Benign prostatic hyperplasia (BPH) and prostatitis are very common diseases in elderly men. The recent researches demonstrate that chronic inflammation is often present in the prostatic tissue of patients with BPH and the platelet-derived growth factor (PDGF) plays an important part in stimulating prostatic cell proliferation by inducing mitogenesis. This article presents a detailed discussion on the relationship between PDGF and BPH.

3.The role of PDGF/PDGFR in the regulation of platelet formation.

Platelet-derived growth factor (PDGF), a potent chemotactic and mitogenic factor, is involved in the regulation of hematopoiesis and platelet production. Our studies demonstrate the presence of functional PDGF receptors (PDGFR) on human megakaryocytes/platelets and CD34(+) cells, and their ability to mediate a mitogenic response. PDGF promotes the ex vivo expansion of human hematopoietic stem (CD34(+)) and progenitor (CD41(+)) cells. More significantly, PDGF enhances the engraftment of human CD45(+) cells and their myeloid subsets (CD33(+), CD14(+) cells) in NOD/SCID mice. PDGF also stimulates in vitro megakaryocytopoiesis via PDGFR and/or the indirect effect on bone marrow microenvironment to produce TPO and other cytokines. It also shows a direct stimulatory effect of PDGF on c-Fos, GATA-1 and NF-E2 expressions in megakaryocytes. We speculate that these transcription factors may be involved in the signal transduction of PDGF on the regulation of megakaryocytopoiesis. PDGF also enhances platelet recovery in mouse model with radiation-induced thrombocytopenia. This radioprotective effect is likely to be mediated via PDGFR with subsequent activation of the PI3K/Akt pathway. It provides a possible explanation that blockage of PDGFR may reduce thrombopoiesis and play a role in imatinib mesylate-induced thrombocytopenia.

4.A preliminary study on the changes of expression of PDGF-beta, PDGFR-beta, TGF-beta 1, TGFR, bFGF and its relationship with the wound age in wound healing.

OBJECTIVE:
To explore the relationship between the expression change of cytokines and the wound age during the healing process of rats skin wound.
METHODS:
Immunohistochemical and image-analysis methods were performed on vital skin wounds(after incision 0.5-168 h am) and postmortem damage(after incision 0.5-6 h pm).
RESULTS:
The expression of the cytokines PDGF-beta, PDGFR-beta, TGF-beta 1, and bFGF in the epithelial cells was already enhanced since 0.5 h am after damage and their strongest expression reaction was seen at 24-96 h am. In addition, the expression of PDGF-beta, PDGFR-beta, TGF-beta 1 and bFGF was also found in the macrophages and the fibroblasts of the granulation tissue, and the expression changes in the postmortem damage group showed that the skin tissue within 0.5-3 h after incision showed immunohistochemical changes but weakly expression and 3 h thereafter no any change was found.
CONCLUSION
The expression characteristics of the above mentioned cytokines in wound repair should be related to the wound age and it reminds therefore that they may be used as immunohistochemical criteria for accurate determining the wound age.

7.A comparative study of PDGF and EGF expression in skin wound healing between human fetal and adult.

OBJECTIVETo explore the differences of PDGF and EGF expression in the wound healing between fatal and adult.
METHODSWith the established animal model of fetal scarless healing and the adult samples, an immunohistochemical technique was used to evaluate the expression of PDGF and EGF in the normal adult skin, normal fetal skin, and the process of their wound healing.
RESULTS1. The expression of the PDGF was not found in the fetal skin, but a mild amount of the PDGF was shown in the epidermis and the upper dermal layer 12 hours and 1 day after the wounding process. In the normal adult skin, expression of PDGF was shown in the dermal fibroblasts, macrophagocytes and blood capillaries, and a strong expression was presented during its wound healing process. 2. In the fetal skin, the expression of the EGF was seen in the epidermis, hair follicles, sebaceous glands and sweat glands, but there were no markedly changes during the wound healing. In the adult skin, a positive stain of the EGF was shown in the basal layer of the epidermis while the mild stain in hair follicles and sweat glands. The level of the expression became gradually decreasing with the time going in the wounded adult skin.
CONCLUSIONThe different expression of growth factors between fetal and adult skin in wound healing may be one of the important reasons that the fetal wound could produce scarless healing.

