1.Relationships between the polymorphism of methylenetetrahydrofolate reductase gene C677T and hypertension, cardiac structure and function
Medical Journal of Chinese People's Liberation Army 2001;0(07):-
Objective Elevated plasma homocysteine(Hcy) level has been associated with the increased risk for cardiovascular disease. The level of plasma Hcy is related to methylene tetrahydrofolate reductase(MTHFR) gene C677T polymorphism. The aim of this study is to evaluate the relationships between MTHFR gene C677T polymorphism and hypertension, cardiac structure and function. Methods Genotype of MTHFR gene C677T polymorphism was detected by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) analysis and decided by electrophoresis and sequencing in 695 patients with essential hypertension and 509 age-matched normal controls.Echocardiography was performed in patients with essential hypertension. Results In hypertension group, the frequencies of TT genotype and T allele were significantly higher than those in control group(26.5% vs 20.6%,48.7% vs 42.4%;P=0.015,0.002).The essential hypertension odds ratio(OR) associated with MTHFR 677T allele was 1.300(95%CI:1.166 to 1.450).Logistic regression analysis indicated that MTHFR gene C677T polymorphism was an independent variable of hypertension(P=0.027).End diastolic dimension(EDD),interventricular septal thickness(IVST),left ventricular posterior wall thickness(LVPWT),left ventricular mass(LVM) and left ventricular mass index(LVMI) were higher in hypertensive patients with T allele than that in those with CC genotype, but all had no statistical significance(P=0.428,0.118,0.064,0.077,0.054).Conclusion The 677T allele in MTHFR gene may be a risk factor for hypertension.Hypertensive patients with MTHFR 677T had a trend of hypertrophy, but the relationship has no statistical significance. The relationship between MTHFR gene C677T polymorphism and left ventricular hypertrophy need to be further studied.
2.Effect of ASODN transfection targeting VEGF-C gene on proliferation and apoptosis of Hela cells
Cancer Research and Clinic 2009;21(7):437-439,443
Objective To explore effect on the proliferation and apoptosis of Hela cells in vitro by using vascular endothelial growth factor-C(VEGF-C) antisense oligonucleotides (ASODN). Methods VEGF-C ASODN was transfected into Hela cells by liposome-mediated; the cells transfected with the oligodeoxynuclecotide (SODN) and saline were used as control groups. The efficiency of transfection was detected by fluorescence microscope. The inhibitive rate of cell growth was detected with MTT methods. Apoptosis and cell cycle were evaluated using FCM. The mRNA expression of VEGF-C was determined by RT-PCR, and the expression of VEGF-C was determined by western blotting. Results VEGF-C ASODN had been transfected into Hela cells by liposome-mediated. The index of apoptosis after transfection 48 h was (19.39±1.81)%. G2/M phase cell increased after transfection, meanwhile the expression of VEGF-C degraded on the level of mRNA and protein (P<0.05). Conclusion Transfeeted with VEGF-C ASODN could down-regulate the expression of VEGF on the level of mRNA and protein, block the cell circle, inhibit cell proliferation and induce apoptosis in Hela cells in vitro.

Chinese Journal of Infectious Diseases 2001;0(06):-
Objective To study the antibiotic resistance mediated by the mar mechanism in clinical isolated Escherichia coli. Methods To analyse the antibiotic resistance pattern and the out membrane protein spectrum of clinical isolated E. coli . The plasmid pNJR3 2 carrying the wild type gyr A gene was transferred into clinical multiple resistant strain LC 1. The resistant pattern and plasmid pattern of LC 1 before and after plasmid transferring were compared. The mar OR of clinical strain LC 1 was cloned and sequenced through PCR. The nucleotides of mar gene were compared with the wild type issued by GenBank. Results Of all the 49 clinical strains , six strains express Mrp5, a mar specific out membrane protein. No changes were detected on susceptibility to fluoroquinolone of clinical strain LC 1 after pNJR3 2 transferred suggesting that gyr A was not the main reason contributing to fluoroquinolone resistance. Sequence analysis showed three mutant spots in mar OR of clinical strain LC 1. Conclusion There does exist mar mechanism in clinical isolated Escherichia coli resistant to fluoroquinolone. The mutation in mar OR may contribute to the mar phenotype of LC 1.
