1.Study on quality standards for Xiegananshen Capsules

Objective: To establish the quality standards for Xiegananshen Capsules. Methods: Poria in this prescription was identified by microscopic examination; Radix Gentianae, Fructus Gardeniae, Radix Angelicae Sinensis and Radix Scutellariae were identified by TLC. Gentiopicrin was determined by HPLC. Results: Poria could be detected by microscopic examination; Radix Gentianae, Fructus Gardeniae, Radix Angelicae Sinensis and Radix Scutellariae could be detected by TLC. Gentiopicrin showed a good linear relationship at a range of 2.6 ~13?g, r=0.9995. The average recovery was 97.8%, and RSD was 1.0%. Conclusion: The established methods are simple, feasible and reproducible. This study provided a method for the quality control of Xiegananshen Capsules.
2.Relationship between open reading frame 26 subtypes in human herpesvirns-8 and Kaposi's sarcoma

Objective To learn the subtypes of open reading frame 26 (ORF26) of human herpesvirus 8 (HHV-8) in patients with Kaposi's sarcoma, and to evaluate the relationship of ORF26 subtypes to clinical types and invasiveness of Kaposi's sarcoma. Methods Thirty-two paraffin-embeded tissue speci-mens of Kaposi's sarcoma were collected in the Department of Dermatology, People's Hospital of Xinjiang Uygur Autonomous Region, from 1996 to 2007. DNA was extracted from these specimens, nested-PCR was used to amplify HHV-8 DNA. PCR products were subjected to bi-directional sequencing after extraction from agarose gels. Phylogenetic analysis was carded out by using the software DNAStar, program Clustal W and PHYLIP package for the determination of ORF26 subtype. Fisher's exact test was performed to evaluate the relationship of ORF26 subtypes to clinical types and invasiveness of Kaposi's sarcoma. Results Of the 32 specimens, 30 were positive for HHV-8 DNA with a positivity rate of 93.75%, and 6 specimens of AIDS related Kaposi's sarcoma were all positive. ORF26 A subtype was detected in the HHV-8 DNA of 17 posi-tive specimens, and C subtype in that of other 13 specimens. Neither the incidence of mucosal lesions nor the distribution of clinical subtypes was of significant difference between patients with ORF26 A subtype and those with ORF26 C subtype (both P > 0.05). Conclusions A and C may be the predominate subtypes of ORF 26 in HHV-8 of patients with Kaposi's sarcoma, and the ORF26 subtype is unrelated to the presence of mucosal lesions in or the clinical types of patients with Kaposi's sarcoma.
3.Oxidative stress induces apoptosis in HepG2 cells

AIM: Direct exposure of cells to reactive oxygen species can induce apoptosis. In this study we investigate how oxidative stress induces cell death in HepG2 cells and characterize the molecular events involved. METHODS: Oxidative stress was created by exposing HepG2 cells to 2 mmol/L H2O2. Apoptosis was determined by analysis of DNA fragmentation by agarose gel electorphoresis. The mitochondrial membrane potential was analyzed using DePsipher fluorescent staining and the expression of cytochrome c in the cytosolic fraction was measured by Western blotting analysis.The caspase activity was detected using fluorometric assay kit by a fluorescence microplate reader. RESULTS: When HepG2 cells were treated with 2 mmol/L H2O2, the cells displayed DNA fragmentation, a typical feature of apoptosis, after 12 h. The mitochondrial membrane potential appeared different in two group of cells. H2O2 -treated cells appeared green fluorescence as early as 4 h, which represents de - energized mitochondria, the untreated cells appeared red fluorescence,a feature of mitochondria with intact membrane potential. In treated cells, the expression of cytochrome c increased and accumulated in cytosolic fraction with treatment time, caspase - 3 activity increased by 6.7 - fold ( P < 0.01 ) at 8 h and caspase -9 activity increased by 3.6 - fold (P < 0.01 ) at 12 h, respectively, however, the activity of caspase - 8 remained unchanged. CONCLUSION: These findings suggest that oxidative stress can induce apoptotic cell death in HepG2 cells, and the mechanism is related to mitochondrial pathway, which activates caspase -9 and- 3, but not caspase -8.
4.The impact of serum uric acid level to CSFP

