2.Studies on the expression of TNF alpha, IL-1 beta, IL-6 and IL-4, IL-10, IL-13 mRNA in lung tissue of rat with acute lung injury induced by oleic acid and lipopolysaccharide

Objective To study the role of pro inflammatory cytokines tumor necrosis factor?(TNF ?),interleukin 1beta(IL 1?),interleukin 6(IL 6) and anti inflammatory cytokines interleukin 4(IL 4),interleukin 10(IL 10),interleukin 13(IL 13) in acute lung injury(ALI). Methods Wistar Rats (200?50g) were randomly divided into the oleic acid(OA) and Lipoplysaccharide(LPS) induced lung injury experimental group and the only OA induced lung injury control group. The LPS(2mg/kg) was injected in rat's tail vein at the 1, 4, 12 and 24 hours after administration of OA(0.2ml/kg, iv). Reverse transcription polymerase chain reaction(RT PCR) were used to detecting the expression of TNF ?, IL 1?, IL 6 and IL 4, IL 10, IL 13 mRNA in the lung tissue of rats in two groups. Results The results showed that the peak expression of TNF ? and IL 1? mRNA at one hour after first hit by oleic acid, and IL 6 mRNA at four hours were observed. Others, such as IL 4, IL 10 and IL 13 mRNA, the highest levels in lung tissue at four hours after lung injury induced by oleic acid. Conclusions The results suggested that TNF ? and IL 1? are presenting early phase of ALI, and that IL 6 played its important role in delayed development. The over expression of anti inflammatory cytokines IL 4, IL 10 and IL 13 may be as a promoters rather than protectors in inflammatory amplification.
3.Effect of Ankle States on Peak Torque of Knee Flexion and Extension for Old Adults

Objective To explore the effect of state of the ankle on peak torque of knee flexors and extensors. Methods 18 adults in community over 65 years old were included. Their peak torque with knee flexion and extension was measured under 4 kinds of action of ankle. Results There was significant difference in peak torque neither flexion nor extension among 4 kinds of action of ankle (P>0.05). Conclusion It is unimportant of the state of ankle for knee training.
4.A primary study of evolution of hepatitis B virus based on motif discovery.

Hepatitis B is a serious infectious disease worldwide, and hepatitis B virus (HBV) is the direct cause of this disease. In recent years, as an essential part of its evolutionary process, HBV mutation has been extensively studied domestically and globally. However, the study on the conserved sequences in HBV sequences is still in its infancy. In this study, we applied multiple EM for motif elicitation (MEME) algorithm to discover HBV motif and proposed a new metric, conservative index (CI), to carry out phylogenetic analysis based on HBV sequences. Then, the constructed phylogenetic tree was subjected to reliability assessment. The results demonstrated that the new metric CI combined with the MEME algorithm can effectively help to discover motifs in HBV sequences and construct a phylogenetic tree based on them and to analyze the evolutionary relationship between HBV sequences; in addition, the possible ancestral sequences of samples may be obtained by conservative analysis. The proposed method is valuable for the exploratory study on large HBV sequence data sets.

6.Effects of different doses of lipopolysaccharide on acute lung injury in rats

Objective To investigate the changes of plasma interleukin 4 (IL 4) in rats with acute lung injury (ALI) induced by lipopolysaccharide (LPS) and to explore the relationship between IL 4 and ALI. Methods A total of 120 Wistar rats were randomly divided into 2, 4, 6, and 8 mg/kg groups according to different doses of LPS administration at each observing time point. A control group (NS), receiving saline injection, was also employed. The indexes of respiratory rate, PaO 2, wet weight/dry weight (W/D) of lung lobes, and pathological changes were observed at 1, 2, 4, and 6 h after injury. The plasma IL 4 content was detected by enzyme linked immunosorbant assay. Results ① Various degrees of ALI in rats were induced by different doses of LPS. Acute respiratory distress syndrome (ARDS) was induced under the condition of LPS ≥6 mg/kg. ② LPS could induce increased plasma IL 4 in rats. The peak value of plasma IL 4 in rats increased significantly under the condition of LPS ≥6 mg/kg. Conclusion ① ALI model can be duplicated by injection of LPS. ② LPS ≥6 mg/kg is the critical dosage for ARDS in rats. ③ The obvious increase of plasma IL 4 may be associated with the occurrence of ALI/ARDS.
7.Characteristics of the acute lung injury induced by oleic acid and lipopolysaccharide in rats

Objective To investigate the characteristics of the acute lung injury induced by oleic acid (OA) and lipopolysaccharide (LPS) in rats. Methods Wistar rats were injured with OA (0.2 mL/kg) and LPS (2 mg/kg) to establish the acute lung injury (ALI) model. The indexes of respiratory rate, PaO 2, wet weight/dry weight (W/D) of lung lobes, plasma IL 13, and pathological changes were observed. Results Acute respiratory distress syndrome (ARDS) was found in rats treated with OA LPS. The level of PaO 2 was lower than 8 kPa at the early stage. Increased death rate, significantly increased water content in the lungs, more serious pulmonary edema and infiltration of inflammatory cells accompanied by formation of hyaline membrane, and significantly increased plasma IL 13 content were also found. There were significant changes of the above indices, especially in OA LPS/4 h group. Conclusion OA LPS might cause serious ALI/ARDS. Injury due to second time low dose OA LPS after the first time injury may induce ARDS, which may be associated with the abnormally increased level of anti inflammatory cytokines. There is a relative sensitivity between the two injuries.

9.Value of Single Photon Emission Computed Tomography Thyroid Imaging for Diagnosis of Congenital Hypothyroidism

Objective To explore the causes of congenital hypothyroidism by single photon emission computed tomography(SPECT) thyroid imaging.Methods Nineteen cases with congenital hypothyroidism were collected,and SPECT thyroid imaging was performed 15 minutes late after injected 99m TcO_ 4- 18.5-37.0 MBq.Results Five cases were found with normal glands,1 case with enlarged thyroid, 2 cases with enlarged thyroid and increased radiation,4 cases with ectopic thyroid,4 with a hypogenetic thyroid,and the last 3 cases showed no thyroid imaging.Conclusions The causes of congenital hypothyroidism can be found by SPECT thyroid imaging,which is important to the diagnosis of congenital hypothyroidism combined with detection of T_3,T_4,TSH.
10.Determination of protocatechuic acid and protocatechualdehyde in Compound Sijiqing Tablets by HPLC

AIM: To establish an external standard HPLC method for the determination of protocatechuic acid and protocatechualdehyde in Compound Shijiqing Tablets(Folium Ilicis Chinensis, etc.). METHODS:Alltima C 18(5?m, 4.6mm?250mm)was used. The mobile phase was consisted of methanol-1% acetate(18∶82). The flow rate was 1.0mL?min -1. The UV detection wavelength was at 280nm. RESULTS: The two components has good resolution. The liner ranges were in the range of 7.6~127.1?g?mL -1 for protocatechuic acid and in the range of 2.62~43.6?g?mL -1 for protocatechualdehyde with the correlation coefficients 0.9997 and 0.9994, respectively. The average recoveries were 99.9% and 97.5% with RSD 2.1%,2.2%(n=5), respectively CONCLUSION: The method is accurative, reproducible and can be used to control the quality of Compound Sijiqing Tablets.
