The rice Rim2/Hipa is a stress-induced transposon superfamily recently identified in Oryza genomes. Genomic variation was found in the Rim2 core region among rice genetic resources/genomes, indicative of high genomic divergence accumulated during the Rim2 evolution. Based on the divergence and quiescent state of the Rim2 elements, a Rim2 paralog display-based fingerprinting approach was developed to effectively identify rice genetic resources and explore their genetic relationships within a set of rice germplasm including 45 accessions ofO. sativa and 8 accessions of its wild relatives O. rufipogon. A dendrogram showed not only clear genetic diversity of rice germplasm, but also considerable genetic differentiation among wild rice resources. The wild rice relatives were either clustered as an independent group, or among the japonica varieties. This Rim2-based fingerprinting approach could also serve as a sensitive tool to identify rice hybrids from their parents, and variety stability, demonstrating its great potential in evolution study ofrice genomes and in rice breeding and seed production.

The comparative analysis of the biological characterization and the genetic background study of EV71 circulating strains is commonly recognized as basic work necessary for development of an effective EV71 vaccine. In this study, we sequenced five EV71 circulating strains, isolated from Fuyang, Hefei, Kunming and Shenzhen city of China and named them FY-23, FY-22, H44, K9 and S1 respectively. The sequence alignment demonstrated their genotypes be C4. The genetic distance of the VP1 gene from these isolates suggested that they were highly co-related with genetic identity similar to other previously reported EV71 strains in China. Additionally, these strains were identified to display some obvious proliferation dynamics and plaque morphology when propagated in Vero cells. However, a distinctive difference in pathogenic ability in neonatal mice was found. Some differences in cross neutralization test & immunogenic analysis were also found. All these results are related to the biological characterization of circulating EV71 strains in China and aid in the development of an EV71 vaccine in the future.

Objective To investigate the dynamic changes of three types of anti-poliovirus neutralizing antibodies and anti-hepatitis A virus (HAV) IgG antibody in children who were immunized with inactivated enterovirus 71 (EV71) vaccine (human diploid cell). Methods Serum samples were collected from the subjects immunized with inactivated EV71 vaccine.Neutralizing antibodies against EV71 and poliovirus were detected by micro-cytopathic effect neutralization test.Enzyme linked immunosorbent assay (ELISA) was used to detect IgG antibody against HAV.Results The geometric mean titers (GMTs) of anti-EV71 neutralizing antibody increased to 4.85 following the first-dose injection of inactivated EV71 vaccine.A significant increase of GMTs (up to 64.37) could be observed 28 days after the second-dose vaccination.Meanwhile, results of the dynamic monitor showed that there were slight fluctuations in the neutralizing antibodies against three types of poliovirus on day 28 (28 days after the first-dose vaccination) compared with those on day 0 (before vaccination) (P<0.05);types Ⅰ and Ⅲ anti-poliovirus neutralizing antibodies on day 56 (28 days after the second-dose vaccination) remained slightly different from those on day 0 (P<0.05), but type Ⅱ anti-poliovirus neutralizing antibody on day 56 had restored to normal level (P>0.05).The level of anti-HAV IgG antibody was stable and no significant difference was found during the observation period (P>0.05).Conclusion This study shows that inactivated EV71 vaccine has no impact on anti-HAV IgG antibody in Children during the two-dose vaccination and in anti-EV71 antibody-producing period, but has slight influence on the anti-poliovirus antibodies. In general, changes in antibody profile do not affect the clinical efficacy of immune response.

Objective:To investigate the effects of resistance exercise on mitochondrial function, muscle attenuation and muscle fiber morphology in quadriceps femoris of aged rats. Methods:The 18-month-old male rats were randomly divided into five groups( n=8, each): control(not exercise), 0% weight-bearing, 30% weight-bearing, 50% weight-bearing, and 70% weight-bearing exercise. After maximum resistance running, intermittent race table resistance exercise with 0%, 30%, 50%, 70% maximum load were performed. The treadmill placement slope was 35°, the running speed was 15 m/min, and exercise was performed every other day.At the end of the 8 th week, the mitochondrial membrane potential of quadriceps femoris muscle was measured, cytosol contents of cytochrome C(Cyt c), apoptosis-inducing factor(AIF), and apoptotic protein(Smac/DIABLO)were measured, and the morphology and structure of muscle fibers were observed. Results:Compared with the control group, the mitochondrial ΔΨmt was increased in the 0%, 30%, 50% and 70% maximum load groups, with a significant difference in the increment in the 0%, 30% and 50% maximum load groups( t=7.412, 5.611, 6.213, all P<0.01).Compared with the 0% maximum load group, the percentage of cells with mitochondrial ΔΨmt was statistically significantly decreased in the 30% maximum load group(10.6%)( t=9.356, P<0.05), while the percentage of cells with mitochondrial ΔΨmt was statistically significantly increased in the 70% maximum load group(10.03%)( t=8.341, P<0.05).Compared with the control group, the contents of Cyt c, AIF and Smac/DIABLO in the cytoplasm of quadriceps femoris of aged rats with 8-week resistance exercise were decreased, among which the contents of Cyt c and Smac/DIABLO were statistically significantly decreased in the three groups of 0%, 30%, and 50% maximum load( t=8.324, 7.516, and 6.871, all P<0.05), as well as the decrement in AIF of the three groups of 0%, 30%, and 50% maximum load was statistically significant( t =9.434, 8.78, and 7.342, all P<0.05).Compared with the control group, the vacuolar area of muscle fibers was extremely significantly decreased in the 0%, 30%, and 50% maximum load groups( t =5.567, 6.784, and 7.432, P<0.01); the protein content in the quadriceps femoris muscle was very significantly increased in the 30%, 50%, and 70% maximum load groups( t =7.478, 6.765, and 4.564, all P <0.01).Compared with the 0% maximum load group, the protein content in the quadriceps femoris muscle was very significantly increased in the 30%, 50%, and 70% maximum load groups( t=9.236, 8.342, and 6.456, all P<0.01). Conclusions:Low and medium weight-bearing resistance exercise can improve the mitochondrial function of quadriceps femoris, reduce the femoral quadriceps mitochondria-released proapoptotic proteins Cyt c, AIF, and Smac/DIABLO, and reduce the incidence of quadriceps femoris apoptosis.Low and medium weight-bearing resistance exercise can increase the protein content of muscle fibers, reduce the vacuolar area of muscle fibers, maintain muscle mass, and delay the occurrence of sarcopenia.
