1.Construction and expression of circular RNA in vitro

Objective To construct encoding RNA that can be cyclized in vitro by using the permuted intron exon(PIE)strategy in the maturation process of eukaryotic mRNA,and transfect it into HEK-293T cells for expression.Methods The sequences of 5'and 3'cyclic arms with groupⅠcatalytic intron,the internal ribosome entry sites(IRES)of Coxsackievirus B3(CVB3)and the target gene were selected to construct the template plasmid. Linearization plasmid template obtained by PCR was used to synthesize linear RNA through in vitro transcription(IVT),which then started in vitro cyclization(IVC)by the addition of cyclization reagents to obtain circular RNA(circRNA). RNA cyclization was confirmed by agarose gel electrophoresis and ribonuclease R(RNase R)digestion. HEK-293T cells were transfected with circRNAs respectively carrying enhanced green fluorescent protein(EGFP),firefly luciferase(Fluc),and influenza virus hemagglutinin(HA)IVR-180 genes,to verify their expression with in vitro.Results With RNA cyclization,the main band of agarose gel electrophoresis became smaller and small fragments appeared. After RNase R digestion,only some circRNA bands remained.HEK-293T cells transfected with EGFP-circRNA showed significant green fluorescence under the fluorescence microscope.The Fluc expression values of HEK-293T cells transfected with Fluc-circRNA were on average 20 times higher than non cyclized RNA,and the relative light unit(RLU)scaled up with the increase of Fluc-circRNA transfection dose. Western blot analysis showed that HA protein was successfully expressed in HEK-293T cells transfected with HA-circRNA.Conclusion In this study,linear RNA was successfully cyclized in vitro and different proteins were expressed,which lays a foundation of the research of new influenza vaccines and mRNA vaccines.
2.MRI compatibility and safety of cochlear implants

The increasing magnetic strengths used for magnetic resonance imaging (MRI) combined with a greater number of MRI patients with cochlear implants (CIs) raises possible safety issues.This paper reviews the interactions between MRI and CI,and assesses MRI compatibility and safety of CIs at 0.2,0.3,1.0,1.5 and 3.0 tesla (T).In a word,CIs represent a significantcontraindication to MRI.Only after careful evaluation of the individual risk and careful preparations can an MRI be safely performed.
3.Discussion of Management Technique Regulations of Medical Consumable Materials

4.Teachers’Practice in Improving the Training Quality of Neurology Clinical Pharmacists

Objective:To improve the training quality of neurology clinical pharmacist. Methods:The experience during the training of neurology clinical pharmacists in recent ten years was summarized. Results:The students were taught in groups according to their majors and promoted in stages according to the teaching plans. Besides,various teaching methods were carried out timely. After tutored by the methods mentioned above,the students could quickly raise their working ability to play good roles of clinical pharmacist and work independently with more confidence. Conclusion:The training quality of neurology clinical pharmacists has been effectively improved by teaching in groups,promoting in stages and providing various teaching methods timely.

6.Study on the effect of genistein and adriamycin on the proliferation of human ovarian cell A2780 and growth of tumor bearing nude mice

Objective To study the effects of genistein and adriamycin on the proliferation of human ovarian cell A2780 and growth of tumor bearing nude mice.Methods The ovarian cancer cell A2780 suspension were inoculated into nude mice (n=40 )to establish the animal models with ovarian tumor.These model rats were randomly divided into four groups,saline group and different genistein concentrations combined with adriamycin treatment group (high-dose,middle-dose and low-dose group)by intraperitoneal injection once a week.After treatment for four weeks,the ovarian tumors in all rats were peeled off and weighted for the measurement of tumor inhibition rate.The expression of c-myc in ovarian tumors were detected by immunohistochemistry.In vitro,after affected by different genistein concentrations combined with adriamycin,the expression of c-myc in A2780 cell of each group were detected by western blot.The proliferation of A2780 cell were determined by MTT method,and the invasiveness of cell by cell adhesion and transwell assay,the apoptosis rate and cell cycle were measured by flow cytometry. Results The tumor inhibition rates in middle-dose (40.26 ± 4.25)% and high-dose groups[,(44.81 ±5.74)%])were significantly lower than saline group (P<0.05).Immunohistochemistry results showed that c-myc monoclonal antibody in middle-dose and high-dose groups can significantly inhibit the activation of c-myc in ovarian carcinoma tissue.Compared with control group,the expression of c-myc in A2780 cell were significantly decreased,the proliferation and invasiveness of cell were inhibited,cells in G0/G1 phase were significantly increased(P<0.05)and in S phase were(P<0.05)decreased,and the apoptosis were induced by genistein combined with adriamycin in middle-dose and high-dose (P <0.05 ).Conclusion Genistein combined with adriamycin in certain doses could inhibit the proliferation and invasiveness of ovarian carcinoma,and induce its apoptosis.The mechanism may be related to the decreased expression of c-myc.
7.Biological role of microRNAs in gastric cancer