8.Changes of serum angiogenesis in patients with chronic mountain sickness.

AIMThe clinical manifestation of chronic mountain sickness (CMS) is polycythemia, pulmonary hypertension and mionectic blood. However, the pathogenesis of it is not identified now. So it is necessary to investigate the effects of the angiogenic growth factors on the pathophysiologic development of CMS.
METHODSThe serum levels of basic fibroblast growth factor (bFGF), platelet-derived growth factor (PDGF) and vascular endothelial growth factor (VEGF) in 13 healthy Tibetan natives (Native), 17 healthy people in Xining (control group) and 35 CMS patients were determined by quantitative sandwich enzyme immunoassay. Meanwhile, the levels of Hb, Hct and SaO2 were determined.
RESULTSThe serum levels of bFGF (107.26 +/- 7.86) ng/L, PDGF (630.18 +/- 9.89) ng/L and VEGF (543.74 +/- 6.76) ng/L in CMS were significantly higher than those in Natives (37.01 +/- 9.16; 292.16 +/- 6.88; 125.51 +/- 7.26) ng/L, and in control group (40.58 +/- 5.34; 287.68 +/- 8.33; 76.26 +/- 4.60) ng/L, respectively (P < 0.01). There was no difference between the natives and the control group in bFGF and PDGF (P > 0.05), while there was predominant difference between the Natives and the control group in VEGF (P < 0.01). There was a predominant positive correlation between the serum levels of bFGF, PDGF or VEGF and hemoglobin concentrations in CMS respectively (P < 0.01). And there were positive relations between angiogenic growth factors each other.
CONCLUSIONThe serum levels of bFGF, PDGF and VEGF in patients with CMS significantly increase, these angiogenic growth factors may play important role on the pathophysiologic development of CMS; the VEGF level likely contributes to the adaptation to plateau hypoxia in healthy Tibetan natives; the elevated bFGF, PDGF and VEGF levels are likely associated with excessive erythropoiesis in CMS.

9.Effect of PDGF-AA on Ca2+i in vascular smooth muscle cells in SHRs.

AIMTo explore the relationship between proliferation and hypertrophy of vascular smooth muscle cells and PDGF-AA and PDGFR-alpha expression in SHRs and the role of [Ca2+]i in it.
METHODSExpress difference of PDGF-AA, PDGFR-alpha, PDGFR-beta in SHR/WKY-VSMC was observed by Western blot. The effect of Ca2+ inhibitor (nimodipine) on proliferation, hypertrophy and free Ca2+ concentration of SHR-VSMC induced by PDGF-AA was observed by Western blot, [3H] incorporation and fluorescent digital image technique.
RESULTSPDGF-AA and PDGFR-alpha expression was markedly increased in SHR-VSMC than in WKY-VSMC, but PDGFR-beta was not different in SHR and WKY-VSMC. PDGF-AA-stimulated PCNA expression, [3H] incorporation and [Ca2+]i increasing were observed in SHR-VSMC. Dose-dependent nimodipine-inhibited PCNA expression, [3H] incorporation and [Ca2+]i increasing induced by PDGF-AA also were observed in SHR-VSMC.
CONCLUSIONSSpontaneously expression increasing of PDGF-AA and PDGFR-alpha in spontaneously hypertension rats (SHRs) may be one of the important factors on vascular reactivity and vascular modeling mediated through proliferation and hypertrophy in SHR-VSMC, and [Ca2+]i play an important role in this process.

10.Research advances on abnormal marrow fibre in leukemia.

The genesis and development of leukemia not only associate to intrinsic factors, but also relate with the fibrous hyperplasia in the bone marrow. This review mainly focuses on the interaction between fiber-producing cells and leukemia cells, the relationship between fibrous hyperplasia and prognosis of leukemia, the regulation of TGF-beta, PDGF and other cytokines, the underlying mechanism of fibrous hyperplasia so as to explore the potential therapeutic targets for improving the prognosis of leukemia.
