2.Relationship between coronary atherosclerosis and aortic pulse pressure in patients with primary hypertension

BACKGROUND: Both pulse pressure (PP) and aortic stiffness are the predictors of coronary artery disease (CAD). The relationship between aortic PP and atherosclerosis may be bi-directional. Some investigations have tested the relationship between coronary arteriongraphy-diagnosed coronary heart disease and aorta pulse pressure (APP).OBJECTIVE: To observe the relationship between coronary atherosclerosis and APP in patients with primary hypertension.DESIGN: Case control observation.SETTING: Department of Cardiology, Xuanwu Hospital, Capital University of Medical SciencesPARTICIPANTS: A total of 300 patients with primary hypertension who received the treatment in the Department of Cardiology, Xuanwu Hospital, Capital University of Medical Science between July 2002 and January 2005. The patients were all untreated hypertensive patients who were referred for a first diagnostic coronary angiography. All of them met the diagnostic criteria of international hypertensive association. Secondary hypertension, myocardial disease, valvular disease of the heart (VDH), heart failure, liver and renal insufficiency and so on were excluded. The patients, 170 male and 130 female, were aged (61±11 )years. Informed consents were obtained from all the patients.METHODS: A total of 300 untreated hypertensive patients were recruited for a first diagnostic coronary angiography. According to whether having coronary artery disease (CAD), the patients were divided into CAD group (143 in total, 92 male, 64%) and non-CAD group (157 in total, 82 male, 52%). Informed consents were obtained from all the patients.The following data were collected: invasive intra-aortic systolic blood pressure (ASBP) and intra-aortic diastolic blood pressure (ADBP), extent of coronary artery disease, and basic clinical materials of the patients. All the observed index were expressed as Mean±SD. Independent sample t test was used in the comparison between two groups, and P < 0.05was set as significant difference.MAIN OUTCOME MEASURES: ASBP, ADBP and BP, ratio of stroke volume (SV), which reflects aortic stiffness, to aortic pulse pressure (APP); The number of branch and stegnotic extent of coronary artery disease (Cases with stegnotic extent < 50% were included in non-CAD group; Basic clinical indexes and routine biochemical indexes of patients after admission (including fasting blood glucose, serum creatinine, blood lipid and so on).RESULTS: The enrolled 300 patients with hypertension all participated in the result analysis. In the whole population, ASBP and PP were significantly higher in CAD group than non-CAD group [(150.3±26.5) vs. (145.6±23.3) mm Hg, P < 0.05;(77.1±22.7) vs. (70.4±19.3) mm Hg, P < 0.05)]. The ratio of APP to SV in CAD group was markedly higher than that in non-CAD group(1.20±0.44) vs. (0.96±0.33), P < 0.05]. Fasting blood glucose of patients in the CAD group was significantly higher than that in the non-CAD group [(1.38±0.27) vs. (1.08±0.28) mmol/L, P < 0.01]. Serum creatinine of patients in the CAD group was significantly higher than that of non-CAD group [(11.98±2.15) vs. (11.19±1.58) μ mol/L, P <0.01]. High-density lipoprotein cholesterol of patients in the CAD group was significantly lower than that in the non-CAD group [(0.54±0.13) vs. (0.62±0.18) mmol/L, P< 0.01].CONCLUSION: Atherosclerosis may further aggravate the aortic stiffness and then cause the increase of APP in patients with primary hypertension.
3.Investigation of House-dust Mite Fauna in School Dormitory in Guangzhou City

Objective To understand the house-dust mite fauna in the school dormitory in Guangzhou city.Methods With the direct smear method and saturation solution of NaCl floatation method,a total of 970 bed-dust samples collected from the five school dormitory buildings were examined,from Oct.to Dec.in 2006.The house-dust mite in the samples were counted and classified.Results The prevalence rate of mite was 98.76%.About 72% of the samples,the number of mites was 500 and 3 000/g dust.Mean number of the mites in bed-dust samples showed a significant difference between the male and female student dormitory,and among five dormitory buildings.The mean number of mites usually decreased as the floor increased.Dermatophagoides pteronyssinus,Dermatophagoides farinae,gamasid mites,scab mites and pyemotidae etc were found in the samples.Conclusion House-dust mites universally exist in bed-dust of the school dormitory in Guangzhou city and the number is larger.Dermatophagoides pteronyssinus,Dermatophagoides farinae are the dominant kinds among the mites.
4.Evaluation of reconstruction of circular ligament with fascia lata for the treatment of old capitulum of humerus dislocation

