2.Advance in the study of poly(lactide-co-glycolide) nano/microparticles as gene vector.

Biodegradable nano/microparticles of poly(D, L-lactide-co-glycolide) (PLGA) is a novel non-viral gene vector, which has many advantages, such as safety, non-immunogenicity, easy of large-scale preparation and well load-capability. Therefore, more and more attentions and researches have been paid on its application. Especially, PLGA has shown enormous potential application value and space in the field of plasmid DNA (pDNA) delivery system. On the basis of the current situation of PLGA nano/microparticles for pDNA delivery, this paper focused on summarizing the current preparation approaches and surface modified methods of PLGA particle, furthermore showing its application in gene therapy and genetic vaccine delivery. These showed that PLGA nano/microparticles have extensive prospect in the development of controlled gene delivery system.
3.Study on a Pre-enrichment Medium for the Simultaneous Recovery of Salmonella spp., Escherichia coli and Staphylococcus aureus

This text involved with a pre-enrichment medium for the simultaneous recovery of Salmonella spp.,Escherichia coli and Staphylococcus aureus.This medium is named buffered saline broth(BSB),which contains peptone 10g,beef exact 3g,disodium hydrogen phosphate 9g,potassium dihydrogen phosphate 1.5g,additive 50g,deionized water 1 000mL,pH7.2.1cfu per one milliliter of Salmonella spp.,Escherichia coli and Staphylococcus aureus in saline were stimutaneously added to 97 mL of BSB,buffered peptone water,lactose broth,nutrient broth,Escherichia coli broth,rappaport-vassiliadis enrichment broth,7.5% sodium chloride enrichment medium,respectively,and incubated for 18h at 37℃.The results showed BSB was the best enrichment medium,in which Salmonella spp.,Escherichia coli and Staphylococcus aureus multiplied at nearly same speed,and reached at 10~6 、10~6 、10~7 cfu/mL,respectively.Multiplex PCR produced specific amplicons of expected sizes,284bp for Salmonella spp.invA gene,622bp for Escherichia coli phoA gene,484bp for Staphylococcus aureus nuc gene.In contrast,the three bacteria couldn't multiply harmoniously in the other six media.So BSB might be considered as the medium,which could enrich above mentioned three bacteria.
4.Surface Electromyography of Lumbar Muscles in Non-specific Low Back Pain Patients during Different Bridging Exercise

Objective To investigate the features of surface electromyography of lumbar muscles in non-specific low back pain (NLBP) patients when they were practicing different types of bridging exercise. Methods 18 healthy subjects (control group) and 18 NLBP patients (NLBP group) were included and asked to practice bridging exercise with single and double feet. The root mean square (RMS) of surface electromyography of both sides of lumbar muscles were recorded. Results No significant difference was found in the control group. The RMS of erector spinae was more in the supported side than in the contra side for NLBP patients in bridging exercise with single foot. There was no significant difference among different time points in the same muscle. Conclusion The contractions of erector spinae in both sides are not equal in asymmetric bridging exercise in NLBP patients.
6.Effect of allergen on function of peripheral blood CD4+T cells in patients with asthma

Objective To investigate the relationship among the proliferation of CD4+T cells, the intracellular levels of interleukin-10 (IL-10) and transforming growth factor-?1(TGF-?1), and allergic bronchial asthma. Methods Dermatophagoides farinae antigen were prepared as allergen. Twenty-five patients with asthma and 15 healthy individuals were enrolled and divided into blank control group, allergen group and self control group, respectively, after venous blood sample collection. The proliferation of CD4+T cells and the distributions of CD4+/IL-10+, CD4+/TGF-?+1 and CD4+/IL-10+/TGF-?+1 were measuredby flow cytometry (FCM). Results The distributions of CD4+/IL-10+, CD4+/TGF-?+1 and CD4+/IL-10+/TGF-?+1 could hardly be detected in the peripheral blood samples of the blank controls of the patients with asthma and healthy ones. In the allergen group of the healthy individuals, the peripheral blood CD4+T cells were significantly proliferated, and the proportions of CD4+T cells andCD4+/IL-10+ cells were much higher than the self control group, while there was no significantly increase in the proportions of CD4+/TGF-?+1 and CD4+/IL-10+/TGF-?+1 subgroups. In the allergen group of those with asthma, the proportions of peripheral blood CD4+, CD4+/IL-10+, CD4+/TGF-?+1 and CD4+/IL-10+/TGF-?+1 cells were not found significantly increased compared with those self controls. 7.Clinical effect of orthokeratology for juvenile with myopia astigmatism and its effects on corneal endothelial cells

