OBJECTIVETo compare the therapeutic effect and toxicity of chemotherapy, used alone or in combined with Shenqi Fuzheng Injection (SFI), for the treatment of advanced colorectal carcinoma (ACRC).
METHODSOne hundred and fifty-two patients with ACRC were equally randomized by digital table, to the treated group, treated by chemotherapy of FOLFOX regimen combined with SFI, and the control group treated by FOLFOX regimen alone. The therapeutic effect and adverse reaction of the treatment in patients were assessed.
RESULTSThe effective rate (CR +PR) was 63.2% (48/76) in the treated group and 46.1% (35/76) in the control group, showing significant difference between the two groups (P < 0.05). The median survival time in the two groups was 31 weeks and 28 weeks respectively. CD4/CD8 ratio was significantly increased in the treated group (1.56 +/- 0.21, 1.64 +/- 0.28, P < 0.05), but significantly decreased in the control group (1.58 +/- 0.22, 1.46 +/- 0.33, P < 0.01). Quality of life in the former group was higher than that in the latter group (P < 0.05). Times/case of nausea, vomiting, leukopenia occurring in the control group was more than those in the treated group A (P < 0.05).
CONCLUSIONBy combining with SFI, some adverse reactions of chemotherapy (such as nausea, vomiting, leukopenia) and its influence on patients' immunity could be alleviated in treating ACRC, which might enhance the efficacy of chemotherapy, and improve the quality of life and prolong the median survival time in patients.

6.Proteasome inhibitor induces substantia nigra degeneration and inclusion body formation in rats
Chinese Journal of Neuromedicine 2009;8(5):455-458
Objective To observe nigral degeneration with inclusion body formation in rats and their behavioral changes after treatment with proteasome inhibitor, and investigate the role ofproteasomal dysfunction in the pathogenesis of Parkinson disease. Methods Lactacystin, a selective proteasome inhibitor, was injected stereotaxically into the lett substantia nigral pars compacta of the rats, and an equal volume of saline was injected in the control group. Spontaneous behavioral abnormalities and apomorphine-induced contralateral rotations of the rats were observed after the injection. The pathological changes in the substantia nigra were detected using Nissl staining, and the expressions of tyrosine hydroxylase (TH) and a-synuclein in the substantia nigra and TH expression in the striatum were investigated by immunohistochemistry. The ultrastructural changes in the substantia nigral pars compacta were observed with transmission electron microscope. Results Seven days after lactacystin injection, the rats exhibited reduced apontaneous activities, tremor, progressive bradykinesia, and apomorphine-induced contralateral rotation. Nissl staining 3 weeks after lactacystin injection revealed significantly reduced neurons in the lett substantia nigra pars compacta and loosened structures of the Nissl bodies;immunohistochemistry demonstrated obvious degeneration in the lett substantia nigra pars compacta, where the TH-positive cells were significantly decreased with enhanced expression of a-synuclein. The number of TH-positive fibers in the striamm was significantly reduced 3 weeks afterlactacystin injection, and electron microscopy revealed the formation of inclusion bodies as a result of protein aggregation. Conclusion Lactacystin injected stereotaxically into the lett substantia nigrai pars compacta can induce substantia nigra degeneration, inclusion body formation and behavioral changes in rats, suggesting the applicability of lactacystin for establishment of animal models of Parkinson disease. Proteasomal dysfunction may play an important role in the pathogenesis of Parkinson disease.

