1.A study on calcification of cultured aortic medial cells in vitro

Objective To investigate calcification of cultured aortic medial cells in vitro and acceleration by 25 hydroxycholesterol or ? glycerophosphate. Methods Aortic medial cells were obtained by explantation, and the calcification was observed by von Kossa staining. Insoluble calcium precipitation in cellular layer was determined by biochemical method , and osteocalcin in the media was analyzed with radioimmunoassay. Results Two different types of primary cells were shown from culture: one with parallel cellular growth and being negative by von Kossa staining, the other cell type formed cellular nodules with positive von Kossa staining. After 28 days of cell passages, cell growth appeared no nodule formation. However, many cellular nodules and positive von Kossa staining were observed in the passaged cells treated with 25 hydroxycholesterol or ? glycerophosphate, and both insoluble calcium 〔(57 80?18 50)?g/pool, (67 50?15 30)?g/pool〕and osteocalcin 〔(0 886?0 063)?g/L, (0 895?0 061)?g/L〕in the medium were significantly increased than that of the untreated cells. Conclusions Cultured aortic medial cells could be divided into two subtypes, one with the characters of smooth muscle cells, the other with the micro vascular pericytes which could calcify the extracellular matrix. 25 hydroxycholesterol and ? glycerophosphate promoted the in vitro calcification, and osteocalcin secretion was increased in the process of calcification of aortic medial, suggesting that osteocalcin might participate in the aortic calcification.
2.Determination of Selenium in Drinking Water by Diaminonaphthalene Spectrophotometry

To establish a spectrophotometric method for detecting selenium in drinking water,some subjects,such as the features of absorption spectrum of the reacting product of selenium and diaminonaphthalene,disturbance of other coexisting ions,detection range,detection limit,precision and accuracy were studied.The results showed that:the maximum absorption wavelength of the reacting product of Se4+and 2,3-diaminonaphthalene was 377 nm,the detection limit,molar absorptivity (ε377) and linear range of this method was 0.018 μg/50 ml,2.7×104 L/(mol·cm)and 0.2~10 μg/50ml respectively.The regressive equation was A=0.077C+0.014 with a correlation coefficient r value of 0.9999.The relative standard deviation (RSD) and recovery rate were <54.7% and 96.5%~106% respectively.This method was simple,sensitive and accurate,could meet the requirement of the routine monitoring of selenium in drinking water.
3.Mediation effects of neuroticism between parental rearing patterns and interpersonal sensitivity

Objective To explore the role of neurotic in parental rearing patterns and interpersonal sensitive.Methods Using stratified sampling method to test 702 middle school students,symptom checklist 90 (SCL90),Egma minnen av bardndosnaupp forstran(EMBU) and Eycenck Personality Questionnaire(EPQ) were used to investigate and take one and a half years of follow-up observation on the sample of 245 students from first grade of junior and senior middle school.Correlated variables were made the correlation analysis,regression analysis,constructing the structure equation model and using tracking data to confirm.Results (1) Neuroticism,father's affective warmth and understanding,father's excessive protection,mother's rejection and deny had a direct effect,and the variation of total symptoms could be explained 40.8% ; father's affective warmth and understanding,father's excessive protection,mother's over-interference and over-protection,mother's rejection and deny had a indirect effect on interpersonal sensitive,and explained 24.0%.(2) Effect analysis showed father's affective warmth and understanding,father's excessive protection,mother's rejection and deny impact on interpersonal sensitive by neuroticism,and father's excessive protection was the largest which accounted for 48.0% of the total effect.The second was father's affective warmth and understanding,accounting for 47.3 %.The last was mother's rejection and deny accounted for 42.7%,and its indirect effect increased with the age(69.1%).All of mother's over-interference and overprotection through the neurotic influence interpersonal sensitive,and the direct effect was 50.3% between neurotic and interpersonal sensitivity.Conclusion Indirect effect of neurotic existed in relationships of parents rearing patterns and interpersonal sensitivity.
4.Three-point bending mechanical properties of the bone in a rat model of osteoporosis after treatment with four kinds of drugs

