2.The relationship between the body mass index and health-related quality of life of 4696 young students aged 18~25 years old

Objective To explore the relationship between the body mass index and health-related quality of life(HRQoL) of young students aged 18~25 years old. Methods 4696 students aged 18~25 years old from nine provinces and municipalities (Jiangsu, Anhui, Gansu, Qinghai, Fujian, Boijing, Jilin, Jiangxi and He'nan) were investigated. The HRQoL of students with different body mass index were compared with rank sum test and lo-gistic regression analysis. Results Obesity and underweight students scored lower in both physical component and mental component scores of SF-36. Compared to students with normal weight, odds ratios (ORs) of impaired HRQoL among obesity students significantly increased in both physical domain (OR=1.86) and mental domain (OR=1.90), while OR of impaired HRQoL among underweight students significantly increased only in physical domain(OR=1.21). Analysis for boys and girls scparately,OR of impaired HRQoL increased in physical domain among underweight boys(OR=1.37), and increased in mental domain among overweight girls(OR=3.85). Con-clnsion Different body mass index have different impact on young students' HRQoL. Both obesity and trader-weight could reduce HRQoL. But obesity have more impact on girls' mental domain than boys, and underweight have more impact on boys' physical domain than girls.
3.The changes of Na~+,K~+-ATPase activity and ? isoforms in rat neurons suffered from global ischemia

Aim To investigate the changes of Na~+,K~+-ATPase activity and ? isoforms in rat neurons suffered from global ischemia.Methods The BCAL rat was induced by bilateral carotid artery ligation.The contents of H_2O,Na~+ and K~+ in brain tissue were detected.The method of spectrophotometry was applied to measure the activities of Na~+,K~+-ATPase.With immunohistochemical assay,the expression of the Na~+,K~+-ATPase ?_1,?_2,or ?_3 isoform was detected in neurons of rat hippocampus and cortex following ischemia.Results In the BCAL rats,the contents of H_2O and Na~+ were increased while the content of K~+ and the activity of Na~+,K~+-ATPase were decreased.The ?_1,?_2 or ?_3 isoform of Na~+,K~+-ATPase distributed in a tissue-specific fashion in neurons of hippocampus and cortex,in which ?_1 and ?_3 were abundant and ?_3 was more than ?_1,but ?_2 was much less.In BCAL rat,?_1 and ?_3 isoforms were significantly decreased in hippocampus and cortex neurons.Conclusion These results suggest that the changes of Na~+,K~+-ATPase ?_1 and ?_3 isoforms may be involved in the global cerebral ischemia

5.Screening anti-human IgG for colloidal gold conjugate with dot immunogold filtration assay

Objective:To screen the appropriate anti-human IgG (secondary antibody) for conjugating with colloidal gold by dot immunogold filtration assay,and put forward the method of using multiple evaluation indicators for secondary antibody comparing and analyzing. Methods:Three different secondary antibodies A,B and C from three different companies were screened. Firstly the titration was used to determine the optimal reaction conditions. Then three secondary antibodies were conjugated to the colloidal gold,tested with a dot immunogold filtration assay for hydatidosis specific antibody detection. The optimal conjugated secondary antibody were compared with the standard indirect enzyme-linked immunosorbent assay. Results:The optimal pH of three secondary antibodies were 8. 5,the binding capacity were 38. 4,24 and 19. 2μg /ml colloidal gold. According to the comprehensive evaluation,the diagnostic effects of sec-ondary antibody B was better than others. Its diagnostic effects in dot immunogold filtration assay was compared with enzyme-linked im-munosorbent assay. The Kappa was 0. 895(P<0. 05) and showed the two methods were in good agreement. Conclusion:The appropriate secondary antibody for conjugating with colloidal gold could be screened by dot immunogold filtration assay and the evaluation indicators which put forward by this study,the screening method would have an important reference value for all kinds of colloidal gold test kit to screen the suitable secondary antibody.
6.Cytotoxic effect and mechanism of bakuchiol and bakuchiol combined with psoralen on HK-2 cell