To investigate the influence of Anxin granules combined with tirofiban on acute myocardial infarction (AMI) Patients after elective percutaneous coronary intervention (PCI). One hundred and twenty AMI patients were randomly divided into treatment group and control group. The patients in the two groups were all given Tirofiban 30mins before PCI . The treatment group was added Anxin granules 30 mins before and after PCI. Tissue factor (TF) and von willebrand factor (vWF) were tested at 6 hours after operation. Syndromatology alteration of traditional Chinese medicine (TCM) and bleeding complications were observed at 4 weeks after operation. Both TF and vWF at 6 hours after operation of the treatment group was lower than the control group significantly (P < 0.01), while the condition of myocardial ischemia at 90 mins after operation of the treatment group was better than control group with significance. The syndromatology alteration of TCM especially spontaneous perspiration and hypodynamia of the treatment group were improved significantly compared to control group 4 weeks after operation. All patients in both groups had no bleeding complications and thrombopenia. The study suggests that Anxin granules combined with tirofiba can improve the clinical efficacy and the endothelial function of AMI patients after PCI with no increase in bleeding events.

OBJECTIVE: To investigate the relationship between the perfusion CT features and the clinicopathologically determined prognostic factors in advanced gastric cancer cases. MATERIALS AND METHODS: A perfusion CT was performed on 31 patients with gastric cancer one week before surgery using a 16-channel multi-detector CT (MDCT) instrument. The data were analyzed with commercially available software to calculate tumor blood flow (BF), blood volume (BV), mean transit time (MTT), and permeability surface (PS). The microvessel density (MVD), was evaluated by immunohistochemical staining of the surgical specimens with anti-CD34. All of the findings were analyzed prospectively and correlated with the clinicopathological findings, which included histological grading, presence of lymph node metastasis, serosal involvement, distant metastasis, tumor, node, metastasis (TNM) staging, and MVD. The statistical analyses used included the Student's t-test and the Spearman rank correlation were performed in SPSS 11.5. RESULTS: The mean perfusion values and MVD for tumors were as follows: BF (48.14+/-16.46 ml/100 g/min), BV (6.70+/-2.95 ml/100 g), MTT (11.75+/-4.02 s), PS (14.17+/-5.23 ml/100 g/min) and MVD (41.7+/-11.53). Moreover, a significant difference in the PS values was found between patients with or without lymphatic involvement (p = 0.038), as well as with different histological grades (p = 0.04) and TNM stagings (p = 0.026). However, BF, BV, MTT, and MVD of gastric cancer revealed no significant relationship with the clinicopathological findings described above (p > 0.05). CONCLUSION: The perfusion CT values of the permeable surface could serve as a useful prognostic indicator in patients with advanced gastric cancer.

Chinese Medical Journal 2008;121(18):1792-1795
BACKGROUNDHyperostosis is a common pathological change among people more than 50 years old; it is connected with many risk factors, which are all indefinite. The aim of this study was to prospectively investigate the correlation between obesity and female hyperostosis.
METHODSTotally 4326 females were included in this study and their basic information including their age, stature, body weight, course of disease, symptoms, medical complications, frequency of exercise and smoking, and X-ray and bone mineral density (BMD) examination results, was carefully collected for a statistical analysis. The t test or chi(2) test was used to evaluate the differences between two groups; an analysis of variance (ANOVA) was used to evaluate the differences among several groups; the relationship between hyperostosis and body mass index (BMI), age, medical complications, exercise, average BMD was analyzed using Logistic regression.
RESULTSThe incidence rate of hyperostosis in obese patients was higher than that in patients with normal weight (P = 0.000). Obesity was relevant to hyperostotic sites (P = 0.000), and the incidence of hyperostosis in one or several sites of the lumbar vertebrae, knee joints, and other sites was higher in obese patients than in patients with normal weight. There was also a difference in the extent of hyperostosis between these two groups. BMI had positive effects on the incidence and degree of hyperostosis, which were also relative to the sites of hyperostosis, and the BMI of patients without hyperostosis were much lower than those of the patients with hyperostosis in their lumbar vertebrae, knee joints, or multiple sites. Obesity, age, and exercise had positive effects on the incidence of hyperostosis (P = 0.002, 0.000, 0.018).
CONCLUSIONSObesity is a significant potential stimulant of hyperostosis, especially hyperostosis in knee joints and multiple sites; keeping fit might be an important way to prevent it.
