1.Effects of Aspirin Combined with Clopidogrel on Related Indexes of Patients with Cerebral Infarction Induced by Middle Cerebral Artery Stenosis in Stable Phase

OBJECTIVE:To investigate the effects of clopidogrel combined with aspirin on related indexes of patients with cerebral infarction induced by middle cerebral artery stenosis in stable phase.METHODS:Clinical data of 127 patients with cerebral infarction induced by middle cerebral artery stenosis in stable phase were analyzed retrospectively and divided into observation group (65 cases) and control group (62 cases) according to therapy drugs.Both groups received routine treatment.Control group was additionally given Aspirin enteric-coated tablet 200 mg orally,once a day.Observation group was additionally given Clopidogrel sulfate tablet 75 mg orally,once a day,on the basis of control group.Both group received treatment for consecutive 1 year.Coagulation function related indexes [D-dimer,PLT,AA-induced platelet inhibition rate,ADP-induced platelet inhibition rate],daily life quality score [modified Fugl-Meyer scale,Barthel indexes,NIHSS score] before and after treatment,the occurrence of 1-year accumulative cerebral infraction recurrence and ADR were observed and compared between 2 groups.RESULTS:Before treatment,there was no statistical significance in above indexes between 2 groups (P>0.05).After treatment,D-dimer level and NIHSS score of 2 groups were significantly lower than before treatment,and the observation group was significantly lower than the control group;AA-induced platelet inhibition rate,ADP-induced platelet inhibition rate,Fugl-Meyer score and Barthel indexes of 2 groups were significantly higher than before treatment,and the observation group was significantly higher the control group,with statistical significance (P<0.05).The recurrence rate of 1-year accumulative cerebral infraction in observation group was significantly lower than control group,with statistical significance (9.23% vs.22.58%,P<0.05).No severe ADR was found in 2 groups during treatment.CONCLUSIONS:Compared with aspirin alone,aspirin combined with clopidogrel in the treatment of cerebral infarction induced by middle cerebral artery stenosis in stage phase can effectively improve coagulation function and quality of daily life,reduce recurrence rate with good safety.
2.Comparison of Therapeutic Efficacy and Safety of Sertraline and Paroxetine in the Treatment of Post Stroke Depression

OBJECTIVE:To compare the therapeutic efficacy and safety of sertraline and paroxetine in the treatment of post stroke depression.METHODS:The information of 91 patients with post stroke depression were analyzed retrospectively and divided into observation group (42 cases) and control group (49 cases) according to therapy plan.Based on routine treatment,observation group was given Sertraline hydrochloride tablet orally with initial dose of 50 mg at first week,once a day,increasing by 50 mg each week,no more than 200 mg.Control group was given Paroxetine tablet with initial dose of 10 mg,once a day,increasing by 10 mg each week,no more than 40 mg.Treatment course of 2 groups lasted for 2 months.Clinical efficacies of 2 groups were observed.NIHSS score,MMSE score,HAMD score,the serum levels of 5-HT,BDNF and S1003 were observed before and after treatment,and the correlation of serum levels of 5-HT,BDNF and S100β with HAMD score were evaluated in observation group after treatment;the occurrence of ADR was recorded.RESULTS:The total response rate of observation group (90.48%) was significantly higher than that of control group (73.47%),with statistical significance (P<0.05).After treatment,NIHSS score,HAMD score and S100β level of 2 groups were significantly lower than before treatment,and HAMD score and S100β level of observation group were significantly lower than those of control group.MMSE score,serum levels of 5-HT and BDNF in 2 groups were significantly higher than before treatment,and the observation group was significantly higher than the control group,with statistical significance (P<0.05).There was no statistical significance in NIHSS score between 2 groups (P>0.05).The serum levels of 5-HT and BDNF were negatively correlated with HAMD score in observation group after treatment,and the level of S100β was positively correlated with HAMD score.There was no statistical significance in the incidence of ADR between 2 groups (P>0.05).CONCLUSIONS:Based on routine treatment,therapeutic efficacy of sertraline is significantly better than that of paroxetine in the treatment of post stroke depression,and can improve cognitive function and depression.The two drugs are similar in improving nerve function and mental state,safety.
3. Effect of TCF21 on proliferation, migration and vascular production of breast cancer cells

Objective:
To investigate the effects of transcription factor 21 (TCF21) on the proliferation, apoptosis, invasion and migration and angiogenesis of human breast cancer cells (MDA-MB-231).
Methods:
TCF21 overexpressing plasmid was transfected into human breast cancer cell line MDA-MB-231 by liposome transfection, and Western blot was used to detect whether the transfection was successful. The effects of TCF21 overexpression on proliferation, apoptosis, invasion and migration and angiogenesis of MDA-MB-231 were detected by MTT, DAPI, Transwell and CAM.
Results:
The expression of TCF21 protein in Western blot showed that the TCF21 overexpression plasmid was successfully transfected into MDA-MB-231 cells. Overexpression of TCF21 inhibited the proliferation of tumor cells and inhibit the proliferation of 48 h cells after transfection. Overexpression of TCF21 inhibited tumor invasion and migration and angiogenesis, and promoted apoptosis of tumor cells. The results of Western blot showed that the protein expression of MMP-9, VEGFA and p-VEGFR2 decreased significantly after overexpression of TCF21.
Conclusions
TCF21 can inhibit the proliferation, invasion and migration, angiogenesis and apoptosis of MDA-MB-231 cells. It is suggested that TCF21 can be used as a potential site for clinical diagnosis and treatment of breast cancer.
