1.Protective effects of limb remote ischemic postconditionting on ischemic stroke rats under hyperglycemia

Aim To study the protective effects of limb remote ischemic postconditioning ( LRIP ) on is-chemic stroke rats under hyperglycemia and explore the mechanisms. Methods Rats were given 50% glucose (6 mL·kg-1 ) by intraperitoneal injection to get acute hyperglycemia model. Then middle cerebral artery oc-clusion ( MCAO) models were created. After blocking middle artery for 1. 5 h and reperfusion for 2 h, behav-ioral testing, infarct size of brain, NO concentration and SOD activity in the serum of those rats were detec- ted. Results LRIP could improve behavioral score, decrease the area of cerebral infarction, increase the concentration of NO and the SOD activity in serum of MCAO rats. Conclusion LRIP can relieve cerebral ischemia-reperfusion injury of MCAO rats under acute hyperglycemia.
2.Clinical observation on muscle regions of meridians needling method in improving upper limb function for children with cerebral palsy of spastic hemiplegia type

Objective: To observe the improving effect of muscle regions of meridians needling method on the upper limb function in children with cerebral palsy of spastic hemiplegic type. Methods: A total of 100 children with cerebral palsy of spastic hemiplegia type were divided into a treatment group and a control group according to the visiting sequence, with 50 cases in each group. The control group was treated with conventional rehabilitation plus conventional acupuncture treatment. The treatment group was treated with conventional rehabilitation plus muscle regions of meridians needling method. The electromyography (EMG) signal values of triceps brachii and pronator teres were detected before treatment, and 3 months and 6 months after treatment. The clinical efficacy was evaluated by Peabody developmental motor scale-fine motor (PDMS-FM) and fine motor function measure (FMFM). Results: Three and six months after treatment, the EMG signal values of triceps brachii and pronator teres, grasping scores and visual-motor integrated scores of PDMS-FM and the FMFM scores in both groups increased to varying degrees compared with the same group before treatment, and the intra-group differences were all statistically significant (all P<0.05). Six months after treatment, the results of the above three items in the treatment group were all better than those in the control group, and the differences between the groups were statistically significant (all P<0.05). Conclusion: Muscle regions of meridians needling method added on the basis of conventional rehabilitation can effectively reduce the muscle tone of upper limb and enhance the muscle strength, and improve the upper limb function in children with cerebral palsy of spastic hemiplegia type. The efficacy is superior to that of the conventional rehabilitation plus conventional acupuncture treatment.
3.Construction and characterization of a pseudorabies virus TK-/gG- mutant.

To construct a TK-/gG- mutant of pseudorabies virus, the gG-detected transfer vector pUSKKBB and genomic DNA of pseudorabies virus TK-/gG-/LacZ+ were co-transfected into IBRS-2 cells. Transfection progeny were plated onto PK-15 cells and incubated for 2 days under methylcellulose. Then the overlay was removed and replaced by 1% low melting point agarose in DMEM supplemented with 150 microg/mL X-gal. After 2 days, white plaques were screened for and purified 4 times. By PCR amplification of gG-deleted gene and LacZ gene, a recombinant virus with TK-/gG- phenotype was confirmed. Sequence of the PCR product revealed that there were 1,176 bp detection in gG gene of the PRV TK-/gG- mutant. Amplifying the gG-deleted gene of different generations of the TK-/gG- mutant showed that the mutant was stable within PK-15 cells. TCID50 assay indicated that the recombinant virus grows well on PK-15 cells. The mice immunized with the TK-/gG- virus showed no sign of abnormality. As a control, all mice inoculated with PRV strain died from the infection. All mice that received TK-/gG- survived after a lethal PRV challenge. However none of the mice injected with phosphate-buffer saline (PBS) survived from the challenge. The above results demonstrated that the recombinant virus could be a candidate marker vaccine strain for eradicating pseudorabies in pig herds.

4.Determination of serum IL-23 and its significance in patients with severe chronic hepatitis B.

