1.An immunohistochemical study of human chorionic gonadotropin in 40 cases of colorectal carcinoma

The localization of the ?-subunits of human chorionic gonadotropin(HCG)in 40 cases of colorectal carcinoma was investigated with PAP method.It was found that 9 cases out of the 40 were positive,in which 2 were Dukes' stage A,4 Dukes' stage B and 3 Dukes' stage C,accounting for 7.7%,44.4% and 60.0% of the total number of the corresponding Dukes' staging of this series.The positive cells were mainly situated in the infiltrating portions and distributed in clusters or diffusely.It is suggested that immunohistochemical study of HCG in colorectal carcinoma can be one of the indices in the monitoring of the malignant biological behaviors of colorectal carcinomas.
2.The significance of immunohistochemical localization of MC5 in the diagnosis and prognosis of colorectal adenocarcinoma

The pattern of immunohistochemical localization of monoclonal antibody MC5 was observed in 215 cases of colarectal adenocarcinoma.It was found that the polarized pattern was predominant in the well-differentiated adenocarcinomas while the deffuse pattern in poorly-differentiated ade-nocarcinomas.Followup study of 133 cases revealed that 5-year sunvival rate was 74.4% in cases with polarized pattern and 36.2% in those with diffuse pattern,a very significant difference between the 2 (P

Objective:To study the expression of P53, Bax, Bcl-2 proteins and the role of cell apoptosis in the formation and development of acute radiation-induced skin ulcers.Methods:A rat model which was locally irradiated with 60 Co γ-rays was used, and the pathological changes were observed for 40 days. Immunohistochemistry and TUNEL assay were performed which enabled the detection of P53, Bax, Bcl-2 and cell apoptosis during the formation and development of radiation skin ulcers.Results: Skin ulcers were found on day 14 after irradiation, and enlarged and deepened gradually during the observation period. P53 was over expressed during days 11 to 40 after irradiation and was localized in vascular endotheliocytes and smooth muscle cells. Bax was moderately positive during days 14 to 21 and weakly positive during days 28 to 35, and was localized in vascular endotheliocytes, some fibroblasts and proliferating keratinocytes. Bcl-2 was weakly positive during days 1 to 11 after irradiation, and was located in keratinocytes, hair follicular cells and some vascular endotheliocytes. Bcl-2 was negative during days 11 to 40.The rate of cell apoptosis, especially of vascular endotheliocytes,wash igher than that in the early process of normal wound healing. Conclusions:After irradiation,the increased expression of the apoptosis-inducing protein P53, Bax and the decreased expression of the apoptosis-inhibiting protein Bcl-2 might be associated with the high rate of apoptotic events, and play important roles in the formation and development of radiation skin ulcers.
4.Comparison of periphery capillary whole blood glucose using BREEZE~(TM)blood glucose meter and venous plasma glucose using laboratory autoanalyzer

Objective To evaluate the precision of BREEZE~(TM) glucose monitor and correlation of glucose measurements between fingertip capillary whole blood glucose(CBG)using BREEZE~(TM) glucose monitor and venous plasma glucose(VPG)using autoanalyzer.Methods All samples of venous plasma and fingertip blood from 188 diabetes or non-diabetes patients were detected for glucose level at fasting,30,60 and 120 min postprandial.BREEZE~(TM) glucose monitor and autoanalyzer measured CBG and VPG respectively.Intra-and inter- coefficients of variations were determined using 10 BREEZE~(TM) glucose monitors with normal,slight high and high glucose levels for the three lots of strips.Results The correlation coefficients between CBG and VPG were all higher than 0.950 at fasting and different postprandial time.98.94% of all measurements were in the A zone when using error-grid analysis.The relevant differences between CBG and VPG were less than 5% at different blood glucose concentrations.The intra-and inter-coefficients of variations of blood glucose values at different blood glucose concentrations using different lots of strips were within 5%.Conclusion BREEZE~(TM) glucose monitor provides high accurate and precise glucose readings on fasting and different postprandial time points over a variety of blood glucose concentrations.
5. Effects of hypobaric hypoxia on respiratory and motor function in spinal cord injury rats

