How to expand and deepen the educational function of library of college and university of traditional Chinese medicine by giving play to its strong points was analyzed in the new model of students'independent study , and measures to improve the information literacy and comprehensive literacy of students were elaborated, such as developing teaching reference books database, performing guidance for different readers, carrying out different activities that make entertainment a medium of education , in order to help students to acquire special knowledge outside of classroom .
2.Spiral CT Diagnosis of Small Hepatocellular Carcinoma

Objective To study the CT features of small hepatocellular cancer.Methods 37 patients with small hepatocellular cancer proved histopathologically underwent spiral CT examinations including plain scans and contrast-enhanced scans.The arterial phase scan,the portal phase scan and delayed phase scan started at 25th second,70th second and 5th minute respectively after injecting the contrast medium with the high pressure syringe,the velocity was 2.5 ~3 ml/s,and the dose was 1.5 ml/kg.Results 73% (27/37) of tumors showed hyperdensity in arterial phase,65%(24/37) of tumors was hypodensity and 35% (13/37) had hyperdensity during portal phase,of which eight showed hypodensity in equilibrium phase.Conclusion Spiral CT can display the features of blood supply of small hepatocellular carcinoma that is of benefit in early diagnosis.
3.Brachial-ankle pulse wave velocity correlates to urinaryprotein in patients with type 2 diabetes mellitus

Objective Urinary protein is closely related to the development and progression of major vascular lesionsin diabetic patients.This study was to investigate the correlation between brachial-ankle pulse wave velocity(baPWV) and urinary protein in patients with type 2 diabetes mellitus ( T2DM) . Methods According to the level of 24-hour urinary albumin ( UAL) , 142 T2DM patients were divided into three groups:control (UA<30 mg, n=84), microalbuminuria(UAL<300 mg, n=26), and clinical albuminuria group ( UAL≥300 mg, n=33) .All the patients underwent examination offasting blood glucose, blood lipid, renal function, HbA1c, islet func-tion, C-reactive protein, blood pressure, and BMI.Their baPWV was determined with an Omron atherosclerosis detector and the correla-tion of baPWV with other factors evaluated by Pearson analysis and multivariate linear regression analysis. Results The baPWVwas significantly higher in themicroalbuminuria([1649.19±229.36]cm/s) and clinical albuminuria ([1759.21±291.05]cm/s) than in the control group ([1522.33±248.37]cm/s) (P<0.05).Pearson analysis showed a positive correlation between 24-hour UAL and baPWV ( r=0.347, P<0.01) and multivariate linear regression analysis manifested the same correlation ( P<0.05) . Conclusion The level of urinary protein is closely correlated with baPWV in T2DM patients.
4.TESTIN tumor suppressor gene in endometrial cancer tissues

ObjectiveTo investigate the expression of TESTIN gene in endometrial carcinoma and explore the functions of this gene in tumor development and progression.MethodsqRT-PCR and immunochemical staining assay were used to determine the mRNA and protein level of TESTIN in the tumor tissues,and the relationship between TESTIN expression and clinical pathology characteristics was analyzed.Results Compared to normal tissue,76.5% (52/68) tumor tissues showed TESTIN reduced ( P < 0.01 ),furthermore,this reduction in the subgroup of endometrioid adenocarcinoma was significant,but it was rarely observed in the subgroup of serous papillary adenocarcinoma.ConclusionsTESTIN was obviously down regulated in endometrail carcinoma,especially in endometrioid adenocarcinoma,which indicated TESTIN played an important role in tumorigenesis of uterine.

