1.An evaluation of status of depression using CES-D and analysis of its influencing factors among male nursing undergraduates

Objective To understand the status of depression and compare with the same age group of Chinese norms of CES-D in urban areas and analyze its potential influencing factors among male nursing undergraduates.Methods In this cross-sectional study,the CES-D and self-made questionnaire were administrated to 102 male nursing undergraduates from two schools of nursing through two-stage random sampling.Results The prevalence of depression was 14.0%,and the score of nursing undergraduates was (11.00±6.37) points,which had no significant difference from the scores of the same age group of Chinese norms in urban areas.Univariate analysis showed that coming from countryside,worse interpersonal relationship,worse career prospect,getting no attention from parents had some significant association with depression.Multivariate analysis showed that worse career prospect and getting no attention from parents associated with lower CES-D scores.Condusions For male nursing undergraduates,efforts should be given from the aspects of families and career prospects to improve their psychological health condition,and reserve qualified resources for nursing profession.
2.A Method of Evaluating the Horizontal Sound Localization Ability in Normal Children Aged 1~3 Years Old

Objective To explore a method of detecting the horizontal localization ability in normal children aged 1~3 years old, and to obtain the minimum audible angle (MAA) at horizontal 0° degree in these children.Methods Using minimum audible angle measure procedure on the basis of conditioned play audiometry and behavioral audiometry methods, sound localization test at horizontal 0° degree was conducted in 37 children aged from 1 to 7 years old with stimulus of different frequencies.Results All 37 children completed the 1 000 Hz tests, 35 children completed the 500 Hz tests, and 30 children completed the 4 000 Hz test.An analysis of 30 children who completed all the tests was performed, 15 of them aged 1~3 years old and the rest aged 4~7 years old.The results for children aged 1~3 years old were as below: 1 000 Hz MAA(0°) =3.57°±1.35°,500 Hz MAA(0°) =5.03°±2.96°,and 4 000 Hz MAA(0°) =5.4°±2.86°, respectively.The results for children aged 4~7 years old were as below: 1 000 Hz MAA(0°) =3.56°±1.48°,500 Hz MAA(0°) =3.5°±2.17°,and 4 000 Hz MAA(0°) =6.13°±4.09°, respectively.There was no significant difference in MAA (0 °) at corresponding frequencies between the two groups.For the children aged 1~3 years old, there was no significant difference in MAA (0 °) among 500 Hz, 1 000 Hz and 4 000 Hz frequencies.For the children aged 4~7 years old, there was significant difference in MAA(0 °) between 500 Hz and 4 000 Hz as well as between 1 000 Hz and 4 000 Hz frequencies.However, there was no significant difference in MAA (0 °) between 500 Hz and 1 000 Hz frequencies.Conclusion MAA test procedure on the basis of conditioned behavioral audiometry and play audiometry could be used to evaluate the ability of sound localization in 1~3 years old children.The frequencies may have effects on children''s sound localization test results and there was no significant difference in MAA (0 °) between the 1~3 years old group and the 4~7 years age group.
3.Effect of hypertonic sugar eyes drops with local to the oxygen therapy for severe corneal edema after cataract surgery

AIM: To explore the effect of hypertonic sugar eyes drops with local to the oxygen therapy for severe corneal edema after cataract surgery.METHODS: Totally 68 patients (68 eyes) with severe corneal edema after phacoemulsification were selected from January 2014 to December 2015 in our hospital, who were aged 53-80(64.45±4.24), including 30 males and 38 females.According to different treatment, they were divided into treatment group (34 cases) given conventional therapy + hypertonic sugar eyes drops with local to the oxygen therapy, the control group (34 cases) given conventional treatment + hypertonic sugar eyes drops.Visual acuity and corneal situation were observed during the treatment.RESULTS: Corneal edema fade time of treatment group was 11.62+0.53d, that of control group was 15.23±0.62d, the difference between the two groups was significant (P<0.05).Preoperative best corrected visual acuity (BCVA) and corneal endothelial cell count had no significantly difference between the two groups while the corneal endothelial cell count were significantly different compared with postoperative of the two groups (P<0.05).At 7d after treatment, the BCVA and corneal endothelial cell count had significantly difference between the two groups (P<0.05), which did not have significant difference at 1mo after treatment (P>0.05).The effective rates of the two groups was significantly different (P<0.05).CONCLUSION: Hypertonic sugar eyes drops with local to the oxygen therapy is effective for severe corneal edema after cataract surgery.
4.Isolation,culture and identification of adipose derived stem cells from human subcutaneous adipose tissues