Objective To investigate the effect of hypobaric hypoxia on the respiratory function of C5 spinal cord injury rats by simulating the environment of low pressure and low oxygen in air transport. Methods 20 male SD rats were randomly divided into 3 groups: sham operation group (n=4), spinal cord injury group (n=8) and spinal cord injury + low pressure group (n=8). Spinal cord injury model was made by compression method. These rats in the three groups were treated with carotid artery catheterization, and were fed for 24 hours. Spinal cord injury + low pressure group was put into 2500 m low pressure environment for 4 hours. Arterial blood gas, blood routine test, HE staining of the spinal cord and lung tissue were detected to evaluate respiratory function, and BBB score was used to evaluate motor function of rats. Results There was no significant difference between the three groups in pH, oxygen partial pressure, carbon dioxide partial pressure, oxygen saturation, glucose, lactate, alkali surplus, buffer alkali and other indicators (P>0.05). The percentage of reticulocyte in spinal cord injury group was lower than that in sham operation group (2.05%±0.69% vs. 2.88%±0.20%, P=0.034), there was no significant difference between spinal cord injury + low pressure group and other two groups (2.37%±0.59% vs. 2.88%±0.20%, P=0.178; 2.37%±0.59% vs. 2.05%±0.69%, P=0.282). There was no significant difference among the three groups in the leukocyte counts and percentage of neutrophils, lymphocytes, monocytes, erythrocytes, hemoglobin and hematocrit (P>0.05). HE staining showed that there was no obvious bleeding and exudative changes in spinal cord in sham operation group, and the tissue structure was intact. The histological integrity of spinal cord injury group and spinal cord injury + low pressure group was significantly different from that of sham operation group, but there was no significant difference between the two groups. There was no significant difference in bleeding, exudation and edema between the three groups, and the alveolar structure was intact. BBB score showed no significant difference between spinal cord injury group and spinal cord injury + low pressure group (P=0.440). Conclusion The respiratory and motor function of C5 spinal cord injury rats would not be significantly affected by 4 hours of low pressure and hypoxia.

8.Cloning and expression analysis of glucose-6-phosphate dehydrogenase 1 (G6PDH1) gene from Chimonanthus praecox.

Glucose-6-phosphate dehydrogenase is main regulatory enzyme for pentose phosphate pathway. To amplify the core sequence of G6PDH gene from Chimonanthus praecox, the primers were synthesized, based on the conserved nucleotide sequence of other reported plant G6PDH genes. The specific primers were designed according to the major fragment. The full length cDNA of the G6PDH1 gene was isolated by the 3' and 5' rapid amplification of cDNA ends approach. Transcript levels of G6PDH1 isoform was measured by real-time quantitative RT-PCR in different tissues and in responds to cold treatment. The G6PDH1 subcellular localization, transmembrane domain, three-dimensional structure, and phylogenetic analysis were predicted by different software to analysis the bioinformatics of G6PDH1 protein. The G6PDH1 cDNA sequence was 2 011 bp in length and consisted of 1 551 bp Open Reading Frame (ORF) , encoding a protein of 516 amino acids. Expression analysis results in different tissues showed that G6PDH1 was primarily observed in flowers and roots, as opposed to the leaves and stems. Cold treatment experiments indicated that cold treatment caused a rapid increase in G6PDH1 expression in flowers within 12 h. The full-length cDNA of G6PDH1 and its expression analysis will play an important role for further study on cold stress responses in Ch. praecox.

10.Primary study on developmental speciality and cultural practices of Amphicarpaea edgeworthii.

OBJECTIVEThe experiments were conducted to study specialty of seed germinating, plant growing and developing and cultural practices of A. edgeworthii.
METHODThe germinating and sowing test, growth habitants, photosensitive reaction of seedling from subterraneous seeds and above ground seeds on A. edgeworthii were studied in this experiment.
RESULT AND CONCLUSIONThe results indicated that (1) The water could not be absorbed by seed from plant above ground; (2) The underground and above ground seeds could normally germinate, grow, blossom and bear fruits, A. edgeworthii sowed in early May, blossomed in early September and matured in the last ten days of October; (3) The seed leaf of seedling grew underground; (4) The underground seed was produced from subterraneous branch stem which developed from node of seed leaf; (5) A. edgeworthii was shade demanding, high-temperature sensible short-day light plant. The flowering could be greatly advantaged by short-day light treatment (12, 10, 8, 6 h). Cultural practices were studied also in this experiment.
