1.The management of duodenal ulcer with acute perforation by laparoscopic surgery

Objective To evaluate the long term effects of laparoscopic surgery on duodenal ulcer with acute perforation. Methods From 1995, Twenty-five patients with duodenal ulcer perforation and diffused peritonitis were diagnosed definitely by TV laparoscopic technique, and the perforations were sutured and repaired by greater omentum under laparoscopy. Six cases of them were performed parietal cell vagotomy simultaneously. After operation the drain was put into abdominal cavity and H2 receptor blocking agent, proton pump inhibitor or plus antihelicabater pylori therapy were used. Results All operations of 25 cases were accomplished under laparoscopy. Whatever operative complications had not happened. Average operation times were 55 minutes. The total hospital days were 6 in average. Postoperative follow-up ranged from 0. 5 -3 years, no duodenal ulcer recurred. Conclusion The laparoscopy adopted in treating duodenal ulcer perforation is a procedure with minimal invasion and scarceness of postoperative complication; its clinical effectiveness is as good as laparotomized operation.
4.Effects of BUSHEN YIZHI DECOCTION on interspace explore learning and memory in rat model with Alzheimer's disease

Object To observe the improving effects of BUSHEN YIZHI DECOCTION (BSYZD) on interspace explore learning and memory in rat model with Alzheimer's disease. Methods Eighty 15-month Wistar rats were induced by ip D -galactose for four weeks and injection of basal nucleus of Meynert with ibotenic acid (IBO) to make AD model, then randomly divided into AD model group, Hup-A treated group, BSYZD (high dose, 12 g/kg?d) treated group and BSYZD (low dose, 6 g/kg?d) treated group, and also normal aged and young groups. After treating for four weeks, Morris water maze was used to assess the improvement of rat interspace explore learning and memory. Results In the interspace explore experiment, the significant differences were observed between the model, Hup-A treated groups and normal aged, young groups (P0.05). Conclusion BSYZD possesses a certain preventive effects on interspace explore learning and memory in AD rat model.
6.Effect of pulse electro-magnetic fields on the repair of osteo chondral fracture in rabbits

Objective:To study the effects of pulse electro-magnetic fields (PEMFs) on the healing of tibial condylar osteochondral in rabbits.Methods:Sixteen rabbits were divided into two groups,and osteochondral fractures of left tibia condyle were created,internal fixation was performed with steel wire.Group 1 was constantly treated with PEMFs six hours per day;group 2 was not treated.The results were studied by gross morphology,light microscopy and elect ro-microscopy.Results:75% and 13% fractures were repaired by hyalin-like cartilage in group 1 and in group 2 respectively.Conclusion:PEMFs are essential to facilitation of the cartilage repair.
7.Cloning and expression of genotype B and C hepatitis B virus in eukaryotic cells

Objective To construct recombinant full length genotype B and C hepatitis B virus (HBV)and to examine HBV DNA replication and hepatitis B surface antigen(HBsAg),hepatitis B e antigen(HBeAg)expressions in Huh7 cells. Methods The full length genotype B and C HBV DNA were extracted and amplified from two HBV infected patients. The recombinant plasmids were constructed by inserting the amplified HBV fragments into the eukaryotic expression vector,pHY106,which were then transfected into Huh7 cells. The cells transfected with blank pHY106 vector were used as control. HBV DNA replication at 72 hours of transfection was detected by Southern blot. The HBV DNA levels in Huh7 cells at 24,48,72,96 and 120 hours of transfection were determined by real-time polymerase chain reaction(PCR).Meanwhile, the HBsAg and HBeAg expression levels in the supernatants at 24,48,72,96 and 120 hours were determined by enzyme linked immunosorbent assay(ELISA).Results The recombinant plasmids expressing genotype B or C HBV DNA were successfully constructed.The HBV replicative intermediates in HBV core particles,including rcDNA dsDNA and ssDNA,were detected by Southern blot.HBV DNA level could reach 8 lg copy/mL which was by real-time PCR. HBsAg and HBeAg levels determined by ELISA peaked at 72 hours after transfection and then declined gradually. Conclusions The recombinant plasmids inserted with genotype B or C HBV DNA are constructed successfully, which can express high levels of HBsAg and HBeAg in Huh7 cells. This system provides a platform for studying the pathogenesis of B and C genotype HBV, the interaction between HBV and host, as well as exploiting new drugs against HBV.
8.Detection and their clinical significance of cytokines levels in serum and synovial fluid from patients with rheumatoid arthritis

Aim To study cytokine(CK) changes of IL-6,IL-8,sIL-2R and TNF-a levels in sera from patients with juvenile rheumatoid arthritis(JRA) and rheumatoid arthritis(RA) and its correlation with the conventional inflammation indexes,erythrocyte sedimentation rate(ESR) and C-reactive protein(CRP). Methods Levels of IL-6,IL-8,sIL-2R and TNF-a in serum from 30 JRA patients and 34 RA patients,in the synovial fluid (SF)from 4 JRA patients,7 RA patients,6 osteoarthritis(OA) patients and 9 meniscitis(MT) patients were detected by sandwich ELISA. Results As compared with control group,① levels of IL-6 and SIL-2R in sera from 30 JRA patients and 34 RA patients and levels of IL-8 in sera from 30 JRA patients possessed marked difference P∨ 0.01 and P∨ 0.05,respectively.② There were significant difference among serum IL-8 and sIL-2R levels from systemic JRA patients, pauciaticular JRA patients,and serum IL-6 levels from polyarticular JRA patients(P<0.01). ③ There were remarkable difference among SF sIL-2R levels from 4 JRA patients and 7 RA patients and in SF IL-6 levels from 7 with RA patients.④ There were obvious correlativity between serum IL-6,sIL-2R levels and changes of serum ESR and CRP from the JRA patients(r=0.532 and r=0.621,respectively). Conclusion ① IL-6 and sIL-2R exhibit a close correlativity with the activity of JRA and RA,which is the main index of activity of JRA and RA diseases.② sIL-2R might not only participate in systemic pathologic process in JRA and RA diseases,but also serves as a major CK in local articular injuries.IL-6 participates in local articular injuries too,and seems to play a more important role in local articular injuries of RA.③ IL-8 may participate in systemic pathologic process of JRA,but play a minor part in local articular injuries.
10.MORPHOMETRIC ANALYSIS ON THE MITOCHONDRIA OF MYOCARDIUM IN EXERCISE TRAINED RATS

Adult male rats were divided at random into two groups and trained on the treadmill running schedules, 60 min/day and 10min/day for group A and B separately. Four weeks later the ultrastructure of the left ventricular myocardium was examined and the specific surface of mitochondria wasmeasured. Comparing to the corresponding control group, the specific surface of mitochondria in the two groups of trained rats was decreased significantly. (5.883+0.12 um and 6.050?0.12 um to 6.564?0.17 um). This means myocardium mitochondria in trained rats were swelling. The reason might be that the myocardium injury was induced by exercise training.