Objective:To establish a method for isolating adipose derived stem cells(ADSCs)from resected human subcutaneous adipose tissues.Methods:ADSCs were isolated,cultured,and expanded from human subcutaneous adipose tissues.Immuno-fluorescent staining of specific molecules.FACS and multi-lineage differentiation induction were used to characterize the obtained ADSCs.Results:ADSCs obtained in this study had the characteristics of stem cells and expressed specific molecules;they also possessed a multi-lineage differentiation potential,which was genetically stable.Conclusion:ADSCs can be isolated from human subcutaneous adipose tissues,which provides a novel and abundant seeding cells for tissue engineering.
5.Construction and Expression of RNase-Resisting His-Tagged Virus-Like Particles Containing FluA/B mRNA.

To prepare virus-like particles containing FluA/B mRNA as RNA standard and control in Influenza RNA detection, the genes coding the coat protein and maturase of E. coli bacteriophage MS2 were amplified and cloned into D-pET32a vector. Then we inserted 6 histidines to MS2 coat protein by QuikChange Site-Directed Mutagenesis Kit to construct the universal expressing vector D-pET32a-CP-His. In addition, the partial gene fragments of FluA and FluB were cloned to the down-stream of expressing vector. The recombinant plasmid D-pET32a-CP-His-FluA/B was transformed to BL21 with induction by IPTG. The virus-like particles were purified by Ni+ chromatography. The virus-like particles can be detected by RT-PCR, but not PCR. They can be conserved stably for at least 3 months at both 4 degrees C and -20 degrees C. His-tagged virus-like particles are more stable and easier to purification. It can be used as RNA standard and control in Influenza virus RNA detection.

6.The relationship between serum level of H-FABP before PCI and prognosis in patients with STEMI

Objective To observe the relationship between serum level of H-FABP before percutaneous coronary intervention (PCI) and prognosis in patients with ST-segment elevation myocardial infarction (STEMI). Methods A total of 104 cases were recruited in this study. PCI was performed 12 h after onset. The concentration of H-FABP was detected before operation. Gensini score system was used to calculate the integral of all patients after coronary angiography. According to Gensini score, patients were divided into three groups, 34 cases in group A (8≤Gensini scores<41), 34 cases in group B (41≤Gensini scores<64) and 36 cases in group C (Gensini scores≥64). Indicators were analyzed statistically including systolic blood pressure (SBP), diastolic blood pressure (DBP), smoking history, H-FABP, left ventricular ejection fraction (LVEF), total cholesterol (TC), triglycerides (TG), high density lipoprotein (HDL-C), low density lipoprotein (LDL-C), fasting blood sugar, diabetes (DM) and hypertension. The patients were followed up for 12 months after operation. According to the occurrence of major adverse cardiovascular events (MACE), patients were divided into MACE group and non-MACE group. The related factors of two groups were statistically analyzed. The multivariate Logistic regression analysis was used to screen the risk factors of MACE for patients with STEMI after emergency PCI. Results There is a gradually increasing tendency in total cholesterol levels among three A, B and C groups (P<0.05). LVEF values were lower in B group and C group than that of A group (P<0.05). There were no significant differences in levels of SBP, DBP, H-FABP, TG, HDL-C, LDL-C, fasting plasma glucose and smoking history, DM, and hypertension between three groups (P>0.05). In 12-month follow-up, the proportion of STEM combined with DM, the time from onset to PCI, the level of TC, the level of H-FABP before operation and Gensini score were significantly higher in MACE group than those of non-MACE group (P<0.05). Logistic regression analysis showed that STEM combined with DM, higher serum level of H-FABP before operation were risk factors of MACE in 12-month after operation (P<0.05). Conclusion For patients with acute STEM combined with diabetes and elevated preoperative serum level of H-FABP, be alert to the occurrence of MACE in 12-month after PCI.
7.Experiences and lessons learned from treatment of unstable pelvic fractures

