2.Plasma D-lactate levels as a useful marker of increased intestinal permeability and gut barrier dysfunction in patients after intraperitoneal operations

Objective To determine the kinetic changes of plasma D-lactate and endotoxin levels in patients after intraperitoneal operations. Methods Blood samples of 63 patients undergoing selective laparotomy were collected 0, 2, 24 and 48h after surgery. According to various criteria, the patients were divided into two groups respectively, i.e. extensive operation or medium sized operation,involvement of gastrointestinal (GI) tract or non-involvement GI tract, and development of symptoms of SIRS or no SIRS after operation. Plasma levels of D-lactate and endotoxin in systemic circulation were determined. Results The analytical data showed that the plasma levels of D-lactate and endotoxin were elevated significantly at 2, 24 and 48h post laparotomy in SIRS patients and major surgery group, and there was also a significant correlation between the plasma levels of D-lactate and endotoxin(?=0.91,P
3.Clinical study on gut barrier dysfunction and bacterial translocation in patients following gastrointestinal surgery

Objective To investigate gut barrier dysfunction and bacterial translocation (BT) from gastrointestinal tract in postoperative patients using polymerase chain reaction (PCR) and plasma D-lactate measurement, and to study the relationship between BT and infections complications. Methods 34 patients undergoing selective gastrointestinal operations were observed. Blood samples were collected prior to and 2, 24, 48 h after surgery for bacterial culture, plasma D-lactate, and LPS measurement. PCR was used to detect microbial DNA. PCR was performed after DNA extraction, with target ?-lactosidase gene of E. coli and 16S rRNA gene of most pathogenic bacteria. All the patients were observed for 30 days for monitoring infectious complications. Results No bacterial DNA was detected before surgery, but it was positive in 7 patients (20.6%) after operation. 4.Value of combined utilization of urinary CysC and NAG in the diagnosis of renal tubules damage

Objective To discuss the diagnostic value of combined utilization of urinary cystatin C (CysC) and β‐N‐acetyl‐glu‐cosaminidase (NAG) in renal tubules injury .Methods Urine samples were collected from 130 cases of patients with renal tubular damage (case group) and 100 cases of health individuals (normal control group) ,respectively .The concentration of CysC and the activity of NAG in urine were detected .Results The sensitivity ,specificity ,and positive predictive value of combined utilization of CysC and NAG in diagnosis of renal tubules damage were 85 .4% ,89 .6% and 82 .1% ,respectively .The concentration of CysC and the activity of NAG in case group after treatment were both significantly different from those before treatment (P<0 .05) .Conclu‐sion Combined utilization of urinary CysC and NAG has a significant value for the diagnosis and the therapeutic monitoring in re‐nal tubules injury .
5.Research on quality parameters of scutellariae radix (formula particles) using on-line NIR in pilot with different extraction process.

The on-line monitor for the changes in the content of baicalin in Scutellariae Radix formula particles during the extraction process was conducted by using near infrared spectroscopy (NIR). High performance liquid chromatography (HPLC) was used as a reference method. Kennard-Stone (KS) was used to divide sample sets, so as to compare different pretreatment methods. The synergy interval partial least squares (SiPLS) was used to screen out modeling wave band to establish partial least-squares models. The relative error method was applied to predict forecast set samples of Scutellariae Radix in three extraction phases. The results showed that the model established by Savitzky-Golay smoothing with 11 points (SG11 points) was the best, with the root mean square with cross validation (RMSECV), root mean square error of correction (RMSEC) and root mean square error of prediction (RMSEP) of baicalin of 0.092 7, 0.134 4 and 0.114 8, respectively, the determination coefficient R2 of greater than 0.99, and the relative error of baicalin content of less than 5%. This indicates that the on-line near infrared reflectance spectroscopy could be applied in on-line monitor and quality control of the extraction process of Scutellariae Radix formula particles.

6.Study on application of electrochemical microsensor detecting NO released from macrophage stimulated by Escherichia coli

Objective To apply nitric oxide(NO) electrochemical microsensor in the real time detection of NO released from RAW 264 .3 cells infected by E .coli ,and to explore the application value of this NO microsensor in the research area of infection im‐munity against bacterium .Methods Taking NO microsensor to detect NO released from RAW 264 .3 cells respectively stimulated by E .coli of different densities and of 1 × 107 mL -1 for different time .Results The level of NO released from RAW 264 .3 cells was enhanced obviously when incubated with E .coli as compared with that of normal cells and the extent of incersase depended on the density of E .coli (P<0 .01) .The released level of NO increased gradually from the beginning and reached its peal at the time of 12 h then decreased slowly when incubated with E .coli of 1 × 107 mL -1 .Conclusion The electrochemical microsensor was applied in the real time detection of NO released from macrophages activated by E .coli successfully .
7.Biocompatiblity of various polypropylene meshes in breast ptosis correction

Objective Implanting surgical meshes to support breast in periareoler mammaplasty shows an excellent prospect.But shrinking of the mesh and the adhesion formation affected the postoperative shape and palpation of the breast.The aim of this study was to compare biocompatible properties of four different polypropylene meshes (Prolene(R) ; Vypro Ⅱ(R) ; Premilene Mesh (R) and Premilene Mesh LP(R)) and to discuss the inclusion criteria of meshes in this technique.Methods The study was carried out in pigs.Four different polypropylene meshes were implanted subcutaneously in belly.90 days after implantation the mesh samples were explanted,and complications,foreign-body reaction,collagen fiber and elastic fiber formation were observed,as well as ultrastructures for the biocompatible properties.Results The biocompatible properties of each mesh were different.The foreign-body reactions determined the biocompatible properties,and affected the postoperative shape greatly.Conclusions Premilene Mesh LP o as a newer low-weight polypropylene mesh,has superior properties to the others.
