1.Application of multi-tumor markers protein chip diagnose system in the diagnosing of ovarian cancer

Objective To investigate the application of Multi-tumor markers Protein Chip Diagnose System in the diagnosing of ovarian cancer. Methods Using the Multi-tumor markers Protein Chip Diagnose System to determine and analyze the concentration values of 12 tumor markers(AFP, CEA, NSE, CA125, CA153, CA242, CA199, PSA, f-PSA, FER, ?-HCG and HGH) in the sera of 53 malignant ovarian cancer patients, 12 ovarian cysts patients and 98 normal persons. Result At least one kind of tumor marker was found higher in 44 sera of the 53 malignant ovarian cancer patients(positive ratio is 83 0%), in 7 sera of the 12 ovarian cysts patients(positive ratio is 58 3%) and 2 sera of the 98 normal persons (negative ratio is 97 9%). NSE、HGH、PSA and f-PSA were first found higher in the sera of ovarian cancer patients. Conclusion The application of Multi-tumor markers Protein Chip Diagnose System in the diagnosing of cancer not only greatly reduces analysis of tumor markers concentration in serum and left the accuracy of diagnosis but may lead new discoveries about the tumor markers.
2.EFFECTS OF Zn~(2+) ON ISOLATED SUPERIOR CERVICAL SYMPATHETIC GANGLION OF RABBITS

The effects of Zn~(2+) on the isolated superior cervical sympathetic ganglion of rabbit were stud ied with sucrese-gap method. It was found thatZn~(2+) significantly inhibited ganglienic action poten-tial with a manner of concentration-dependence.The IC-50 value was 1. 48 mmol/L. The effects ofZn~(2+) were antagonized by neostigmine and 1-cys-tein, reduced under high concentration of Ca~(2+) andenhanced under low concentration of Ca~(2+). It wasalso found that Zn~(2+) inhibited ganglionic N wave,less affected p wave and without any effect on LNwave.
3.EFFECTS OF Zn~(2+) ON THE NEUROMUSCULAR TRANSMISSION

Experiments were carried out on the isolatedrat phrenic nerve diaphragm preparations and chickbiventer cervicis preparations. Zn~(2+) at cumulativeconcentration inhibited muscle contractile responseto indirect stimulation without affecting the con-ductivity of nerve. The inhibiting. action of Zn~(2+) onthe muscle contractile response to indirect stimula-tion was stronger than that on contractile responseto direct stimulation. Zn~(2+) did not affect the Achsensitivity of the chick biventer cervicis muscleThe effects of Zn~(2+) on neuromuscular transmis-sion were antagonized by Ca~(2+) and L-cystein.
4.EFFECTS OF Cu~(2+) ON ISOLATED SUPERIOR CERVICAL SYMPATHETIC GANGLION OF RABBITS

The effects of Cu2+ on superior cervical sympathetic ganglion of rabbits were studied by the sucrose-gap method. Cu2+ at cumula- tive concentration inhibited ganglion action potentials. IC50 of Cu2+ was 0.26 mmol/L. The effects of Cu2+ were antagonized by neostig-miae and L-cystein, while enhanced under 1/2[Ca2+] and reduced under 3[Ca2+] .Cu2+ inhibited ganglionic N wave and P wave,without affecting LN wave.
5.Clinical features of Kennedy desease(Report of 3 cases)

Objective To explore the clinical features of Kennedy disease .Methods The clinical data of 3 patients with Kennedy disease was respectively analyzed .Results All three patients were middle-aged male and had a chronic onset .Patients were mainly presented with the muscle weakness and fasciculation in the proximal limb and bulbar , and the symptoms grow progressively .Two patients were hyperplasia of mammary glands , 3 patients had high levels of serum creatine kinase , and 2 patients'serum testosterone level was increased .EMG detected a widespread neuronal damage in all three cases , and the sensory conductions were abnormal in 2 patients.Repeat numbers of CAG in exon 1 of androgen receptor gene of 2 patients were tested, and they were 57, 47 respectively.Conclusions The clinical features of Kennedy disease are muscle weakness and androgen insensitivity syndrome in male patient and EMG presented with motor neuron damaged .Repeat number of CAG in exon 1 of androgen receptor gene is increased.
7.Effect of human growth hormone on cell cycle kinetics in colonic cancer cells