BACKGROUND:Mechanical property test is an important method for assessing the therapeutic effect of drug therapy in osteoporosis animal models. But there are rare reports on the three-point bending mechanical test for evaluating the therapeutic effects of a variety of drugs on a female rat model of senile osteoporosis. OBJECTIVE: To explore the interventional effects ofDanqi particles, premarin, ipriflavone and αD3 in a rat model of osteoporosis through the three-point bending mechanical test. METHODS:Forty-eight female Wistar rats were randomized into six groups, and animal models of senile osteoporosis were made in al rats except for those in the normal control group. Then, the rats were givenDanqi particles (0.9g/kg/d), ipriflavone (1 mg/kg/d), αD3 (0.1 mg/ka/d) and premarin (0.3 mg/kg/d) in theDanqi, ipriflavone, αD3 and premarin groups, respectively. An electronic universal testing machine was used to perform the three-point bending mechanical test on the bilateral tibiae of rats. RESULTS AND CONCLUSION: Compared with the model group, the maximum load, maximum stress, maximum bending moment, maximum stress, elastic modulus of the tibia were significantly higher in theDanqi, ipriflavone and premarin intervention groups (P < 0.05). There were no significant differences in the maximum load, maximum stress, maximum strain and modulus of elasticity between αD3 and model groups (P > 0.05). There were also no significant differences in the maximum load, maximum stress and maximum strain between the Danqi and normal control groups (P> 0.05). These findings indicate thatDanqi, ipriflavone and premarin interventions have good achievements in the three-point bending mechanical test, and theDanqi particles have the best intervention effects. αD3 has no obvious effects on the three-point bending mechanical performance.
5.metabonomics research on coronary heart disease patients of phlegm turbidity syndrome and qi deficiency syndrome.

OBJECTIVETo study the correlation between Chinese medical types of coronary heart disease (CHD) [i.e., phlegm turbidity syndrome (PTS) and qi deficiency syndrome (QDS)] and their metabolites.
METHODSRecruited were 65 CHD patients including 37 cases of PTS and 28 cases of QDS. Serum endogenous metabolites in the two syndrome types were determined by gas chromatograph-mass spectrometer-computer (GC/MS), and their differences between their metabolic profiles analyzed.
RESULTSMore than 100 chromatographic peaks were totally scanned. Chromatograms obtained was matched with mass spectrum bank, and finally we got the category contribution value of 46 kinds of substances. Results of MCTree analysis showed patients of PTS and patients of QDS could be effectively distinguished. Compounds contributing to identify the two syndromes were sequenced as serine, valine, 2 hydroxy propionic acid. Comparison of metabolites showed contents of serine and 2 hydroxy propionic acid were higher in patients of PTS than in patients of QDS (P<0.05).
CONCLUSIONThe differences in the metabonomics of CHD TCM syndrome types could provide material bases for TCM syndrome differentiation of CHD, indicating that metabonomics technologies might become a new research method for TCM syndrome typing.

6.Different doses vitamin D to bone metabolism in obese persons after Roux-en-Y gastric bypass

Objective To determine the effect of different doses vitamin D supplementation to the change of bone metabolism in 6 months after Roux-en-Y gastric bypass ( RYGB) .Methods The change of body weight and body mass index ( BMI ) in 36 patients in 6 months after RYGB WAS analyzed.Then,the effect of low-dose (n=11,400 IU/day) and high-dose (n=12,1 600 IU/day) vitamin D to the serum calcium,25-hydroxyvitamin D and bone density were observed ,and 15 patients as control .Results In 6 months after RYGB,the body weight and BMI were decreased ,and the differences were statistical significance (P<0.05).The serum calcium,25-hydroxyvitamin D and bone density were increased in high-dose group,and the differences were statistical significance (P<0.05).Conclusion High-dose (1 600 IU/day) vitamin D supplementation is effective to the bone loss of patients undergoing RYGB .
7.Comparison and analysis of ASSR test in sensorineural hearing loss children with cochlear lesion and retrocochlear lesion.

OBJECTIVE:
To compare the auditory steady-state response (ASSR) test data of severe sensorineural hearing loss children with cochlear lesion and those with retrocochlear lesion and find some specific phenomena of retrocochlear lesion in ASSR, then to improve diagnostic accuracy in clinical practice.
METHOD:
Between 2008 to 2012. 96 children (179 ears) were diagnosed with "retrocochlear auditory nerve lesion" and recieved ASSR test (" retrocochlear lesion" group). Eighty-one (143 ears) the same age children (143 ears) were diagnosed as "cochlear auditory nerve lesion" and selected them as the "cochlear lesion" group. Twenty-six (50 ears) normal hearing children at the same age who had ASSR test records selected from the pediatric hearing center database of our hospital were selected as "normal control" group. Compare the difference of ASSR threshold, ASSR elicit rate and ASSR audiogram among the three groups.
RESULT:
(1) ASSR threshold: Compared each frequency threshold in ASSR test with cochlear.lesion group, the retrocochlear lesion group with wave V and wave I had no significant difference in 500 Hz and 1000 Hz but had significant difference in 2000 Hz and 4000 Hz. As for the retrocochlear lesion group without wave V and wave I, there was a decline in 500 Hz, 1000 Hz, 2000 Hz and 4000 Hz compared with cochlear lesion group. (1) ASSR elicit rate: When compared to retrocochlear lesion group, the cochlear lesion group had a significance lower elicit rate in the four frequency. (3) Number of elicit reactions in ASSR test . In the retrocochlear lesion group, the elicit reactions in all the four frequency in ASSR test was significantly higher than ochlear lesion group. (4) ASSR audiogram type. The proportion of rising curve audiogram in retrocochlear lesion group was 26.83% (with wave I and wave V), 40% (with wave I and without wave V ) and 33.80% (without wave I and wave V).
CONCLUTION
(1) Children with a severe hearing loss in ABR test and a rise type audiogram in ASSR test should be most possibly considered as retrocochlear lesion. (2) ASSR threshold cannot be used in determinate the severity of hearing loss in children with retrocochlear lesion.