OBJECTIVETo investigate the level of the serum IL-23 and its correlation with serum biochemical indices of liver function tests and HBV DNA load in patients with chronic severe hepatitis B.
METHODSFifty patients with chronic severe hepatitis B (severe hepatitis group) and 18 healthy controls (control group) were enrolled in the study. The serum IL-23 expression level was detected by ELISA method. The correlation between IL-23 and ALT, AST, TBil, HBV DNA load was analyzed using Pearson's correlation analysis, respectively.
RESULTSSerum IL-23 expression level in severe hepatitis group was significantly higher than that of control group (P < 0.05). Serum IL-23 level was positively correlated with serum ALT, AST, respectively (P < 0.05), but was not correlated with TBil and HBV DNA load, respectively (P > 0.05).
CONCLUSIONSerum IL-23 expression level was increased in patients with chronic severe hepatitis B and was associated with the severe of inflammation. We, therefore, believe that IL-23 might be involved in the pathogenesis of chronic severe hepatitis B.

5.A randomized, single-blind, parallel-controlled and multicentre study:compare the efficacy and safety of domestic and imported human recombinant FSH in WHO group Ⅱ anovulatory infertility

Objective To evaluate the efficacy and safety of domestic human recombinant FSH (rhFSH) in women with anovulation of WHO groupⅡ. Methods A randomized, blind, parallel-controlled, non-inferiority and multicenter study was performed. A total of 534 admitted to 13 hospitals from May 2008 to August 2009. There were 531 women with ovulatory disorder was included in the statistical analysis, were randomly divided into test group (domestic rhFSH, n=352) and control group (imported rhFSH, n=179). Percentage of cycle with mature follicle, ovulation rate, clinical pregnancy rate, multiple pregnancy rate, ovarian hyperstimulation syndrome (OHSS) and adverse events were observed. Results No statistical significant differences (P>0.05) were observed between the two groups in terms of the efficiency on mature follicle [91.8%(323/352) versus 88.8%(159/179)], ovulation rate [91.3%(295/323) verus 90.6%(144/159)], clinical pregnancy rate [19.2%(62/323) verus 18.2%(29/159)], the number of the follicles<14 mm, the level of serum LH and progesterone, the thickness of endometrium on the day of hCG administration. The number of follicle≥18 mm and 14 mm≤follicle<18 mm and the level of serum estradiol on the day of hCG in the test group were significantly higher than those in the control group (P<0.05). The number of days of rhFSH administration in the test group was significantly less than that in the control group [(9.8±2.2) versus (11.4± 0.6) days, P<0.05], the dosage of rhFSH was significantly lower than that in the control group [(879 ± 419) versus (1 043 ± 663) U, P<0.05]. The multiple pregnancy rate in the test group was significantly higher than that in the control group [21% (13/62) versu 10% (3/29), P<0.05]. The incidence of OHSS and adverse events were similar between the two groups (P>0.05), and no other adverse events were observed in test group during treatment. Conclusion Ovarian stimulation with domestic rhFSH is effective, safe and economical in women with anovulation of WHO groupⅡ.
7.The measurement and significance of serum chemokine RANTES in patients with chronic hepatitis B.

OBJECTIVETo investigate the relationship of the serum chemokine RANTES level in patients with chronic hepatitis B among different clinical severity and to explore its possible reasons.
METHODS144 patients with chronic hepatitis B were divided into mild-moderate (46), serious (51) or severe group (47) according to the different clinical severity and 18 normal cases were taken as normal control. The serum level of chemokine RANTES was detected with an ABC-ELISA assay. Statistical analysis was performed on the software of SPSS 13.0.
RESULTSThe serum chemokine RANTES levels in patients with chronic hepatitis B (2227.06 +/- 790.80, 5878.49 +/- 3334.58, 3482.77 +/- 2315.62 ng/L in mild-moderate, serious and severe group respectively) were significantly higher than that in the normal control (329.46 +/- 152.00 ng/L). The differences between each two hepatitis groups were also statistically significant (P < 0.05) and serum chemokine RANTES level in serious group was highest among them.
CONCLUSIONSerum chemokine RANTES level in patients with chronic hepatitis B elevates significantly and it might play an important role in the pathogenesis of chronic hepatitis B.

