1.High Expression of mdlA Gene in Pichia pastoris and Study of Properties of the Recombinant Lipase

The mono-and diacylglycerol lipase(MDGL)-encoding gene(mdlA) was inserted into methanol-inducible expression vector pPIC9K.The linearized recombinant plasmid was uransformed into chromosome of Pichia pastoris GS115 strain by electroporation.Some high-copy transformants(His~(+)Mut~(+)) were picked up by G-418 and conformed by PCR.SDS-PAGE analysis indicated efficient expression of recombinant MDGL.Some enzymatic properties of the recombinant MDGL were also determined.The activity of the recombinant MDGL was up to 325U/mL under the optimal conditions and no activity was detected towards olive oil.The amount of fatty acid produced by the catalysis of recombinant MDGL and triacylglycerol lipase had a increase of 93.5% over triacylglycerol lipase lonely.

How to grasp the developing direction of Traditional Chinese Medicine(TCM) theory is the kernel of the TCM subject.In studying the theory of TCM,the three key propositions,how to keep the self main body,how to show the theoretic value and how to perform the durative innovation,must be answered.For keeping the main body,it must be clarified in unscrambling the origin,and be hold in the changeable reform,and be stand out under the scientific background.For showing the theoretic value of TCM,its academic value must be embodied in life(sciences),its leading value must be displayed during the subject development,and its guiding value must be emerged in clinical practice.For performing the durative theoretic innovation,it must be extended during inheriting the classical theory,and be improved in clinical practice,and be created in interdisciplinary crossing.Only in this way,would the basic theory of TCM acquire a breakthrough.
3.Research on diagnosis standard of phlegm-dampness constitution of TCM

Classification is a foundational problem in TCM Constitution Theory,phlegm-dampness constitution is one kind of elementary constitutions type.To normalize and deepdeepen research on classification of Constitution,we generalize the characteristics and establish diagnosis standard of phlegm-dampness constitutions.In our study,we examined gene expression profiles of peripheral blood cells by using Affymetrix GeneChip Human Genome U133A,revealed special expressed genes of phlegm-dampness constitutions.
4.Effects of professional benefit recognition to stress coping of tuberculosis nurses

Objective To investigate the effects of professional benefit recognition to stress coping of tuberculosis nurses. Methods A total of 82 tuberculosis nurses were assigned into experimental and control groups by random digits table method with 41 cases each. The experimental group was given four times of professional benefit recognition interventions in additional to regular training, while control group was just given regular training. The two groups were asked to complete the Professional Benefit Recognition questionnaires and Stress Coping Style questionnaires before the intervention and 1 month after the intervention, respectively. Results Before the intervention, the scores of the Professional Benefit Recognition questionnaires and Stress Coping Style questionnaires between two groups had no significant difference (P>0.05). After the intervention, the Professional Benefit Recognition questionnaires scores of experimental group increased from ( 2.93 ± 0.40) points to (3.81±0.38) points that was significantly better than that of control group(2.99±0.39) points, and there were significant differences (t=49.337,-60.834, P<0.05). The immature and mixed Stress Coping Style questionnaires scores of experimental group: self- accusation, illusion, avoidance, rationalization were (0.51 ± 0.13) , (0.51 ± 0.24), (0.56 ± 0.21), (0.51 ± 0.16) points, which were significantly lower than before the intervention (0.70 ± 0.09), (0.65 ± 0.10), (0.67 ± 0.14), (0.67 ± 0.08) points and that of control group (0.63 ± 0.16), (0.65 ± 0.18), (0.68 ± 0.18), (0.64 ± 0.13) points, with the difference being significant (t=-4.071--2.541, P<0.05). The mature coping scores of the experimental group:problem solving, appealing were (0.70 ± 0.10), (0.67 ± 0.13) points, which were significantly higher than that of control group (0.63 ± 0.12), (0.58 ± 0.15) points and that of experimental group before the intervention (0.55 ± 0.12), (0.51 ± 0.11) points, with the difference being significant (t=2.308, 2.098, P <0.05). Conclusions Professional benefit recognition interventions can improve professional benefit recognition and stress coping style, is beneficial to nurses' physical and mental health and nursing quality.
5.THE PREGNANCY IN PATIENTS WITH PROSTHETIC HEART VALVE (REPORT OF FIVE CASES)

It is still a controversy whether a patient with prosthetic heart valve can have a normal pregnancy. Here we report five cases of such patients, of which two with a tissue valve and one with a mechanical valve. Their heart functions were of grade I. They were carefully monitored by both the obstetrician and cardiologist till the term. The pregnancy periods were uneventful, but two babies were delivered by cesarean section and one was vaginal delivery. The patient with an artificial mechanical valve was given oral anticoagulant every day and vitamin K1 injected before the operation. Both the mothers and babies had no complications. Twenty-four hours later, the oral anticoagulant was resumed and puerperium was still normal. Two other patients with a mechanical heart valve were pregnant, their heart functions were of grade II and one of them suffered from complications. They were not in a condition for continuous pregnancy, and so, operations were performed. Therefore, women with a prosthetic heart valve can have children if their management is closely supervised and extreme care taken with the use of oral anticoagulant.
6.Testing interaction between two know proteins by ras recruitment system