Objective To discuss the clinical treatment of the unstable pelvic fractures. Methods A retrospective study was conducted based on the clinical data of 32 patients with unstable pelvic fractures who were admitted to the Third Hospital of Hebei Medical University from June 2008 to June 2010.There were 20 patients with type B injury and 12 with type C injury according to AO classification scheme.Of all, four patients were treated conservatively, one was treated with emergency external fixation followed by angiographic management, and the remaining 27 patients received selective surgeries. The surgeries were carried out at days 4-43 after trauma by using the external fixators in seven patients, the plates in nine, the iliosacral screws in four, the plate plus external fixator in four, the iliosacral screw plus external fixator in two and the iliosacral screw plus plate in one, respectively. Results The follow-up for an average 15 months among 30 patients showed bony union in 27 patients, malunion in two and nonunion in one. The clinical outcomes of 29 patients was assessed with the Majeed' s scoring system, which showed excellent result in 26 patients, good in nine and poor in three, with excellence rate of 90%. Secondary injuries caused by unstable fractures occurred in two patients and iatrogenic injuries caused by fracture reduction in two.Conclusions Unstable pelvic fractures should be treated surgically as early as possible to restore the stability of the full pelvic ring and prevent the secondary injury, while the conservative treatment is unreliable.Attentions should be paid to avoidance iatrogenic injuries, especially nerve injuries, during the course of treatment.
8.The clinical and pathological characteristics of breast ductal carcinoma in situ and with microinvasion

Objective To study the clinical and pathological characteristics of breast ductal carcinoma in situ (DCIS) and with microinvasion (DCIS-MI). Methods From June 1999 to June 2003, a total of 41 patients with DCIS and DCIS-MI patients were retrospectively studied. Results Pathology varies between DCIS type Ⅰ, type Ⅱ and DCIS-MI patients. Mammographic calcification was the only presenting symptom in 9.76% of patients with DCIS accounting for 50% of these cases. The accordance rate of mammography with final diagnosis was 80%, and 33.3% of mammography in DCIS-MI type 2 showed mass or chaos of gland construction. The initial symptom in 57% of patients was nipple discharge. For fiberoptic ductoscopy, the accordance rate was 95.65%. There were two local recurrent cases in DCIS-MI type 2. Conclusion Breast ductal carcinoma in situ and with microinvasion had their own characteristics, including different prognosis.
9.A retrospective analysis of high risk factors for nonunion of femoral neck fractures

Objective To retrospectively study postoperative nonunion of femoral neck fractures so as to identify high risk factors for their nonunion.Methods A total of 965 patients with complete clinical data out of the 1 323 patients with femoral neck fractures treated by internal fixation with cannulated compression screws from January 2003 to January 2011 were included in the study.Logistic muhiple regression method was used to analyze five factors including patients' age,gender,injury mechanism (whether high energy injury or not),fracture type (with or without displacement) and medical comorbidities [American Society of Anesthesiologists Physical Score (ASAS) grade] and their correlation with fracture healing to investigate the high risk factors for the nonunion of femoral neck fractures.Results Quantitative assay of the factors affecting healing of femoral neck fractures was ranged as follows:medical comorbidities (ASAS grade),injury mechanism (whether high energy injury or not),fracturc type (with or without displacement),gender and age.Conclusion Age > 50 years,females,displacement fracture,high energy injury,and ASAS grade above Ⅲ are high risk factors for nonunion of femoral neck fractures.
10.Affinity maturation of a single chain antibody against VEGFR2 by hydrophilic shuffling.

Abstract: This study is to improve the affinity of scFv-AK404R against VEGFR2. The secondary mutational library was constructed by hydrophilic shuffling in CDR3 region of the heavy chain. VEGFR2-specific screening was performed by phage display technology and the protein of mutants was expressed in periplasm of E.coli HB2151 and purified by affinity chromatography. The affinity constant of scFvs was measured by competitive ELISA, and the structure of scFvs was analyzed by bioinformatics. The result showed that a library with 6.4x10(5) scFv members was established by electro-transformation. Two mutated clones with high absorbance value were isolated after screening. After purification by affinity chromatography, electrophoretically pure scFv proteins were obtained. The competitive ELISA showed that the affinities of WZ01 and WZ02 were three times higher than that of the parental AK404R, and bioinformatics analysis showed that the enlarged contact surface and fitted closely with KDR3 surface may be the reasons for improved affinity. These results suggest that introducing hydrophilic amino acids to the heavy chain CDR3 region is an effective approach to improve the affinity of scFv.
