3.Establishment of the National references for Sabin vaccine produced in Vietnam

A study on the establishment of the effective standard for the national standard samples of vaccine of single vaccine type 1, 2 and 3 and triple vaccine (Sabin) produced in Vietnam was carried out. The sesults have shown that the National standard sample had a high unified and stable properties that meet the requirement of the world health organization (WHO). The National standard sample were used to evaluate the quality of sabin vaccine produced in Vietnam which showed that 450 batches (100%) sabin vaccine produced in the scientific center for the vaccine production during 1995-1999 obtained the standard of WHO.

4.The relation between oral immunization with poliomyelitis vaccine and the evolution of poliomyelitis epidemic in North Vietnam

All children in suspicion of poliomyelitis from 28 provinces of North Vietnam (from 1993 to 1999) were enrolled in study performed with a form of inquiry concerning all related personal health documents and reports. Results shown that all children suffered from poliomyelitis with isolated wild poliovirus accounted for 73.4% were primarily non-immunized or under-dose-immunized. Conclusion recommended the continuation of oral immunization among all under-one year old children in combining with the providing two doses of oral vaccine per year for under five years old children

5.Monitoring the potency of oral polio vaccine (OPV) in the field in 1999

In 1999, 49 vaccine samples had been collected from the field in routine immunization days. The potency of the samples had been verified in National Centre of Control of Medio-Biological Products, Ha Noi. The results showed that the potency of all the vaccine samples is at the acceptable limit, type 2 is the most stable and type 3 is the least stable one

7.Study of three kinds of primary immunization schedules with poliovirus vaccine.

Objective: To compare the immunogenicity of three kinds immunization programs with poliovirus vaccine. Methods: Healthy infants aged 2 months or over were selected and divided into three groups by complete randomization method. Basic immunization with Sabin inactivated poliovirus vaccine(sIPV) and bivalent oral poliovirus vaccine(bOPV) were completed. Three kinds of basic immunization procedures were 1sIPV+2bOPV,2sIPV+1bOPV and 3sIPV, respectively.Two qualified serums that before basic immunization and 28-42 days later were collected, and measured the poliovirus neutralizing antibody with microcell neutralization method. To compare the difference by analysis of variance, rank test and χ2 test. Results: After the basic immunization, 205 subjects of the positive conversion rate of poliovirus neutralizing antibodies of types Ⅰ, Ⅱ and Ⅲwere all higher than 97.00%, and the positive rates were all higher than 98.00%, the geometric mean titer (GMT) of neutralizing antibody was significantly higher than that before basic immunization in three groups.There were significant differences in the positive rate and GMT before and after basic immunization of typeⅠ, Ⅱand Ⅲ in the three (P<0.05). The highest GMT in three groups after basic immunization were all typeⅠ, followed by type Ⅲ, and the lowest in type Ⅱ. The GMT of type Ⅱin 2sIPV+1bOPV and 3sIPV groups were both higher than that in sIPV+2bOPV group. Conclution: After three kinds of basic immunization, the poliovirus neutralizing antibodies of serum were all at high levels in three groups, which could form an effective immune barrier against poliovirus. The immunogenicity of three kinds of basic immunization programs were all well, but there were certain differences of neutralizing antibodies among three kinds basic immunization programs. The immunogenicity in 2sIPV+1bOPV and 3sIPV groups against typeⅡpoliovirus were better than that in 1sIPV+2bOPV group.

8.Evaluation of the documentation for OPV production in the year of 2006

Background: Good Manufacturing Practice (GMP) is a important part of quality assurance (QA). Implementation of GMP request to establish the documentation system and should be reviewed as well as evaluated strictly. Documentation is important because it helps the competent person make decisions whether to finish the product or not and it is used as a base for inspection. In the year of 2006, 100% OPV production line in Center for research and production of vaccines and biologicals (POLYVAC) had established the document system. Objectives: To evaluate of the documentation for OPV production in the year of 2006 in POLYVAC. Subjects and method: Documentation for OPV production in 2006 were evaluated by standard operating procedure (SOP) (recommended by WHO). The standards included blank space; erase; code and unit of measurement; revision; no signage and others. Results and Conclusion: Documentation is different between departments. Onlymonkey ranch in Reu island applied 100% SOP. 776 errors were found. Among them blank space (32.2%), erase (27%), code and unit of measurement (21%), revision (5%), no signage (12%) and others (3.6%). The results were the basis of promoting implementation of GMP in vaccine manufacturing in POLYVAC.

9.Determination of adventitous agent in monkeys and monkey kidney cells culture using for OPV production in 2006.

Background: In 2006, Center for research and production of vaccines and biologicals \ufffd?Ha Noi produced 10 million doses of poliomyelitis vaccine met quality standard which given by World Health Organization (WHO). The monkeys for vaccine production were selected and monkey kidney cells were tested arcording to the WHO requirements (the strong moykeys with no adventitous agents). Objectives: To assess the rates of mokeys (at the Reu island in Quang Ninh province) with no adventitous agent infection and the cells using to produce type 1 poliomyelitis vaccine in 2006. Subjects and method: The study was performed by using microneutralization technique on 39 Macaca mulatta mokeys with weights were 1.5 to 2.5 kg. Results: 11 out of 39 monkeys (28,21%) are negative to SIV, SV40, poliomyelitis viruses, foamy virus as well as tuberculin test. 100% lots of primary monkey kidney cells culture produced in 2006 are free from adventitous agents arcording to the WHO requirements. Conclusion: Center for research and production of vaccines and biologicals - Ha Noi implemented strictly under the WHO guidelines on selection of monkeys and monkey kidney cells for OPV production from Sabin strains.

10.Investigation on circulation and mutation of Sabin poliovirus strains isolated from AFP cases and from OP-vaccineated healthy children

During the period from 1999 to 2000, 12 poliovirus isolates were obtained from acute flaccid paralysis (AFP) cases and 2 from healthy children who have been vaccineated with Sabin's oral polio vaccine. These isolated have been examined by PCR-RFLP. The results showed that the isolates were genetically identical to the regional Sabin's poliovirus strains
