of reperfusion (T1),nicotinamide-adenine dinucleotide (NAD+) concentrations were measured using enzyme-linked immunosorbent assay to reflect the degree of mPTP opening.The myocardial infarct size was determined at T3 by TTC staining.Results Compared with group C, LVDP, HR, ±dp/dtmax and CF were significantly decreased, and LVEDP was increased at T2,3, and the concentrations of LDH, CK-MB and Tn-T in the coronary effluent, myocardial infarct size at T3, and NAD+ concentrations in the coronary effluent at T1 were increased in group I/R (P<0.05).Compared with I/R and HL groups, LVDP, ±dp/dt CF and HR were significantly increased, and LVEDP was decreased at T2,3, and the concentrations of LDH, CK-MB and Tn-T in the coronary effluent, myocardial infarct size at T3, and NAD+ concentrations in the coronary effluent at T1 were decreased in group H (P<0.05).Conclusion Hydromorphone postconditioning can reduce myocardial I/R injury in isolated rat hearts, and the mechanism is related to inhibition of mPTP opening.
6.Efficacy of hydromorphone for patient-controlled intravenous analgesia after hip replacement in elderly patients

Objective To evaluate the efficacy of hydromorphone for patient-controlled intravenous analgesia (PCIA) after hip replacement in elderly patients.Methods Seventy patients, aged 65-75 yr, weighing 40-70 kg, of American Society of Anesthesiologists physical status Ⅰ or Ⅱ , undergoing elective unilateral hip replacement, were randomly divided into either analgesia with hydromorphone group (group H, n =35) or analgesia with fentanyl group (group F, n=35).After a loading dose of 20 μg/kg of hydromorphone was injected at the beginning of skin closure, a PCA pump was connected, and the PCIA solution contained hydromorphone 0.25 mg/kg and tropisetron 10 mg in 120 ml of normal saline in group H.After a loading dose of 1 μg/kg of hydromorphone was injected at the beginning of skin closure, a PCA pump was connected, and the PCIA solution contained fentanyl 25 μg/kg and tropisetron 10 mg in 120 ml of normal saline in group F.The PCA pump was set up with a 0.5 ml bolus dose, a 15 min lockout interval and the background infusion at a rate of 2 ml/h in both groups.Tramadol 0.5 mg/kg was injects intravenously as rescue analgesic, and visual analogue scale score was maintained ≤ 3.The Ramsay sedation score,the number of attempts and the number of tramadol administration were recorded at 24 and 48 h after operation.The adverse effects within 48 h after operation and patient's satisfaction with analgesia were recorded.Results There was no significant difference in the Ramsay sedation score, the number of attempts and the number of Tramadol administration between H and F groups.Compared with group F, the incidence of adverse effects such as postoperative nausea, vomiting, respiratory depression, drowsiness, urinary retention, was significantly decreased, and the level of satisfaction with analgesia was increased in group H.Conclusion Hydromorphone provides accurate efficacy for PCIA after hip replacement in elderly patients, with fewer adverse effects and higher level of patient' s satisfaction.
7.Clinical research of Wanyao-Tingli manipulation in the treatment of instability of lumbosacral joint causedto low back pain and activity

Objective To observe the effect of Wanyao-Tingli manipulation in the treatment of instability of lumbosacral jioint causedto low back pain and activity. Methods From March 2009 to May 2012, 130 patients with Instability of Lumbosacral Joint were recruited and randomly divided into atreatment group(63 cases)and acontrol group(67 cases). The treatment group was given Wanyao-Tingli Manipulation, and the control group was received Votalin Futalin Cream. The therapeutic effect was observed in both groups after continuous treatment for 7 days, different time record of low back pain score and activity was observed after treatment at 1,3,5,7 days of metrics. Results After1, 2, 3, 4 times treatment, the VAS score of and treatment group VAS score, the efficacy of lumbar flexion activity of treatment group were significantly better than the control group (t=71.619, 57.166, 56.329, 56.632;72.579, 55.330, 38.418, 25.889, P<0.01);After 1, 2 times treatment, the treatment group lumbar extension activity and flexor activity of treatment group comparison with the control group significantly improved compare with the control group(t=26.372, 22.092, 13.418, 16.373, P<0.01). Conclusion Wanyao-Tingli Manipulation can significantly relieve improve low back pain and improve activity in the treatreatment ofing instability of lumbosacral joint with immediate significant effect.
8.Comparison of cognition and attitude towards the elderly family nursing among college students and nurses

Objective To investigate the cognition and attitude towards the elderly family nursing among nurses and college students in Chengde, and analyze its influencing factors. Methods A total of 1000 college students and nurses in Chengde were investigated by self-designed questionnaire about their cognition and attitude to elderly family nursing. Results Nurses' cognition and attitude towards the elderly family nursing was better than nursing college students, medical students were better than non-medical students; and there was no statistical significance about their expectation for government's senior citizen service among different groups. Conclusions More attention should be paid to family members' attitudetowards the elderly home care, and should strengthen the elderly family care through various way.

Objective To observe the protective effect and mechanism of 2,3,5,4 '-tetrahydroxystilbene-2-O-beta-D-glucoside ( THSG) on atherosclerosis in ApoE konck-out mice. Methods A total of 24 ApoE knock-out mice were randomly divided into normal control group (n=8), model control group (HFD, high-fat diet, n=8) and treated group (THSG, 20 mg· kg-1, i. g. , n=8). The atherosclerosic plaque of aorta wall and aorta root were measured by oil red O staining;The expression of lectin-like oxidized low-density lipoprotein receptor-1 (LOX-1) in human umbilical vein endothelial cells (HUVEC) through C-reaction protein ( CRP) was studied by Western blotting. Results The atherosclerosis plaque in normal control group was not observed. The lipid accumulation decreased in the aorta and the plaque areas in the aortic sinus in THSG treated-group compared with model control group. Moreover, THSG down-regulated CRP-induced LOX-1 expression in HUVEC. Conclusion The atheroscletosis plaque in ApoE knock-out mice was decreased by THSG. The mechanism might be related to the inhibition of the expression of LOX-1 protein.
