1.Assessing drug targeting of Yougui Pill, Zuogui Pill, and their disassembled prescriptions using infrared thermography.

OBJECTIVETo dynamically assess drug targeting of Yougui Pill (YP) and Zuogui Pill (ZP) using infrared thermography.
METHODSIn this self-control experiment, five healthy volunteers were recruited. By using infrared thermography 10 to 11 thermal images of different body locations were taken from each participant after they took warm water, YP, ZP, and their dissembled prescriptions at 30, 70, 100, 130, and 160 min, respectively. The heat values in the lower quadrant abdomen, uterus, Du channel, and Shenque (CV8) were statistically analyzed after scanning for 125 times.
RESULTSAdministration of YP and its disassembled prescriptions enhanced the heat value of the locations of the Du channel and Shenque (CV8), but did no enhance the heat value of the lower quadrant abdomen at 30 min. Administration of ZP and its disassembled prescriptions reduced the heat value in the locations of the lower quadrant abdomen, uterus, Du channel, and Shenque (CV8) at each time point.
CONCLUSIONThe drug targeting of ZP and YP focused on the locations of the Du channel and Shenque (CV8), not on the locations of the lower quadrant abdomen or uterus.

2.THE HEALTH FUNCTION OF FERMENTED RICE BRAN EXTRACT BY BACILLUS NATTO

Objective:To explore the resource of rice bran by comparing antioxidative activities and growth promotion of Bacillus bifidus between supernatant of formented rice bran(RBF) by Bacillus natto and water extract of rice bran(RBW) . Method:The reducing capacity the?OH and ??O 2 scavenging capacity and the inhibitory effect to oxidize lard were determined in vitro. The growth promotion for Bacillus bifidus by photodensity with simulated condition was investigated in vivo. Results:RBF and RBW had antioxidant activity in vitro. The IC50 of scavenging?OH and ??O 250 of RBF was 3.55 mg/ml and 23.5 mg/ml,0.3 and 10 folds higher than that of RBW respectively. In inhibiting oxidation of lard,RBF had a little higher antioxidative activities than RBW,near VE. RBF and RBW could promot growth of Bacillus bifidus by 65.2% and 17.8% respectively. By enzymatic digestion,the promotive rate of RBF was still 51.6%. Conclusion:RBF had higher antioxidant activity and growth promotion to Bacillus bifidus.

5.Effect of estrogen deficiency on the expression levels of nuclear factor-kappa B and interleukin-17 in periodontal tissues

BACKGROUND:There is a very close relationship between osteoporosis and periodontal disease in postmenopausal women, but the mechanism remains unclear. OBJECTIVE:To investigate the expression of nuclear factor-κB in the alveolar bone andinterleukin-17 in the serum and gingiva in the mouse model of osteoporosis caused by ovariectomy. METHODS:Female mice aged 3 months were randomly divided into ovariectomy and sham operation groups. At 6 months after surgery, the mouse models were evaluated histologically on the submandibular bone and thigh bone stained with hematoxylin and eosin. In the submandibular bone, the expression levels of OCN and Runx2 were detected by RT-PCR, and the expression level of nuclear factor-κB was detected by immunohistochemical staining and western blot assay. Besides, the expression level of interleukin-17 in the serum and gingival homogenate was evaluated using Cytometric Beads Array. RESULTS AND CONCLUSION:The thigh bone in the ovariectomy group revealed the thin cortical bone, enlarged marrow cavity, and increased resorption lacunae, as well as fewer, thinner trabeculae with lower density and irregular structure. Compared with the sham operation group, the expression levels of OCN and Runx2 in the alveolar bone were decreased in the ovariectomy group. The activation of nuclear factor-κB (P65)appeared with P65 positive expression in the submandibular bone in the ovariectomy group, and the relative expression level was higher than that in the sham operation group. The serum level of interleukin-17 in the ovariectomy group was higher than that in the sham operation group, but the level in the gingival tissue showed no significant difference between the two groups. These results indicate that estrogen deficiency after ovariectomy can activate nuclear factor-κB signal pathway to play a role in periodontal osteolysis. However interleukin-17 in the local periodontal tissue may not be a key cytokine to damage the periodontal tissue.