Abstract?AIM:To investigate the clinical effect of orthokeratology for 400 juvenile with myopia astigmatism and its effects on corneal endothelial cells.?METHODS:Four hundred patients(800 eyes), of whom the average age was 11.5 ±2.3 years old, 239 male, 161 female, were divided into two groups: orthokeratology group and spectacles group. Parameters including efficacy data ( uncorrected visual acuity, corneal curvature, axial length and diopter ) and corneal endothelial cell data ( count of endothelial cell, endothelial cell density, fluorescein staining and central corneal thickness) were observed at 1d, 1, 6, 12 and 24mo after wearing.? RESULTS: The visual acuity of spectacles group recovered to normal after wearing, that of orthokeratology group recovered to normal at 1mo after wearing.At 2a after wearing, the corneal curvature, diopter of orthokeratology group decreased significantly (40.09 ±0.31D, 0.23 ±0.06D respectively); while those of spectacles group increased, the differences between the two groups were significant (P<0.05).The axial length of the two groups increased slightly at 1mo after wearing ( P>0.05 ) compared to those before wearing. At 2a after wearing, the axial length of the two groups were 23.96 ± 0.38mm, 26.49±0.88mm respectively (P<0.05).At 2a after wearing, central corneal thickness was 527.33 ± 27.69mm, 526.98±26.89μm(P>0.05).The count of endothelial cell and endothelial cell density both decreased after wearing without significant differences (P>0.05).?CONCLUSION: Orthokeratology has less effect on the corneal endothelial cells, no obvious adverse reactions and can control the prognosis of myopia.
8.Significance of Changes of Plasma Levels of Adrenomedullin and Endothelin-1 in Infants with Congenital Heart Disease Complicated with Congestive Heart Failure

Objective To investigate the roles of adrenomedullin(ADM) and endothelin-1(ET-1) in left-to-right shunt congenital heart disease(CHD) complicated with congestive heart failure(CHF).Methods Plasma ADM and ET-1 concentrations were measured by radioimmunoassay in heart failure stage and recovery period of 18 infants with left-to-right shunt CHD.Also 20 healthy infants acted as control group were measured.Results Plasma concentrations of ADM and EF-1 were significantly higher in heart failure stage of left-to-right CHD infants than those in their recovery period,and both in their heart failure stage and in their recovery period were higher than that in control group(P
9.Isolation and purification of canine islets

Objective To investigate factors influencing islet isolation and purification in dogs and to explore how to obtain massive purified viable islets from canine pancreas.To provide experimental evidence for pancreatic islet transplantation in clinical practice.Methods Islets were isolated from pancreas of 22 dogs by modified automated techniques,followed by purification using continuous density gradients.The purity of islets was determined by dithizone (DTZ) staining,and the function of islets was evaluated by glucose stimulating insulin release test.Results The crude islets yield was ( 155 040 ±310) IEQ/pancreas while the yield was (74 200 ±185) IEQ/pancreas after purification.The average purity was (89.4 ±2.6) %.The recovery rate was(47.8 ±1.3) %.Islet viability was(93.0 ± 1.7)%.The modified techniques improved both the yield and the quality in islet isolation and purification.The islets were morphologically intact after purification.Glucose stimulating insulin release test showed that the difference of the secreted insulin concentration had statistical significance ( P <0.001 ) between low concentration glucose group and high concentration glucose group.Conclusion It is of great value for clinical research to obtain enough islets through islet isolation and purification in dogs.
10.Comparison of direct and indirect three-dimensional placement methods of facial soft tissue

Objective To compare the reproducibility and reliability of facial soft tissue landmarks using a direct and an indirect placement methods in facial three-dimensional (3D) soft tissue assessment.Methods 3D coordinates of 37 soft tissue landmarks were obtained respectively in 35 normal healthy volunteers from 3D optical capture system and Materialise's interactive medical image control system (Mimics).Landmarks were affixed on the cutaneous surface (direct method) and marked on the 3D reconstructed craniofacial model (indirect method).Intra-class correlation coefficients (ICCs) and paired t-tests were used to compare the reliability and reproducibility of facial landmarks between two placement methods.Results For the direct placement method,86.5 % landmarks had an ICC higher than 0.75.For the indirect placement method,75.7% landmarks had an ICC higher than 0.75.The inter-observer variability using the direct placement method was (1.259±0.566) mm,which was significantly smaller (P<0.05) than (1.420±0.638) mm obtained by the indirect placement method.Conclusions Compared with indirect method,direct method has higher reliability and reproducibility in evaluating facial soft tissue.
