2.Treating postoperative right ventricular failure by using milrinone and prostaglandin E1
Chinese Journal of Thoracic and Cardiovascular Surgery 1995;0(05):-
Objective To study the effect of milrinone(Mil) and prostaglandin E1(PGE1) on postoperative right ventricular failure(RVF). Methods 28 patients with congenital heart diseases and pulmonary hypertension undergoing cardiopulmonary bypass (CPB) were randomly divided into four groups: control group (group I), receiving Mil (Group II), receiving PGE1 (Group III), and receiving both Mil and PGE1(group IV). The hemodynamic indexes of all the patients, including mean pulmonary artery pressure (PAP), mean arterial blood pressure (MAP), cardiac index(CI), systemic vascular resistance (SVR) and pulmonary vascular resistance(PVR), were measured and compared. Intubation time and side effects were observed. Results Compared with the other three groups, postoperative PAP、PVR and SVR in group IV deceased and CI increased significantly. The intubation time was also shorter in group IV. Conclusion Postoperative RVF can be treated efficiently by using Mil and PGE1.

AIM:To investigate the effect of tanshinone IIA on the apoptosis of human umbilical vein endothe -lial cells (HUVECs) after high glucose treatment.METHODS:The cell viability was determined by MTT assay. The cell apoptotic rate was examined by flow cytometry with Annexin V-FITC/PI double staining .The expression of Bcl-2 and Bax, and the release of mitochondrial cytochrome C ( Cyt C) were analyzed by Western blotting .RESULTS: Tanshinone IIA significantly inhibited high glucose-induced decrease in cell viability and increased the cell apoptosis .Additionally, after tanshinone IIA treatment , Bax expression and the release of mitochondrial Cyt C were significantly inhibited , while Bcl-2 expression was increased .CONCLUSION: Tanshinone IIA prevents high glucose-induced endothelial cell apoptosis via mitochondria-dependent pathway .

Objective To study the relationships between hemoglobin concentration, cytokine and cardiac function in patients with ischemic cardiomyopathy. Methods Hemoglobin concentration, sernm TNF-α,IL-6 in 121 patients with ischemic cardiomyopathy were measured, left ventricular ejection fraction(LVEF)by echocardiography, according to hemoglobin concentration, these patients were distributed to the anemic group and the non-anemic group, contrasting to 27 healthy persons. Results The levels of serum TNF-α,IL-6 in the anemic patients group were higher than those in the non-anemic group subjects, but LVEF Wag lower. With the increase of anemia severity, the levels of serum TNF-α,IL-6 significantly increased;there was a positive correlation between LVEF and the levels of serum TNF-α,IL-6(P<0.01),and a negative correlation between LVEF and Hb concentration. Conclusion The levels of cytokine promote the pathophysiological procedure between cardiac failure and anemic in ICM.

Alzheimer's disease (AD) is a complex neurodegenerative disorder which seriously causes the dementia in elderly people and afflicts millions of people worldwide. Drug discovery for Alzheimer's disease therapy has been a hot research area and a big challenge, in which development of acetylcholinesterase (AChE) inhibitors design was the most active and some AChE inhibitors are commercially available for AD medication already. However, practical using of commercial AChE inhibitors showed their limited usefulness and related adverse effects. Thus, it is extremely urgent to find novel AChE inhibitors with higher potency and less adverse effects. Based on the accurate crystallographic studies about AChE, strategies for multi-binding site AChE inhibitors have been formed, followed by design of the multi-target directed ligands. In this review, the structures and binding modes of commercial AChE inhibitors were briefly discussed, together with the development of AChE inhibitor design for AD therapy: from multi-binding site inhibitors to multi-target directed ligands.

To produce specific antibodies against malachite green ( MG) , one special hapten was synthesized and characterized, and conjugated to carrier protein as immunogen. The immunogen showed excellent reactogenicity and immunogenicity. One specific monoclonal antibody (mAb, named MG-DA4-C7) with high sensitivity and specificity for MG in indirect competitive enzyme-linked immunoassay ( icELISA ) was screened. The isotype was IgG1 and the light chain was κ type. After optimization of ELISA conditions, the proposed icELISA showed a 50% inhibition value ( IC50 ) of 0. 96 μg/L, a linear range ( IC20-IC80 ) of 0. 1-8. 1 μg/L and a limit of detection ( LOD, IC10 ) of 0. 05 μg/L for determination of MG. The assay showed cross-reactivity of 18. 1%, 26. 5% with crystal violet and brilliant green, respectively, and negligible cross-reactivity with other metabolites of MG (<0 . 1%) . The average recoveries of MG from spiked fish samples were from 87. 3% to 107. 3%. Good correlation (R2=0. 999) was obtained between the results of icELISA and those of liquid chromatography-tandem mass spectrometry analysis. The proposed icELISA is suitable for the determination of MG in fish samples in a simple and sensitive manner.

Objective To detect the expression and function of ZNRF3 in different kinds of thyroid cancer tissues and cells. Methods Immunohistochemistry was used to detect the expressions of ZNRF3 protein in 35 cases of papillary thyroid carcinoma and 10 cases of poorly differentiated thyroid carcinoma. The expressions of ZNRF3 gene in TPC-1 and 8505C were detected by RT-PCR,and the cell lines were derived from papillary thyroid carcinoma and poorly differentiated thyroid carcinoma respectively. After silenced ZNRF3 gene expression with lentivirus, the proliferation ability of TPC-1 cells were detected with CCK-8,the invasion and metastasis ability of TPC-1 cells were detected with Transwell. Results According to results of immunohistochemistry and RT-PCR,the expressions of ZNRF3 in papillary thyroid carcinoma cells and tissues were higher than those in poorly differentiated thyroid carcinoma cells and tissues, the differences were statistically significant (4.83±0.44 vs.3.13 ±0.59,t =2.20,P <0.05;1.01±0.06 vs.0.21±0.04,t =11.80,P<0.01).After ZNRF3 geng silencing, according to the results of CCK8,the proliferation ability of TPC-1 cells was significantly enhanced in 72 h,the difference was statistically significant (0.96 ± 0.10 vs.0.64 ± 0.05,t =3.19,P < 0.05);and according to the results of Transwell, the TPC-1 cell's invasion (0.12 ± 0.01 vs.0.09 ±0.00,t =5.48,P<0.01) and migration (0.22 ±-0.01 vs.0.17 ±0.01,t =4.58,P <0.05) also increased, the differences were statistically significant. Conclusion The expression of ZNRF3 in papillary thyroid carcinoma is higher than that in poorly differentiated thyroid cancer.ZNRF3 is tumor suppressor gene in the thyroid tumors.