Objective:Introduce yeast two-hybrid system RRS screening system through interaction between two known proteins,Pak and Chp,in order to apply this system on research of the interaction between other proteins.Methods:Select the yeast strain cdc25-2 for single clone that displays low reversion rate,and prepare the competences of the yeast by LiAc.Only transform the plasmid Met425-Myc-Ras-Pak into the yeast to eliminate its self-activation.Co-transform the plasmids Met425-Myc-Ras-Pak and Ura-M-ChpAc into the yeast,set up some controls,observe their growth on different mediums at 36℃.Results:The cdc25-2 cells which are used to prepare competences have no reversion.After the transformation of the plasmid Met425-Myc-Ras-Pak into the yeast,there is neither toxicity nor self-activation.Only when the two plasmids which can express the two corresponding known proteins are co-transformed,and only on the medium suitable for their expression,the cdc25-2 cells can grow at 36℃.Conclusion:(1)The RRS screening system verifies the interaction between Pak and Chp correctly.So it can be used for research of the interaction between other known proteins.(2)We can construct a bait of an known protein,then use the RRS screening system to look for the interacted proteins.
7.Current Situation and Progress of Study on Pharmacology Scutellaria Baicalensis Georgi

We reviewed the domestic studies on Scutellaria Baicalensis Georgi.Scutellaria Baiealensis Georgi has a variety of pharmacological effects,and the main effects are anti-microbial,anti-inflammatory and anti-tumor functions.Scutellaria Baicalensis Georgi has good developing prospects.Up to now,Chinese researches on Scuteilaria baicalensis Georgi have gained gratifying results,but some aspects are still existing defects such as the studies on active ingredients and formula of Scutellaria baicalensis Georgi have not been deeply studied.We propose that human genomics and proteomics researching results should be adopted in the study of Scutellaria Baicalensis Georgi,together with the advanced technologies of bio-chip and biological chromatography being used to elucidate mechanisms at molecular level.
8.The current situation of minimally invasive skills in the treatment of choledocholithiasis

Objective To discuss the minimally invasive skills in the treatment of choledocholithiasis.Methods A retrospective analysis was made to sum up the minimally invasive methods in the treatment of choledocholithiasis,reinforcing sufficient cognition to the treatment of the disease.Results The minimally invasive methods in the treatment of choledocholithiasis included endoscopic sphincterotomy,endoscopic papillaryballoon dilation,fibercholedochoscopy,laparoscopic common bile dnctexploration and laparoscopy combined with endoscopy in treating choledocholithiasis.Conclusion The conception of the treatment of choledocholithiasis was renewed by djfferent minimally invasive skills.With the development of the medical apparatus and instruments,the minimally invasive skills will play an important role in the treatment of choledocholithiasis.
9.Clinical pathologic analysis of the eat-scratch diseases

Objiective To explore clinical pathologic characteristics, diagnosis and differential diagnosis of the cat-scratch diseases. Methods 2 patients with cat-scratch diseases were analyzed by histological observation and Warthin-Starry argentaffine fibers stain. Results One patient had a clearly medical history bitten finger by a roam a-bout cat. Histologically, the nodular was composed of epithelioid cells, multinuclear giant cells with a central necrot-ic. Conclusions The common etiology of the cat-scratch disease were Bartonella hensellae. These lesions greagly re-semble those of lymphogranuloma venereum and non-caseous tuberculosis. The diagnosis and differential diagnosis was based on the history, a specific cutaneous test and Warthin-Starry argentaffine fibers stain.
10.Study on DNA damage and cell apoptosis in the blood vessel endothelial cell of diabetic rats

Objective To explore the relationship of DNA damage and cell apoptosis in the blood vessel endothelial cell of diabetic rats. Methods Streptozotocin was used to make diabetic rats, 16 rats were random divided into diabetic group and control group. DNA damage was measured by comet assay and cell apoptosis was detected by flow eytometry. Results DNA damage score was 29.5 ± 6.7 and 8.6±3.6 in diabetic rats and controls, the rate of cell apoptosis was respectively 10.9±3.5 and 4.4 ± 1.2. It had a significant difference between two groups in DNA damage and cell apoptosis(P < 0.05). The rat of cell apoptosis had strongly positive correlation with DNA dam-age scores (r = 0.82, P < 0.05). Conclusion DNA damage and cell apoptosis are found in diabetes mellitus, cell apoptosis may be asso-ciated with DNA damage. The roles of DNA damage and cell apoptosis in diabetes mellitus need further research.