8.A clinical study on the efficacy of HBeAg-positive chronic hepatitis B patients treated with adefovir dipivoxil for 4 years.

OBJECTIVETo observe the efficacy and safety on the efficacy of HBeAg-positive chronic Hepatitis B patients treated with adefovir dipivoxil for 4 years.
METHODSNinety-five patients with HBeAg-positive chronic hepatitis B were treated with adefovir dipivoxil 10 mg per day orally. The patients were observed before and after treatment for their serum levels of ALT and HBV DNA, the new increasing rates of serum ALT normalization, HBV DNA clearances, HBeAg loss, HBeAg seroconversion and adverse drug events.
RESULTSAt 4 years on study, the rates of ALT normalization, HBV DNA clearances, HBeAg loss, HBeAg seroconversion and HBV DNA rebound were 89.5%, 63.2%, 47.4%, 41.1% and 8.0%, respectively. No drug related to renal function impairment was found during the treatment, eight patients had adverse drug events but all were mild.
CONCLUSIONAdefovir dipivoxil could effectively inhibit HBV replication, normalize ALT and enhance transformation from HBeAg to HBeAb for cases with naive and treated-first patients. The efficacy were increased with prolongation of the treatment period. It is safe and has a good tolerance.

9.Detection of hepatitis C core antigen in intravenous drug addictions.

OBJECTIVETo study the status of detection of hepatitis C core antigen in intravenous drug addictions, and discuss the foreground of the hepatitis C core antigen ELISA test system.
METHODSHCV core antigen, HCV RNA quantity, anti HCV-IgG, HBsAg were analysis in all the plasma samples taken from 93 cases of intravenous drug users.
RESULTSThe specialty and sensitivity of HCV core antigen in intravenous drug addictions 100% -54% separately. When HBsAg were positive, the sensitivity of HCV core antigen was 38%, while HBsAg negative, the sensitivity of HCV core antigen was 69% (P < 0.01).
CONCLUSIONThe detections of HCV core antigen showed high specialty but low sensitivity in intravenous drug addictions. The positive rate has positive relation with HCV RNA virus logarithm quantity. Coinfection with HBV are the interfere factor of HCV core antigen detection. In screening experimentations, the detection of HCV core antigen in plasma may be applied as supplement method for anti-HCV-IgG. It can also be used to monitor viremia in HCV infection.

10.Efficacy of two nucleoside analogs to treat resistant HBeAg-negative chronic hepatitis B.

OBJECTIVETo observe the therapeutic effect and safety of entecavir and adefovir in the treatment of lamivudine-resistant HBeAg-negative chronic hepatitis B.
METHODSSixty-five patients with lamivudine-resistant HBeAg-negative chronic hepatitis B were randomly divided into two groups. The entecavir treatment group included 33 patients, who were administrated entecavir 1.0 mg/d. The adefovir treatment group included 32 patients, who were administrated adefovir dipivoxil 10 mg/d. Changes in serum HBV DNA, liver functions, phosphocreatine kinase, creatinine and adverse reaction were dynamically monitored.
RESULTSAt the end of the 12th, 24th, 48th week of treatment, the rates of serum ALT normalization of the entecavir treatment group were higher than that of the adefovir treatment group, but there wasn't statistically difference between two groups until the end of the 48 th week of treatment (P > 0.05). The rate of sera to turn negative for HBV DNA of the entecavir treatment group was significantly higher than that of the adefovir treatment group at the end of the 12th week. Moreover, the difference was statistically significant (P<0.05).
CONCLUSIONBoth entecavir and adefovir dipivoxil might have a good response to lamivudine-resistant HBeAg-negative chronical hepatitis B. Entecavir could achieve better therapeutic effects.