8.Nasopharyngeal Carriage Status of Opportunistic Pathogens in Old People and Its Significance

OBJECTIVE To investigate the nasopharynx carriage status of opportunistic pathogens in non-infectious old people.METHODS The totally of 592 pharyngeal samples from old persons were collected.RESULTS The isolated rates of main opportunistic pathogens were Haemophilus parainfluenzae,Stomatococcus mucilaginosus,Streptococcus pneumoniae and Klebsiella pneumoniaa.The isolated rate of single opportunistic pathogen was 45.8% and that of multiple opportunistic pathogens was 3.9%.The total carriage rate of opportunistic pathogen was 49.7%.The carriage rate of G+ or G-bacteria was 13.2% or 86.8%,respectively.The isolated rates of 2 multiple pathogens,S.pneumoniae and K.pneumoniae were increased with aging.The isolated rate of K.pneumoniae was higher in persons with more than 3 kinds of underlying diseases.CONCLUSIONS Aging,pharyngeal carriaging of S.pneumoniae,H.parainfluenzae,K.pneumoniae,et al and with multiple underlying diseases might be the risk factors for elder people to suffer from infectious respiratory disease.Isolating pharyngeal opportunistic pathogen among non-infection people is necessary and significant for prevention and treatment of infectious respiratory disease.
9.Metabonomics analysis of effects induced by PCBs and high fat diet combined expo-sure in SD rats

Objective To explore the effect on metabolism of glucose and lipids, potential toxicity mechanism and possible biomarker candidates by analyzing urine metabonome changes of rats after oral administration of polychlorinated biphenyl congeners( PCBs) and high fat diet alone or in combination.Methods Male SD rats were divided randomly into control group,high fat diet group, PCBs group and combination group of PCBs and high fat diet.Urine samples were collected after 6-week treatment, 1 H NMR spectra were performed and analyzed by principal component analysis ( PCA) . Results The PCA scores plot of urine 1 H NMR data showed that the combined group could be easily distinguished from the other three groups, suggesting great difference in metabolism.The loading plot of the PCA revealed significant increase in the levels of lactate, glucose, creatine, 2-hydroxy-isovaleric acid and reduction in the levels of citrate, succinate, taurine, hippurate and trimethylamine oxide ( TMAO) in the combined exposure group after six-week exposure.Conclusion The altered levels of metabolites induced by combined exposure of PCBs and high fat diet may be related to the injury to mitochondrial function, reduction of energy metabolism in tricarboxylic acid cycle (TAC).These effects possibly lead to perturbations in the metabolism of glucose, lipid and amino acids.The altered metabolites may be considered biomarker candidates of toxicity induced by exposure to PCBs and high fat diet.
10.Effects of oxidative stress on the expression of endothelial-leukocyte adhesion molecule-1 in porcine trabecular meshwork cells.

OBJECTIVETo investigate the effects of oxidative stress on the expression of endothelialleukocyte adhesion molecule-1 (ELAM-1) in porcine trabecular meshwork cells and to elucidate the relationship between the expression of ELAM-1 and IL-1alpha.
METHODSPrimary cultured porcine trabecular meshwork cells were cultured without serum and treated with 1 mmol/L H2O2 with or without pretreatment of IL-1ralpha of different concentrations. The expression of ELAM-1 was tested by immunocytochemistry.
RESULTSELAM-1 immunocytochemistry staining was positive in the H2O2-treated porcine trabecular meshwork cells. It was negative in the porcine trabecular meshwork cells pretreated with IL-1alpha of high concentrations (180 microg/ml and 600 microg/ml).
CONCLUSIONSOxidative stress can induce the expression of ELAM-1 in porcine trabecular meshwork cells. In the in vitro cell system, IL-1ralpha of high concentration can inhibit the expression of ELAM-1 induced by oxidative stress.