Objectives:The effects of human growth hormone (hGH) on cell cycle kinetics were studied in human colonic cancer cell line, LS-174-T cell. Methods: The cells were cultured in RPMI-1640 medium for 24 hours with hGH at concerntration of 50, 100, 200 ng/ml with or without cisplatin. Cell cycle kinetics was analyzed by flow cytometry. Results: The significant effects of hGH on cell cycle kinetics were found in the cell line. Percentage of S phase increase (P<0.01) and percentage of G2/M phase decreased (P<0.01).DNA Index (DI) didn't increase.The cells had a lot of apoptosis and percentage of S phase decreased (P<0.01) with addition of cisplatin. Conclusions: In vitro hGH may induce the proliferation of colonic cancer cells. Cisplatin may suppress the stimulatory effects of hGH on S phase and increase the apoptosis of colonic cancer cells.
8.Study on optimizing human acellular dermal matrix and fluorescence labeling the Co-cultured fibroblasts

Objective To optimize human acellular dermal matrix(ADM) and evaluate its biological characters. Methods Human skin was treated with hypertonic saline followed by NaOH maceration(group A), hypertonic saline followed by sodium dodecyl sulfate (SDS) detergent(group B) or Dispase Ⅱ followed by Triton X-100(group C), the resulting ADM were sectioned, and then were stained by special immunohistochemistry method. The cytotoxicity of them were evaluated by methyl thiazolyl tetrazolium (MTT) colorimetry and then cell compatibility was analyzed by cell culture;The optimized ADM resulted was choosen for use. Fibrablasts(FBs)were transfected with adenovirus vector encoding green fluorescent protein gene(Ad-GFP)and the growth of them on the optimized ADM was observed by fluorescent microscopy. Results Collagen and elastic fibers can still be observed in three kinds of ADM. The cells in dermis can be disintegrated both in group A and C, but not in group B. The cytotoxicity scores of the ADM prepared in group A and B were grade 0 or grade 1, while that of group C was more than grade 1.The ADM prepared by NaCl-NaOH maceration had good biocompatibility. There was statistical difference in adhering number of NIH3T3 cells in group A and B. NIH3T3 cells grew well in group A and the resulted ADM was optimized. FBs transfected with Ad-GFP grew well in the optimized ADM. Conclusion The ADM prepared by NaCl-NaOH maceration was a good tissue engineering biomaterial with a little cytotoxicity and rich in resouce.
9.Protective effect of safflower solution on warm ischemia reperfusion injury of liver

Objective To study the protective effect and mechanism of safflower solution on warm ischemia reperfusion injury(IRI) of liver in rats.Methods Male Sprague-Dawley rats were randomly divided as 4 groups: Group S(sham group);group I/R(IRI group);group IPC(ischemic preconditioning group);group SPC(safflower solution preconditioning group).The rats were sacrificed at 24 h after reperfusion,serum levels of ALT and AST were measured and HE staining of liver tissues were made to detect rat liver histological changes and grade liver IRI(Suzuki score);apoptosis were monitored by TUNEL;TNF-?,MIP-2 and ICAM-1 mRNA of liver were measured by RT-PCR;NF-?B gene products of liver were detected by Western blotting.The mean value of all the assay tests was compared.Results After 24 h reperfusion,compared with group I/R,the liver function(ALT and AST) in group SPC and group IPC was significantly improved(P

Objective To study the value of magnetic resonance imaging (MRI)in diagnosis of cubital tunnel syndrome (CuTS). Methods We studied the findings of electrophysiological examination and MRI in 30 elbows of 23 patients with CuTS and 15 elbows of 1 5 controls.We observed the motor conduction velocity (MCV),cross sectional area (CSA)and relative signal intensity (RSI)of the ulnar nerve acrossing the elbow at the point of largest size proximal to the entrapment point (CSA1 ,RSI1 )and the entrapment point (CSA2 ,RSI2 ).Then we calculated the ratio of CSA (CSAR=CSA1/CSA2 ),and the ratio of RSI (RSIR=RSI1/RSI2 ).Results The value of CSA1 and RSI1 was significantly greater than CSA2 ,RSI2 in the patient group (P<0.05).The value of CSA1 ,RSI1 , CSAR and RSIR in patients were significantly larger than that in controls (P<0.05).MCV was negatively correlated with CSA1 and CSAR in the patient group (r=-0.62,r=-0.53).There were no correlation between MCV and RSI1 ,RSIR in the patient group. The area under ROC curve of CSAR was the largest 0.94 (95% CI,0.83-1).The optimum cutoff point of CSAR was 1.83.The CSAR had sensitivity of 93.3% and specificity of 80% in diagnosis of CuTS.Conclusion MRI combined with the electrophysiologi-cal examination shows a high accuracy of locating the entrapment point of ulnar never lesion at the elbow.The CSAR of the ulnar nerve is the best MRI parameters in diagnosis of CuTS.