[Objective]To evaluated the effect of reconstruction of circular ligament with fascia lata for the treatment of old capitulum of humerus dislocation.[Method] Twenty-four cases were included in this study with ranged age from 5 to 14 years old.Surgical operation time was 6 months to 5 years after injury.All cases received recontruction of circular ligament with fascia lata,without ulna recection.Internal fixation was used to fix the humeroradial joint.[Result]Eleven cases were followed up for and 5.9 years(ranged,5-7 years).Six cases showed rotation restriction,with average of 69?.Four cases showed flexion-extension restriction with an average of 21?.Sublocaiton was found in 1 case.Unlar nerve injury was found in 2 cases.Kirschner wire broken was found in 4 cases.Simotaneous radial nerve injury was found in 1 case.[Conclusion] It should be taken very carefully if patient's age is over 13 years old.Fascia lata is satisfactory replacement for circular ligament.

7. Effect of matrine on sensitivity of A549/DDP cells to cisplatin by inhibiting microRNA-192

Objective: To investigate the effect of matrine on increasing the sensitivity of A549/DDP cells to cisplatin by regulating microRNAs. Methods: Data mining was performed on a microarray dataset (accession ID: GSE43249) from GEO database using bioinformatics analysis. Then microRNAs which related to the sensitivity of A549 cells to cisplatin were selected as candidate microRNAs. A549 and A549/DDP cells were cultured in vitro. The expression changes of candidate microRNAs were confirmed by qRT-PCR. The effect of matrine on the level of candidate microRNAs was also determined by qRT-PCR in A549/DDP cells. The level of microRNA-192 (miR-192) in A549/DDP cells was up-regulated by transfecting sequence of miR-192mimic. The CCK-8 assay was used to observe the effect of matrine and (or) miR-192 up-regulation on the sensitivity of A549/DDP cells to cisplatin. Results: Data mining Results: showed that there were 11 microRNAs with significant changes in A549/DDP cells compared with cisplatin-sensitive A549 cells (P < 0.05). The changes of miR-192 and miR-194 expression were confirmed in vitro. Compared with untreated A549/DDP cells, matrine treatment reduced the level of miR-192 in A549/DDP cells (P < 0.05). Matrine treatment increased the sensitivity of A549/DDP cells to cisplatin. Up-regulation of miR-192 could antagonize this effect and induce the resistance of A549 cells to cisplatin. Conclusion: Matrine may increase the sensitivity of A549 cells to cisplatin by inhibiting miR-192.

10.Intermedin inhibits pulmonary collagen synthesis in rats with pulmonary hypertension induced by high pulmonary blood flow

[ ABSTRACT] AIM:To explore the regulatory effect of intermedin ( IMD) on pulmonary collagen synthesis and ac-cumulation in rats with pulmonary hypertension induced by high pulmonary blood flow.METHODS: Healthy male SD rats (n=20) were randomly divided into control group (n=7), shunt group (n=7) and shunt with IMD group (n=6).The shunting of abdominal aorta and inferior vena cava was produced in rats of shunt group and shunt with IMD group.After 8 weeks, IMD was administered into the rats of shunt with IMD group subcutaneously by mini-osmotic pump for 2 weeks.Mean pulmonary artery pressure (mPAP), relative medial thickness (RMT) of pulmonary arteries, contents of hydroxyproline, collagen type I and III, bone morphogenetic protein-2 (BMP-2), and the mRNA expression of procollagen I and III in lung tissues were measured and compared.RESULTS:Compared with control group, mPAP and RMT of medium and small pul-monary arteries in the rats of shunt group were significantly increased.Meanwhile, the lung hydroxyproline, collagens I and III and BMP-2 contents, and the mRNA expression of lung procollagen I and III were all significantly increased compared with control group.However, IMD significantly decreased mPAP, alleviated the changes of pulmonary vascular micro-struc-ture, decreased the collagen accumulation and pulmonary tissue homogenate BMP-2 contents, and inhibited the mRNA ex-pression of procollagen I and III in the lung tissue of shunting rats.CONCLUSION:IMD plays a protective role in the de-velopment of pulmonary hypertension and pulmonary vascular structural remodeling induced by high blood flow by inhibiting pulmonary collagen synthesis and accumulation, possibly in association with the BMP-2 pathway.