OBJECTIVEThis study aims to investigate and compare the toxic effects of four types of metal oxide (ZnO, TiO(2), SiO(2,) and Al(2)O(3)) nanoparticles with similar primary size (∼20 nm) on human fetal lung fibroblasts (HFL1) in vitro.
METHODSThe HFL1 cells were exposed to the nanoparticles, and toxic effects were analyzed by using MTT assay, cellular morphology observation and Hoechst 33 258 staining.
RESULTSThe results show that the four types of metal oxide nanoparticles lead to cellular mitochondrial dysfunction, morphological modifications and apoptosis at the concentration range of 0.25-1.50 mg/mL and the toxic effects are obviously displayed in dose-dependent manner. ZnO is the most toxic nanomaterials followed by TiO(2), SiO(2), and Al(2)O(3) nanoparticles in a descending order.
CONCLUSIONThe results highlight the differential cytotoxicity associated with exposure to ZnO, TiO(2), SiO(2), and Al(2)O(3) nanoparticles, and suggest an extreme attention to safety utilization of these nanomaterials.

7.Effect of crocin on expression of SIRT1/PGC-1αin hippocampus under hypoxia at high altitude in rats

Objective To observe the effect of crocin pretreatment on the learning memory ability, the ultrastructure and SIRT1/PGC-1α expression in hippocampus under hypoxia at high altitude in rats. Methods 96 male SD rats were randomly divided into Hypoxia model group and Hypoxia treatment group with 48 in each.The group rats in treatment group were injected with Crocin once daily(50 mg/kg) and the model group were injected with the same dose of 0.9%NaCl for 3 days,and then they were shifted to high al-titude under hypoxic environment and then the samples were taken at 1 d,3 d,5 d,7 d.Morris water maze test was used to observe learning and memory behaviors of rats.The level of SIRT1/PGC-1αprotein was detected by Western blot and the level of mRNA was detected by RT-PCR.Results Compared with the model group (1d(37.55±1.34)s,3d(46.51±6.19)s,5d(42.12±2.73)s),treatment group showed significant less time in looking for platform in Morris water maze experiment at (1d(33.58±2.23)s,3d(30.19±3.35)s and 5d (37.10±1.48)s;all P<0.05),and reduced damage in hippocampal tissues of ultrastructure.The results of RT-PCR showed that the mRNA levels of SIRT1 and PGC-1α in hippocampus of rats were higher in treat-ment group (1d(0.24±0.01),(0.34±0.02);3d(0.27±0.01),(0.37±0.02);5d(0.26±0.08),(0.26± 0.03)) than those in model group(1d(0.23±0.02),(0.32±0.03);3d(0.12±0.02),(0.19±0.02);5d(0.20 ±0.01),(0.24±0.03);all P<0.05),and there was no significant difference at 7d in two groups(P>0.05). Western blot showed that the protein expression levels of SIRT1 and PGC-1α in hippocampus of rats were higher in treatment group (1d(0.72±0.02),(0.72±0.02);3d(0.76±0.03),(0.75±0.021);5d(0.54± 0.03),(0.74±0.01)) than those in control group (1d(0.53±0.04),(0.62±0.04);3d(0.15±0.02),(0.13± 0.02);5d(0.23±0.03),(0.21±0.02);all P<0.05),and there was no significant difference at 7d between groups(P>0.05).Conclusion Crocin pretreatment can ameliorate mitochondrial damage and improve the a-bility of learning and memory under hypoxia at high altitude in rats,which may be achieved through increased expression of SIRT1 and PGC-1α in the hippocampus.
8.Construction of Ischemic Cardiomyopathy Model via Ligation of Left Anterior Descending Artery by using Diannan Miniporcine