ObjectiveTo investigate on the impact of the serum uric acid (UA) level coronary slow flow phenomenon(CSFP).MethodsSelected 30 cases of coronary slow flow patients as CSFP group,30 cases of patients with coronary heart disease,30 cases of patients with angiographically normal as the control group.The serum UA level was measured in three groups.The CSFP group and the control group were measured left anterior descending,circumflex and right coronary artery TIMI frame count(TFC) value.To analyze the mean TFC value of CSFP patients and the relationship between the serum UA level.ResultsCompared with the control group,the left anterior descending,circumflex and right coronary TFC results were significantly higher(all P < 0.05) in the CSFP group.The mean serum UA level of CSFP group ,control group and CHD group were(340.79 ± 79.40)μmol/L, (220.45 ± 82.34)μmol/L,(42625 ± 87.02) μmol/L, the difference was statistically significant (all P < 0.05).The mean serum UA level and TFC value of CSFP group was positively correlated(r = 0.943, P = 0.007).ConclusionElevated level of serum UA is closely related to the occurrence and development of the CSFP.
5.Atrioventricular nodal ablation for patients with chronic atrial fibrillation

Objective: To evaluate effects of atrioventricular nodal ablation, pacemaker and pharmaco logic agents on ventricular rate control. Methods:Three patients with chronic sustained atrial fibrillation ( AF) were treated by atrioventricular nodal ablation. Fast-slow junctional cardiac rhythm in this course meant the ablation was successful. Unless the ablation was able to get Ⅱ? Ⅱ type or Ⅲ? AVB, R-R intervals ≥3 s, ventricular rate≤40 bpm, it would be canceled. Otherwise implantation of a permanent pacemaker was performed. Results:One patients having complete atrioventricular block and a ventricular rate of 38 -40 bpm had implantation of a permanent pacemaker. Two patients having no atrioventricular block, prolongation of R-R intervals and ventricular rate of 90 - 120 bpm were given antiarrhythmic drugs. Conclusion: Patients with chronic sustained AF can be treaded with atrioventricular nodal ablation, pacemaker and pharmacologic agents effectively.
6.Determination of linoleic acid and ricinoleic acid in Anchan Emulsion by GC

Objective: To establish a determination for Ricinus communis L.and Carthamus tinctorius L.could in Anchan Emulsion. Methods: The determinations were carried out by GC. Chromatographic conditions were: using Chromosorb DEGS as a stationary phase column temperature 180 ?C , flame ionization detecter.Rusults:The content limit of Linoleic acid wasn't lower than 20%, The content limit of Ricinoleic acid wasn't lower than 20%.Conclusion: The established methods is simple, feasibl and reproducible. This study provides a method for the quality control of Anchan Emulsion.
7.Effects of intensive insulin therapy on non-specific inflammatory factors in patients of critical illness complicated with hyperglycemia