OBJECTIVE To study the nephrotoxicity induced by bakuchiol alone and bakuchiol combined with psoralen and to explore its mechanism. METHODS The cytotoxicities of bakuchiol and bakuchiol combined with psoralen were investigated using human renal tubular epithelial cell lines (HK-2), in presence or absence of hepatic S9 mixture. The HK-2 cells were exposed to culture medium alone (blank control), 0.5% DMSO (vehicle control), aristolochic acid Ⅰ (AAⅠ;positive control), psoralen 5 μmol·L~(-1) group, bakuchiol 5,10,20,30 and 40 μmol·L~(-1) groups, and bakuchiol+psoralen (20+5), (30+5) and (40+5)μmol·L~(-1) groups, respectively. The cell viabilities were examined by MTT assay; cell membrane injuries were examined by detecting lactate dehydrogenase (LDH) release rate; and the morphological changes in HK-2 cells were observed with contrast microscope. The rate of cell apoptosis was detected by AnnexinⅤ/PI staining, and cell cycle was detected by PI staining with flow cytometry. RESULTS No cytotoxicity was found in psoralen 5 μmol·L~(-1) group. The HK-2 cell viabilities were significantly reduced after 4, 24, 48 and 72 h of exposure to either bakuchiol 20, 30 and 40 μmol·L~(-1)groups or bakuchiol+psoralen (20+5), (30+5) and (40+5)μmol·L~(-1) groups in a time- and concentration-dependent manner. The IC_(50) values of bakuchiol were (26.4±4.8), (21.8±0.6) and (24.1±0.8)μmol·L~(-1) for 24, 48 and 72 h exposure, respectively. The cytotoxicity of bakuchiol was significantly decreased in presence of hepatic S9 mixture. The LDH release rate of HK-2 cell increased significantly after 24 h of exposure to bakuchiol 20,30 and 40 μmol·L~(-1) or bakuchiol+psoralen groups. With the concentration and time increasing, the HK-2 cells became more and more contracted and rounded. In bakuchiol 40 μmol·L~(-1) or bakuchiol+psoralen (20+5), (30+5) and (40+5)μmol·L~(-1) groups, HK-2 cells showed apoptotic characters. In bakuchiol or bakuchiol+psoralen groups, apoptotic cells significantly increased and cells in G2 phase markedly decreased. CONCLUSION Bakuchiol has a significant cytotoxicity in HK-2 cells, and combined with psoralen can not decrease its toxicity. The cytotoxicity of bakuchiol is significantly reduced in the presence of hepatic S9 mixture. The possible mechanisms of the renal cytorotoxicity of bakuchiol are as follows: ① direct damage to the cell membrane; ② inducing cell apoptosis; ③ inhibiting intracellular DNA synthesis and block cell mitosis and proliferation.
7.Health-related quality of life and its affecting factors in the elderly individuals: data from 9 provinces of China

Objective To investigate the health-related quality of life (HRQOL), demographic characteristics, and health behaviors of the Chinese elderly to find out high-risk population and behaviors. Methods Data was collected from a cross-sectional survey performed in Jiangsu, Anhui, Gansu,Qinghai, Fujian, Beijing, Jilin, Jiangxi, and Henan province. MOS SF-36 was used for HRQOL assessment, t test was used for HRQOL comparison between the elderly and the general population. Multiple stepwise linear regression analysis was used to evaluate the affecting factors. Results The HRQOL among the Chinese elderly were PF ( Physical Functioning) 79 ± 21, RP ( Role-Physical ) 68 ± 40, BP ( Bodily Pain) 72 ± 23, GH ( General Health) 57±22, VT ( Vitality ) 69 ± 20, SF ( Social Functioning) 79 ± 23, RE ( Role-Emotional ) 72 ±40, M H ( Mental Health)76 ± 18, which were lower than Sichuan Norm on 7 dimensions ( P < 0. 05 ; except on M H dimension) and lower than Hangzhou Norm on 5 dimensions (P < 0. 05 ;except on GH, VT, and MHdimensions). The important affecting factors included physical exercise, education level, medical history of chronic disease, age, race, marriage, body mass index ( BMI ), sleeping habits, and gender. Conclusions Compared with the general population, the HRQOL in the elderly might be lower. The health policy and community heahhcare services should focus on the elderly individuals with insufficient exercise, lower education level, chronic disease history, and ethnic minority, or widows. Health education should encourage them to improve physical exercise and sleeping behavior.
8.Effects of renovascular hypertension on contractile/ diastolic function,calcium transient and calcium sensitivity in rat ventricular myocytes