MicroRNAs (miRNA) are a class of small non-coding RNA molecules that are rich in content and are highly con-served. miRNA can regulate vital biological processes, including cell division, differentiation, and cell apoptosis. Their expression is dysregulated in many diseases. miRNA are essential in the initiation and development of various cancers. Recent miRNA studies have gradually moved from basic molecular studies of cancer onto areas of clinical application. The use of miRNA as biomarkers for cancer diagnosis and target molecules for cancer treatment is increasingly being identified. This review summarizes the potential applications of miRNA as biomarkers for diagnosis and prognosis of gastric cancer, drug response, and susceptibility prediction.
8.Diagnosis of isolated anomalous origin of right pulmonary artery from ascending aorta by echocardiography

Objective To evaluate the value of echocardiography in patients with isolated anomalous origin of right pulmonary artery from ascending aorta. Methods The eehoeardiographie results of five patients with isolated anomalous origin of right pulmonary artery from ascending aorta were reviewed retrospectively. Results Severe pulmonary hypertension occurred in all the patients,besides accompanied with patent duetus arterious, no other congenital malformations combined. Three patients underwent operation. Patent ductus arterious was missed in one patient's diagnosis. Another patient was missed isolated anomalous origin of right pulmonary artery from ascending aorta in several echoeardiographic examinations, once misdiagnosed as complete transposition of the great arteries. Conclusions There are many anatomic and hemodynamic similarities in patients with isolated anomalous origin of right pulmonary artery from ascending aorta. Grasping their common charaeters and exploring multi-position and multi-section are helpful to make correct diagnosis.
9. Research progress on intestinal flora interact with Chinese materia medica

There are a huge number of microorganisms in human intestinal, and they are closely related to human health. Their characteristics are huge amount and great variety, if the intestinal flora are imbalance, it could cause a variety of diseases. Traditional Chinese medicine can balance the intestinal flora, meanwhile intestinal flora can metabolize Chinese materia medica (CMM) to effective component. The research on CMM and the intestinal flora has great significance to realize the modernization of CMM. In this paper, we summarized the recent years’ research reports and its achievements on the interaction of intestinal flora and CMM, and provide a reference for further research about the relationship of intestinal flora and CMM.
10.Effect of perioperative blood transfusion on outcome of patients in orthopedic surgery

Objective To analyze the effect of perioperative blood transfusion in orthopedic surgery.Methods A total of 201 patients who underwent an orthopedic surgery in our hospital in October 2015 to September 2016 were reviewed.Hemoglobin concentrations,hematocrit,postoperative hospital stay,infection and other related data were analyzed.Results 90 patients were transfused with blood and in 111 patients were not during orthopedic surgery (massive blood transfusion group including 17 cases;non massive blood transfusion group including 94 cases).Hemoglobin concentrations of transfusion group (103.09±16.37)g/L was lower than the non transfusion group (115.79± 13.68)g/L before liying hospital,postoperative hospitalization time of transfusion group (12.012±8.36) was longer than that of non transfusion group (16.14 ± 10.47)days.Postoperative infection rate in non transfusion group was 1.11%,and in transfusion group was 15.32%,in massive transfusion group is 52.94%.Conclusion Perioperative blood transfusion is not conducive to the recovery of hemoglobin level patients in orthopedic surgery,prolong hospital stay and increase the rate of infection after surgery.Perioperative blood transfusion makes it difficult to support the outcome of the disease
