1.Mechanism of immunosuppressive effects of triptolide

Triptolide (TL) is a major active component extracted from Chinese traditional herb Tripteryginm wil-fordii Hook. In this article, the effects of TL on mixed lymphocyte culture (MLC),suppressor T cell(Ts) activity, delayed type hypersensitivity (DTH) reaction, IL - 2 secretion activity and Th/Ts ratio were evaluated, In vitro TL 0. 5~10 ?g ? L-1suppressed one-way MLC,Lymphocytes induced in first MLC with TL of 5,10 ?g ? L-1 suppressed the second MLC after irradiation with 3000 rad 60Co source. This suggests that TL may induce Ts cell-s. In vivo, DTH reaction sensitized to dinitrofluo-robenzene (DNFB) monitored by the increase in weight of the ears challenged with antigen was suppressed by TL at doses of 0. 12 to 0. 50 mg ? kg-1(ip,qd?5) and the IL -2 activity secreted by spleen cells of these mice was inhibited at doses of 0. 25 and 0. 5 mg ? kg-1. TL 0. 25, 0. 5 mg ? kg-1(ip,qd?8) also had prominent suppres-sive effects on enhanced DTH reaction which was induced by cyclophosphamide(250 mg ? kg-1,ip). Th/Ts ratio of mouse thymus cells were reduced after given 0. 25 mg ? kg-1 of TL for 5 consecutive days. The above-mentioned data suggest that TL has suppressive effects on cellular immunity function and the suppressive mechanism may be related to the suppression of Th cells and IL-2 secretion activity and the induction of Ts cells.
2.A STUDY ON THE NUTRITIVE VALUE OF INFANT STAPLE FOOD SPRAYS IN RATS

Three infant staple food sprays were studied. Preparation A contained 17.5% cow's milk powder, preparations B and C contained 30% and 26% soybean powder respectively, preparations A and B both contained 22% cane sugar; while C was without cane sugar. Other components in these 3 sprays were mainly rice powder and a little amount of soybean oil and whole egg powder. 3 preparations were fed to 3 groups of male weaning rats for 4 weeks. As indicated by parameters such as body weights gained, FE, CE and PER, preparation C had a significantly higher value than A and B. A and B got a practically similar results. Those effects were also true in a repletion test. The BV and nitrogen balance values were similar among these 3 preparations. There was no prominent effect on serum osmotic pressure in rats fed 22% cane sugar for 4 weeks, but had a lower caloric utilization in group A and B as compared with group C
3.Determination of amodiaquine in human plasma by high performance liquid chromatography

AIM:To establish HPLC method for the determination of amodiaquine in human plasma.METHODS: Amodiaquine and internal standard(hydroxychloroquine) were analyzed on C18 column(150 mm ×4.6 mm, 5 μm) with methanol: water:triethylamine: orthophosphoric acid (21:77.5:1:0.5 )as mobile phase at the flow rate of 1.0 mL · min-1The UV detector was set at 294 nm. RESULTS: The retention times of amodiaquine and internal standard were 5.82, 8.56 min, respectively. The calibration curve was linear in the range from 10 to 1 000 μg ·L- 1 ( r = 0.999 8, n = 9 ). The limit of quantitation was5 μg· L-1. The extraction recovery was between 75.5 % and 82.7 %, and the methodological recovery was between 97.0 % and 104.8 %. The intra-day and inter-day RSD were less than 6.0 % and 7.5 %, respectively. CONCLUSION: This HPLC method is simple, sensitive and suitable for pharmacokinetic study of amodiaquine.
4.Content Determination of Gatifloxacin in Human Plasma by HPLC

OBJECTIVE: To establish a HPLC method for content determination of gatifloxacin in human plasma.METHODS: The Analytical column was C18,the mobile phase consisted of acetonitrile-30mmol/L ammonium acetate-triethylamine-orthophosphoric acid (20∶80∶1.0∶0.7) with a flow rate of 1.0ml/min,the detection was performed at UV 294nm.RESULTS: The calibration curve was linear in the concentrations ranging from 0.1 to10.0?g /ml (r=0.9 992).The detection limit was 0.05?g/ml.The intraday RSD was less than 8% and interday RSD was less than 10%.The average recovery was (101.67?3.06)%.CONCLUSION: The method is simple, sensitive, accurate and suitable for determination of gatifloxacin in human plasma and pharmacokinetic study.