Objective To construct a large animal model of ischemic heart disease induced by acute myocardial infarction (AMI) . Methods Male Diannan Mini-pigs were selected (n=8), weight 20 ± 4 Kg. After general anesthesia and endotracheal intubation, blood pressure and electrocardiograph were monitored. Parasternal incision was taken and thoracotomyv was performed at 4th-to-5th intercostalgap. Pericardium was opened and left anterior descending artery was ligated. Echocardiography was performed at baseline, postoperative 28 and 60 day. Euthanasia and heart explanted was conducted at 60 day of experimental pig. After gross examination of the heart, the histological examination was used to assess myocardial infarction by using H&E staining. Results All of the pigs were presented with ventricular arrhythmias in varying degrees after ligation of the left anterior descending artery, of whom 2 died of refractory ventricular fibrillation, the mortality was 25%. Echocardiography assessment showed the loss left ventricular function, the thinning of left ventricular apical and the enlargement of left ventricule. Histological examination revealed that myocardial fibrosis and fibrous scar were formed in myocardial infarction area. Conclusion The ligation of left anterior descending artery through thoracotomy under direct vision is a reliable strategy for construction of AMI porcine model and the experimental porcine can develop ischemic cardiac dysfunction progressively.
9.Research of dendritic cells pulsed with HPV peptide inducing specific CTL in vitro

Objective To explore the potential of autologous dendritic cells (DC) pulsed with HLA-A201-binding peptide E613-21(KLPDLCTEL) and E786-94(TLGIVCPI)in inducing specific T cells respouse in vitro.Methods Cervical carcinoma patients with positive HLA-A201 were enrolled and their monocytes isolated and induced into dendritic cells and pulsed with HLA-A201-binding peptide E613-21 and E786-94.PBLs were primed by DCs every week for thee times.The cytokine level of supernatant of CTLs was tested by ELISA.The percentage of special CTLs was tested by flow cytometry.The specific killing effect of CTLs was tested by MTT.Results the numbers of DCs of eleven cervical carcinoma patients were (10.79±0.88) ×106(100 ml peripheral blood).CDllc+HLA-DR+(97.15±2.41)%,CD80+(84.28+5.39)%,CD83 +(85.17±5.06) %,CD86 + (97.74+0.87) %.Proliferation index of PBLs primed by DCs three times was 15.4± 1.5.Cytokine levels including IL-2,IL-12,IFN-γ and TNF-α were obviously higher than nonpriming PBLs[(2551.9+195.3) pg/ml,(554.9±64.0) pg/ml,(2416.9±281.7) pg/ml,(632.4 +71.1)pg/ml,respectively] (P<0.05),but IL-10 was no significant difference between priming CTLs and nonpriming CTLs.The average percentage of special CTLs was obviously higher than control group[(6.32±1.54)%,P<0.05].The killing effect of CTLs was obviously higher than control group(P<0.05).Conclusion Dendritic cells pulsed with peptide E613-21 and E786-94 can induce special CTLs in vitro and stimulate CTLs secret cytokines.This will provide science basis for research of therapeutic HPV vaccine.
10.Gleevec induces apoptosis in K562 cells through activating caspase-3.

The present study is to elucidate the mechanisms underlying Gleevec-induced apoptosis of chronic myeloid leukemia (CML) K562 cells in vitro. The apoptotic cell death and cell cycle distribution after Gleevec treatment and the effect of PDCD4 siRNA on Gleevec-induced apoptosis of K562 cells were analyzed by flow cytometry. The effect of Gleevec on p-Crkl, caspase-3, PARP and PDCD4 protein levels, and the knockdown efficacy of PDCD4 siRNA were detected by Western blotting. The results showed that Gleevec dramatically suppressed the phosphorylation level of Crkl in a dose-dependent manner and induced significant apoptosis and G0/G1 cell cycle arrest of K562 cells in time- and dose-dependent manners. In addition, Gleevec activated caspase-3 and its downstream substrates PARP, and the caspase pan inhibitor Z-VAD-FMK (50 micromol x L(-1)) markedly reduced Gleevec-induced apoptosis from 47.97% +/- 10.56% to 31.05% +/- 9.206% (P < 0.05). Moreover, Gleevec significantly increased the protein expression of programmed cell death 4 (PDCD4). PDCD4 knockdown by siRNA reduced Gleevec-induced apoptosis from 46.97% +/- 14.32% to 42.8% +/- 11.43%. In summary, Gleevec induced apoptosis in K562 cells via caspase-3 activation.