Objective To observe the effects of intensive insulin therapy on C-reactive protein (CRP) ,interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α ) in the patients of critical illness complicated with hyperglycemia and its incidence of side effect. Methods Two hundred and nine patients of critical illness complicated with hyperglycemia were randomly divided into intensive insulin therapy group (106 patients,blood glucose maintained at a level of 4.4-6.1 mmol/L) and conventional insulin therapy group ( 103 patients, blood glucose maintained at a level of 9.0-11.1 mmol/L). Serum levels of CRP,TNF- α and IL-6 were determined on 0,24,48,72 h respectively after ICU admission. Results The levels of blood glucose of both groups reached the target level. The incidence rates of the hypoglycemia had no significant difference between two groups [6.60 % (7/106) vs. 4.76% ( 3/63 ),P > 0.05]. After 72 h treatment, serum level of CRP in intensive insulin therapy group was significantly lower than that in conventional insulin therapy group (P < 0.05 ). After 24,48 and 72 h treatment, serum level of IL-6 in intensive insulin therapy group was significantly lower than that in conventional insulin therapy group (P < 0.05 ). After 48 and 72 h treatment, serum level of TNF-αin intensive insulin therapy group was significantly lower than that in conventional insulin therapy group (P < 0.05). Conclusion Intensive insulin therapy can significantly decrease the levels of non-specific inflammatory factors in patients of critical illness complicated with hyperglycemia, which brings beneficial effect to the patients.
8.Expressions of Kaposi′ s sarcoma-associated herpesvirus type 8-associated microRNAs k12-1 and k12-12 in Kaposi′s sarcoma and their significance

Objective To measure the expressions of Kaposi′s sarcoma-associated herpesvirus type 8 associated-microRNAs k12-1 (kshv-miR-k12-1)and k12-12 (kshv-miR-k12-12)in Kaposi′s sarcoma tissue, and to assess their relationship with pathological stage and lesion area of Kaposi′s sarcoma, HIV infection, and human herpesvirus type 8 (HPV-8)infection. Methods Totally, 18 paired tissue specimens stored in liquid nitrogen from Kaposi′ s sarcoma lesions and paralesional skin were collected. Total RNAs were extracted from these specimens by using Trizol reagent, and reversely transcribed into cDNA. SYBR Green real-time fluorescence-based quantitative PCR was performed to measure the expressions of kshv-miR-k12-1 and kshv-miR-k12-12 in these specimens. The relationship of kshv-miR-k12-1 and kshv-miR-k12-12 expressions with the pathological stage and lesion area of Kaposi′s sarcoma, HIV and HPV-8 infections was analyzed. Results Compared with paralesional normal skin, Kaposi′s sarcoma lesions showed significantly increased expressions of kshv-miR-k12-1 (2-ΔΔCt: 1.016 ± 1.645 vs. 0.029 ± 0.019, t = 2.542, P = 0.016)and kshv-miR-k12-12 (2-ΔΔCt: 2.104 ± 1.973 vs. 0.102 ± 0.093, t = 4.301, P = 0.000). There were no significant differences in the expressions of kshv-miR-k12-1 or kshv-miR-k12-12 between patients with HIV or HPV-8 infection and those without, among patients with different pathological stages of Kaposi′s sarcoma, or among patients with different lesion areas (all P > 0.05). Conclusion Both kshv-miR-k12-1 and kshv-miR-k12-12 are highly expressed in Kaposi′s sarcoma, but neither of their expressions is related to HIV or HPV-8 infection, pathological stage or lesion area of Kaposi′s sarcoma.
9.Analysis of cause and correlative factors of death within short-term in initially lucid patients with intracerebral hemorrhage

Objective To analyze the correlative factors and cause of death within short-term in initially lucid patients with intracerebral hemorrhage (ICH).Methods 441 patients with spontaneous ICH admitted within 24 hours of onset and with Glasgow Coma Scale (GCS) score more than 9 on admission were enrolled. The demographic characteristics, clinical features at onset, CT, electrocardiograph (ECG) and laboratory findings, medical complications and 27 days outcome were recorded. Univariate and multivariate analysis were performed.Results 24 (5.4%) patients died winthin 27 days after onset, 14 (58.3%) died of brain herniation and central respiratory failure, another 10 (41.7%) of systemic complications. Multivariate analysis demonstrated that age was 75 years old or more, urinary incontinence at onset, peripheral white blood cell count more than 10.0?10 9/L on admission, blood glucose more than 7.0 mmol/L and abnormal ECG were independent correlative factors of death.Conclusions The short-term outcome of initially lucid patients with ICH is favorable in general, albeit with a relatively low mortality. Brain herniation and central respiratory failure were main cause of death, and they have independent correlative risk factors.