6.Reversal effect of cinobufacine on adriamycin resistancehuman breast cancer cells

Aim To investigate the reversal effect of cinobufacine(Cino)on adrimycin(ADM)resistant human breast cancer cell line MCF-7/ADM.Methods The cytotoxic effect of Cino or ADM and the sensitivity of ADM to cells was determined by MTT assay.The intracellular concentration of ADM was detected by HPLC. The expression of P-glycoprotein(P-gp)was examined by flow cytometric(FCM) .Results The maximum non-toxic dose Cino(15 mg?L-1) increased the sensitivity of ADM in MCF-7/ADM,decreased the IC50 of ADM in MCF-7/ADM from 38.14 mg?L-1 to 12.93 mg?L-1, and significantly increased the intracellular concentration of ADM in MCF-7/ADM and reduced the expression of P-glycoprotein.Conclution The results showed that Cino can partially reverse multidrug resistance(MDR)of MCF-7/ADM cells and the mechanism might be associated with the increase of intracellular accumulation of ADM and the reduced expression of P-glycoprotein(P-gp)in MCF-7/ADM cells.

8.Prenatal diagnosis of X-linked adrenoleukodystrophy in 20 pedigrees

Objective:To summarize the characteristics of genetic variation and prenatal diagnosis in pedigrees with X-linked adrenoleukodystrophy (X-ALD) and elucidate the value of prenatal diagnosis in preventing the birth of children with X-ALD.Methods:Twenty pedigrees, clinically diagnosed with X-ALD in Peking University First Hospital from November 2012 and March 2019, were included in this retrospective study. Genomic DNA was extracted from peripheral blood and amniotic fluid or chorionic villi samples of probands and their families for detecting variants in ATP-binding cassette subfamily D member 1 ( ABCD1) gene using polymerase chain reaction (PCR)-Sanger sequencing. Linkage analysis was also performed on five microsatellite markers near ABCD1 gene to exclude maternal contamination. Characteristics of ABCD1 gene variants and prenatal diagnosis of X-ALD pedigrees were summarized by descriptive statistics. Results:Twenty ABCD1 gene variants were identified in the 20 pedigrees. The variants in three probands that were not detected by next-generation sequencing were identified by PCR-Sanger sequencing. Among the mothers of the 20 probands, 17 carried ABCD1 variants and three did not. We performed 24 prenatal diagnoses on 20 pregnancies (24 fetuses) and identified eight fetuses with variants who were finally terminated. The 16 cases without variants were born alive. The validation results obtained after termination or delivery were consistent with those performed prenatally. Conclusions:No hotspot variants in ABCD1 gene are detected in these X-ALD patients and most variants are maternally inherited. PCR-Sanger sequencing is an effective method for detecting ABCD1 variants. Prenatal diagnosis for mothers who had a body with X-ALD could prevent another one from birth.

10.Changes of Pancreatic Islets Functions and Insulin Resistance Index in Children with Severe Stress

Objective To compare the saccharometabolism with the pancreatic islets functions and insulin resistance index in children with severe stress. Methods Thirty children with severe stress and 30 healthy children in control group were tested. The levels of fasting blood glucose (FBG), fasting insulin (FINS) and fasting C - peptide (FCP) were detected by radioimmunoassay respectively and insulin sensitivity index (ISI), insulin resistance index (IR) and fasting blood cell function index (FBCI) were calculated statistically. Results There were significant differences between the children with severe stress and the normal controls in the levels of FINS, FCP and FBG,(all P0.05). Conclusion There is insulin resistance with the significant decrease in the insulin sensitivity index and significant increase in insulin resistance index in the children with severe stress, which may cause the disorder in glucose metabolism in children with severe stress.