Aim To observe effects of hypertension on contractile/diastolic function and calcium transient in rats ventricular myocytes. Methods The model of one-kidney-one-clip (1k1c) hypertensive rat was pre-pared by partially ligating the left renal artery and removing the right kidney. Left ventricular myocytes were enzymatically isolated. Then the contraction and calcium transient of a single cell from both normal and renovascular hypertensive rats were observed by a video-based motion edge-detection system simultaneously. Effects of calcium in various concentrations on contractile/diastolic function and calcium transient in ventricular myocytes from renovascular hypertensive rats were assessed in the same way. Results Compared with normal cardiac myocytes, the shorting amplitude and the contractile and diastolic velocity were increased significantly in 1k1c hypertensive rat cardiac myocytes. However their intracellular calcium in contractile and diastolic periods, the extent of calcium transient and the parameters of intracellular calcium dynamics were unchanged. But the extracellular calcium of different concentrations could shift the Fura-2 fluorescence ratio-cell shorting amplitude curve from hypertension rat myocytes to the left compared with that from normal rats. Conclusions The hypertension increases the contractility of rat cardiac myocytes, which is due to raising their sensitivity to calcium.
9.Effect of droperidol, midazolam conscious-sedation on im-plicit memory in epidural anesthesia

Objective: To compare the effects of droperidol, midazolam sedation on implicit memory in patients under epidural anesthesia. Methods: Thirty- two patients with ASA I 一 II un-dergoing general surgery operations with epidural anesthesia were divided into 2 groups,group D received droperidol 2.5mg and fentanyl 0.05mg as sedative, group M received midazolam 3mg and fentanyl 0.05 mg as sedative. The explicit memory and implicit memory scores were gained by methods of process dissociation procedure, and the patients' assessment of alertness/sedation scales were graded simultaneouly. Results: The median of patient' assessment of alertness/ seda-tion scale in group D and group M were 4 and 3 respectively. Patients in group D had higher explicit memory scores and lower implicit memory scores than those in group M. But the implicit memory had not disappeared completely in group D. Respiratory depression emerged when increasing the dosage of droperidol. Conclustion: Droperidol can specifically depress the implicit memory, but it results in respiratory depression when the dosage of droperidol increased. It limits the usage of droperidol to eliminate the implicit memory completely.
10.Effect of Different Does of Simvastatin in Reducing Serum Lipid

Objective:Comparing the effect of different doses of simvastatin in lowering the serum lipid.Methods:79 patients were randomized into group A and group B,and were given simvastatin 10 mg*d-1 (group A) and 20 mg*d-1 (group B),respectively for a total of 8 weeks.Results:Comparing with baseline,in group A,TC,TG,LDL-C were decreased by 23.4%,20.0% and 30.7%,respectively (P<0.01); HDL-C was increased by 17.5%.The content of serum TC,TG and LDL-C was decreased to the normal range in 12.8%,28.2% and 15.4% of the patients in group A.For the group B,TC,TG,LDL-C were decreased by 32.7%,22.8% and 42.8%,respectively (P<0.01); HDL-C was increased by 13.7%.The content of serum TC,TG and LDL-C was decreased to the normal range in 65.0%,57.5% and 65.0% of the group B patients.Conclusion:Oral intake of 20 mg of simvastatin once a day can effectively reduce the serum lipid.The patients can well tolerate and no obvious side effect was observed in our study.