8.Imageology study of sellar turica and its potential role on growth of pituitary adenoma

Objective To study imageology anatomy concerned of sellar turica and explore its potential role on growth of pituitary adenomas. Methods According to classification of sinus sphenoidalis,103 normal imageology data of sellar turica region were analyzed, including 83 cases with entire saddle type sinus sphenoidalis and 20 cases anti-saddle type sinus sphenoidalis. Furthermore, the clinical data of 45 cases of pituitary adenomas with extension into sinus sphenoidalis were retrospectively studied. Results The relationship between sellar floor's shapes and sinus sphenoidals'types was that the larger the sinus sphenoidalis was, the deeper the floor was(98.8%). The relationship between sellar floor's shapes and the superior surface of hypophysis was that most of depressed sellar floors were companied with depressed superior surfaces of hypophysises(93.8%). The anteroposterior shapes of sellar turica in midsagittal can be divided into 4 types such as bag-type, kettle-type, tube-type, and sifter-type. 45 patients suffering from pituitary adenoms with extension into sinus sphenoidalis all had entire saddle type sinus sphenoidalis. Conclusions The imageology anatomy concerned of sellar turica could explain the phenomena of pituitary adenomas with extension toward sellar floor and sinus sphenoidalis. Referring to the literatures concerned, it indicated that the imageology anatomy was the key factor of affecting the growth of pituitary adenomas.
9.Survey and analysis in organizational commitment and influencing factors of clinical nurses at tertiary hospital in Shenzhen

Objective To study the organizational commitment and influencing factors of clinical nurses at tertiary hospital in Shenzhen. Methods 347 clinical nurses from a tertiary hospital were select-ed by stratified random sampling and investigated and analyzed for organizational commitment with ques-tionnaires. Results Total score for organizational commitment of clinical nurses was (60.45±9.99),in which, from the highest to the lowest, the scores for five aspects were normative commitment (14.49±2.97), affective commitment (13.50±3.57), ideal commitment (11.69±3.20), economic commitment (11.27±3.23), opportunity commitment (9.50±3.30),respectively, their influencing factors included age, nursing age, mari-tal status, professional tide. Conclusions The organizational commitment of clinical nurses is at higher-middle level, their influencing factors are multi-dimensional, demographic characteristics have different im-pact on five aspects of organizational commitment.
10.Dynamic contrast-enhanced MRI in serological negative early stage rheumatoid arthritis compared with osteoarthritis in hand

Objective To investigate the value of dynamic contrast-enhanced magnetic resonance imaging ( DCE-MRI) in differentiating serological negative early stage rheumatoid arthritis (RA) from osteoarthritis (OA)in hand. Methods Plain MRI, enhanced MRI of whole hand and DCE-MRI of single slice were performed in 53 patients suspected of RA or OA and 18 healthy volunteers. Twenty-three of them were diagnosed as RA,including 18 cases of serological negative RA and 18 of them were diagnosed as OA after 3 to 6 months follow-up. The contrast rate,slope, thickening of synovial membrane were measured on DCE-MRI in three groups and the MRI findings were also were detected in both RA and OA groups. The contrast rate and slope of synovial membrane were compared among three groups using rank sum test. The thickening of synovial membrane were compared among three groups using variance analysis. MRI signs of RA and OA group were evaluated with rank sum test Results The dynamic contrast rate of synovial membrane in RA group,OA group and control group was(100.78±61.96)%,(40.44±15.43)% and (23.56±9.14)%,respectively.Individually,RA group to OA group,u=3.101,P=0.002;RA group to control group,u=4.669,P=0.000;OA group to control group,u=3.482,P=0.000.The slope of contrast curve of synodal membrane of RA group,OA group and control group was 72.50°±13.34°,45.39°±9.94°,14.56°±5.75°,respectively.Individually,RA group to OA group,u=8.002,P=0.000;RA group to control group,u=17.102,P=0.000;OA group to control group,u=9.100,P=0.000.The synovial membrane thickening of RA group,OA group and control group was(3.3±0.5),(2.8±0.7)and (1.4±0.6)mm,respectively.Individually,RA group to OA group,q=2.622,P=0.011;RA group to control group,q=9.583,P=0.000;OA group to control group,q=6.961,P=0.000.Conclusion The quantitative index of DCE-MRI,including contrast rate and synovial membrane thickening may provide useful information for differentiating OA from semlogical negative early stage RA.The contrast rate and thickening of synovial membrane in RA group are higher than those in OA group.Many signs of MRI Can help differentiate OA from RA.
