1.Effects of Sufentanil in inhibiting Hep3B cell viability and promoting cell apoptosis in liver cancer

Objective To investigate the effects and mechanism of Sufentanil on Hep3B cell viability in liver cancer.Methods Sufentanil of different concentrations (0.01,0.1,1,5,10,1 5 μmol/L)was used to treat Hep3B cells.The changes of cell cycle,apoptosis and protein expression were detected to explore the potential mechanisms by MTT method,flow cytometry and Western blot,respectively.Results Reduced viability of Hep3B cells,arrested cell cycle in the G0/G1 phase,decreased expression of survivin protein,and increased expression of caspase-3 protein were observed with the increase of Sufentanil concentration. Conclusion Sufentanil inhibited the viability of Hep3B cells through affecting cell cycle,promoting cell apoptosis and changing protein expressions of survivin and caspase-3.
2.Efficacy of nalbuphine in preventing shivering after neuraxial anesthesia in patients undergoing cesarean section

Objective To evaluate the efficacy of nalbuphine in preventing shivering after neuraxial anesthesia in patients undergoing cesarean section.Methods Eighty American Society of Anesthesiologists physical status Ⅰ or Ⅱ patients,aged 20-35 yr,weighing 55-80 kg,undergoing elective cesarean section under neuraxial anesthesia,were divided into nalbuphine group (group N,n=40) and control group (group C,n=40) using a random number table method.After delivery,nalbuphine 0.1 mg/kg was intravenously injected immediately before clamping the umbilical cord in group N,and the equal volume of normal saline was given instead in group C.Ramsay sedation score was recorded before giving nalbuphine,at 5 min after giving nalbuphine,and at the end of surgery.The development of shivering was recorded from the end of nalbuphine administration until the end of surgery,and the shivering intensity was estimated using Wrench grading.The development of over-sedation,nausea and vomiting,bradycardia,hypotension and dizziness was recorded from the end of nalbuphine administration until the end of surgery.Results Compared with group C,Ramsay sedation scores were significantly increased at 5 min after giving nalbuphine and at the end of surgery,the incidence of shivering was decreased,the shivering intensity was reduced (P<0.05),and no significant change was found in the incidence of adverse reactions in group N (P> 0.05).Conclusion Nalbuphine can prevent the occurrence of shivering after neuraxial anesthesia in patients undergoing cesarean section.
3.The mechanism of cilostazole alleviating intestinal mucosal barrier dysfunction in type 2 diabetic mice

【Objective】 To explore the effect of cilostazol on intestinal barrier function in type 2 diabetes (T2DM). 【Methods】 The GSE142153 dataset was downloaded from GEO database to analyze gene changes in diabetic patients. Eight-week-old male db/db mice and control m/m mice were randomly divided into m/m+cmc, m/m+cilo, db/db+cmc, and db/db+cilo groups. Mice in different groups were given cilostazol and corresponding solvents for 4 weeks. We detected the levels of serum sCD40L and the expression of CD40 in intestinal tissue, and evaluated the mice’s intestinal barrier function by examining intestinal permeability, water content, bacterial number, and tight junction protein expression in different groups. 【Results】 Differential expressed genes were enriched in platelet activation and endothelial barrier function pathways in diabetic patients. Compared with those in the control group, the levels of serum sCD40L in db/db diabetic mice elevated significantly, and the CD40 expression, permeability, water content and bacterial number in intestinal tissue increased obviously, while the expression of tight junction protein decreased. Cilostazol treatment in diabetic mice decreased the levels of serum sCD40L and CD40, and alleviated significantly the intestinal barrier dysfunction. 【Conclusion】 Cilostazol attenuated the damage of intestinal barrier function in T2DM, and its protective effect may be related to the inhibition of platelet activation in diabetic mice.
4.Comparison of the effects of remimazolam and propofol on postoperative nausea and vomiting in patients undergoing painless prostate biopsy based on propensity score matching

【Objective】 To investigate the effects of remimazolam and propofol on postoperative nausea and vomiting (PONV) in patients undergoing painless prostate biopsy, so as to optimize the anesthesia protocols. 【Methods】 A retrospective analysis was conducted on the clinical data of 1217 patients who underwent painless prostate biopsy in our hospital during Jan. 2023 and Jun.2024. Among them,1093 patients met the inclusion criteria and were divided into two groups: the remimazolam group (n=294) and the propofol group (n=799). After 1∶1 propensity score matching, with 267 patients in either group, a comparison was conducted regarding the incidence of PONV and anesthesia recovery time. 【Results】 Before propensity score matching, the remimazolam group had older age [66(53, 83) years vs.63(49, 78) years] , higher body mass index (BMI) [25.30(21.83, 29.23) vs.24.46(20.79, 28.91)] , larger intraoperative use of sufentanil [9(8, 10)μg vs.7(6, 9) μg] , higher intraoperative use rate of ondansetron (55.4% vs.47.6%), and longer surgical duration [16(14, 20) min vs.15(13, 17) min] , with statistically significant differences (P<0.05). There were no statistically significant differences in the aforementioned factors between the two groups after propensity score matching (P>0.05). Before propensity score matching, the incidence of PONV was higher in the remimazolam group than in the propofol group (17.7% vs.11.5%,P=0.007), while after propensity score matching, the incidence of PONV did not differ significantly between the two groups (12.7% vs.17.2%,P=0.146). Before and after propensity score matching, the anesthesia recovery time was significantly shorter in the remimazolam group than in the propofol group [3(2, 4) min vs. 7(4, 10) min,P<0.001] . 【Conclusion】 Compared with propofol, remimazolam does not increase the incidence of PONV in patients undergoing painless prostate biopsy but can shorten anesthesia recovery time.
